The study compared the cardiorespiratory, hemogasometric and sedative effects of the combination of midazolam (0.41mg/kg) and butorphanol (0.31mg/kg) plus detomidine (157µg/kg) (DTMB) or dexmedetomidine (36µg/kg) (DXMB) in collared peccaries. Collared peccaries (n= 20) were divided into two groups, either DTMB or DXMB. The variables (FC, f, PAM, SpO2, EtCO2 and TR) were evaluated after application of the drugs. Sedation was assessed by visual analogue scale, muscle relaxation, posture and auditory response. ANOVA followed by paired t-test (parametric) and Mann Whitney Rank Sum Test (non-parametric) with P< 0.05 were performed. No statistical difference was observed for the latency period. A significant increase was observed between groups for the variables f, PAM and SpO2 with higher values for DTMB and EtCO2 with higher values for DXMB. The two groups presented a reduction in HR and lactate concentration, and an increase in bicarbonate concentration. SpO2 remained below 90% throughout the experiment in both groups. The animals of the two groups presented deep sedation and maximum muscle relaxation. It is concluded that the two protocols tested provided adequate sedation and could be indicated for chemical containment of collared peccaries.(AU)

RESUMEN

Oocyte in vitro maturation (IVM) is the first step of the in vitro reproductive technologies that enables mature oocytes to be generated ex vivo and after used for embryo production. In this sense, the establishment of culture environment, as oocyte incubation time, is essential for the success of the IVM. Therefore, the study was carried out to investigate the relationship between the meiotic potential and the IVM times of collared peccary oocytes, wild mammals of great commercial and ecological interest. Thus, ovaries were collected of females derived from captivity and transported to the laboratory within 1 hour of slaughtering. The oocytes derived from follicles (3-6mm in diameter) were recovered by aspirated and sliced. Good quality oocytes (evenly granulated cytoplasm with a least one layer of surrounding cumulus cells) were selected and subjected to culture in TCM 199 supplemented with 10µg/mL FSH, 10% FBS and 100µM cysteamine at 38.5°C, 5% CO2 and maximum humidity for 24 or 48 hours. After the incubation period, the nuclear status, the presence of first polar body and the expansion of cumulus cells of oocytes were assessed. The data obtained were analyzed by Fisher exact test (P<0.05). A total of four sessions (2-3 females per session) were performed, resulting in eighteen aspirated and sliced ovaries with normal morphological characteristics. An oocyte recovery rate of about 83.1% (59/71) was obtained with 3.3 oocytes/ovary and 2.3 viable oocytes/ovary. After different incubation times, differences (P<0.05) were observed in 24 and 48 hours for expansion of the cumulus cells (38.1% vs. 100%), presence of first polar body (52.4% vs. 90.5%) and nuclear status in second metaphase (19.0% vs. 76.2%), respectively. In conclusion, 48 hours is suitable time for the in vitro maturation of oocytes derived from collared peccaries when compared to the time of 24 hours, according to the meiotic potential observed. Additional studies should be conducted to improve the quality of the oocyte culture environment, as medium composition, aiming to obtain viable mature oocytes for other in vitro biotechnologies.(AU)

Animal-plant interactions in Neotropical forests are complex processes. Within these processes, mid-to large-sized mammals consume fruits and seeds from several species; however, because of their size these mammals are overhunted, resulting in defaunated forests. Our objective was to evaluate and compare seed removal and survivorship in a forest with no hunting, a forest with moderate or reduced hunting, and a forest with higher hunting pressure. We examined the interaction between Astrocaryum gratum and white lipped peccary (Tayassu pecari) to tease apart the defaunation process. To isolate and evaluate mammal seed removal rates and to identify the causes of mortality on A.gratum, under the three different hunting pressures forests, we used exclosures in each one. In four different forest-patches for each forest, we positioned a block-treatment consisting of three exclosures (total exclusion, peccary exclusion, and control), randomly distributed 5m apart and the block-treatments spaced 50-75m apart from one another. We established 15 treatments in total for each patch (5 blocks per patch). There were 20 blocks within each forest type. For total exclusion, all vertebrates were excluded using galvanized wire mesh exclosures. The second, the peccary exclusion, was designed to stop peccaries from entering treatment units, providing access only to small vertebrates; larger mammals were able to access the treatment unit by reaching over the sides and the open top; finally, the Control allowed full access for all mammals. Fresh A. gratum fruits were collected from the forest floor under different adult trees throughout the study area. In each exclosure treatment, twenty A.gratum seeds were placed, and their removal was recorded. In total, 3 600 seeds were analyzed. Seed survival was lower in unhunted forest compared to areas with moderate hunting and forest with a higher hunt pressure, supporting the hypothesis of the importance of mammals in seed removal. From the initial 400 seeds left for each control exclosure in each type of forest, there was a significant difference between the seed removal; 1.75% seeds in the unhunted forest remained; 43.5% in the moderately hunted forest, and 48.5% in hunted forest. The main cause of seed mortality was white lipped peccaries; while in the forests without them, the main removal was caused by rodents and a higher insect infection was observed in the heavily hunted forest. Our results indicated that defaunation affects seed survivorship. Rev. Biol. Trop. 63 (1): 57-67. Epub 2015 March 01.

