RESUMEN

@#Castleman disease is a rare and complicated disease. A case of Castleman disease (CD) in a 40-year-old woman on the neck was reported and literature review was made to understand the occurrence, clinical characteristics, diagnosis and differential diagnosis, treatment, and prognosis of Castleman disease. Because of the rarity of CD, its occurrence in the general population and the typical clinical manifestations are still lacked. There are two types of CD, namely localized CD and multicentric CD, both of them are not easy to be diagnosised. CT scans in preoperative is helpful for the clinical diagnosis, while histopathological examination and lymph node biopsy are beneficial for the early diagnosis. The treatment result is based on the histopathological examination and lymph node biopsy. The prognosis is different with the type of CD. For the localized CD, the prognosis is usually good, while for the multicentric CD is bad. Both of localized CD and multicentric CD have a certain reoccurrence rate.

Objective:To explore the predictive value of serum thyroglobulin autoantibody (TgAb) for recurrence / metastasisin of thyroglobulin (TG)-negative and TgAb-positive patients with differentiated thyroid carcinoma(DTC) after thyroid ablation.Methods:The clinical data of 57 patients with DTC,undergoing complete resection of thyroid tissue,TG negative and TgAb positive,who were reviewed in Liaocheng People's Hospital during April 2013 to April 2015,were selected and divided into recurrence/metastasis group (n=20) and no recurrence/metastasis group (n=37).The TG and TgAb levels in the two groups were detected and compared by the electrochemical luminescence method,the sensitivity,specificity,positive predictive value and negative predictive value of TgAb in the diagnosis of DTC recurrence / metastasis were analyzed;the independent risk factors of DTC recurrence / metastasis were analyzed by Logistic regression.Results:The TgAb levels(72~3850 IU/mL) in the recurrence/metastasis group was higher than that(18~3638 IU/mL) in the no recurrence/metastasis group,the difference was statistically significant (P<0.05).The sensitivity,specificity,positive predictive value,negative preictive value of TgAb in the diagnosis of recurrence/metastasis of DTC were 85.71%,83.33%,75.00%,90.91%,respectively.Logistic Regression analysis showed that the 100≤ TgAb <204 IU/mL,204≤ TgAb≤ 1000IU/mL,and > 1000IU/rnL levels of TgAb were the independent risk factors of rucurrence/metastasis ofDTC (OR=1.267,2.853,6.791,P<0.05).Conclusion:TgAb can be used as evaluation of an important index of the recurrence/metastasis of patients with DTC when serum thyroglobulin (TG) was negative and TgAb was positive after thyroid ablation.The higher the TgAb levels,the more probability of the recurrence/metastasis.

RESUMEN

Purpose To study the clinicopathologic features of inflammatory myofibroblastic tumor (IMT) of long bone.Methods HE and immunohistochemistry of EnVision two-step were used to observe the clinical,radiological,histological and immunophenotype features of IMT of bone.The literatures were reviewed.Results 4 cases of IMT of bone were respectively located in the tibia (2 cases) and femur (2 cases).Histologically,the lesions were characterized by collagen-rich and spindled to plump myofibroblast-like cells and a variable admixture of inflammatory cells.Immunohistochemical study showed that the vimentin,SMA,actin,H-caldesmon and CD34 were positive.Conclusion The IMT is a rare and locally aggressive tumor.The diagnosis should combine the histological characters with immunohistochemical results and should be differentiated from the other tumors and tumor-like lesions.

