RESUMEN

The drug delivery system with "gatekeeper" is designed to achieve a stable entrapment state of the payload under normal physiological conditions through the gatekeepers. With tumor microenvironment or stimulation of exogenous factors, the gatekeeper is detached or altered to promote the responsive release of the drug. In this paper, we focus on applications of stimuli-responsive linkages and stimuli-responsive groups, and review research progresses of drug delivery system with "gatekeeper" developed over the past 10 years.

RESUMEN

OBJECTIVES: Since 2003 South Korea's suicide rate has remained the highest among Organization for Economic Cooperation and Development countries. Gatekeeper training is a major strategy in the national suicide prevention plan in Korea. In this study, we evaluated the effect of the Korean standardized suicide prevention program on the gatekeeper's intervention, developed by the Korea Association for Suicide Prevention and implemented by the Korea Suicide Prevention Center. METHODS: A telephone survey was conducted in order to assess awareness and intervention for the high risk group. Out of the 35825 people who completed the educational course in 2013, 800 gatekeepers were selected by proportional quota sampling. RESULTS: Of the 800 subjects, 93% answered that people in the high risk group of suicide sent diverse signals implying suicide ; 19.9% of the gatekeepers found actual high risk suicidal people, 95% of whom listened to why they came to think about suicide and 71.5% of listeners gave information about suicide prevention and referred them to mental health professionals. CONCLUSION: The Korean standardized suicide prevention program may be helpful to the gatekeepers in playing an adequate role in intervention, but further study is needed in order to evaluate long-term educational effects.

RESUMEN

PURPOSE: This study aims to investigate the effects of a peer gatekeeper training program on existential spiritual well-being, interpersonal relationship with friends, and suicide probability for middle school students. METHODS: The participants were fifty students who were at a vocational middle school in D city from october 13, 2012 to november 10, 2012. To test the effects of the peer gatekeeper training program, the participants were divided into two groups in the way of matching distribution, an experimental group(24) and a control group(26). Data were analyzed by Chi-square test, t-test, and Fisher exact probability test using the SPSS/WIN 12.0 program. RESULTS: After attending the peer gatekeeper training program, significant differences between the experimental group and the control group were found in existential spiritual well-being(t=2.80, p=.007) and interpersonal relationship with friends scores(t=2.67, p=.010). However, there was no statistically significant difference in suicide probability between the two groups. CONCLUSION: The results indicate that the peer gatekeeper training program enhance existential spiritual well-being and interpersonal relationship with friends of middle school students. Therefore, this program could be a good strategy that teachers and community mental health nurses can use for suicide prevention of middle school students.

RESUMEN

PURPOSE: This study was done to investigate the effects of a Peer Gatekeeper Training Program on self-esteem, interpersonal relationships with friends and suicide probability for high school students. METHODS: The recruited participants were forty-three students a vocational high school in G city. The study was done from August 31, 2012 to September 25, 2012. To test the effects of the Peer Gate Keeper Training Program, the participants were divided into two groups, an experimental group (22) and a control group (21). The Peer Gate Keeper Training Program was provided to the control group after the posttest was completed. Data were analyzed using chi2-test, t-test, and Fisher exact test with the SPSS/WIN 18.0 program. RESULTS: After attending the Peer Gate Keeper Training Program, significant differences were found in self-esteem, interpersonal relationships with friends, and suicide probability scores between the experimental group and the control group (t=3.24, p=.002) (t=2.16, p=.036) (t=-2.95, p=.006). CONCLUSION: The results indicate that the Gate Keeper Training Program enhances self-esteem and interpersonal relationships with friends and reduces suicide probability in high school students. Therefore, this program is a good strategy that teachers and community mental health nurses can use for suicide prevention in high school students.

RESUMEN

PURPOSE: This study was done to evaluate the effects of the peer suicide prevention gatekeeper program for use with middle school students. METHODS: The research design was a quasi-experimental research of a nonequivalent control group. The gatekeeper program was used as the experimental treatment with the students from May 14 to June 11, 2010. The 54 research participants were assigned to the experimental (23) or control (31) group. The gatekeeper program was administered four times to the experimental group. Data analysis was done with SPSS 12.0 for t-test and ANCOVA. RESULTS: There were significant changes in the level of problem solving competence. There were no significant changes in the level of depression, and suicidal ideation. CONCLUSION: These results indicate that the Gatekeeper program is an effective nursing intervention to increase the level of problem solving competence in middle school students.

RESUMEN

To investigate the RNA interference (RNAi) effect induced by vector-derived small interfering RNA (siRNA) targeting the three gatekeeper genes (Rad52, Ku70, Ku80) and screen the more effective target sites from candidates for further research, by using siRNA design tools online,we selected 2 candidate sequences directed to every gatekeeper gene. According to the sequences, six vector-derived siRNAs (denoted psiRNA1-6) and one mocking psiRNA7 were constructed. Among them, psiRNA1 and psiRNA2 targeted Rad52, psiRNA3 and psiRNA4 to Ku70, psiRNA5 and psiRNA6 to Ku80. The mocking psiRNA7 was used as control. After sequence identification, the seven plasmids were transfected into HepG2 cell line. siRNA-induced silencing of gatekeeper genes was determined by using RT-PCR at RNA level and Western Blot at protein level. The results showed that the six plasmids specifically targeting the coding region of gatekeeper genes were successfully designed and constructed. To some extent, the six plasmids could reduce the expression of target gene.Comparatively, the plasmid-derived siRNA psiRNA1, psiRNA4 and psiRNA5 were more effective than their counterparts. The results suggest that the gene silencing efficiency of siRNA is different,depending on their targeted region, and siRNA may provide us with practical tools for further study on the three gatekeeper genes, i.e. Rad52, Ku70, Ku80.
