RESUMEN

Chromium (VI) a highly toxic metal, a major constituent of industrial waste. It is continuously release in soil and water, causes environmental and health related issues, which is increasing public concern in developing countries like Pakistan. The basic aim of this study was isolation and screening of chromium resistant bacteria from industrial waste collected from Korangi and Lyari, Karachi (24˚52ʹ46.0ʺN 66˚59ʹ25.7ʺE and 24˚48ʹ37.5ʺN 67˚06ʹ52.6ʺE). Among total of 53 isolated strains, seven bacterial strains were selected through selective enrichment and identified on the basis of morphological and biochemical characteristics. These strains were designated as S11, S13, S17, S18, S30, S35 and S48, resistance was determined against varying concentrations of chromium (100-1500 mg/l). Two bacterial strains S35 and S48 showed maximum resistance to chromium (1600 mg/l). Bacterial strains S35 and S48 were identified through 16S rRNA sequence and showed 99% similarity to Bacillus paranthracis and Bacillus paramycoides. Furthermore, growth condition including temperature and pH were optimized for both bacterial strains, showed maximum growth at temperature 30ºC and at optimum pH 7.5 and 6.5 respectively. It is concluded that indigenous bacterial strains isolated from metal contaminated industrial effluent use their innate ability to transform toxic heavy metals to less or nontoxic form and can offer an effective tool for monitoring heavy metal contamination in the environment.

Snow trout (Schizothorax plagiostomus) is an economically important freshwater fish, mostly found in northern areas of water reservoirs of Pakistan. The current study was conducted in River Swat to analyze the bioaccumulation of heavy metals (Pb, Cr, Ni, and Zn) in tissues of Schizothorax plagiostomus. Tissues were extracted and dissolved in perchloric acid (HClO4) and nitric acid (HNO3) along with hotplate. The heavy metals, zinc (Zn), lead (Pb), chromium (Cr), and Nickel (Ni) were determined using Perkin Elmer 2380 atomic absorption spectrophotometer. Results shows great variation in the content of the metal related to tissue type and sampling sites. A high concentration of bioaccumulation was reported at Charbagh, whereas lowest at Odigram: Charbagh>Landakai>Odigram. In the same way, Cr was the most accumulated heavy metal followed by lead, nickel, and Zinc.

Wet detoxification has traditionally been seen as the most promising technology for treating chromium-contaminated sites. However, the addition of chemicals in the wet detoxification process not only increases the cost but also introduces extra pollutants. Moreover, the chromium-containing slag may be re-dissolved in the form of Cr(VI), and the increased concentration of Cr(VI) results in a serious "returning to yellow" phenomenon in the chromium-contaminated sites, causing undesirable secondary pollution. Microbial remediation is a promising technology to address the re-dissolution of chromium-containing slag after wet detoxification, and this article reviews the advances in this area. Firstly, the toxicity, current situation and conventional technologies for treating the chromium-containing slag were briefly summarized. The mechanisms of the inevitable re-dissolution of chromium-containing slag after wet detoxification were summarized. Three main mechanisms, namely bioreduction, biosorption and biomineralization, which are involved in the environmental-friendly and efficient microbial remediation technology, were reviewed. The variation of microbial species and the succession of microbial community during the bioremediation of chromium-contaminated sites were discussed. Finally, future research directions were prospected with the aim to develop long-term, stable and sustainable technologies for remediating the chromium-contaminated sites.

RESUMEN

Abstract The present study aimed to evaluate the potential for using toxicity assays with sediment samples for the detection of water pollution caused by the discharge of tannery effluents into water bodies and its application to environmental forensic investigation. The study included ecotoxicological evaluation of sediments, survey of benthic organisms in the field, as well as chromium, cadmium and lead dosage which provided data for a sediment quality triad evaluation. The sediment samples showed acute and chronic toxicity to the bioindicators, low biodiversity of benthic macrofauna and high chromium concentration, reaching up to 4365 mg.Kg–1. A close relationship was observed between the separate results of ecotoxicological sediment evaluation and the sediment quality triad. The sediment ecotoxicological assessment proved to be applicable to tracking sources of contamination related to tanneries and similar activities in environmental forensics.

