Background: The bioethanol produced from biomass is a promising alternative fuel. The lignocellulose from marginal areas or wasteland could be a promising raw material for bioethanol production because it is present in large quantities, is cheap, renewable and has favorable environmental properties. Despite these advantages, lignocellulosic biomass is much more difficult to process than cereal grains, due to the need for intensive pretreatment and relatively large amounts of cellulases for efficient hydrolysis. Therefore, there is a need to develop an efficient and cost-effective method for the degradation and fermentation of lignocellulosic biomass to ethanol. Results: The usefulness of lignocellulosic biomass from wasteland for the production of bioethanol using pretreatment with the aid of ionic liquids of 1-ethyl-3-methylimidazolium acetate and 1-ethyl-3-methylimidazolium chloride was evaluated in this study. The pretreatment process, enzymatic hydrolysis and alcoholic fermentation lasted a total of 10 d. The largest amounts of bioethanol were obtained from biomass originating from agricultural wasteland, in which the dominant plant was fireweed (Chamaenerion angustifolium) and from the field where the common broom (Cytisus scoparius) was the dominant. Conclusions: The plants such as fireweed, common broom, hay and goldenrod may be useful for the production of liquid biofuels and it would be necessary in the further stage of research to establish and optimize the conditions for the technology of ethyl alcohol producing from these plant species. Enzymatic hydrolysis of biomass from agricultural wastelands results in a large increase in fermentable sugars, comparable to the enzymatic hydrolysis of rye, wheat, rice or maize straw.

This study evaluated the effect of different seeding rates and cutting heights on production and composition of millet. Two randomized block design experiments were performed. In the first trial, 32 parcels (1.2 × 1.2 m) were used for evaluation of seeding rate of 24, 35, 50 and 60 kg/ha. Three cuts were carried out to evaluate plant height, fresh and dry mass yield, dry matter (DM), neutral detergent fiber (NDF), crude protein (CP) and acid detergent lignin (ADL) concentration and in situ DM and NDF digestibility. In the second trial, 20 parcels (1.2 × 3.0 m) were used to evaluate 20, 30, 40 and 50 cm of cutting heights for two growing cycles, performing the evaluations previously listed. Seeding rate linearly increased plant height, fresh and dry mass production, and tended to linearly decrease dry matter in situ digestibility, without effects on forage composition. Cutting height had no effect on crude protein (CP) concentration, but linearly increased plant height, dry mass production, DM content and tended to decrease NDF in situ digestibility and ADL content. Thus, the use of the highest evaluated seeding rates (60 kg/ha) and cutting height (50 cm) increased forage production; however, the use of the lowest residual height increased forage quality.(AU)


Os efeitos de diferentes taxas de semeadura e altura de corte na produção e composição bromatológica do milheto foram avaliados em dois experimentos executados em delineamento em blocos casualizados. No primeiro experimento, foram utilizadas 32 parcelas (1,2 x 1,2 m) e avaliadas as taxas de semeadura de 24, 35, 50 e 60 kg/ha. Três cortes foram realizados para avaliar a altura das plantas, a produção de massa fresca e seca, os teores de matéria seca (MS), fibra em detergente neutro (FDN), proteína bruta (PB) e lignina, assim como a digestibilidade in situ da matéria seca e FDN. No segundo experimento, 20 parcelas (1,2 x 3,0 m) foram usadas para avaliar o efeito das alturas de corte de 20, 30, 40 e 50 cm, sobre as variáveis previamente citadas. De acordo com o aumento da taxa de semeadura, houve aumento linear da altura das plantas e da produção de massa seca e fresca e tendência de redução linear da digestibilidade in situ da matéria seca, sem afetar a composição da forragem. A altura de corte não afetou a concentração de proteína bruta, mas aumentou linearmente a altura das plantas, a produção de massa seca e o teor de matéria seca e tendeu a reduzir a digestibilidade in situ da FDN e a concentração de lignina. Assim, o emprego de maiores taxas de semeadura (60 kg/ha) e de altura de corte (50 cm) aumentam a produção de forragem, enquanto que menores alturas de corte aumentam a qualidade da forragem.(AU)