Objective To evaluate the value of spiral CT in diagnosis and treatment of lung lesions in clini-cal work.Methods A retrospective analysis of 100 patients with pathological diagnosis of lung lesions patients were randomly divided into the control group (CT normal scan)and observation group (CT -D post -processing tech-niques).Lungs lesions,hilar and mediastinal lymph node metastasis sensitivity,specificity and accuracy of lymph node metastasis of both group were analyzed.Results In the control group,the diagnosis of lung lesions showed sensitivity of 75%,specificity of 50% and accuracy of 72%;while in the observation group the sensitivity was 80%,specificity was 65 % and accuracy rate was 85%.The differences between the two groups were statistically signficant(χ2 =3.854,4.604,5.007,all P <0.05).50 cases of pathologically verified as lung cancer patients with hilar lymph node metastasis in the control group showed diagnostic sensitivity of 60%,specificity of 72%,accuracy of 70%;while diag-nosed hilar lymph node metastasis in the observation group showed sensitivity of 76%,specificity of 84 percent and accuracy rate of 84%.The differences between the two groups were statistically signficant(χ2 =5.882,4.196,5.534, all P <0.05).50 cases of pathologically verified as lung cancer patients with mediastinal lymph node metastasis in the control group,showed diagnostic sensitivity of 62%,specificity of 57% and accuracy of 60%;in observation group the sensitivity was 72%,specificity was73% and accuracy was 72%.The differences between the two groups were statisti-cally signficant(χ2 =4.342,5.626,4.583,all P <0.05).Conclusion The spiral CT for diagnosis of lung lesions positioning has good effect,and can be widely used in the diagnosis of lung lesions.

Objective To study the effect of the exogenous recombinant human FGF-basic with different concentrations upon inducing human adipose-derived stem cells (hASCs) to differentiate into adipose cells,and the optimum concentration of exogenous rh-bFGF by experimental research.Methods hASCs were isolated and extracted by enzymatic digestion from the liposuction aspirate.hASCs using adipogenic supplement were divided into experimental group and blank group:the experimental group of adipogenic supplement was divided into adding the exogenous rh-bFGF 10 ng/ml,20 ng/ml and 40 ng/ml,the blank group of adipogenic supplement was cultured without exogenous rh-bFGF.MTT method was used to detect the adipocytes proliferation.The oil red O staining was used in the qualitative analysis on the time of newly forming adipocyte cells.Western blot was used to detect the effects of rh-bFGF on the expression of lipid droplets surface protein CIDEC at different stages during the culture.Results The experimental group could obviously shorten the period of inducing hASCs to differentiate into adioicytes,and promote the proliferation of adipocytes.The formation rate and the proliferation of adipocytes in the group adding 40 ng/ml rh-bFGF were superior to those in the experimental group else and blank group.The average time of the newly formed lipid droplets by adding 40 ng/ml rh-bFGFwas (11.5±1.9)h.The average absorbance of cell proliferation by adding 40ml rh-bFGF was 0.52 ±0.10.The CIDEC expression quantity of adding 40 ng/ml rh-bFGF group was also superior to that in the experimental group and blank group.Conclusions rh-bFGF in hASCs adipogenic supplement could promote the proliferation of adipocytes and dramatically accelerates the program of hASCs differentiating to adipocytes,in which the optimum concentration of rh-bFGF is 40ng/ml.

RESUMEN

Applying cellular transplant methods in the treatment of peripheral nerve injury has become a new hot spot in tissue engineering and drawn much attention. Bone marrow stromal cells (BMSCs) grow fast and can be easily purified in vitro. Besides, they have the capability of differentiating into Schwann cells under specific conditions. For all the reasons above, BMSCs are considered good succedaneum of Schwann cells and new seed cells for tissue engineered peripheral nerve. In this review, the physiological functions and bionomics of Schwann cells and BMSCs are introduced, as well as various methods inducing BMSCs into nerve tissue cells and the foreground of the research.

The bulge of hair follicles contains a mixed population of stem cells,including epidermal stem cells,neural crest stem cells,as well as another type of stem cells which can be induced into melanocytes.Bulge stem cells have the ability to divide,proliferate and are pluripotent as other adult stem cells.The neural crest stem cells can differentiate in vitro into neurons,glia and melanocytes under proper induction.Meanwhile,it was proved that implanting these cells into the gap region of damaged nerves could greatly enhance the regeneration rate of nerve and restoration of nerve function.In this review,distribution of the stated stem cells in hair follicle bulge and their characteristics of differentiating into neurons were discussed,as well as their possible application in regenerative medicine.

