RESUMEN

Objective To explore the role of Notch signaling pathway in the matrix synthesis of osteoarthritis cartilage cell and its possi -ble mechanism.Methods Selected the femoral condylar cartilage of 8 patients who were admitted into our hospital and treated with knee joint replacement as the observation group,while the normal femoral condyle cartilage of one patient with above-knee amputation was selected as the control group.The specimens were given histological examination or cell isolation culture and detection.The cultured cells were divided into 4 groups,namely the normal cartilage cells,OA cartilage cells,OA cartilage cells with γ-secretase inhibitor DAPT,OA cartilage cells with recombinant human proteins Delta4.Then immunohistochemistry and Western blot were used for detecting the expression of Notch -1,Notch-3, Jagged-1,Jagged-2 and HES5 in chondrocytes in vitro.Results The expression of Notch signaling pathway and the phenotype of cartilage cells changed in the osteoarthritis.The expression of Notch-1,Jagged-1,Jagged-2 and HES5 were activated.γ-secretase inhibitor DAPT (20 μmol/L)could inhibit the expression of Notch-1,HES5,Jagged-1,and Notch-3,while it had no obvious effect on the expreesion of Jag-ged-2.Recombinant human proteins Delta 4(100 ng/μL)could promote the expression of Notch-1,Jagged-2,and HES5,and it had no obvi-ous effect on the expreesion of Jagged-1 and Notch-3.Conclusion The expression of Notch signaling pathway of cartilage cells changed in the osteoarthritis.DLL4 can activate the Notch signaling pathway and DAPT can inhibit the Notch signaling pathway.

RESUMEN

Cartilage apoptosis functions a lot in OA occurrence.Its apoptosis has two ways of NO and Fas,mainly controlled by Bcl-2 gene family,Bax,P53,ICE and c-myc gene family;the degeneration of cartilage cell base can induce apoptosis.It shows cartilage apoptosis function in OA occurrence,helpful to prevention and treatment of OA.

RESUMEN

Objective: To study the effect of interleukin 1(IL 1?) on the metabolism of osteoarthritic and normal mature condylar chondrocytes in temporomandibular joints, and investigate the role of IL 1? in the pathogenesis of temporomandibular joint osteoarthritis. Methods: The primary generation of osteoarthritic and normal condylar chondrocytes cultured in the monolayer condition was treated with 20 ?g?L -1 recombined human interleukin 1? (rhIL 1?), and then collected to be detected with RT PCR method for the cellular metabolism including mRNA expression of type Ⅱ collagen, aggrecan, collagenase, insulin like growth factor 1 (IGF 1), and transforming growth factor?1(TGF?1). Results: The normal mature condylar chondrocytes showed the obviously decreased mRNA expression of type Ⅱ collagen and aggrecan after the intervention of exogenous IL 1?, but less influence could be found for the collagenase expression. The osteoarthritic condylar chondrocytes exhibited the decreased mRNA expression of type Ⅱ collagen and collagenase under the effect of IL 1?, while the cellular mRNA expression of aggrecan didn’t change obviously. The intervention of exogenous IL 1? didn't show an obvious influence on the cellular expression of endogenous growth factors such as IGF 1 or TGF ?1 for both the normal and osteoarthritic condylar chondrocytes cultured in vitro. Conclusion: IL 1? could not only disturb the expression of cartilage matrix molecules by the normal condylar chondrocytes, which lead to the lesion of condylar cartilage, but also worsen the abnormal cartilage matrix environment within the osteoarthritic condylar cartilage.

RESUMEN

We present a case of a 64-year-old man with chondrosarcoma complicated with intratumoral hemorrhage. Cartilage cell tumors within the cranium are very rare. Only less than 0.2% of all intracranial tumors. However chondrosarcoma with intratumoral hemorrhage has not been reported. We report a rare case of chondrosarcoma with intratumoral hemorrhage in parasella region with a literatures review.

RESUMEN

We experienced a case of falcine mesenchymal chondrosarcoma in a 22-year-old pregnant woman. Cartilage cell tumors within the cranium are very rare, only less than 0.2% of all intracranial tumors. Because a few examples of mesenchymal chondrosarcoma in this locttion have been reported, we reviewed previous reports cases, to determine the natural history of intracranial chondrosarcomas. The distinguishing features of this rare tumor are compared with previous cases of itracranial tumors derived from cartilage.

RESUMEN

By using the method of chondrocytes in monolayer culture in vitro we observed the effect of free radicals generated by the xanthine oxidase and xanthine on DNA, matrix proteoglycan (PG), collagen synthesis and ultrastructure of human embryo chondrocytes. Our results showed that the synthesis of DNA, PG and collagen of the chondrocytes were obviously inhibited and the membranes and subcellular organelles of the chondrocytes were damaged strongly. On the bases of these results we suggest that oxygen free radicals from poymorphonuclear leukocytes and phagocytes would be the important factors of cartilage injury in arthritis.

RESUMEN

In this paper, We present the method of using glucuronate as a parameter to determine the growth of cartilage cell in the culture media. Using this method, we observed the metabolic condition of cartilage cell and knew the effect of the serum with the various types and different concentration to the growth of cartilage cell. The. experimental method was used for the first passage chondrocyte of rabbit. We put the cartilage cell in the culture media containing different concentration of serum from calf and human; then, observed the growth of cartilage cell under the microscope, collected the medium in given time; and determined the amount of glucuronate in the medium. Our results indicated that the first passage chondrocyte of rabbit can grow in the medium containing serum of calf and human in 5%~15% Also, the study shows that it is possible to determine the amount of glucuronate in the medium as a parameter to respond the growth of the cartilage cells.

RESUMEN

Objective To evaluate the security and biocompatibility of a new kind of absorbable high-strength PDLLA-CPP composite, which is used for internal fracture fixation. Methods According to the national standard of the procedure of appraise in food security and toxicology, micronucleus test of bone marrow cell, spermatic aberration test, chromosomal aberration test, and teratogenic test were measured with IP maximal tolerance dose of maceration extract from PDLLA-CPP composites. Meanwhile, chondrocyte culture with PDLLA-CPP composite was used to evaluate the effect of PDLLA-CPP composite on cartilage cell proliferation. Results Micronucleus test of bone marrow cell, spermatic aberration test, chromosomal aberration test, and teratogenic test were not affected by IP maximal tolerance dose of PDLLA-CPP maceration extract. PDLLA-CPP composite also had no cytotoxicity on cartilage cell proliferation. Conclusion PDLLA-CPP is a new and safe medical material with good biocompatibility and absorbability.