O objetivo foi avaliar as características agronômicas e químico-bromatológicas de dois híbridos de milho Bt (30F35H e CD397YH) ensilados com inoculante enzimobacteriano. Os teores de FDN foram semelhantes para todas as frações de planta, já o teor de FDA diferiu quanto à planta inteira e colmo, enquanto a lignina diferiu em relação à planta inteira, colmo e sabugo. A DIVMS não apresentou diferença entre os híbridos em nenhuma das frações. As silagens foram produzidas em silos experimentais (aproximadamente 200kg). As concentrações de MS, EE, FDN, NDT e DIVMS não mostraram diferença entre as silagens dos híbridos avaliados. Já os teores de MM, PB, FDA e lignina diferiram. Não houve diferença entre as silagens dos híbridos para os valores de NDT estimado e para a DIVMS. Também não foi observado efeito do inoculante sobre os valores de CHT, CNF, FDN e DIVMS. Conclui-se que o híbrido Pioneer foi superior ao Coodetec em produtividade de MV ha-1, porém a composição nutricional das silagens não diferiu na concentração de NDT e digestibilidade avaliadas em ovinos. Não houve efeito do uso de inoculante na digestibilidade da matéria seca e da fração fibra em detergente neutro das silagens.


The aim was to evaluate the agronomic characteristics and chemical composition of the two corn hybrids (30F35H and CD397YH) ensiled with enzymatic bacterial inoculants. NDF were similar for all plant fractions, since the ADF content differed as to the whole plant and stem, lignin differed in relation to the whole plant, stem and cob. IVDMD did not differ among treatments in any of the fractions. The silages were produced in experimental silos (approximately 200kg). The concentrations of MS, EE, NDF, IVDMD and TDN showed no difference between the silages of hybrids. Since the levels of MM, CP, ADF and lignin differed; there was no difference between hybrids for silage TDN and IVDMD. There was also no effect of the use of inoculants on the values of CHT, NFC, NDF and IVDMD. It is concluded that Pioneer was superior to Coodetec productivity of MV-1 ha. The nutritional composition of silages did not differ in the concentration of TDN and digestibility in sheep assessed. There was no effect of using inoculants on the digestibility of dry matter and neutral detergent fiber content of the silage.

Oligosaccharides and dietary fibres are non-digestible food ingredients that preferentially stimulate the growth of prebiotic Bifidobacterium and other lactic acid bacteria in the gastro-intestinal tract. Xylooligosaccharides (XOS) provide a plethora of health benefits and can be incorporated into several functional foods. In the recent times, there has been an over emphasis on the microbial conversion of agroresidues into various value added products. Xylan, the major hemicellulosic component of lignocellulosic materials (LCMs), represents an important structural component of plant biomass in agricultural residues and could be a potent bioresource for XOS. On an industrial scale, XOS can be produced by chemical, enzymatic or chemo-enzymatic hydrolysis of LCMs. Chemical methods generate XOS with a broad degree of polymerization (DP), while enzymatic processes will be beneficial for the manufacture of food grade and pharmaceutically important XOS. Xylooligomers exert several health benefits, and therefore, have been considered to provide relief from several ailments. This review provides a brief on production, purification and structural characterization of XOS and their health benefits.

