O presente estudo analisou um binômio de tempo-temperatura alternativo para ser utilizado na pasteurização lenta sobre a inativação da fosfatase alcalina no leite caprino. Sua eficiência foi demonstrada pela contagem padrão em placas, e foi feita a comparação no processamento de leite refrigerado e congelado. Foram utilizados 18 tratamentos em leite caprino cru (nove em leite refrigerado e nove em leite congelado). Estes foram acondicionados em frascos de 300 mL, pasteurizados a 60, 63 e 65°C durante 10-20-30 minutos, e testadas às enzimas fosfatase alcalina e peroxidase. A contagem padrão em placas (CPP) e coliformes a 35 e 45°C foi feita nas amostras cruas e em cada tratamento, em duplicata. Após a pasteurização, todos os tratamentos apresentaram: não crescimento de microrganismos mesófilos, coliformes com <0,3 NMP/mL, prova de fosfatase negativa e peroxidase positiva. A pasteurização foi eficiente para melhorar a qualidade microbiológica do leite tanto refrigerado quanto congelado. Todos os binômios avaliados apresentaram resultados satisfatórios para alcançar os parâmetros preconizados em legislação, sugerindo-se o menor binômio (60°C por 10 min). Não houve diferença entre as formas de armazenamento das amostras: refrigerada ou congelada.


The objective of this study was to investigate an alternative time-temperature binomial to be used in the slow pasteurization on the alkaline phosphatase inactivation in the goat milk. Its efficiency was demonstrated with the standard counting in plates, and also refrigerated and the frozen milks processing were compared. Eighteen treatments were used in the raw goat milk (nine refrigerated milk and nine frozen milk). They were packed in 300 mL-flasks, pasteurized at 60-63-65°C for 10, 20, 30 minutes, and then tested for alkaline phosphatase and peroxidase enzymes. The standard counts in plates (CPP) and coliforms at 35°C and 45°C were performed in the raw samples and in the every treatment, in duplicate. After the pasteurization process, all of the treatments showed: no growth of mesophilic microorganisms, coliforms with <0.3 MPN / mL, negative phosphatase and positive peroxidase tests. The pasteurization was efficient to improve the microbiological quality of the milk either refrigerated or frozen. All of the evaluated binomials presented satisfactory results to reach the recommended parameters preconized in the legislation, suggesting the smaller binomial (60°C for 10 min). There was no difference between the samples storage form, either refrigerated or frozen.

A presente comunicação objetivou avaliar a quantificação do caseínomacropeptídeo (CMP), bem como diferenciá-lo (devido à adulteração com soro) do pseudo-CMP (devido à proteólise bacteriana) em amostras de leite cru coletadas nos domicílios do sul do Brasil. Os resultados reforçam a necessidade de práticas higiênicas durante a ordenha e estocagem do leite. As amostras de leite estudadas não estavam adulteradas por adição de soro, mostrando que a análise por cromatografia de exclusão por tamanho deve ser complementada a fim de revelar a identidade do peptídeo (CMP ou pseudo-CMP). A contagem bacteriana total (TBC) também se mostrou útil como indicador da contaminação do leite por micro-organismos proteolíticos, uma vez que uma relação diretamente proporcional entre TBC e pseudo-CMP foi estabelecida.(AU)

ABSTRACT Wild plant species with potential for use as ornamental green foliage are often reported. Nevertheless, there are few studies evaluating morphological and anatomical aspects associated with this use. Preliminary trials suggest that the species of Sphaeradenia (Sander ex Mast.) Harling, a Cyclantaceae that occurs from Costa Rica to Peru, is suitable to be used as fresh foliage because of attractiveness of their leaves in vase and its durability without showing tissue damage, compared with other genera of this family. Through the study of anatomical characters, it is possible to predict the potential of a species that can serve as ornamental foliage. In this study, three foliar anatomical features (thickness of the cuticle and leaf, and percentage of fibers in the mesophyll) of seven Cyclanthaceae species were evaluated, using anatomical techniques and image analysis with the LUCIA® and ImageJ softwares. Statistical analysis of the results suggests that this high durability is associated with the thickness of the cuticle and mesophyll, but not with the amount of fibers in the leaf.

O conhecimento da anatomia da folha é crucial para o entendimento da adaptação das plantas ao ambiente. O objetivo deste estudo foi descrever a anatomia da lâmina foliar de 11 espécies lenhosas, frequentes nas savanas do extremo norte da Amazônia, com ênfase na identificação de atributos adaptativos a ecossistemas abertos, sujeitos a forte insolação e déficit hídrico sazonal. Amostras de folhas foram coletadas e processadas segundo técnicas usuais para estudos de anatomia e histoquímica. Bowdichia virgilioides, Byrsonima coccolobifolia, By. crassifolia, By. verbascifolia, Casearia sylvestris, Curatella americana, Erythroxylum suberosum, Himatanthus articulatus, Miconia albicans, Roupala montana e Xylopia aromatica apresentaram caracteres típicos de plantas heliófilas e xerófilas, como cutícula espessa e estômatos predominantes na face abaxial, além de forte investimento em tecido fotossintético. Em oito das onze espécies, o parênquima paliçádico (PP) ocupa 50% ou mais do espaço do mesofilo. Curatella americana, mesofilo isobilateral, e Bo. virgilioides, mesofilo homogêneo, foram as espécies com maior investimento em PP (~80% e 100%, respectivamente). Além disso, destaca-se a presença de hipoderme (Bo. virgilioides e X. aromatica) ou de epiderme estratificada, densos indumentos, idioblastos cristalíferos e extensões da bainha de feixes. Em síntese, este conjunto de atributos estruturais protege a lâmina foliar contra o excesso de luminosidade, aumenta a resistência mecânica, minimiza a transpiração e contribui para manutenção do balanço hídrico da planta, favorecendo, portanto, o estabelecimento destas espécies nas savanas sazonais do norte da Amazônia.


