RESUMEN

Crop residues decomposition are controlled by chemical tissue components. This study evaluated changes on plant tissue components, separated by the Van Soest partitioning method, during cover crop decomposition. The Van Soest soluble fraction was the first to be released from the crop residues, followed by cellulose and hemicellulose. Lignin was the crop residue component that suffered the least degradation, and for certain crop residue types, lignin degradation was not detected. The degradation of the main components of crop residues (soluble fraction, cellulose, hemicellulose and lignin) is determined by the chemical and structural composition of each fraction.


A decomposição de resíduos culturais é controlada pela composição química do tecido vegetal. O objetivo deste estudo foi avaliar as alterações que ocorrem nos componentes do tecido vegetal, separados pelo fracionamento de Van Soest, durante a decomposição de plantas de cobertura. A fração solúvel foi a primeira a ser liberada dos resíduos culturais, seguida pela celulose e hemicelulose. A lignina foi o componente dos resíduos culturais de menor degradação, sendo que em alguns resíduos culturais não foi possível detectar a degradação deste componente. A degradação dos principais componentes dos resíduos culturais (fração solúvel, celulose, hemicelulose e lignina) é determinada pela composição química e estrutural de cada uma destas frações.

RESUMEN

The shortage of medical resources promotes medical treatment reform, and smart healthcare is a promising strategy to solve this problem. With the development of Internet, real-time health status is expected to be monitored at home by using flexible healthcare systems, which puts forward new demands on flexible substrates for sensors. Currently, the flexible substrates are mainly traditional petroleum-based polymers, which are not renewable. As a natural polymer, cellulose, owing to its wide range of sources, convenient processing, biodegradability and so on, is an ideal alternative. In this review, the application progress of nanocellulose in flexible sensors is summarized. The structure and the modification methods of cellulose and nanocellulose are introduced at first, and then the application of nanocellulose flexible sensors in real-time medical monitoring is summarized. Finally, the advantages and future challenges of nanocellulose in the field of flexible sensors are discussed.

Avicel is made of a mixture of microcrystalline cellulose (MCC) and carboxymethyl cellulose (CMC), and used for virus plaque assay. The avicel in common use is produced by FMC Biopolymer. Due to the relatively fixed proportion of MCC and CMC, avicel in common use is not suitable for plaque determination experiment of all types of viruses. In this study, we evaluated the effect of avicel made of different proportions of MCC and CMC on virus plaque assay, and developed an improved avicel virus plaque assay featured with simple and convenient operation, good practicability and high stability. To generate avicel overlays with different proportions of MCC and CMC, twelve different 2×avicel solutions were prepared. Their overall viscosity and bottom viscosity were measured to evaluate the ease of operation. The results showed that most of the 2×avicel solutions (except the 4.8% MCC+1.4% CMC and 4.8% MCC+1.0% CMC group) were easy to absorb and prepare nutrient overlap than 2×CMC solution. In order to find the best scheme to detect the titer of porcine epidemic diarrhea virus (PEDV), these avicel overlay solutions with different proportion of MCC and CMC were used as a replacement in the standard plaque assay. By comparing the size, clarity, stability and titer accuracy of virus plaque, we identified that 0.6% MCC and 0.7% CMC was the most preferable composition of avicel overlay for PEDV plaque assay. In conclusion, we developed an improved virus plaque assay based on avicel, which may facilitate the research of virus etiology, antiviral drugs and vaccines.

Gluconacetobacter xylinus is a primary strain producing bacterial cellulose (BC). In G. xylinus, BcsD is a subunit of cellulose synthase and is participated in the assembly process of BC. A series of G. xylinus with different expression levels of the bcsD gene were obtained by using the CRISPR/dCas9 technique. Analysis of the structural characteristics of BC showed that the crystallinity and porosity of BC changed with the expression of bcsD. The porosity varied from 59.95%-84.05%, and the crystallinity varied from 74.26%-93.75%, while the yield of BC did not decrease significantly upon changing the expression levels of bcsD. The results showed that the porosity of bacterial cellulose significantly increased, while the crystallinity was positively correlated with the expression of bcsD, when the expression level of bcsD was below 55.34%. By altering the expression level of the bcsD gene, obtaining BC with different structures but stable yield through a one-step fermentation of G. xylinus was achieved.