Neste trabalho, 2 diferentes procedimentosforam utilizados para a extração de lignina a partir do licor negro, caracterizando-se quimicamente as amostras de lignina obtidas em relação a umalignina comercial, visando a aplicação no desenvolvimento de sensores voltamétricos. A análise elementar, espectroscopia na região do infravermelho, análise térmica e caracterização eletroquímica do material mostraram que, tanto a origem da lignina quanto a metodologia de obtenção da mesma, a partir do licor negro, podem fornecer materiais com propriedades químicas distintas, embora possuam comportamento eletroquímico similar. Observou-se, também, que a lignina só pode ser armazenada na forma sólida,devido à oxidação pelo oxigênio dissolvidodurante o tempo em que a solução mãe é armazenada.No entanto, a oxidação dalignina é necessária quando se tem por objetivo o desenvolvimento de sensores voltamétricos, devido a predominância de carbonos sp2na estrutura químicaoxidada, condição em que se obtém maior condutividade. Constatou-se também a necessidade de utilizar um transdutor metálico para o desenvolvimento de eletrodos qumicamente modificados com este material, visto que os eletrodos de carbono modificados com lignina oxidada ou não oxidada não apresentaram atividade eletroquímica. Devido à pequena porcentagem de enxofre existente na estrutura química, a lignina oxidada tende a se organizar pelos grupamentos SH quando na presença de ouro, expondo os grupamentos quinônicos eletroativos. A lignina oxidada ainda foi utilizada no preparo de eletrodo de pasta de carbono com nanopartículas de ouro, na qual a lignina oxidada impregnada no grafite atua como redutor "in-situ" do ouro, permitindo o preparo de um sensor voltamétrico versátil, capaz de realizar a determinação de ácido ascórbico, dopamina, nitrito e iodato. No que tange ao comportamento eletroquímico de fármacos e estudos de interação fármaco-DNA, eletrodos de carbono foram modificados com DNAdupla fita com a finalidade de monitorar a interação DNA-Gemcitabina.O fármaco não apresentou atividade eletroquímica tanto na região positiva quanto na região negativa de potencial. A interação do mesmo com o DNA promove a condensação/agregação das duplas fitas do DNA em uma primeiraetapa, seguida da clivagem do nucleosídeo guanosina, formando guanina livre. O comportamento eletroquímico de leflunomida e sulfasalazina, dois fármacos aplicados ao tratamento da artrite reumatóide, foi estudado e mecanismos de oxidação foram propostos para cada fármaco


The chemical properties of samples of lignin, which were precipitated from black liquor using two different methodologies (precipitation with CO2 and H2SO4), were studied and the results compared to those obtained from a commercial lignin sample in order to prepare voltammetric sensors. The elementary analysis, infrared spectroscopy, thermal analysis and electrochemical characterization of the material demonstrated that both, the source of lignin and the precipitation method from the black liquor, can provide lignin samples with different chemical properties, although the electrochemical behavior of all samples has been the same. Lignin could only be stored in solid form as lignin in the black liquor is slowly and quantitatively oxidized by dissolved oxygen, preventing the extraction procedures. However, the lignin as extracted from black liquor cannot be used to modify solid electrodes due its high resistivity. The previous oxidation of the all material was necessary when the aim was its application on the sensors development. The electrical conductivity in the oxidized lignin was achieved, probably due to the predominance of sp2 hybridized carbon atoms, which improved orbital overlapping in the material. In addition, it was necessary to use a metallic transducer to produce electrodes modified with films of lignin with good electrochemical activity. The films drying time was also important parameter, which suggested a specific organization of lignin macromolecules over the electrode surface. Due to the small percentage of sulfur in the material, the oxidized lignin tended to be organized by the SH groups in the presence of metallic substrates, exposing its electroactive quinone groups. The oxidized lignin was further used to prepare carbon paste electrodes modified with gold nanoparticles, in which the impregnated oxidized lignin on graphite acted as an "in situ" reducing agent towards HAuCl4.The resulting composite allowed the preparation of a versatile voltammetric sensor, capable of detecting ascorbic acid, dopamine, nitrite and iodate. Regarding the electrochemical behavior and drug interaction studies DNA-molecule, carbon electrodes were modified with double strand DNA with the purpose of monitoring Gemcitabine-DNA interaction. The drug showed no electrochemical activity both, in the positive and the negative potential. The Gemcitabine-DNA interaction promoted condensation / aggregation of the double strand DNA in a first step, followed by cleavage of the nucleoside guanosine in the form of free guanine. In addition, the electrochemical behavior of sulfasalazine and leflunomide, two pharmacological compounds applied to the treatment of rheumathoid arthritis, were studied and their oxidation mechanisms were proposed

