RESUMEN

Objective To explore the value of ultrasound elastography in the non-invasive monitoring of liver elasticity of stable recipients at different stages after liver transplantation. Methods Clinical data of 73 stable recipients after liver transplantation were collected. According to the time after liver transplantation, all patients were divided into the early group (n=25) and medium-to-long group (n=48). In addition, 38 healthy subjects were assigned into the control group. The ultrasound indexes and liver function indexes were statistically compared among each group. The ultrasound elastography indexes of liver and spleen were analyzed, and their correlation with liver function indexes was analyzed. Results Compared with the control group, the ultrasound indexes, alanine aminotransferase (ALT), γ-glutamyl transferase (GGT) and alkaline phosphatase (ALP) levels were significantly increased in the early group (all P < 0.05), and the ultrasound indexes in the medium-to-long group were significantly increased, whereas the GGT level was significantly decreased (all P < 0.05). Compared with the early group, the right oblique diameter of liver, ALT, GGT and ALP levels were significantly decreased in the medium-to-long group (all P < 0.05). Compared with the control group, the sound touch elastography (STE) and sound touch quantify (STQ) values of liver and STE value of spleen in the early group and medium-to-long group were significantly increased (all P < 0.05). Compared with the early group, the ultrasound elastography indexes of liver and spleen in the medium-to-long group were remarkably decreased (all P < 0.05). The ultrasound elastography indexes of liver were weakly correlated with the ALT, aspartate aminotransferase (AST) and GGT levels, significantly correlated with ALP level. The STE value of spleen was weakly correlated with the ALP level. The STE value of liver was significantly correlated with the STQ value of liver. The STE and STQ values of liver were weakly correlated with the STE value of spleen. Conclusions The characteristics of liver elasticity in stable recipients after liver transplantation are various among different stages. Persistent monitoring of liver elasticity may provide a novelnon-invasive monitoring method during follow-up after liver transplantation.

Objective To assess the applicability of sound touch elastography ( ST E) and sound touch quantify ( ST Q ) in measuring liver and spleen stiffness . Methods One hundred and eighteen healthy volunteers were included and underwent ST E and ST Q . T he success rate ,variability and reproducibility of ST E and ST Q were analyzed . T he accurate sampling size and number of tests for liver ST Q were also analyzed . Results T he success rates ,variability ,reproducibility of ST E and ST Q in liver were 97 .5% and 99 .2% ,8 .7% and 12 .0% ,0 .917 and 0 .916 , respectively . While those with spleen were 76 .3% and 66 .9% ,12 .4% and 16 .4% ,0 .847 and 0 .706 ,respectively . The sampling size of 1 .5 cm×1 .0 cm yield the lowest variability ( 8 .5% ) ( F =6 .562 , P =0 .002) ,and there was no significant difference between results of detecting 5 times and 10 times( P =0 .571) . T he liver and spleen stiffness of ST E were 5 .75 kPa ( 95%CI :5 .60-5 .91 kPa) and 15 .58 kPa ( 95% CI :14 .99 -16 .16 kPa) . Conclusions The measurement of liver stiffness using both ST E and ST Q have a high success rate and low variability . However ,ST E is better than STQ in measuring spleen stiffness .

Objective To activate partial thromboplastin time (APTT) indicating the coagulation state in intrinsic coagulation system and the efficacy of anticoagulant drugs, and develop a bioassay method to quantify anticoagulating bioactivity in Chuanxiong Rhizome and related Chinese patent medicines for quality assessment. Methods Chuanxiong Rhizome powder was extracted in ethanol and water, respectively. The mixed extract was used as sample to quantify the level of extended APTT in vitro. The blood was taken from the heart of rabbit. The agglomerating time was determined after adding APTT reagent in plasma. The prolongation rate of APTT was chosen as biomarker for anticoagulating bioactivity. Sodium ferulate was chosen as reference. The amount of anticoagulating bioactivity was quantified in Chuanxiong Rhizome extract by the Amount reaction of parallel line analysis (2.2) method. Moreover, the amounts of anticoagulating bioactivity were quantified in the nine Chuanxiong Rhizome samples including Chuanxiong Rhizome raw materials, decoction pieces, and related Chinese patent medicines. Results Both sodium ferulate and Chuanxiong Rhizome extract showed significant anticoagulating bioactivity (P < 0.01). The reliability test for quantifying the level of anticoagulating bioactivity in solidum ferulate and Chuanxiong Rhizome extract was passed through, and the measured value was valid. The correlation coefficient was 0.995 5 (n = 5) between the concentration of solidum ferulate in the range of 1 — 5 mg/mL and their rates of anticoagulating bioactivity. The RSD for the amounts of anticoagulating bioactivity was 9.34% (n = 6) by six replicated tests with the confidence limit rate of 15.98% (n = 6). The amounts of anticoagulating bioactivity were significant difference among various types of Chuanxiong Rhizome samples, i.e. 5.431 — 7.620 U/g for five Chuanxiong Rhizome raw materials, 5.910 and 3.017 U/g for Chuanxiong decotion pieces and processed slice with yellow wine, 14.516 and 29.035 U/g for Tongmai Granules and Xuefu Zhuyu Pills. Conclusion The developed method can accurately quantify the level of anticoagulating bioactivity in Chuanxiong Rhizome raw materials, decoction pieces and related Chinese patent medicines, and assess their quality.

RESUMEN

Objective To study the effects of standardized-lifestyle intervention on the type 2 diabetic elder patients.Methods The study group enrolled 270 individuals with type 2 diabetes mellitus randomly by using the model of “Zhiji Health Care” and the software of the lifestyle and disease prevention system.Parameters were collected during a six month period for the evaluation,such as fasting plasma glucose(FPG),postprandial glucose(PPG),cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),body mass(BMI),glycosylated hemoglobin(HbAlc),blood pressure,daily intake,total physical activity and effective physical activity and other indicators.Results After six months' standardized lifestyle intervention:(1)in the type 2 diabetic patients,the parameters of weight,BMI,waist circumference,systolic and diastolic blood pressure were significantly decreased(t values were:23.82,25.91,19.56,23.37 and 19.53,respectively,all P < 0.01); FPG,PPG,HbA1c,TC,TG and LDL-C were significant decreased compared with that before intervention(t values were: 12.06,11.82,9.74,6.98,5.79 and 6.10,respectively,all P < 0.0l).Meanwhile,HDL-C was significantly increased(t =2.51,P < 0.05).(2)the total daily intake was significantly reduced compared with that before intervention.The effective exercise burden was significantly increased,compared with that before intervention(P < 0.01).Conclusion Standard lifestyle management helps to control the blood glucose,blood pressurem,body weight and other risk factors,with reduction of complications and medical costs.

RESUMEN

Objective To find a rapid and simple method for quantifying the amount of eluted proteins in chromatography. Methods The method was developed by utilizing the principle of the absorbance of protein at 280 nm and the integral function of the software UNICORN for AKTA FPLC, namely, the protein quantity was evaluated based on its A_ 280 and peak area. Results The relation between peak area and quantity of the protein was linear correlation. The protein quantity could be expressed as X=Y/A. The Y represented the peak area (mAU?ml) of the interested fraction, and A represented the measurable value (mAU) of 1 g/L interested protein through the FPLC 280 nm cell and X was the quantity of the interested protein (mg). The method was used in the purification of recombinant human zona pellucida-3 protein (rhZP3)and the quantity of the purified rhZP3 evaluated from the formula was confirmed by Lowry method. Conclusion The method is easy, rapid, repeatable and practicable to quantify the protein in chromatography.
