Abstract Current study investigated the effectiveness of different macrophytes as culture media for Ankistrodesmus gracilis in laboratory conditions. Significant difference (p < 0.05) was reported in cell density with regard to conventional culture medium (CHU12) and macrophytes culture media. Mean cell density in NPK, Eichhornia crassipes and E. azurea media was higher (p < 0.05) than in conventional culture medium. Chlorophyll-a was higher than 1 g.L-1, except in CHU12 (0.7 ± 0.4 g.L-1) and T. domingensis (0.8 ± 0.3 g.L-1) media. Nitrate decreased sharply as from the 7th-day of the experiment. Ammonium and total phosphorus were highest in culture media and ranged between 0.4 g.L-1 (P. cordata medium) and 1.7 g.L-1 (CHU12 medium) for ammonium, and between 0.8 g.L-1 (CHU12 medium) and 1.9 g.L-1 (T. domingensis medium) for total phosphorus. Results revealed inorganic fertilizer and macrophytes combined with vitamins may be effective as culture media and strongly supports the growth of Ankistrodesmus gracilis, since cell density and biochemical composition are similar to or higher than conventional culture medium (CHU12). Macrophyte is a tool for aquaculture since biological wastes may be used with nutrients to improve the cultivation of microalgae.


Resumo O presente estudo investigou a eficácia na utilização de diferentes macrófitas como meio de cultura para Ankistrodesmus gracilis em condições laboratoriais. Foi observada diferença significativa (p < 0,05) entre a densidade celular em relação aos meios de cultura convencional (CHU12) e de macrófitas. Os meios de cultura com Eichhornia crassipes, E. azurea e NPK apresentaram densidade celular maiores (p < 0,05) que o meio de cultura convencional. Os teores de clorofila-a foram superiores a 1 g.L-1, exceto nos meios de cultura CHU12 (0.7 ± 0.4 g.L-1) e de T. domingensis (0.7 ± 0.3 g.L-1). O nitrato diminuiu acentuadamente a partir do 7º dia de experimento. Dentre os nutrientes, amônia e o fósforo total foram os mais elevados nos meios de cultura, variando entre 0.4 g.L-1 (meio de P. cordata) a 1,7 g.L-1 (meio CHU12) para amônia e, entre 0.8 g.L-1 (meio CHU12) e 1,9 gL-1 (meio de T. domingensis) para o fósforo total. Os resultados do presente estudo mostraram que o uso de fertilizantes inorgânicos e macrófitas, combinados com vitaminas, pode ser eficaz como meio de cultura no crescimento de Ankistrodesmus gracilis, uma vez que a densidade celular e a composição bioquímica foram semelhantes ou superiores ao meio de cultura convencional (CHU12). As macrófitas são ferramentas a serem adotadas na aquicultura, desde que os resíduos biológicos podem ser usados como nutrientes para melhorar o cultivo de microalgas.

Abstract Extraction of compounds from microalgae requires cell disruption as a pretreatment to increase extraction yield. Botryococcus braunii is a microalga with a significant content of carotenoids and other antioxidant compounds, such as chlorophylls. Cell disruption of B. braunii using CO2 rapid depressurization was studied as a pretreatment for the extraction of carotenoid and chlorophyll pigments. We studied the effect of temperature (21–49 °C) and pressure (6–13 MPa) during static compression on pigment recovery with supercritical CO2 at 40 °C, 30 MPa and solvent flow of 4.7 L NPT/min. Within the experimental region, the extraction yield of carotenoids and chlorophylls increased by 2.4- and 2.2-fold respectively. Static compression conditions of high pressure and low temperature increased the extraction of carotenoids and especially chlorophylls. We selected 21 °C and 13 MPa as the cell disruption condition, which produced 1.91 g/kg d.s. of carotenoids and 14.03 mg/kg d.s. of chlorophylls. Pretreated microalga gave a 10-fold higher chlorophyll extraction yield compared to the untreated sample. While for carotenoids and tocopherols were 1.25 and 1.14-fold higher, respectively. Additionally, antioxidant activity of pretreated microalga (33.22 mmol TE/kg oil) was significantly higher than the value for the untreated samples (29.11 mmol TE/kg oil) (p ≤ 0.05). Confocal microscopy images showed morphological differences between micro-colonies with and without disruption treatment, suggesting that partial cell disruption by rapid depressurization improved the extraction of microalga compounds.