The efficiency of enzymatic hydrolysis of cellulose can be improved by various pretreatments of the substrate. In order to increase the efficiency of enzymatic saccharification of the wheat straw, we determined the effect of different pretreatments on the physical structure, chemical components and enzymatic saccharification of wheat straw. Our results showed that combination of grinding and sodium hydroxide (NaOH) treatment had high effect on the enzymatic hydrolysis of wheat straws. The optimal pretreatment condition was to grind the wheat straws into the sizes of 120 meshes followed by treatment with 1.0% NaOH for 1.5 h (121°C/15psi). Under this condition, the cellulose content of wheat straw was increased by 44.52%, while the content of hemicellulose and lignin was decreased by 44.15% and 42.52%, respectively. Scanning Electronic Microscopy and infrared spectrum analyses showed that significant changes occurred in the structure of wheat straws after pretreatment, which is favorable for the hydrolysis and saccharification. Cellulase produced by Penicillium waksmanii F10-2 was used to hydrolyze the pretreated wheat straw and the optimal condition was determined to be 30 h of enzymatic reaction under the temperature of 55°C, pH 5.5 and substrate concentration of 3%.

Fourteen strains of Grifola frondosa (Dicks.) S. F. Gray, originating from different regions (Asia, Europe and North America) were tested for lignin degradation, ligninolytic enzyme activities, protein accumulation and exopolysaccharide production during 55 days of cultivation on oak sawdust. Lignin degradation varied from 2.6 to7.1 percent of dry weight of the oak sawdust substrate among tested strains. The loss of dry matter in all screened fungi varied between 11.7 and 33.0 percent, and the amount of crude protein in the dry substrate varied between 0.94 to 2.55 percent. The strain, MBFBL 596, had the highest laccase activity (703.3 U/l), and the maximum peroxidase activity of 22.6 U/l was shown by the strain MBFBL 684. Several tested strains (MBFBL 21, 638 and 662) appeared to be good producers of exopolysaccharides (3.5, 3.5 and 3.2 mg/ml respectively).

The production of manganese peroxidase (MnP) from Bacillus pumilus and Paenibacillus sp. was studied under absence and presence of the inducers indulin AT, guayacol, veratryl alcohol, lignosulfonic acid and lignosulfonic acid desulfonated. Indulin AT increased the activity of B. pumilus MnP up to 31.66 U/L after 8 h, but no improve was observed for Paenibacillus sp., which reached maximum activity (12.22 U/L) after 20 h. Both MnPs produced by these microorganisms were purified in phenyl sepharose resin and the proteins from crude extracts were eluted in two fractions. However, only the first fraction of each extract exhibited MnP activities. Tests in different pH and temperature values, from pH 5.0 to pH 10.0 and 30 ºC to 60 ºC, respectively, were carried out with the purified MnP. The maximum activity reached for B. pumilus and Paenibacillus sp. MnPs were 4.3 U/L at pH 8.0 and 25 ºC and 11.74 U/L at pH 9.0 and 35 ºC, respectively. The molar masses determined by SDS-PAGE gel eletrophoresis were 25 kDa and 40 kDa, respectively, for the purified enzyme from B. pumilus and Paenibacillus sp.

This study presents new and alternative fungal strains for the production of ligninolytic enzymes which have great potential to use in industrial and biotechnological processes. Thirty autochthonous fungal strains were harvested from Bornova-Izmir in Turkiye. In the fresh fruitbody extracts laccase, manganese peroxidase and lignin peroxidase activities, which are the principal enzymes responsible for ligninocellulose degradation by Basidiomycetes, were screened. Spores of some of the basidiomycetes species such as Cortinarius sp., Trametes versicolor, Pleurotus ostreatus, Abortiporus biennis, Lyophyllum subglobisporium, Ramaria stricta, Ganoderma carnosum, Lactarius delicious ve Lepista nuda were isolated and investigated optimum cultivation conditions in submerged fermentation for high yields of ligninolytic enzyme production. In addition, isolated fungal strains were monitored on agar plates whether having the capability of decolorization of a textile dye Remazol Marine Blue.