Background: Fuels and chemicals from renewable feedstocks have a growing demand, and acetone, butanol and ethanol (ABE) are some relevant examples. These molecules can be produced by the bacterial fermentation process using hydrolysates generated from lignocellulosic biomass as sugarcane bagasse, one of the most abundant sources of lignocellulosic biomass in Brazil. It originates as a residue in mills and distilleries in the production of sugar and ethanol. Results: In the present work, two strategies to generate hydrolysates of sugarcane bagasse were adopted. The fermentation of the first hydrolysate by Clostridium acetobutylicum DSM 6228 resulted in final concentrations of butanol, acetone and ethanol of 6.4, 4.5 and 0.6 g/L, respectively. On the other hand, the second hydrolysate presented better results (averages of 9.1, 5.5 and 0.8 g/L, respectively), even without the need for nutrient supplementation, since key elements were already present in the medium. The productivity (QP) and yield (YP/S) of the solvents with second hydrolysate were 0.5 g/L•h-1 and 0.4 g/g, respectively. Conclusions: The results described herein open new perspectives for the production of important molecules from residual lignocellulosic biomass for the fuel and chemical industries within the context of second-generation biorefinery.

Background: Pretreatment of lignocellulosic biomass is essential for using it as a raw material for chemical and biofuel production. This study evaluates the effects of variables in the chemical pretreatment of the Arundo biomass on the glucose and xylose concentrations in the final enzymatic hydrolysate. Three pretreatments were tested: acid pretreatment, acid pretreatment followed by alkaline pretreatment, and alkaline pretreatment. Results: The amounts of glucose and xylose released by the enzymatic hydrolysis of the Arundo biomass obtained from acid pretreatment ranged from 6.2 to 19.1 g/L and 1.8 to 3.1 g/L, respectively. The addition of alkaline pretreatment led to a higher yield from the enzymatic hydrolysis, with the average glucose concentration 3.5 times that obtained after biomass hydrolysis with an acid pretreatment exclusively. The use of an alkaline pretreatment alone resulted in glucose and xylose concentrations similar to those obtained in the two-step pretreatment: acid pretreatment followed by alkaline pretreatment. There was no significant difference in 5-hydroxymethylfurfural, furfural, or acetic acid concentrations among the pretreatments. Conclusion: Alkaline pretreatment was essential for obtaining high concentrations of glucose and xylose. The application of an alkaline pretreatment alone resulted in high glucose and xylose concentrations. This result is very significant as it allows a cost reduction by eliminating one step.

Resumo Os novos materiais restauradores em desenvolvimento devem evitar danos aos tecidos dentários. Portanto, o objetivo deste estudo foi avaliar a biocompatibilidade de uma marca comercial de cimento de ionômero de vidro convencional (CIV) modificado com microfibras de celulose (CIV+MC) ou nanocristais de celulose (CIV+NC) através da implantação de três amostras em tecido subcutâneo na região dorsal de 15 ratos Rattus norvegicus albinus. Cada rato recebeu um exemplar de cada cimento, resultando nos seguintes grupos (n=15): Grupo CIV (controle, n=15), Grupo CIV+MC e Grupo CIV+NC. Nos intervalos de 7, 30 e 60 dias os animais foram sacrificados e os seguintes aspectos foram avaliados histologicamente: tipo de células inflamatórias, fibroblastos, vasos sanguíneos, macrófagos, células gigantes, tipo de reação inflamatória e espessura da cápsula (µm). Estes eventos foram quantitativamente classificados conforme os escores: (-) ausente, (+) suave, (++) moderado e (+++) intenso. Os resultados foram analisados estatisticamente pelo teste Kruskal-Wallis e pós-teste Mann-Whitney. Aos 7 dias, o Grupo CIV+NC demonstrou um nível mais elevado de reparação tecidual porque havia maior quantidade de fibroblastos (p=0,022) e uma menor quantidade de macrófagos (p=0,008) e células mononucleares (p=0,033). Neutrófilos e células gigantes estavam ausentes em todos os períodos experimentais. Aos 60 dias, o Grupo CIV+NC apresentou cápsula de tecido fibroso com espessura mais reduzida (26,72±2,87 µm) em comparação ao Grupo CIV+MC (41,21±3,98 µm (p=0,025). No geral, todos os materiais apresentaram satisfatória biocompatibilidade, no entanto, o cimento de ionômero de vidro modificado com nanocristais de celulose proveu reparação tecidual mais avançada comparativamente aos demais materiais avaliados.