Collared peccaries (Peccary tajacu) are among the most hunted species in Latin America due the appreciation of their pelt and meat. In order to optimize breeding management of captive born collared peccaries in semiarid conditions, the objective was to describe and correlate the changes in the ovarian ultrasonographic pattern, hormonal profile, vulvar appearance, and vaginal cytology during the estrus cycle in this species. During 45 days, females (n=4) were subjected each three days to blood collection destined to hormonal dosage by enzyme immunoassay (EIA). In the same occasions, evaluation of external genitalia, ovarian ultrasonography and vaginal cytology were conducted. Results are presented as means and standard deviations. According to hormonal dosage, six estrous cycles were identified as lasting 21.0 ± 5.7 days, being on average 6 days for the estrogenic phase and 15 days for the progesterone phase. Estrogen presented mean peak values of 55.6 ± 20.5 pg/mL. During the luteal phase, the high values for progesterone were 35.3 ± 4.4 ng/mL. The presence of vaginal mucus, a reddish vaginal mucosa and the separation of the vulvar lips were verified in all animals during the estrogenic peak. Through ultrasonography, ovarian follicles measuring 0.2±0.1 cm were visualized during the estrogen peak. Corpora lutea presented hyperechoic regions measuring 0.4±0.2 cm identified during luteal phase. No significant differences (P>0.05) between proportions of vaginal epithelial cells were identified when comparing estrogenic and progesterone phases. In conclusion, female collared peccaries, captive born in semiarid conditions, have an estral cycle that lasts 21.0±5.7 days, with estrous signs characterized by vulvar lips edema and hyperemic vaginal mucosa, coinciding with developed follicles and high estrogen levels.(AU)

Biological studies are necessary for the management of wildlife in captivity, and knowledge of reproduction is one of the important features for increasing production. The objective of the research was to determine the age at which male collared peccaries reach puberty. Testicular samples of 15 animals, aged 7 to 16 months, distributed into five groups (G1, G2, G3, G4 and G5) were used. The testes showed considerably increased weight, length and width (p < 0.05) from G1 to G3, whereas, from this group onward, the development of this organ was slower. There was positive correlation (p < 0.001) between the following testicular parameters: weight and length (r = 0.97), weight and width (r = 0.88), length and width (r = 0.92). Regarding the diameter of seminiferous tubules, an increase was observed (p < 0.05) from G1 to G4. The total number of spermatogenic cells increased significantly (p < 0.05) until G3 and then it stabilized. There was also positive correlation between testis weight and tubular diameter (r = 0.99, p < 0.001), and testis weight and spermatogenic cells (r = 0.98, p < 0.001). The number of Sertoli cells decreased significantly (p < 0.05) from G1, when they were undifferentiated as support cells, to G5, when they occurred together with the complete line of spermatic cells. The results demonstrate that the reproductive development of peccaries can be classified into the following stages: impuberty (G1, 7-8 months); pre-pubertal (G2, 9-10 months); pubertal (G3, 11-12 months); post-pubertal 1 (G4, 13-14 months); and post-pubertal 2 (G5, 15-16 months). Based on the histological analyses, puberty in the male collared peccary was determined to occur between 11 and 12 months of age.