Este estudo apresenta novas cepas de fungos produtores de enzimas ligninolíticas com potencial de aplicação em processos industriais e biotecnológicos. Trinta cepas de fungos autóctones foram obtidos em Bornova-Izmir, Turquia. Os extratos frescos dos corpos de frutificação foram submetidos à triagem de atividade de lacase, manganês peroxidase e lignina peroxidase, que são as principais enzimas de degradação de ligninocelulose pelos Basidiomycetes. Foram isolados esporos de Cortinarius sp, Tramnetes versicolor, Pleorotus ostreatus, Abortiporus biennis, Lyophyllum subglobisporium, Ramaria stricta, Ganoderma carnosum, Lactarius delicius ve Lepista desnuda, investigando-se as condições ótimas de cultivo em fermentação submersa para produção de enzimas ligninolíticas com elevado rendimento. Além disso, as cepas fúngicas isoladas foram monitoradas em placas de ágar quanto a capacidade de descoloramento do corante têxtil Remazole Marine Blue.

The expression of the enzymatic system produced by basidiomycetous fungi, which is involved in the degradation of xenobiotics, mainly depends on culture conditions, especially of the culture medium composition. Trametes villosa is a strain with a proven biotechnological potential for the degradation of organochlorine compounds and for the decolorization of textile dyes. The influence of glucose concentration, addition of a vegetable oil-surfactant emulsion, nature of the surfactant and the presence of manganese and copper on the growth, pH and production of laccase, total peroxidase and manganese-dependent peroxidase activities were evaluated. In general, acidification of the medium was observed, with the pH reaching a value close to 3.5 within the first days of growth. Laccase was the main activity detected under the different conditions and was produced throughout the culture period of the fungus, irrespective of the growth phase. Supplementation of the medium with vegetable oil emulsified with a surfactant induced manganese-dependent peroxidase activity in T. villosa. Higher specific yields of laccase activity were obtained with the addition of copper.


A expressão do sistema enzimático produzido por fungos basidiomicetos envolvido na degradação de xenobióticos é bastante dependente das condições de cultivo, principalmente da composição do meio de cultivo. Trametes villosa CCB176 é uma linhagem com comprovado potencial biotecnológico para degradação de compostos organoclorados e descoloração de corantes têxteis. Foi avaliada a influência da concentração de glicose, adição de emulsão de óleo vegetal e surfactante, natureza do surfactante e os metais manganês e cobre no crescimento, pH e na produção das atividades de lacase, de peroxidases totais e de peroxidase dependente de manganês. No geral, ocorreu acidificação do meio com pH atingindo valor próximo a 3,5, nos primeiros dias de crescimento. Lacase foi a principal atividade detectada nas diferentes condições e sua produção se deu durante todo o período de cultivo do fungo, independente da fase de crescimento. Suplementação do meio com óleo vegetal emulsificado com surfactante resultou em indução da atividade de peroxidase dependente de manganês produzida por T. villosa. Maiores valores de rendimento específico da atividade de lacase foram propiciados pelo cobre.

RESUMEN

The present study was undertaken with the objective to study the characteristics of the effluent of Century Pulp and Paper Mill, Lalkuan (Uttarakhand) in different seasons. The variations in the physicochemical characteristics were observed and monitored up to 12 months at three different sites. Mean values of temperature, pH, chlorides and total phenols of the effluent were found below, whereas colour, BOD5, COD and lignin concentrations were above the minimum national standards (MINAS).