Developing new restorative materials should avoid damage to tissue structures. This study evaluated the biocompatibility of a commercial dental glass ionomer cement (GIC) mechanically reinforced with cellulose microfibers (GIC+CM) or cellulose nanocrystals (GIC+CN) by implantation of three test specimens in subcutaneous tissue in the dorsal region of 15 Rattus norvegicus albinus rats. Each rat received one specimen of each cement, resulting in the following groups (n=15): Group GIC (Control), Group GIC+CM and Group GIC+NC. After time intervals of 7, 30 and 60 days, the animals were sacrificed and the following aspects were histologically evaluated: type of inflammatory cells, fibroblasts, blood vessels, macrophages, giant cells, type of inflammatory reaction and capsule thickness (µm). These events were scored as (-) absent, (+) light, (++) moderate and (+++) intense. The results were statistically analyzed by Kruskal-Wallis test and Mann-Whitney post test. At 7 days, Group GIC+NC showed more favorable tissue repair because quantitatively there were more fibroblasts (p=0.022), fewer macrophages (p=0.008) and mononuclear cells (p=0.033). Polymorphonuclear neutrophils and giant cells were absent in all experimental periods. At 60 days, test specimens in Group GIC+NC were surrounded by a fibrous tissue capsule with reduced thickness (26.72±2.87 µm) in comparison with Group GIC+CM (41.21±3.98 µm) (p=0.025). In general, all biomaterials showed satisfactory biocompatibility, but glass ionomer cement modified with cellulose nanocrystals showed a more advanced tissue repair.

Native rhizobia are ideal for use as commercial legume inoculants. The characteristics of the carrier used to store the inoculants are important for the survival and symbiotic potential of the rhizobia. The objective of this study was to investigate the effects of peat (PEAT), perlite sugarcane bagasse (PSB), carboxymethyl cellulose plus starch (CMCS), and yeast extract mannitol supplemented with mannitol (YEMM) on the survival, nodulation potential and N2 fixation capacity of the native strains Sinorhizobium mexicanum ITTG R7T and Rhizobium calliandrae LBP2-1T and of the reference strain Rhizobium etli CFN42T. A factorial design (4 × 3) with four repetitions was used to determine the symbiotic potential of the rhizobial strains. The survival of the strains was higher for PEAT (46% for strain LBP2-1T, 167% for strain CFN42T and 219% for strain ITTG R7T) than for the other carriers after 240 days, except for CFN42T kept on CMCS (225%). All the strains kept on the different carriers effectively nodulated common bean, with the lowest number of nodules found (5 nodules) when CFN42T was kept on CMCS and with the highest number of nodules found (28 nodules) when ITTG R7T was kept on PSB. The nitrogenase activity was the highest for ITTG R7T kept on PEAT (4911 μmol C2H4 per fresh weight nodule h−1); however, no activity was found when the strains were kept on YEMM. Thus, the survival and symbiotic potential of the rhizobia depended on the carrier used to store them.