Heavy metals participate in chemical reactions of fish metabolism, which makes these elements a major focus on contamination studies. The objective of this study was to determine the concentrations of heavy metals Cd, Cr, Cu, Pb, Zn and Hg in muscle tissue of fish of Cassiporé River Basin (State of Amapá) and environment, aiming at assessing the risk of contamination. The concentrations of Cd in Plagioscion squamosissimus (1.061 µg g-1) and Pb in Poptella compressa (4.280 µg g-1) had values that exceed the boundaries established by Brazilian law. The Cr concentration in P. compressa (0.937 ± 0.5 µg g-1), Pimelodella cristata (0.463 ± 0.03 µg g-1) and Cyphocharax gouldingi (0.364 ± 0.12 µg g-1) and Hg in P. squamosissimus (0.670 µg g-1), Pseudoplatystoma fasciatum (0.630 µg g-1), Hoplias malabaricus (0.570 µg g-1 e Serrasalmus rhombeus (0.548 µg g-1) were above the legal limit. The water column showed high concentrations of heavy metals in all analyzed points, showing a contamination of this river basin. Therefore, the high concentrations of heavy metals in the environment and muscle tissue of fish indicate a high degree of contamination in the Cassiporé river basin and risk to human health.

Chromium pollution is produced in connection with industrial processes like in tanneries. It has been suggested that bioremediation could be a good option for clean up. The stress effect of variable chromate levels, pHs and growth temperatures on biochemical parameters of two Cr(VI) reducing bacterial strains Pseudomonas aeruginosa Rb-1 and Ochrobactrum intermedium Rb-2 was investigated. Transmission electrone microscopy (TEM) was performed to study the intracellular distribution of Cr(VI). It was observed that initial stress of 1000 µgmL-1 caused significant enhancement of all studied biochemical parameters at pH 7.0 and growth temperature of 37 °C showing great bioremediation potential of the strains. Transmission electron microscopy revealed that the distribution of chromium precipitates was not uniform as they were distributed in the cytoplasm as well as found associated with the periplasm and outer membrane. Fourier transform infrared spectroscopy showed the possible involvement of carboxyl, amino, sulpohonate and hydroxyl groups present on the bacterial cell surface for the binding of Cr(VI) ions. Cr(VI) stress brought about changes in the distridution of these functional groups. It can be concluded that the investigated bacterial strains adjust well to Cr(VI) stress in terms of biochemical parameters and along that exhibited alteration in morphology.

RESUMEN

An understanding of the characteristics of occupational lung cancer is important to establish policies that prevent carcinogen exposure and to compensate workers exposed to lung carcinogens. This study analyzed the characteristics of occupational lung cancers in workers who were compensated under the Industrial Accident Compensation Insurance Law between 1994 and 2011. A total of 179 occupational lung cancers were compensated. The main carcinogenic exposure was asbestos, followed by crystalline silica and hexavalent chromium. The mean exposure duration and latency were 19.8 and 23.2 yr. The most common industry was manufacturing, followed by construction and transportation. The most common occupation was maintenance and repair, followed by foundry work, welding, painting, and spinning or weaving. Although asbestos was predominant carcinogen, the proportion of these cases was relatively low compared to other developed countries. Proper surveillance system is needed to monitor occupational lung cancer and improve prevention measures.

RESUMEN

BACKGROUND: Patch testing remains the gold standard method for the identification of the etiologic agent of allergic contact dermatitis. However, it is a subjective, time-consuming exam whose technique demands special care and which presents some contraindications, which hamper its use. In a recent study, we showed that the proliferation assay can suitably replace patch testing for the diagnosis of chromium allergy, which had been previously demonstrated only for nickel allergy. In this study, we try to refine the method by reducing the incubation period of cultures for lymphocyte proliferation assays in response to chromium. OBJECTIVE: Develop an alternative or complementary diagnostic test for chromium allergic contact dermatitis. METHODS: We compared the production of 9 cytokines (IFN-γ, IL-2, IL-4, IL-5, IL-10, IL-12, IL-13, IL-17 and RANTES) between 18 chromium-allergic patients and 19 controls. RESULTS: Chromium increased the production of IFN-y, IL-5, IL-2 and IL-13 in allergic patients, but only IL-2 and especially IL-13 helped discriminate allergic patients from controls. The sensitivity, specificity and accuracy found with IL-13 were about 80%. CONCLUSIONS: IL-13 and IL-2 detection may be used to diagnose chromium allergy in 2-day cultures. However, in general, the 6-day cultures seem to be superior for this purpose. .

RESUMEN

This study explored the reduction of adenosine triphosphate (ATP) levels in L-02 hepatocytes by hexavalent chromium (Cr(VI)) using chi-square analysis. Cells were treated with 2, 4, 8, 16, or 32 μM Cr(VI) for 12, 24, or 36 h. Methyl thiazolyl tetrazolium (MTT) experiments and measurements of intracellular ATP levels were performed by spectrophotometry or bioluminescence assays following Cr(VI) treatment. The chi-square test was used to determine the difference between cell survival rate and ATP levels. For the chi-square analysis, the results of the MTT or ATP experiments were transformed into a relative ratio with respect to the control (%). The relative ATP levels increased at 12 h, decreased at 24 h, and increased slightly again at 36 h following 4, 8, 16, 32 μM Cr(VI) treatment, corresponding to a "V-shaped" curve. Furthermore, the results of the chi-square analysis demonstrated a significant difference of the ATP level in the 32-μM Cr(VI) group (P < 0.05). The results suggest that the chi-square test can be applied to analyze the interference effects of Cr(VI) on ATP levels in L-02 hepatocytes. The decreased ATP levels at 24 h indicated disruption of mitochondrial energy metabolism and the slight increase of ATP levels at 36 h indicated partial recovery of mitochondrial function or activated glycolysis in L-02 hepatocytes.