The adsorption study on removal of copper metal from aqueous solution using untreated marine green algae namely Chaetomorpha Antennina sp. was carried out under various experimental conditions. The effects of contact time, pH of the solution, average biosorbent particle size, biosorbent dosage and initial concentration of metal ion have been investigated by following the batch adsorption technique. 30 minutes of biosorption time was found to be sufficient to reach equilibrium rate of sorption for copper ion. The rate of copper removal was directly correlated to biomass amount and contact time. Biosorption of metal ion was pH dependent and the results indicated the optimum pH for the removal of copper as 6.0. The highest biosorption capacity was found to be 25.78 mg of metal ion per gram of biosorbent at initial concentration of 20 mg/L copper ion and increased for higher initial concentrations. The biosorption capacity increased with increase in the amount of biosorbent used for removal. Adsorption data was modled with Freundlich, Langmuir, Redlich- Peterson and Dubinin-Radushkevich adsorption isotherms. Isotherm studies showed that the data best fitted with the Langmuir isotherm model. The kinetic data corresponded well with the pseudo-first order equation, suggesting that the biosorption process is presumably chemisorption

Large-scale lab culture of Ankistrodesmus gracilis and Diaphanososma birgei were evaluated by studying the biology and biochemical composition of the species and production costs. Ankistrodesmus gracilis presented exponential growth until the 6th day, with approximately 144 x 10(4) cells.mL-1, followed by a sharp decrease to 90 x 10(4) cells.mL-1 (8th day). Algae cells tended to increase again from the 11th day and reached a maximum of 135 x 10(4) cells.mL-1 on the 17th day. D. birgei culture showed exponential growth until the 9th day with 140 x 10² individuals.L-1, and increased again as from the 12th day. Algae A. gracilis and zooplankton D. birgei contain 47 to 70 percent dry weight protein and over 5 percent dry weight carbohydrates. The most expensive items in the context of variable costs were labor and electricity. Data suggested that temperature, nutrients, light availability and culture management were determining factors on productivity. Results indicate that NPK (20-5-20) may be used directly as a good alternative for mass cultivation when low costs are taken into account, promoting adequate growth and nutritional value for cultured A. gracilis and D. birgei.


O objetivo deste trabalho foi avaliar o cultivo em larga escala de Ankistrodesmus gracilis e Diaphanososma birgei em laboratório através do estudo da biologia das espécies, composição bioquímica e custo operacional de produção. A. gracilis apresentou um crescimento exponencial até o sexto dia, ao redor de 144 x 10(4) células mL-1. Logo em seguida, sofreu um brusco decréscimo apresentando 90 x 10(4) células mL-1 (oitavo dia). A partir do décimo primeiro dia, as células algais tenderam a crescer novamente, apresentando um máximo de 135 x 10(4) células mL-1 no 17º dia. No cultivo de D. birgei, foi observado o primeiro pico de crescimento no nono dia com 140 x 10² indivíduos L-1, aumentando novamente a partir do décimo segundo dia. A alga clorofícea A. gracilis e o zooplâncton D. birgei possuem aproximadamente 50 e 70 por cento de proteína (PS), respectivamente, com teor de carboidrato acima de 5 por cento. A eletricidade e mão de obra foram os itens mais dispendiosos e, de acordo com os dados obtidos, a temperatura, nutrientes, disponibilidade de luz e manejo do cultivo, foram fatores determinantes sobre a produtividade. Os resultados indicam que o meio NPK (20-5-20) pode ser utilizado diretamente como uma alternativa de cultivo em larga escala, considerando o baixo custo de produção, promovendo adequado crescimento e valor nutricional para A. gracilis e D. birgei.

Food availability may affect metal toxicity for aquatic organisms. In the present study, the influence of high, medium and low densities of the algae Pseudokirchneriella subcapitata (10(6), 10(5) and 10(4) cells.mL-1, respectively) on the chronic toxicity of chromium to the cladoceran Ceriodaphnia dubia was investigated. C. dubia was exposed to a range of chromium concentration from 2.71 to 34.04 µg.L-1 and fed with algae at various densities. In another experiment, the green alga was exposed to chromium concentrations (94 to 774 µg.L-1) and supplied as food in different densities to zooplankton. The survival and reproduction of the cladoceran were measured in these toxicity tests. The IC50 for Cr to P. subcapitata and metal accumulated by algal cells were determined. The results of a bifactorial analysis (metal versus algal densities) showed that metal toxicity to zooplankton was dependent on algal densities. Significant toxic effects on the reproduction and survival of C. dubia were observed at 8.73, 18.22 and 34.04 µg.L-1 Cr when the test organisms were fed with 10(6) cells.mL-1 of P. subcapitata. Although the chlorophyta retain low chromium content, a decrease in the reproduction and survival of C. dubia occurred when they were fed with high algal density contaminated with 774 µg.L-1 Cr. It was concluded that high algal density have an appreciable influence on chromium toxicity to daphnids.


