To observe the effect of pIgR on Salmonella Enteritidis induced inflammation in jejunum and ileum in Chicken,7 day Hyline chickens were taken orally with Salmonella Enteritidis (SE) and killed after 1 d,3 d,7 d and 14 d.The mRNA expression of Toll-like receptor 4(TLR4),MyD88,TRAF6,NF-κB and pIgR was detected by real-time RT-PCR and pIgR protein level was detected by Western blot.The results showed that TLR4 signaling pathway was activated and mR-NA level of the pIgR in jejunum and ileum was enhanced (P＜0.01) and protein level of the pIgR in jejunum and ileum was up-regulated by SE.The study proved that TLR4 signal pathway on mucosal cell surface of jejunum and ileum was activated and expression of pIgR was up-regulated and gut mucosal immunity of chicken was strenghtened.

Objective:To study the influence of TNF-? on the expression of secretory component (SC) in Caco-2 cells.Methods:Immunocytochemistry,ELISA,Western blot and quantitative real-time PCR were used to test SC-positive cells of Caco-2,free SC in culture supernatants,protein and mRNA expression of SC in the cells.Results:The increase of SC-positive cells,free SC in culture supernatants,SC protein expression and expression of SC genes in Caco-2 cells was found under stimulation of TNF-? treatment compared with control(P

Objective:To generate rabbit anti-human Fc?/?R and Poly Immunoglobulin Receptor(pIgR) polyclonal antibodies.Methods:The EC12 domain of human Fc?/?R and the D1 domain of pIgR were expressed as including bodies in Escherichia coli.The purified proteins were used to immunize rabbits to generate polyclonal antibodies.The immune sera were characterized by ELISA and Western blot analysis,then absorbed with cross-action and purified by antigen linked beads.Results:The titers of antiserum were 1∶6 4000 and 1∶25 6000,separately.There was little cross-reaction between the two immune sera.Conclusion:The polyclonal antibodies against human Fc?/?R and pIgR without cross-reaction were successfully generated and could be useful tools to study the relationship between the two receptors.