Los pilares terapéuticos del niño con shock séptico se mantienen en el tiempo, sin embargo, se han incorporado nuevos conceptos, siendo importante que el pediatra y el intensivista tengan conocimiento a cabalidad de ellos. La reanimación con fluidos es una intervención fundamental, no obstante, aún no se ha establecido un tipo de fluido ideal, presentando cada uno limitaciones específicas, no existiendo evidencia sobre la superioridad de un tipo de fluido. Si a pesar de una adecuada resucitación con fluidos persiste el shock, el inicio de inótropos y/o vasopresores está indicado. En caso de refractariedad al uso de vasopresores, nuevos fármacos vasoactivos pueden ser empleados y el uso de hidrocortisona debe considerarse en niños con sospecha de insuficiencia suprarrenal. Existe controversia respecto a la transfusión de glóbulos rojos o el nivel óptimo de glucemia, no existiendo consenso en el valor umbral para el uso de estos hemocomponentes o el inicio de insulina, respectivamente. Asimismo, la utilización de la hemofiltración de alto volumen (HFAV)aún permanece controversial, requiriendo mayores estudios para su recomendación en forma rutinaria en el curso de un shock séptico refractario. El soporte nutricional es primordial, ya que la desnutrición es una grave complicación que debe ser prevenida y tratada adecuadamente. El objetivo de la presente revisión es entregar una actualización en los más recientes avances en tratamiento del shock séptico en la población pediátrica.


Essential therapeutic principles in children with septic shock persist over time, although some new concepts have been recently incorporated, and fully awareness of pediatricians and intensivists is essential. Fluid resuscitation is a fundamental intervention, but the kind of ideal fluid has not been established yet, as each of these interventions has specific limitations and there is no evidence supportive of the superiority of one type of fluid. Should septic shock persists despite adequate fluid resuscitation, the use of inotropic medication and/or vasopressors is indicated. New vasoactive drugs can be used in refractory septic shock caused by vasopressors, and the use of hydrocortisone should be considered in children with suspected adrenal insufficiency, as it reduces the need for vasopressors. The indications for red blood cells transfusion or the optimal level of glycemia are still controversial, with no consensus on the threshold value for the use of these blood products or the initiation of insulin administration, respectively. Likewise, the use of high-volume hemofiltration is a controversial issue and further study is needed on the routine recommendation in the course of septic shock. Nutritional support is crucial, as malnutrition is a serious complication that should be properly prevented and treated. The aim of this paper is to provide update on the most recent advances as concerns the treatment of septic shock in the pediatric population.

In this study, conventional restorative glass ionomer cement (GIC) was modified by embedding it with mechanically processed cellulose fibers. Two concentrations of fibers were weighed and agglutinated into the GIC during manipulation, yielding Experimental Groups 2 (G2; 3.62 wt% of fibers) and 3 (G3; 7.24 wt% of fibers), which were compared against a control group containing no fibers (G1). The compressive strengths and elastic modulus of the three groups, and their diametral tensile strengths and stiffness, were evaluated on a universal test machine. The compressive and diametral tensile strengths were significantly higher in G3 than in G1. Statistically significant differences in elastic modulus were also found between G2 and G1 and between G2 and G3, whereas the stiffness significantly differed between G1 and G2. The materials were then characterized by scanning electron microscopy (SEM) and energy dispersive X-ray spectroscopy (EDS). Heterogeneously shaped particles were found on the G2 and G3 surfaces, and the cement matrices were randomly interspersed with long intermingled fibers. The EDS spectra of the composites revealed the elemental compositions of the precursor materials. The physically processed cellulosic fibers (especially at the higher concentration) increased the compressive and diametral tensile strengths of the GIC, and demonstrated acceptable elastic modulus and stiffness.

The frequent contamination of water resources with drugs comprises one the most important environmental problems. In order to avoid serious disturbances for aquatic life, efficient and economically viable procedures should be developed for removing common pollutants, as paracetamol. From these considerations, the present work evaluated the efficiency of sugar cane bagasse (SCB) and vegetable sponge (VS), two natural adsorbents commonly found in Brazil, for retaining paracetamol molecules dispersed in aqueous solutions. Thus, systems composed of glass columns and peristaltic pumps were optimized and, for pH, the best value was 7.0. After optimisation, adsorption isotherms were built and it was possible to calculate the MAC F values for SCB (120.5 ug/g) and VS (37.5 ug/g). Additionally, real matrices of pretreated water, from a municipal plant for water catchment, were enriched with paracetamol at 5 uM and passed through glass columns packed with SCB, VS and activated carbon (AC). The results showed that SCB was more attractive than AC in terms of price and efficiency (60 percent against 45 percent adsorption, respectively), while VS was responsible for removing 40 percent of paracetamol dissolved in the enriched water samples. Thus, the proposed natural adsorbents can be classified as viable materials to remove paracetamol from water used for human consumption.

