RESUMEN

OBJECTIVE:To investigate penetrative effects of a penetration enhancer and multiple penetration enhancers combi-nation with different proportions on Euodia rutaecarpa superfines cataplasm,so as to optimize enhancer and their concentrations. METHODS:Modified Franz diffusion cell was employed with isolated mice abdominal skin as barrier. HPLC method was adopted to detect the effects of a penetration enhancer (propanediol, azone, oleic acid), multiple penetration enhancers (propanedi-ol-azone,propanediol-oleic acid,propanediol-azone-oleic acid),their proportion and amount on accumulative permeation quantity (Qn) of evodiamine and rutaecarpin in Euodia rutaecarpa superfines cataplasm. RESULTS:The penetrative effect of a penetration enhancer propanediol was significantly better than that of other one penetration enhancers and multiple penetration enhancers;the higher proportion of propanediol in multiple penetration enhancer system was,the better penetrative effects of evodiamine and rutae-carpin had. Using 3%,5 % and 7 % propanediol as enhancer,Q36 h of evodiamine were 11.290,14.332 and 13.537 μg/cm2,and those of rutaecarpin were 11.965,14.856 and 13.901 μg/cm2. CONCLUSIONS:The penetrative effect of 5% propanediol is the best,and can be used as enhancer for E. rutaecarpa superfines cataplasm.

RESUMEN

Objective: To apply the high performance liquid chromatography (HPLC) method to the determination of four ingredients including paeoniflorin, berberine, evodiamine, and rutaecarpin in Wuji Pellet. Methods: Using Thermo-ODS2-Hypersil column (250 mm × 4.6 mm, 5 μm) and taking acetonitrile-0.05 mol/L monopotassium phosphate as mobile phase for elution, the conditions were as follows, flow rate of 1.0 mL/min, UV detector wavelength of 225 nm, and column temperature of 25 °C. Results: The standard curves of paeoniflorin, berberine, evodiamine, and rutaecarpine showed a good linear relationship in 0.0124-0.124, 0.0349-0.349 mg/mL, and 0.6-6, 0.5-5 μg/mL. The average recovery rates were 99.45%, 97.69%, 98.18%, and 98.46%, respectively. Conclusion: The method is of high sensitivity and nice reproducibility, and it could be used for the simultaneous determination of the four ingredients in Wuji Pellet. © 2013 Tianjin Press of Chinese Herbal Medicines.

RESUMEN

AIM: To establish HPLC fingerprint of Sishen Pill and a determination of main component of Sishen Pills(Fructus Evodiae, Semen Myristicae, Fructus Psoraleae, etc.) by HPLC. METHODS: The samples were extracted with the solvent methanol. The separation was performed on YWG-C 18(4.6mm?250mm, 10?m) column with water- methanol-acetonitrile as a mobile phase gradient elution. The flow rate was 1.0mL?min -1, the column temperature was at 30℃ and the wavelength of UV detector was at 225nm. RESULTS: There are 21 higher peaks and 12 marker peaks in the fingerprint of eight batches of samples. For evodiamine, the regression equation was Y=147.37X+1186.32, r=0.9999, the liner range was from 5 to 1000?g?mL -1 and the recovery was 99.83%. For rutaecarpin, the regression equation was Y=101.19X+279.81, r= 0.9999, the liner range was from 4.5 to 900?g?mL -1. The recovery was 98.72%. CONCLUSION: The quality standard of Sishen Pill was established firstly. It can be used for quality cantrol of Sishen Pill.