Malignant mesothelioma and lung cancer are representative examples of occupational cancer. Lung cancer is the leading cause of cancer death, and the incidence of malignant mesothelioma is expected to increase sharply in the near future. Although information about lung carcinogen exposure is limited, it is estimated that the number of workers exposed to carcinogens has declined. The first official case of occupational cancer was malignant mesothelioma caused by asbestos exposure in the asbestos textile industry in 1992. Since then, compensation for occupational respiratory cancer has increased. The majority of compensated lung cancer was due to underlying pneumoconiosis. Other main causative agents of occupational lung cancer included asbestos, hexavalent chromium, and crystalline silica. Related jobs included welders, foundry workers, platers, plumbers, and vehicle maintenance workers. Compensated malignant mesotheliomas were associated with asbestos exposure. Epidemiologic studies conducted in Korea have indicated an elevated risk of lung cancer in pneumoconiosis patients, foundry workers, and asbestos textile workers. Occupational respiratory cancer has increased during the last 10 to 20 yr though carcinogen-exposed population has declined in the same period. More efforts to advance the systems for the investigation, prevention and management of occupational respiratory cancer are needed.

RESUMEN

The Mandinga Lagoon in the Mexican State of Veracruz is an important ecological zone that produces 32% of the oyster output in the state of Veracruz, the main oyster producer in Mexico. Samples of water, sediment, and oysters were collected in 2003 and 2004 to study heavy metal pollution. Metal concentrations were determined in water, soil, and oyster tissues from fresh and detoxified Crassostrea virginica, and histology samples were analyzed. Metal (Cr, Cd, and Pb) concentrations in water were within the Mexican legal limits. The recorded values in sediments corresponded to those not producing biological effects (ERL). In the tissues, the highest concentrations corresponded to Pb, above 5.84 μgg-1 dry weight (d.w.); Cd was of 2.23 μgg-1 d.w., and Cr above 6 μgg-1 d.w. The metal levels detected in oysters exceeded the maximum permissible limits (MPL) for Cd and Pb, and oysters were unable to eliminate the concentrations of the bioaccumulated metals during the detoxification stage. The histopathological analysis revealed lesions in the digestive gland, edema, atrophy of epithelia in the digestive tubules, the presence of brown vesicles, hemocytic reaction, and necrosis. During detoxification, a higher number of epithelia were observed in the tubules, as well as an increase in brown vesicles and hemocytic reaction. Forty seven percent of oysters presented histopathological lesions related to metal concentrations. It is important to monitor metal concentrations, to detect the source of pollution, andto evaluate the effects on organisms to establish culture areas and adequate criteria for the exploitation of such an important fishery resource. RESUMEN

Hexavalent chromium (Cr(VI)), a commonly used industrial metal, is a well known human lung carcinogen. Epidemiology and animal studies suggest that the particulate Cr(VI) compounds, specifically the water insoluble compounds, are the more potent carcinogens; however, the carcinogenic mechanism remains unknown. Here we summarize recent Cr(VI)-induced human tumour, in vivo, cell culture and in vitro studies and put the data into context with three major paradigms of carcinogenesis: multistage carcinogenesis, genomic instability, and epigenetic modifications. Based on these studies, we propose a mechanism for chromate carcinogenesis that is primarily driven by the genomic instability paradigm.

RESUMEN

Food availability may affect metal toxicity for aquatic organisms. In the present study, the influence of high, medium and low densities of the algae Pseudokirchneriella subcapitata (10(6), 10(5) and 10(4) cells.mL-1, respectively) on the chronic toxicity of chromium to the cladoceran Ceriodaphnia dubia was investigated. C. dubia was exposed to a range of chromium concentration from 2.71 to 34.04 µg.L-1 and fed with algae at various densities. In another experiment, the green alga was exposed to chromium concentrations (94 to 774 µg.L-1) and supplied as food in different densities to zooplankton. The survival and reproduction of the cladoceran were measured in these toxicity tests. The IC50 for Cr to P. subcapitata and metal accumulated by algal cells were determined. The results of a bifactorial analysis (metal versus algal densities) showed that metal toxicity to zooplankton was dependent on algal densities. Significant toxic effects on the reproduction and survival of C. dubia were observed at 8.73, 18.22 and 34.04 µg.L-1 Cr when the test organisms were fed with 10(6) cells.mL-1 of P. subcapitata. Although the chlorophyta retain low chromium content, a decrease in the reproduction and survival of C. dubia occurred when they were fed with high algal density contaminated with 774 µg.L-1 Cr. It was concluded that high algal density have an appreciable influence on chromium toxicity to daphnids.