Being controlled release dosage forms, tablets allow an improved absorption and release profiles of Ofloxacin. The fact that drugs with fine particles size can be compressed well after wetting, so in our research studies Ofloxacin controlled release matrix tablets were prepared by wet granulation technique. In order to investigate the potential of Ethyl cellulose ether derivatives as a matrix material, Ofloxacin formulations with different types and grades of Ethocel were prepared at several drug-to-polymer ratios. The method adopted for in vitro drug release studies was USP Method-1 [rotating Basket Method] by Pharma test dissolution apparatus using phosphate buffer 7.4 pH as a dissolution medium. Various Kinetic models were employed to the formulations for the purpose of determination of release mechanism. A comparative study was performed between the tested Ofloxacin-Ethocel formulations and a standard reference obtained from the local market. FI dissimilarity factor and f2 similarity factor were applied to the formulations for the checking of dissimilarities and similarities between the tested formulations and reference standard

Chilli fruit is highly susceptible to anthracnose infection at the stage of harvest maturity, due to which the fruit yield in the leading commercial variety Byadgi is severely affected. Field studies on screening of several varieties for resistance to anthracnose have shown that a variety of chilli AR-4/99K is resistant to anthracnose infection. In many crops, resistance to fungal attack has been correlated with PGIP activity in developing fruits based on which transgenic varieties have been developed with resistance to fungi. The present study was carried out to determine whether anthracnose resistance in AR-4/99K was due to the increased levels of PGIP alone and/ or due to differences, if any, in the properties of PGIP. Hence, a comparative study of the properties of polygalacturonase inhibitor protein (PGIP) isolated from fruits of anthracnose resistant chilli var AR-4/99K and a susceptible variety Byadgi was conducted with the objective of utilizing the information in genetic transformation studies. Both the PGIPs from anthracnose resistant and susceptible varieties of chilli exhibited similarities in the elution pattern on Sephadex gel, DEAE cellulose, PAGE and SDS-PAGE. The two PGIPs were active over a wide range of pH and temperature. Both PGIPs showed differential inhibitory activity against polygalacturonase (PG) secreted by Colletotrichum gleosporoides, C. capsici, C. lindemuthianum, Fusarium moniliforme and Sclerotium rolfsii. The inhibitory activity of PGIP from both resistant and susceptible varieties was the highest (82% and 76%, respectively) against the PG from Colletotrichum capsici, a pathogen causing anthracnose rot of chilli, while the activity was lower (1.27 to 12.3%) on the other fungal PGs. Although PGIP activity decreased with fruit maturation in both the varieties, the resistant variety maintained a higher activity at 45 days after flowering (DAF) as compared to the susceptible variety which helped it to overcome the infection by anthracnose as against the susceptible variety (Byadgi) in which PGIP activity was drastically reduced at maturity. The molecular mass of PGIP as determined by SDS-PAGE was found to be 37 kDa. N-terminal sequence analysis of the PGIP showed the first six amino acid residues from N-terminal end were Asp-Thr-His-Lys-Ser-Glu (DTHKSE), respectively. The similarities in properties of the two PGIPs support the earlier findings that resistance of AR-4/99K to anthracnose fungus is a result of its higher PGIP activity at maturity.

