RESUMEN

<p><b>AIM</b>The production of interspecific germline chimeras between chicken and quail were attempted employing the dissociated cells derived from the blastodermal central disk (stage X) and the germinal crescent region of embryo (stage 7-8).</p><p><b>METHODS</b>The central disk (CD) of the area pellucida in chicken blastoderm (stage X) and the germinal crescent region (GCR) of embryo (stage 7-8) were dispersed and injected into the subgerminal cavity of quail blastoderm (stage X). Injected eggs were incubated for 7 days or to hatching. The donor chicken DNA was detected by the polymerase chain reaction.</p><p><b>RESULTS</b>In day-7 embryos, chicken DNA was detected in 5 gonads and 9 brains from 53 survived embryos received chicken CD cells, and 1 gonads and 6 brains from 27 survived embryos received chicken GCR. Chicken DNA was also detected from the semen of one adult male hatched from eggs received chicken GCR cells.</p><p><b>CONCLUSION</b>CD and GCR cells as the donors showed the possibility to produce the interspecific germline chimera, but further studies are needed to make necessary improvement.</p>

RESUMEN

Aspirin (acetylsalicyclic acid) was dissolved either in normal saline or in phosphate buffer and was used in two doses to find out whether teratogenic potential of aspirin in chick blastoderm model is due to its acidic property or due to drug action. Drug was injected sub-blastodermally by window technique in fresh embryonated eggs after 17 hours of incubation at 39 degrees C. Eggs were re-incubated and harvested at 40 hours. Normal development of embryos was seen with normal saline and percentage of normal embryos with 30 micrograms (pH-3.19) and 120 micrograms (pH-2.64) aspirin was 31.7 and 4.9 respectively. Buffer produced 80.8% normal embryos and buffered 30 micrograms (pH-6.87) and 120 micrograms (pH-6.69) aspirin produced 67.7% and 30.8% normal embryos respectively. Changing the pH of aspirin to near neutral decreased the defect induced by aspirin but a significant effect of aspirin was observed at higher dose which could be independent of pH action.

RESUMEN

Four hundred and forty four full primitive streak stage chick embryos were cultured in vitro and 261 were transplanted with 1, 3 or 5 Hensen's nodes in the area opaca. Irrespective of the number of grafts, neural induction was observed in 90% cases. The development of control and grafted embryos and the size of blastoderm area were monitored at the time of grafting and after 20 hr. We find that the induced neural tissue and differentiated tissue of graft-origin neither fuse with the host embryonic axis, nor retard its development.

RESUMEN

Primitive streak stage chick blastoderms were cultured in vitro up to 30 hr by New's technique. Chick blastoderms reaching stages 4 to 12 in vitro cultures and in ovo were harvested and homogenized to release cell nuclei. Fluorescent ethidium bromide-stained nuclei in homogenates were counted in Neubauer's chamber and the size of total blastoderm cell population was determined. Linear regression analysis revealed that both in ovo and in vitro chick blastoderm cell population grows in a biphasic manner with comparable cell population doubling times and the morphogenesis is not affected in vitro during the culture period.

RESUMEN

Teratoma of the testis is a type of mixed tumor made up of various kinds of tissue representing the three primitive layers of blastoderm. Recently we experienced two cases of testicular teratoma in 23-year-old male patients, so we report these two cases of testicular teratoma with review of literatures.
