RESUMEN

Abstract The aim of this study was to assess the effects of methanol extract of G. verum on redox status of isolated heart of spontaneously hypertensive rats after ischemia. Twenty-four Wistar albino rats were divided into three groups: untreated control rats and rats that received 125 and 250 mg/kg G. verum extract for 4 weeks per os. Index of lipid peroxidation (measured as TBARS) and parameters of antioxidative defence system such as level of reduced glutathione (GSH) and activities of catalase (CAT) and superoxide dismutase (SOD) were spectrophotometrically determined in heart homogenate. The index of lipid peroxidation in heart tissue was lower in both treated groups compared to the control group. On the other hand, the activity of SOD was significantly higher after consumption of both doses, while the activity of CAT was significantly higher only after treatment with a higher dose of extract. Based on our results we might conclude that 4-week treatment with methanol extracts of G. verum has the potential to modulate myocardial redox signaling after ischemia, thus significantly alleviating cardiac oxidative stress and exerting dose-dependent antioxidant properties. Future studies are certainly necessary to fully clarify the role of this plant species in myocardial I-R injury.

RESUMEN

<p><b>OBJECTIVE</b>To study the influence of C-terminal domain of ADAMTS13 on its cleaving activity.</p><p><b>METHODS</b>The full-length wild-type (WT) and C-terminal domain truncated type (TT, TSP8 + CUB domains were deleted) of human ADAMTS13 recombinant protein were transfected into and permanent expressed on Hela cells. Western blot and R-CBA were used to directly detect the activities of the two recombinant proteins under the static and stressed condition respectively. ELISA was used to compare the binding abilities of the two proteins by coating with vWF.</p><p><b>RESULTS</b>The recombinant proteins were identified by Western blot with anti-his-tag or anti-ADAMTS13 antibodies. With pretreatment of 1.5 M urea, the enzyme activity of TT was significantly higher than that of WT, and so did in binding ability with vWF While, only WT could cleave vWF under high stress.</p><p><b>CONCLUSION</b>The distal carboxyl-terminal TSP8 together with CUB domains of ADAMTS13 may affect the enzyme activity by regulating the binding of ADAMTS13 to vWF in different conditions, and they are very important for the enzyme activity under high stress force condition.</p>

RESUMEN

<p><b>OBJECTIVE</b>To study the compounds from the whole plant of Galium aparine var. tenerum.</p><p><b>METHOD</b>The various column chromatographic techniques were applied to isolate the chemical constituents. MS, NMR and 2D-NMR spectroscopic techniques were uesd to identify chemical structures.</p><p><b>RESULT</b>Six compounds were isolated from the EtOAc-soluble part of the 95% ethanol extract of the plant, and their chemical structures were identified as 1-(4-hydroxyphenyl)-ethanone (1), vanillic acid (2), 3,4-dihydroxybenzoic acid (3), p-hydroxycinnamic acid (4), gallic acid (5), 4-hydroxytruxillic acid (6).</p><p><b>CONCLUSION</b>The NMR data of compound 6 were completely assigned by 2D-NMR techniques, including 1H-1H COSY, HMQC and HMBC spectra. This was the first time to report isolation of the compound 6 from natural product. The compounds 1-5 was isolated from the genus Galium for the first time.</p>

RESUMEN

<p><b>OBJECTIVE</b>To investigate the chemical constituents of the extract of Galium verum.</p><p><b>METHOD</b>The compounds were isolated by chromatography and identified by spectral data.</p><p><b>RESULT</b>The eleven compounds obtained were identified as (+)-pinoresinol 4,4'-O-bis-beta-D-glucopyranoside (1), epipinoresinol (2), (+) -medioresinol (3), isorhamnetin (4), isorhamnetin 3-O-alpha-L-rhamnopyranosyl-(1-6)-beta-D-glucopyranoside (5), diosmetin (6), diosmetin 7-O-beta-D-glucopyranoside (7), quercetin-3-O-beta-D-glucopyranoside (8), ursolic acid (9), ursolic aldehyde (10) and rubifolic acid (11).</p><p><b>CONCLUSION</b>Compounds 1-5 and 9-11 were isolated from this genus for the first time.</p>
