RESUMEN

Rat model of blood stasis syndrome was prepared by subcutaneous injecting of epinephrine hydrochlorid,then the model rats were administrated by Yunnan Baiyao for 15 days. Blood rheology,coagulation function and histopathology were chosen as indicators to evaluate the successful replication of blood stasis syndrome model and the treatment effect of Yunnan Baiyao. UPLC-Q-TOF-MS was used to rapidly analyze the serum samples of blood stasis syndrome rat after 15 days Yunnan Baiyao treatment,Progenesis QI software was employed to identify the alkaloids components. The results showed that Yunnan Baiyao reduced the plasma viscosity and whole blood viscosity of rats with blood stasis syndrome,prolonged thrombin and prothrombin time,reduced fibrinogen content,and effectively improved pathological state such as inflammatory cell infiltration,blood stasis,congestion and edema of various organs in rats with blood stasis syndrome. Seven alkaloids components from Aconitum kusnezoffii,including karacolidine,senbusine B,isotalatizidine,karakoline,denudatine,talatisamine and chasmanine were found in the rat serum after Yunnan Baiyao treatment. Based on the effectiveness of Yunnan Baiyao in the treatment of blood stasis syndrome induced by epinephrine hydrochloride in rats,alkaloids components from the root of A. kusnezoffii absorbed into blood after Yunnan Baiyao treatment were clarified rapidly and accurately with the help of UPLC-Q-TOF-MS. Karacolidine,senbusine B,isotalatizidine,karakoline,denudatine,talatisamine and chasmanine are the pharmacodynamic material basis of the root of A. kusnezoffii for activating blood circulation and removing blood stasis.

Objective: The methodology of serum chemistry was applied to screening the effective constituents in Ziziphi Spinosae Semen (ZSS). The combination of serum chemistry with network pharmacology might be a potent method for exploring the material basis of traditional Chinese medicine (TCM). Methods: A RSLC-Q Exactive Orbitrap-HRMS method was established for detecting the components in dosed serum after oral administration of the extract of ZSS. By comparative analysis of the chemical profiles of blank serum and dosed serum, the prototype compounds and metabolites in ZSS may be discovered. Firstly, the active target proteins of insomnia were searched in the DrugBank database and analyzed in String database. Cytoscape was used for the reconstruction and visualization of protein-protein interaction networks (PPI). Secondly, the targets of constituents absorbed into blood were searched in the TCMSP and CoolGeN database. Finally, the network of "constituents absorbed into blood-target" was constructed and analyzed using Cytoscape software. Results: Fifteen compounds including 6 flavonoids and 1 metabolite, 5 triterpenoids saponins and 2 alakoids and 1 metabolite from ZSS were discovered. Combined serum chemistry results, network pharmacology analysis revealed that 5 compounds including zizyphusine, spinosin, apigenin, jujuboside A and coclaurine mightserve as the material basis for ZSS. Triterpenoids saponins were corresponding to GABAA targets, while alakoids were corresponding to melatonin target. Conclusion: This work provides a novel strategy for investigation of effective components by combination of serum chemistry and network pharmacology. This attempt should be helpful for screening in vivo effective components for ZSS.

RESUMEN

The method of UHPLC-LTQ-Orbitrap mass spectrometry coupled with higher energy collision dissociation (HCD) was established to rapidly analyze the constituents absorbed into blood after oral administration of steroidal saponins from Radix Ophiopogonis. A total of 31 constituents, including 13 furostanol steroidal saponins and 18 spirostanol steroidal saponins, were characterized based on the accurate mass measurements, fragmentation patterns, chromatographic retention times, and diagnostic product ions. Among them, 8 compounds were unambiguously identified by comparison with their corresponding standards. The results provide comprehensive insights and guidance for elucidation of material basis of Radix Ophiopogonis activity.

Objective: To study the blood components in Zhizi Dahuang Decoction (ZDD) and to further explore its pharmacodynamic material basis. Methods: UFLC fingerprints of serum in rats after ZDD treatment were established with serum pharmacochemistry method. The UFLC profiles of serum samples collected after the treatment of ZDD, ZDD without one of the component herbs and single crude herb were compared, and the transitional constituents to blood and their metabolites were analyzed. Results: Twenty-one constituents absorbed into rat blood were detected after oral administration of ZDD, 12 of which were metabolites. Other nine were prototype constituents, three of which were recognized as geniposide, aloeemodin, and rhein by comparing with reference substances, respectively. Conclusion: The constituents of ZDD absorbed into blood are preliminarily identified, which could lay the foundation for the illumination of its pharmacodynamic material basis.

Objective:For the further studies on the physical foundation of medicinal effi cacy of decoction of Fructus Schisandrae,to study its constituents absorbed into blood of rats after oral administration.Methods:Serum pharmacochemistry of Chinese medicine was used,based on the established high performance liquid chromatography analytical method of plasma effective constituents,to identify the constituents originated from the preparation and their metabolites in the serum by comparing fi ngerprint in vitro with that in vivo.Results:Five constituents have been detected in the blood,they were compound 1,2,3,4,5.Conclusion:Compound ⅣandⅤexist originally in Fructus Schisandrae decoction(including schisandrin),compound 1,2and 3 might be the metabolites of original constituents.