RESUMEN

In the present study sequential anaerobic and aerobic treatment in two step bioreactor was performed for removal of colour in the pulp and paper mill effluent. In anaerobic treatment, colour 50%, lignin 62%, COD 29%, absordable organic halides (AOX) 25% and phenol 29% were reduced in eight days. The anaerobically treated effluent was separately applied in bioreactor in presence of fungal strain, Paecilomyces sp., and bacterial strain, Microbrevis luteum. Data of study indicated reduction in colour 80%, AOX 74%, lignin 81%, COD 93% and phenol 76 per cent by Paecilomyces sp. where as Microbrevis luteum showed removal in colour 59%, lignin 71%, COD 86%, AOX 84% and phenol 88% by day third when 7 days anaerobically treated effluent was further treated by aerobic microorganisms. Change in pH of the effluent and increase in biomass of microorganism's substantiated results of the study, which was concomitant to the treatment method.

RESUMEN

The present study reports on the treatment of pulp and paper mill effluent by Phanerochaete chlysosporium and the same has been compared at two different pH, 5.5 and 8.5. At both the pH, colour, COD, lignin content and total phenols of the effluent significantly declined after bioremediation. However, greater decolourisation and reduction in COD, lignin content and total phenols were observed at pH 5.5. Such bioremediated effluent of pulp and paper mill could gainfully be utilized for crop irrigation.

Foram utilizados dois restos de cultura, resíduo da cultura de milho (RM) e resíduo da cultura de soja (RS) submetidos a extraçäo com solventes orgânicos para isolamento da lignina. O efeito da lignina sobre a digestibilidade da fibra foi verificado por meio do estudo de digestibilidade in situ. Encontraram-se valores de taxa de degradaçäo da matéria seca (MS) inferiores para RM em relaçäo a RS, respectivamente, 18,38 por cento e 41,94 por cento. As taxas de degradaçäo ruminal, obtidas no modelo linear até 24 horas de incubaçäo para hemicelulose (Hem) e celulose (Cel) em RM e RS, foram 12,99 por cento e 3,30 por cento para RM; 44,68 por cento e 36,33 por cento para RS

RESUMEN

O trabalho teve por objetico isolar e caracterizar microscopicamente as ligninas de resíduos agrícolas provenientes das culturas de soja (RS) e de milho (RM), bem como verificar o efeito das ligninas sobre a degradaçäo dos tecidos dos resíduos RM e RS. Por meio da microscopia eletrônica de varredura, estudou-se a ultra-estrutura das ligninas e dos tecidos vegetais de RM e RS. Verificou-se que as ligninas da palha moída do RM e da RS säo morfologicamente diferentes (ultra-estruturas diferentes) e que o desaparecimento dos tecidos digestíveis também é diferente, sendo mais prejudicado no RM

RESUMEN

Este estudo foi realizado com o objetivo de se identificar o tipo e a quantidade de fibras presentes no farelo de arroz. Amostras em triplicata de farelo de arroz fresco, fornecido pela máquina de beneficiamento ARAWAL de Botucatu, foram analisadas pelos Métodos de Fibra Detergente ácido (FDA) e Fibra Detergente Neutro (FDN). As amostras analisadas apresentaram níveis elevados de FDN (27,4 por cento), predominando os conteúdos de hemiceluloses (17,3 por cento) e lignina (20,9 por cento). Por outro lado, o conteúdo de celulose (2,64 por cento) foi menor do que relatado na literatura. Os dados do presente estudo demonstram que o farelo de arroz possui alta concentraçäo de fibras, predominando as do tipo insolúvel como hemiceluloses e lignina, as quais atuam como protetoras no desenvolvimento de câncer de cólon.

RESUMEN

We report the application of a method which combines digestion with pancreatin and neutral detergent treatment in the analytical study of dietary fiber from cassava. The use of pancreatin previous to the detergent extraction enabled rapid filtration, thus giving more reproducible results for neutral detergent fiber (NDF). Acid detergent fiber (ADF), hemicellulose, lignin and pectin were also determined. The values obtained for NDF (4.65 per cent ) and pectin (1.17 per cent ) are very important, considering their role in the digestive process