Chromium [Cr] is the most important element used to plate other metals and electroplating factories are still considered to be the most important industries which pollute the environment to this metal. This paper describes a study conducted to determine the efficiency of reverse-osmosis [RO] as a membrane technique for removal of Cr from wastewater samples synthetically prepared to be similar to electroplating effluents. The RO unit used in this study was a spiral wound module of 2521 TE made by a Korean CSM company. Synthetic wastewater samples containing Cr ions at various concentrations of 1 to 20 mg/L were prepared and subjected to treatment by RO, and quantitative analysis had been accomplished by a colorimetric method. Evaluation of optimized conditions of treatment had also been carried out by determining the effects of changing operating pressure, temperature and pH[s] of samples. Optimum conditions of Cr-treatment by RO in 10 g/L initial Cr concentration were determined to be in the pH range of 6 to 7 and in temperature of about 25°C at an applied pressure of 200 psi. Considering the efficiency of Cr removal which was as high as 99% at the optimized conditions it could be concluded that RO membrane process may be selected and developed as an effective alternative for treatment of metal-contaminated effluents of electroplating and similar industries

RESUMEN

Iron is essential to virtually all organisms but it can be toxic in excess. High concentration of iron and other trace elements could restricted bacterial growth and modify their metabolic pattern as well. However, this study aimed to find out the influence of iron, chromium, cadmium and synergism or antagonism between these elements on growth of a gram negative bacterium. In the series of experiments, E. coli has been cultured in a nutrient broth which supplemented with Fe[+2], Fe[+3], Cr[+3], Cd[+2] alone or in combination with together, at 37°C for 5 h. Bacterial growth was measured every half an hour using spectrophotometer. Findings obtained from this study indicated that bacterial growth reduced at presence of 1 mM/L concentration of Fe[+3] and 0.5 mM/L Fe[+2] in comparison with control. Growth of the bacteria was completely inhibited by 1 mM/L concentration of iron [II]. Chromium has also inhibitory effects on growth of the bacteria and cadmium is very toxic. Cr[+3] and Cd[+] have antagonism effect with iron on the growth of bacteria. This work suggested that trace elements could interact in their metabolism in bacteria. It has also concluded that toxic effects of trace element could be another view against pathogenic bacteria particularly in complex with antibacterial activity of various antibiotics

RESUMEN

High concentration of iron and other trace elements could restrict bacterial growth and modify their metabolic pattern as well. However, this study aimed to find out the influence of iron, chromium, cadmium and synergism or antagonism between these elements on the growth of a gram positive bacterium. In a series of experiments, Bucillus cereus was cultured in a nutrient broth which supplemented with Fe[+2], Fe[+3], Cr[+3], Cd[+2] separately, or in combination with each other, at 37°C for 5 hours. Bacterial growth was measured every half - hour, using spectrophotometer. The results indicated that bacterial growth rate reduced in the presence of 0.5 mM/L concentration of Fe[+2] or Fe[+3], in comparison with control and the growth of bacteria was inhibited by 1 mM/L concentration of iron. The results also revealed that Fe [III] as well as Fe [II] was toxic for bacteria. Chromium had partial inhibitory effects on the growth of bacteria and cadmium was very toxic. Cr[+3] and Cd[+] had antagonistic effect with iron on the growth of bacteria. Data obtained here provide a potentially interesting conceptual advance in toxic effects of trace elements on pathogenic bacteria

RESUMEN

Chromium compounds are potent toxic and carcinogenic substances. With respect to toxicity, hepatic and renal toxicity have been reported both in workers and in animals exposed to chromium (VI). Chromium (VI) compounds induces DNA damage in vivo and in cultured cells as well as the cytotoxicity evaluated by the leakage of lactate dehydrogenase. The present study reports the cytotoxicity of chrome platers who are employed from 8 to 25 years in electroplating industries at Coimbatore, Tamilnadu. Blood samples were collected and estimated for glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), creatine phosphokinase (CPK) and total protein in the serum. The study revealed that there is a significant elevation in the level of LDH, ALP, CPK and transaminases and a decrease in total protein in serum. The results of the study suggests that chromium (VI), a hepatotoxic chemical may perhaps damage the plasma membrane resulting in leakage of enzymes in to the serum of chromeplaters.