The sustainability and production of collared peccary (Pecari tajacu) has been studied in the last few years; however, further information on its reproduction is necessary for breeding systems success. Understanding folliculogenesis aspects will contribute to effective reproductive biotechniques, which are useful in the preservation and production of wildlife. The aim of this study was to evaluate the ovarian folliculogenesis in collared peccary. Ovaries from six adult females of collared peccary were obtained through ovariectomy and analyzed. These were fixed in aqueous Bouin’s solution and sectioned into 7μm slices, stained with hematoxilin-eosin and analyzed by light microscopy. The number of pre-antral and antral follicles per ovary was estimated using the Fractionator Method. The follicles, oocytes and oocyte nuclei were measured using an ocular micrometer. Results showed that the length, width, thickness, weight, and the gross anatomy of the right and left ovaries were not significantly different. However, the mean number of corpora lutea was different between the phases of the estrous cycle (p<0.05), with the highest mean in the luteal phase. Primordial follicles were found in the cortex; the oocytes were enveloped by a single layer of flattened follicular cells. In the primary follicles, proliferation of the follicular cells gave rise to cuboidal cells (granulosa cells). The secondary follicle was characterized by two or more concentric layers of cuboidal cells (granulosa), beginning of antrum formation, and the presence of pellucid zone and theca cells. Antral follicles were characterized by a central cavity (antrum), the presence of cumulus oophorus and theca layers (interna and externa). In the right ovary, the values of the primordial and primary follicles were similar, but significantly different from the secondary ones (p<0.05). In the left ovary, significant differences were observed between all follicles in the follicular phase (p<0.05); the mean number of primordial and primary follicles was similar in the luteal phase. The mean number of pre-antral follicles and antral follicles in the follicular phase was higher in the left ovary (p<0.05). The mean number of antral follicles in the luteal phase was similar in both ovaries. We also found significant differences in mean diameter of preantral follicles, oocyte, granulosa layer and oocyte nucleus during the estrous cycle. In the antral follicles a significant difference was observed only in follicular diameter (p<0.05). The predominance of active primordial and primary follicles was found in both phases; otherwise the secondary follicles and antral follicles showed a high degree of degeneration. The results obtained in the present work will strengthen the development of biotechnology programs to improve the productive potential and conservation of the collared peccary.

We studied the role of white-lipped and collared peccaries (Tayassu pecari and Pecari tajacu) as seed predators and dispersers in the Atlantic rainforest of Brazil. The Atlantic rainforest ecosystem is highly threatened and has experienced dramatic declines in its populations of large mammals. Local extinctions can disrupt essential plant-animal interactions such as seed dispersion and seed predation. We tracked seeds from time of consumption to germination to assess the direct impact peccaries have on seed survival. We offered fruits of 20 species found in the Atlantic rainforest to the peccaries. Seeds were categorised as intact, scarified, ingested or defecated, and germination tests were performed. The overall impact by both peccary species was similar. Seeds were sometime scarified by mastication, always with fatal consequences. Most seeds that were consumed were destroyed during ingestion and digestion. Only small seeds (<10 mm) were found in the feces and germination tests suggest a positive effect from the passage through the guts. Peccaries clearly have a double role as both seed predators and as small seeds dispersers, which is a specialised role within the granivore/frugivore community of the Atlantic rainforest.

We described the macroscopic and quantitative anatomy of the trachea of the Giraffe (Giraffa camelopardalis rothschildi). The trachea of one juvenile male giraffe (25 months of age) weighing 754 kg was used in this study. The length of the neck was 125 cm. The trachea had 107 cm in length on its cervical part and 18 cm on its thoracic part. The total number of cartilage was 87.74 at the neck and 13 at the thorax. The general shape of the duct was mostly circular. The separation of the dorsal ends of the tracheal cartilages was pronounced in the first half of the cervical trachea, reducing caudally and overlapping in the thoracic trachea. In the caudal part the trachea had a tracheal bronchus for the cranial lobe of the right lung and the end of the trachea was divided into two main bronchi, where the left was larger in diameter.

We determined the potential nutritional levels in 25 species of plants, and in earthworms, that constitute part of the diet of white-lipped peccary (Tayassu pecari) in Corcovado National Park, Costa Rica, from January 1998 to March 1999. The highest content of fat and energy was found in seeds of the Myristicacea family. The highest content of calcium was found in vegetative parts of Dieffenbachia spp. Nutritious contents differed among plant parts (seeds, fruits, stems and leaves). Fat and energy content were larger in seeds and fruits, whereas the largest content of protein was found in fruits and leaves. Mineral content also differed among plant parts. Calcium, potassium and magnesium were higher in leaves whereas copper and zinc were higher in seeds. Differences of diet between white-lipped peccaries in Corcovado and in other tropical regions of Latin America could be partially explained by our results. We found several species with higher fat and energy content than palms, which can explain the low consumption of palm seeds in Corcovado. It is possible that the regular consumption of stems and leaves of some species is related to their high mineral content. Seasonality of reproduction in Corcovado seems to be related not only to fruit availability but also to the nutritional quality of food.

White-lipped peccaries are non-seasonal breeders in South America, but little is known about their reproduction in Central America. There are few studies about the sex ratio of this species in the field. We studied the reproduction and sex ratio of white-lipped peccaries during 200 hours of field observation of four radiomarked and two unmarked herds, from July 1996 to April 1997, in Corcovado National Park, Costa Rica. Sex ratio data of three additional, radiomarked herds observed in 1998 were also included. We recorded numbers of mountings, presence of newborns and numbers of nursing interactions. The peccaries showed a distinct reproductive seasonality, with one mating peak in February and another in July. The greatest number of newborns and the peak in nursing activity were observed during July and August, when fruit availability for the peccaries was high. The adult sex ratio was significantly female biased (1.4:1-1.8:1), also in contrast with South American populations.