Objective To investigate the function of bone-marrow mesenehymal stem eells(BMSCs) differ-entiating into neuron-like cells in vitro after transfected by human brain-derived neurotrophie factor (BDNF) gene. Methods Human BDNF genes were cloned and recombinant pcDNA3. 1(-)-BDNF plasmids were construc-ted. BMSCs from five guinea pigs were isolated and cultured while their morphologies were observed by microscope. The surface antigen was detected by flowcytometry. BDNF genes were transfected into BMSCs with electroporation, and the transfected BMSCs were induced by ratinoie acid(RA)after bolted by Geneticin-418 (G418), then the dif-ferentiated BMSCs were identified by immunocytochemistry. Results The culture ceils demonstroted the typical mor-phology and surface antigen of BMSCs. The transfected cells expressed neuron- specific enolase(NSE), Nestin and glial fi-brillary acid protein(GFAP) also secreted BDNF. Conclusion BMSCs transfected by BDNF genes can differentiate into neu-ron- like cells in vitro, electroporation can enhame the transfection efficiency, RA can promote cell induction.

RESUMEN

Bone marrow stem cells(BMSCs) can differentiate into cardiomyocytes when induced with proper reagent. The cardiomyocytes from BMSCs have cardiomyocyte phenotypes with the expression of cardiac-specific antigen and receptor. BMSCs differentiate into cardiomyocytes in host myocardial microenvironments. Mechanism of the differentiation is unclear and need to be studied further.

Objective:To study the conditions of in vitro proliferation and differentiation of rat bone marrow stromal cells(MSC) for its application in transplantation.Methods:MSC were obtained by flushing the shaft with DMEM using a sterile syringe,dissociated by density gradient centrifuge,and cultured in the medium containing DMEM and 10% fatal bovine serum(FBS).Then we added neural growth factors into the medium and observed their influence on MSC proliferation,differentiation and survival by phase contrast microscopy.Immunocytochemistry was used to identify cell types.Results:MSC cultured in medium containing DMEM/ 10% FBS/ neural growth factors proliferated quickly.The neuron-like cells appeared at 7-9 d with the expression of neurone specific enolase(NSE) at(52?9.2)% and glial fibrillary acidic protein(GFAP) at(15?7)%,and could survive more than 7 days;In contrast,cells induced by ?-mercaptoethanol(BME) died gradually after 24 hours.Conclusion:MSC can not only proliferate and differentiate into neuron-like cells effectively in the medium containing neural growth factors but also survive longer.

Objective To study whether noggin alone can induce human bone marrow mesenchymal stem cells (MSCs) to differentiate into neural cells for the treatment of neural degenerative diseases.Methods Human bone marrow MSCs were primarily cultured and then divided into control,Ad group of empty adenovirus vector and Ad-noggin group of adenovirus vector with noggin gene.Their morphological changes were observed with fluorescence microscopy and laser confocal microscopy,and the results of 48 h,4,7,10 d were analyzed with statistical methods.Results Both the vector made human bone marrow MSCs carry green fluorescence.Ad group didn’t show any morphological changes.While for Ad-noggin group,a few neural-like cells appeared in 48 h after transfection.The number of such cells was increased,and there were some neurites around the cells after 4 d.The cells amplified greatly after 10 d.Compared to those of 7 d after transfection,the soma and dendritic diameters of Ad-noggin group were significantly increased after 10 d (P

Objective To investigate the affect of body positions on biochemical indexes. Methods By autogenous contrast and cross matched survey, 107 volunteers divided into 3 season patches of winter, spring and summer, blood samples were drawn from the same part in both standing and lying positions。From19 persons, blood samples were collected respectively after standing and sitting for 15 min, lying for 15 min and 30 min and then sitting for another 15 min。 The blood samples were analyzed for 32 biochemical indexes on analyzer。Results 25 biochemical indexes in sitting position were significantly different from those in lying position (P0。05)。Conclusions Changing body position can result in obvious physiological variation of 28 biochemical indexes, particularly of those related to protein. Such result may lead to abnormality in some marginal values. It suggests body position should not be neglected in analyzing laboratory data.