Knowledge of the anatomical characteristics of the leaf blade is crucial to the understanding of plant adaptation to the environment. The objective of this study was to describe the leaf anatomy of 11 woody species of common occurrence in the open savannas of the northern edge of the Amazon. The focus of the study was on the identification of leaf adaptive features to cope with high irradiances and seasonal water deficits. Leaf samples were fixed and processed by the usual methods for anatomical and histochemical studies. Bowdichia virgilioides, Byrsonima coccolobifolia, By. crassifolia, By. verbascifolia, Casearia sylvestris, Curatella americana, Erythroxylum suberosum, Himatanthus articulatus, Miconia albicans, Roupala montana and Xylopia aromatica showed leaf anatomical traits typical of heliophilous and xerophilous plants such as thick cuticle, stomata prevailing on the abaxial surface, strong investment in photosynthetic tissue. In eight of the eleven species the palisade parenchyma (PP) occupied 50% or more of the mesophyll. Curatella americana, with isobilateral mesophyll, and Bo. virgilioides, with homogeneous mesophyll were the species with the highest investment in PP (~80% and 100%, respectively). Leaves were also characterized by the presence of hypodermis (Bo. virgilioides and X. aromatica) or stratified epidermis, dense indumenta, crystalliferous idioblasts and bundle sheath extensions. This distinctive assortment of anatomical traits helps protecting the leaf blade against excessive irradiances, increases mechanical strength, minimize transpiration and contribute to the maintenance of leaf water balance. Overall they favor the establishment of these species in the seasonal savannas of northern Amazon.

Realizou-se estudo com o objetivo de analisar as alterações na anatomia foliar de Gallesia integrifolia (Spreng) Harms e Schinnus terebinthifolius Raddi quando cultivadas em ambientes de sombra moderada ou densa, simulando as condições naturais encontradas em sistemas agroflorestais tradicionais do sul da Bahia, Brasil. Plantas das duas espécies, com aproximadamente um ano de idade, foram cultivadas em casa de vegetação sob quatro níveis de sombreamento (25%, 17%, 10% e 5%). Estudos anatômicos do limbo foliar foram realizados a partir de material incluído em parafina e seccionado em micrótomo rotativo. Os diferentes níveis de sombreamento ocasionaram alterações na estrutura do mesofilo de ambas as espécies, com diferenças significativas na espessura do parênquima paliçádico, limbo foliar, e densidade estomática. Nas condições em que o experimento foi realizado os resultados obtidos indicaram que G. integrifolia apresenta maior capacidade de aclimatação a ambientes de sombra moderada e densa do que S. terebinthifolius, sendo mais indicada para o cultivo em sistemas agroflorestais pré-estabelecidos.


This study was conducted with the aim of analyzing the changes in the leaf anatomy of Gallesia integrifolia (Spreng) Harms and Schinnus terebinthifolius Raddi when grown in environments with moderate to dense shade, simulating the natural conditions found in traditional agroforestry systems in southern Bahia, Brazil. Plants of both species, being approximately one year-old, were grown at four irradiance levels (25%, 17%, 10% and 5%) in a greenhouse. Leaf anatomical studies were made from material embedded in paraffin and sectioned on a rotary microtome. The different irradiance levels caused a shift in the mesophyll anatomy of both species, with significant differences in palisade parenchyma thickness and leaf stomatal density. In the conditions of this experiment, the results indicated that G. integrifolia has a higher capacity for acclimatization to moderate to dense shade than S. terebinthifolius, and it is thus more suitable for cultivation in pre-established agroforestry systems.

The changes of cell ultra structure as well as Ca2+ homeostasis involved in the drought-induced maize leaf senescence was investigated. Meanwhile, many indicatives of leaf senescence including thiobarbituric acid reactive substance (MDA), electrolyte leakage (EL), and chlorophyll along with soluble proteins were also detected during the process. The Polyethylene glycol6000(PEG6000)-incubated detached leaves showed a slight increase in the MDA content and electrolyte leakage during the first 30 min of our detection, which was corresponded to an unobvious alteration of the cell ultrastructure. Other typical senescence parameters measured in whole leaf exhibited a moderate elevation as well. Thereafter, however, the EL and MDA rose to a large extent, which was correlated with a dramatic damage to the cell ultrastructure with concomitant sharp decrease in the chlorophyll and soluble proteins content. The deposits of calcium antimonite, being an indicator for Ca2+ localization, were observed in the vacuoles as well as intercellular spaces in the leaves grown under normal condition. Nevertheless, after PEG treatment, it was revealed a distinct increment of Ca2+ in the cytoplasm as well as chloroplasts and nuclei. Moreover, with long-lasting treatment of PEG to the detached leaves, the concentration of Ca2+ as described above showed a continuous increment which was consist with the remarked alteration of physiological parameters and severe damage to the ultrastructure of cells, all of which indicated the leaf senescence. Such drought-induced leaf senescence might result from a loss of the cell's capability to extrude Ca2+. All above findings give us a good insight into the important role of Ca2+ homeostasis in the process of leaf senescence accelerated by the drought stress.