RESUMEN

A gene encoding a -1,3-1,4-glucanase (CelA) belonging to family 5 of glycoside hydrolases was cloned and sequenced from the Bacillus subtilis A8-8. The open-reading-frame of celA comprised 1499 base pairs and the enzyme was composed of 500 amino acids with a molecular mass of 55 kDa. The recombinant -1,3-1,4 glucanase was purified by GST-fusion purification system. The pH and temperature optima of the enzyme were 8.0 and 60oC, respectively. The enzyme was stable within pH 6.0-9.0. It was stable up to 60oC and retained 30% of its original activity at 70oC for 60 min. It hydrolyzed lichenan, CMC, xylan, laminarin, avicel and pNPC, but was inactive towards cellobiose. The enzyme activity was markedly activated by Co2+ and Mn2+, but was strongly inactivated by Fe3+. The truncated gene, devoid of cellulose-binding domain (CBD) showed 60% of activity and bound to avicel.

The surface of coir pith, an agricultural solid waste was modified using a cationic surfactant, hexadecyltrimethylammonium bromide (HDTMA) and the modified coir pith was investigated to assess the capacity for the removal of phosphate from aqueous solution. Optimum pH for maximum phosphate adsorption was found to be 4.0. Langmuir and Freundlich isotherms were used to model the adsorption equilibrium data. Kinetic studies showed that the adsorption obeyed second order kinetics. Thermodynamic parameters were evaluated and the overall adsorption process was spontaneous and endothermic. Effect of coexisting anions has also been studied. The feasibility of using spent adsorbent as fertilizer carrier to control phosphate release was also investigated.

Biotransformation of rifamycin B to rifamycin S using two strains of C. lunata namely NCIM 716 and NMU grown on various solid substrates viz., grass, paper, jowar/wheat straw, bran and bagasse was studied. Almost complete biotransformation efficiency of rifamycin B at 0. 06 mM concentration was observed within 24 hr. Among these two strains, C. lunata NMU showed 90% of biotransformation and higher rate of cellulose utilization on solid substrates vis-à-vis reference strain. Cellulase activity of both strains was also studied for exoglucanase, endoglucanase and beta-glucosidase. Column bioreactor studies with bagasse revealed further improvement in biotransformation efficiency of C. lunata NMU.

The effect of pH, nutrient and aeration was studied on the removal of colour and reduction of BOD, COD and heavy metals with addition of readily available source of nitrogen and phosphorus in concentration of 1.0 g/L. Recalcitrant compound was effectively degraded by active microbial consortia. The isolated bacteria were identified as Pseudomonas putida (S1), Citrobacter sp. (S4) and Enterobacter sp. (S5). These organisms not only decolourised effluent upto 97% but reduced BOD, COD, phenolics and sulfide upto 96.63, 96.80, 96.92 and 96.67% respectively within 24 hrs of aeration and the heavy metals were removed upto 82-99.80%. The TSS and TDS were sharply reduced due to degradation. The absorption maxima was also decreased to 90%. However, in control without the microbial consortium no noticeable change was produced.

RESUMEN

Chewable tablets containing low dosage flouride content were prepared using two varities of celluloses and their in vitro parameters were evaluated. An eighteen month clinical trial revealed that both these formulations were effective in controlling the caries. However, ethyl cellulose is proved to be superior to methylcellulose as a controlled release matrix material in controlling caries. Thus this study recommends ethylcellulose matrix tablets containing low flouride content is an efficacious and cost effective drug device in controlling dental caries.

Production of beta-galactosidase by Sclerotium rolfsii NCIM 1084 was studied under submerged fermentation conditions. The enzyme was produced extracellularly and constitutively on glucose. The enzyme production was enhanced when galactose, raffinose, cellobiose, sucrose, xylose, maltose, cellulose and pectin were used as carbon sources. Cellulose and diammonium hydrogen phosphate were best carbon and nitrogen sources, respectively. Surfactants such as Sag, Paraffin oil, Tween 20 and Tween 80 increased the enzyme production. Maximum yield of beta-galactosidase obtained was 3.8-4.2 nkat/ml. The optimum pH, optimum temperature and molecular weight of the beta-glactosidase were 2.7, 60 degrees C and 2,21,000 daltons, respectively. The enzyme is an aryl beta-glactosidase and did not hydrolyse lactose. The Km value for o-nitrophenyl beta-D-galactoside was 3.7 mM. Galactose and 2-mercaptoethanol inhibited the enzyme.