A disponibilidade de alimento pode afetar a toxicidade dos metais para os organismos aquáticos. No presente estudo, a influência de alta, média e baixa densidades da alga Pseudokirchneriella subcapitata (10(6), 10(5) e 10(4) céls.mL-1, respectivamente) sobre a toxicidade crônica do cromo para o cladócero Ceriodaphnia dubia foi investigada. C. dubia foi exposta a uma faixa de concentração de cromo de 2,71 a 34,04 µg.L-1 e alimentada com diferentes densidades algais. Em outro experimento, a alga foi exposta a concentrações de cromo (94 a 774 µg.L-1) e oferecida como alimento ao cladócero em diferentes densidades. A sobrevivência e a reprodução do cladócero foram medidas nesses testes de toxicidade. O valor da CI50 do cromo para P. subcapitata e a concentração de metal retido pelas células algais foram determinados. Os resultados da análise bifatorial (metal versus densidades algais) mostraram que a toxicidade do metal ao zooplâncton foi dependente da densidade algal. Efeitos tóxicos significativos na sobrevivência e reprodução de C. dubia foram observados a 8,73, 18,22 e 34,04 µg.L-1 Cr quando o organismo-teste foi alimentado com 10(6) céls.mL-1 de P. subcapitata. Apesar de a alga reter pouco cromo, decréscimo na sobrevivência e na reprodução de C. dubia ocorreu quando alimentada com alta densidade algal contaminada com 774 µg.L-1 Cr. Concluiu-se que alta densidade algal tem uma considerável influência na toxicidade do cromo aos dafinídeos.

Nonspecific acid phosphatases are a group of enzymes whose activity increases the availability of exogenous and endogenous orthophosphate either through extra- or intracellular hydrolysis of phosphate compounds. Our study demonstrates the activity of acid phosphatases in the filamentous freshwater alga Stigeoclonium tenue. These enzymes were detected following a cerium-based method in which cerium was used as an orthophosphate-capture reagent. In thalli from S. tenue from the natural environment, acid phosphatases were found in the longitudinal cell wall,plasmalemma, and vacuole. In thalli from Bold's Basal Medium culture, these enzymes were found mainly in the plasmalemma; they were scarce in the cell wall. In the thalli grown in phosphate-enriched culture medium, enzymes were found only in the plasmalemma. The low availability of orthophosphate in the medium seems to induce the transport of these enzymes to the cell wall. Its abundance, on the contrary, seems to attenuate this response without affecting the localization of acid phosphatases in the plasmalemma.

The chemical composition of green seaweed, Monostroma undulatum, Wittrock, growing in the Southern Argentina coast, was studied. Samples were collected in Puerto Deseado, province of Santa Cruz (47 degrees 45'L.S., 65 degrees 55'L.W.), from October to December 1999 and 2000. It has been analyzed six sample during this period. Algae were washed with sea water and dried at room temperature for 24 hs. Moisture, nitrogen, lipids and ashes were determined according to AOAC; fiber (total, soluble and insoluble), according to Lahaye. After mineralization with nitric acid, sodium and potassium were determined by flame photometry, calcium by complexometric method, and phosphorus by Gomori's method. The ranges expressed per 100 g dry algae were: protein (Nx6.25): 12.89-21.85; ashes (g): 33.92-40.05; lipid (g): 0.32-1.47; total fiber (g): 14.36-19.6; digestible carbohydrates (calculated by difference) (g): 20.86-32.48; sodium (g): 7.39-13.11; potassium (g): 1.38-3.18; calcium (mg): 149-226; phosphorus (mg): 190-447; Vitamin C (mg): 159-455. These results show that this green seaweed is an important source for protein, fiber, macronutrients minerals and vitamin C, during the macroscopic period. There was an important fluctuation that must be taken into account to consider the commercial collection to use it in human nutrition.

RESUMEN

Cold water extracts of marine green algae Codium dwarkense and C. tomentosum were precipitated with different molar concentrations of KCl and were subjected to anion exchange and gel filtration chromatography. Both the species yielded sulphated arabinan through bioassay-guided purification and both were chemically identified as a polymer of alpha-L-arabinofuranose. Products were assayed for their blood anticoagulant activity using PT, APFT and TT tests and found that they differed in the potency of activity though they are chemically identical. Bioassay-guided purification of cold water extract of C. tomentosum yielded sulphated arabinan and sulphated arabinogalactan.