Objective:To study methods for inducing CD34+ cells of human bone marrow to differentiate into T cells in vitro to provide theory and method basics for the investigation of activity of T cells derived from psoriatic bone marrow CD34+ cells and establish a technological platform to investigate T lymphopoiesis activity of hematopoietic cells.Methods:Bone marrow CD34+ hematopoietic progenitor cells were isolated by immunomagnetic cell selection and induced differentiate into T cells in the bone in the marrow and thymic stromal microenvironment.Immunfluorescence dying method and flow cytometry analysis were performed to detect CD1-CD3+ cells,CD3+CD4+CD8-cells and CD3+CD4-CD8+ cells dynamically.Results:In the first week,the non-adhension cells were composed mostly of immature CD1+CD3-cells and CD1+CD3+ cells and small proportion of mature CD1-CD3+ cells.In the following analysis,the proportion of immature cells rapidly decreased and CD1-CD3+ cells increased.After 1 week culture,CD4+CD8+ double positive T cells and a small population CD4+CD8-and CD4-CD8+ could be detected among the CD3+ cells.In the following culture,the proportion of CD4+CD8+ double positive T cells decreased significantly and single positive T cells increased gradually.However,small proportion of mature T cells could be detected in the early stage and cann't be found after 4 weeks in the culture system without thymic stromal cells.Conclusion:Mature single positive T cells can develop from CD34+ hematopoietic progenitor cells in the bone marrow and thymic stromal microenvironment and the thymic stromal cells are vital for T lymphopoiesis.

RESUMEN

There are different kinds of peripancratic diseases that we can hardly differdntiate from pancreatic tumors preoperatively. Seventeen cases of peripancreatic lesions, in which preoperative diagnosis was made as pancreatic tumor during the last six year period were reviewed retrospectively. They were neurogenic tumor(4 cases), tuberculosis of peripancreatic lymph node(2 cases), recurred gastric cancer (2 cases), Castlemann's disease(2 cases), gallbladder cancer(1 case), splenic hematoma(1 case), mesenteric lymphangioma(1 case), lyphoepithelial tumor(1 case), omental cyst(1 case), pheochromocytoma(1 case) and teratoma(1 case). All of them were operated and confirmed histologically. In conclusion, for correct preoperative diagnosis and proper therapy, the application of additional diagnostic measures such as CT or ultrasound guided aspiration cytology, diagnostic laparoscopy, and endoscopic ultrasound should be considered when the diagnosis is not conclusive. The disease entities we mentioned in this paper should be excluded preoperatively or intraoperatively.

Using the prime b(special to M. tuberculosis) to amplify 21 mycobacterium and 13 nonmycobacterium, the products amplified were detected by agarose gel electrophoresis. The sensitivity was 50fg at annealing temperature 61℃,and only M. tuberculosis, M. gastri were amplified, those results showed: the clinical isolated mycobacterium can be differentiated and detected quickly and efficiently by using the prime b(using the prime a when necessary) to amplify 16S~23S rDNA spacer sequence of mycobacterium.

ObjectiveTo study the possibility of differentiation of human umbilical cord mesenchymal stem cells(HUCMSCs) into human male germ cells,and to explore a new source of cells for the treatement of male infertility.MethodsHUCMSCs were transplanted into the seminiferous tubules of the testis of infertile mice by microinjection.The survival rate,migration and germ cell markers of HUCMSCs in the mice testis were detected via immunohistochemistry,immunofluorescence and confocal laser scanning. ResultsHUCMSCs can survive in the mice testis for at least 120 days,and they can migrate from the lumens to the basement membrane.Immunofluorescence showed that HUCMSCs can further differentiate in the mice testicular environment,and express the germ cell marker.ConclusionsHUCMSCs can survive,migrate and differentiate into early male germ cell-like cells in the infertility mice testis after transplantation.

Objective In order to investigate the effect of nutrition and induction of Sertoli cell on cultured neural stem cells(NSCs) from embryonic mesemcephalon in the rat. Methods Sertoli cells from the testis and the neural precursors(NPs) from the ventral mesencephalon in embryonic SD rat(E13) were co-cultured.The detections of immunoreactivities of nestin,?-Ⅲ tubulin and tyrosine hydroxylase(TH) were made by immunocytochemistry after 5, 7, 10 and 14 days in vitro. Results The number of NPs with nestin-positive was decreasing along the time of the co-culture,while the ?-Ⅲ tubulin-positive cells and TH-positive cells were increasing.The number of ?-Ⅲ tubulin-positive cells was quite high at the 7th day and the TH-positive cells were at the highest level at the 10th day in vitro.At any stage,the numbers of TH-positive cells were higher in the co-culture,compared with those in the mono-culture.Conclusion The neural precursors from the ventral mesencephalon in embryonic rat can be directly induced to differentiate into the dopaminergic progenitors by co-cultured with the Sertoli cells.
