RESUMEN

Aim To investigate the regulatory effect of morphine postconditioning in the LSG on remodeling after myocardial infarction. Methods SD rats were randomly divided into four groups: sham operation group (Sham), myocardial infarction group (MI), myocardial infarction + saline group (Control) and myocardial infarction + morphine postconditioning group (MI + Morphine) . The rat MI model was constructed by ligating the left anterior descending coronary artery, and then morphine was given to the LSG by percutaneous posterior approach. After four weeks, the changes of cardiac function in rats were detected by ultrasound. Masson staining was used to detect fibrosis changes; the expression of Collagen I and Collagen III protein was detected by Western blot. The mRNA expression of ANP and BNP was detected by RT-qPCR. The expression of JJLOR in LSG was detected by immunofluorescence. The concentration of catecholamine in plasma and myocardial tissue was detected by ELISA. Results Compared with the sham group, the cardiac function of the MI group was significantly impaired, the myocardial tissue showed significant fibrosis changes, and the concentration of catecholamine in plasma and myocardial tissue significantly increased. Compared with the control group, the MI + Morphine group reduced myocardial fibrosis collagen deposition in rats after MI, inhibited the expression of ANP and BNP in myocardial tissue, reduced the concentration of catecholamine, and improved the cardiac function of MI rats. Immunofluorescence results showed that JJLOR was expressed in LSG after MI and increased after morphine postconditioning. Conclusions This study shows that morphine postconditioning in the LSG has a protective effect on myocardial remodeling after myocardial infarction. The mechanism may be related to the activation of JJLOR in the LSG by morphine and the reduction of catecholamine release from sympathetic nerve endings.

RESUMEN

Objective To evaluate the effect of stellate ganglion block (SGB) on cellular immune function in diabetic rats. Methods Healthy male Sprague-Dawley rats, aged 3 months, weighing 240-280 g,were used in this study.Diabetes mellitus was induced by intraperitoneal 1% streptozotocin 60 mg/kg and confirmed by blood glucose ≥ 16.7 mmol/L 3 days later.Forty-eight rats with diabetes mellitus were randomly divided into 2 groups (n=24 each) using a random number table:diabetes mellitus group (group DM) and group SGB.Another 24 healthy rats, aged 3 months, were selected and served as control group (group C).At 1 week after successful establishment of the model, unilateral transection of cervical sympathetic trunk (TCST) was performed in group SGB,while the right cervical sympathetic trunk was only exposed in C and DM groups. Before TCST (T0) and on 1,3,7 days after TCST (T1-3),6 rats were randomly selected from each group, and blood samples were collected from the inferior vena cava for determination of the blood glucose, plasma norepinephrine (NE) concentrations (by enzyme-linked immunosorbent assay),and levels of T lymphocyte subsets CD3+,CD4+ and CD8+ in whole blood (using FACSCalibur flow cytometer).C D4+/CD8+ratio was calculated. The rats were weighed before sacrifice, and the rats were sacrificed to obtain the thymus which was weighed. The thymus index (thymus weight/body weight) was calculated. Results Compared with group C,the blood glucose was significantly increased, and the levels of CD3+ and CD4+ in whole blood,CD4+/CD8+ ratio, and thymus index were significantly decreased at T0-3 (P<0.05),and no significant change was found in CD8+ levels in DM and SGB groups (P>0.05),the plasma NE concentrations were significantly decreased at T1-3 in group SGB (P<0.05),and no significant change was found in plasma NE concentrations in group DM (P>0.05).Compared with group DM,the blood glucose and plasma NE concentrations were significantly decreased, and the levels of CD3+ and CD4+ in whole blood,CD4+/CD8+ ratio, and thymus index were significantly increased at T1-3 (P<0.05),and no significant change was found in CD8+ levels in group SGB (P>0.05).Conclusion SGB can improve the cellular immune function in diabetic rats.

RESUMEN

Objective To investigate formalin-induced nociceptive behavior and c-fos expression of interbrain following stellate ganglionic block (SGB) in rabbits. Methods Catheters were inserted closely to right stellate ganglia in rabbits by operation. One week later,twenty-four rabbits were randomly divided into three groups, each group containing 8 animals. groups A,B and C received 0.5ml of normal saline,3% formalin and 3% formalin stimulation by intraplantar injection , respectively. 10 minutes before stimulation,0.5 ml of 0.25% bupivacaine was administered via the catheter in group B, while in groups A and C 0.5 ml of normal saline was applied. Nociceptive behavior was observed for 1 hour using weighted pain score. 2 hours after stimulation, whole interbrain was removed to immunohistochemically detect the expression of c-fos. Results Formalin-induced nociceptive behavior in phase 2 was relieved following SGB, c-fos expression level of hypothalamus in group B was significantly lower than that in group C(P0.05). Conclusion SGB could relieve formalin-induced nociceptive behavior, and downregulate formalin-induced c-fos expression of hypothalamus, which may be related to the mechanisms of SGB for the treatment of inflammatory pain.

RESUMEN

The two-way projection of the intrinsic cardiac neurons in the rat has been studied by means of CB-HRP retrograde tracing and double-labelling with fluorescent tracers. CB-HRP was injected into the right stellate ganglian of the 20 rats. CB-HRP positive neurons were found in the intrinsic cardiac ganglia, moreover, most of the labelled cells localized in the ganglia which were nearby the epicardium of posterior wall of the atria and the root of large vessles. Bb and PI were injected the right stellate ganglia and the wall of the cardiac ventricle of 30 rats respectively, PI-Bb double labelling neurons were found in the posterior wall of the atria for the first time. The present study demonstrated that a few of intrinsi ccardiac neurons give off two long processes, one of which projects to the ventricle, and the other one extends to the stellate ganglion. They may be sensory and may participate the composition of vegetative nervous reflex pathway of heart outside CNS.
