RESUMEN

The androgenic regulation of the eleven enzymes of glycolytic pathway and two key enzymes of HMP pathway was studied in the initial segment, caput, corpus and cauda regions of the epididymis and in the vas deferens of rhesus monkey. The specific activities of enzymes were expressed as units of activity per mg DNA. Results in control animals indicate a marked difference in energy metabolism of epididymis and vas deferens. However, the epididymal duct itself did not show much regional variation in enzyme activities along its length. All the enzymes of the two pathways studied showed significant reduction after castration, the regulatory enzymes being affected more severely. On treatment with exogenous dihydrotestosterone, most of these enzymes showed stimulation in castrated monkeys, but none of them could be restored to normal level. The stimulation of these enzymes on treatment with exogenous dihydrotestosterone varied along the epididymal duct itself being maximum in the initial segment and minimum in the cauda region. The changes in the vas deferens were less marked as compared to the epididymis following castration and androgen replacement.

RESUMEN

Impact of altered serum prolactin status on enzymes involved in glycoprotein metabolism in epididymal tissue of matured monkeys was studied. Hyperprolactinemia (ovine prolactin-250 micrograms/kg body weight/day for 30 days) significantly inhibited the specific activities of dolichylphosphate mannosyl transferase, dolichylphosphate glucosyl transferase and galactosyl transferase, in the epididymal tissues. However, it had an enhanced effect on epididymal glycosidases such as beta-galactosidase, beta-N-acetyl glucosaminidase, beta-N-acetyl galactosaminidase, alpha-mannosidase and alpha-L-fucosidase. Hypoprolactinemia (bromocriptine mesylate-1-mg/kg body weight/day for 30 days) on other hand had no significant effect on the specific activities of both, glycosyltransferases and glycosidases, in the epididymal tissues. The results suggest that hyperprolactinemia inhibits epididymal glycoprotein metabolism by impairing the incorporation of oligosaccharide units into proteins with enhanced degradation. This may have adverse effect on events leading to sperm maturation in epididymal environment.

RESUMEN

Neste trabalho säo estudadas, histoquimicamente, as localizaçöes enzimáticas das enzímas NADH-diaforese e SHD no epitélio e estroma do edpidídimo do gato. Uma reaçäo forte e uniforme da NADH foi observada no epitélio do revestimento do corpo e da cauda do órgäo e uma reaçäo fraca, desta enzima, foi notada na cabeça epididimária. Quanto ao estroma ductular esta reatividade, à NADH, foi fraca ao nível da cabeça e média aos níveis do corpo e cauda epididimários. No concernente às reatividades da SDH, estas foram fracas no epitélio e muito fracas no estroma epididimário em todos os segmentos do ducto

Se investigó la respuesta aguda de las lipasas de lipoproteínas (LPL) de corazón y tejido adiposo epididimal de rata a una sobrecarga de triglicéridos (TG) administrada por vía endovenosa en forma de Intralipid 10%. En los animales ayunados previamente se observó una depresión significativa (p<0,01) en la actividad de la enzima de corazón al final de la primera hora que siguió a la administración de TG y una recuperación de dicha actividad al final del experimento (2h). En los animales alimentados hubo aumento mantenido de la actividad LPL de corazón todo el tiempo, hasta hacerse significativo al final de la segunda hora (p<0,02), acompañado de una disminución rápida de TG circulantes. La enzima de tejido adiposo epididimal de ratas ayunadas experimentó un aumento significativo en su actividad, detectado a los 60 min (p<0,05), que se mantuvo hasta el final del experimento (p<0,01). En las ratas alimentadas se produjo también un aumento (no significativo) en la actividad de LPL de tejido adiposo

Studies on the distribution of Na+, K+-dependent, Mg2+ dependent and Ca2+ dependent Adenosine Triphosphatase (ATP-ases) in the testes, epididymis, seminal vesicles and prostate glands of mature bonnet monkeys were carried out with and without Testosterone propionate (TP) treatment. Comparatively, the Ca2+ dependent ATP-ase was very active in the testes, caput and cauda epididymis and prostate of control animals. However, the Mg2+-dependent ATP-ase activity was predominant in the seminal vesicles. In all the genital tissues the Na+, K+ dependent ATP-ase exhibited low activity compared to other ATP-ase systems. On TP treatment at 1 mg/kg body wt. dose for 30 consecutive days to the second group of animals, all classes of ATP-ases drastically decreased in the testes, cauda epididymis, seminal vesicles and prostate. While in caput epididymis the Mg2+-dependent ATP-ase was stimulated, the Na+, K+-dependent ATP-ase was decreased both in the caput and corpus epididymis by the hormone treatment. The present study reveals the general inhibitory influence on the ATP-ase systems and thereby ionic transport after long term TP administration.
