Abstract This research aimed to conduct a systematic review and metanalysis to compare the frequency of cell damage in crack users and nonusers, through Micronucleous (MN) test in buccal mucosa cells. A comprehensive search was carried out on MEDLINE via PubMeb, Web of Science, LILACS and the grey literature without restrictions. It was included case-control studies that report the frequency of micronuclei in the oral mucosa of adult crack users and nonusers. A review protocol was registered with PROSPERO (CRD42018115672), and conducted in accordance with the PRISMA guidelines for the report of this systematic review. Furthermore, study quality was evaluated using an adapted Newcastle-Ottawa Scale for cross-sectional studies.The original search yielded 27 references, after eligibility criteriaonly five articles were included. The number of micronuclei was higher in crack users compared to nonusers. Also, secondary outcomes: binucleated cells, nuclear buds, pyknosis, karyorrhexis and karyolysis had higher prevalence in crack users.Crack use is associated with genotoxic and mutagenic effects because there is a higher frequency of micronuclei in exfoliated cells of crack users. In addition, MN test proved to be a goodbiomarker to assess the mutagenic impact of crack use in oral epithelium.


Resumen Esta investigación tuvo como objetivo realizar una revisión sistemática y un meta-análisis para comparar la frecuencia de daño celular en usuarios de crack y sin crack, a través de la prueba de micronúcleos (MN) en células de la mucosa bucal. Se realizó una búsqueda exhaustiva en MEDLINE a través de PubMeb, Web of Science, LILACS y la literatura gris sin restricciones. Se incluyeron estudios de casos y controles que informaron la frecuencia de micronúcleos en la mucosa oral de usuarios adultos de crack y sin crack. Se registró un protocolo de revisión con PROSPERO (CRD42018115672), y se realizó de acuerdo con las pautas de PRISMA para el informe de esta revisión sistemática. Además, la calidad del estudio se evaluó mediante una escala Newcastle-Ottawa adaptada para estudios transversales. La búsqueda original arrojó 27 referencias, después de los criterios de elegibilidad se incluyeron un total de cinco artículos. El número de micronúcleos fue mayor en los usuarios de crack en compa ración con los usuarios sin crack. Además, los resultados secundarios de células binucleadas, yemas nucleares, picnosis, cario- rrexis y cariólisis tuvieron una mayor prevalencia en los usuarios de crack. El uso de crack se asocia con efectos genotóxicos y mutagénicos porque hay una mayor frecuencia de micronúcleos en las células exfoliadas de los usuarios de crack. Además, la prueba de MN demostró ser un buen biomarcador para evaluar el impacto mutagénico del uso de crack en el epitelio oral.

Abstract Introduction: Car painters are routinely exposed to organic solvents classified as carcinogenic and mutagenic substances. Objective: To characterize the population susceptibility and evaluate the genotoxic effects of exposure to organic solvents. Methods: A cross-sectional study comparing a group of car painters exposed to organic solvents with a non-exposed group. CYP2E1 polymorphisms and the presence of micronuclei in lymphocytes were determined. Results: One hundred twenty-two workers participated in the study: 62 who worked in car paint shops and were exposed to solvents, and 60 who were not exposed. There were statistically significant differences between the two groups regarding micronucleated cells and nucleoplasmic bridges frequencies (p=0.042 and p=0.046, respectively; exact likelihood ratio). Significant differences were found at the interaction between the CYP2E1 genotype c1c1 and occupational exposure to solvents, with higher frequencies of micronuclei (p= 0.013) and micronucleated cells (p= 0.015). However, when the frequencies of micronuclei, micronucleated cells and nucleoplasmic bridges in the exposure group were compared between the c1c1 and c2c2/c1c2 allele groups of the CYP2E1 polymorphism, statistically significant differences were found. Conclusions: This study confirms that when workers with CYP2E1 polymorphisms, specifically the c1c1 genotype, are exposed to organic solvents, they are more likely to have somatic cell mutations, a condition associated with increased susceptibility to diseases such as cancer


Resumen Introducción: Los pintores de vehículos automotores están rutinariamente expuestos a agentes como los solventes orgánicos, capaces de producir efectos mutágenos y carcinógenos. Objetivo: Caracterizar la susceptibilidad poblacional y evaluar los efectos genotóxicos debidos a la exposición a solventes orgánicos. Métodos: Estudio de corte transversal que comparó a un grupo de pintores de carros expuestos a solven tes orgánicos con un grupo de personas no expuestas. Fueron determinados tanto los polimorfismos de CYP2E1 como la presencia de micronúcleos en linfocitos. Resultados: Participaron 122 personas, 62 trabajadores de talleres de pintura de autos expuestos a solventes y 60 personas no expuestas. Con relación al cuestionario Q 16, 32% de los expuestos refirieron síntomas sugestivos de neurotoxicidad. Las frecuencias de células micronucleadas y de puentes nucleoplásmicos fueron significativamente mayores en los expuestos que en los no expuestos: p= 0.042 y p= 0.046, respectivamente, Razón de verosimilitud exacta). Fueron halladas diferencias significativas en la interacción de CYP2E1 (c1c1) y la exposición ocupacional a solventes, con mayores frecuencias de micronúcleos (p= 0.013) y de células micronucleadas (p= 0.015). Conclusiones: Este estudio reafirma que los trabajadores expuestos a solventes orgánicos con polimorfismos de CYP2E1, específicamente con genotipo c1c1, tienen mayor probabilidad de presentar mutaciones en las células somáticas, condición asociada con una mayor susceptibilidad a enfermedades como el cáncer

ABSTRACT BACKGROUND: Gastric cancer is known as the fourth most common cancer. Current treatments for cancer have damaged the sensitive tissues of the healthy body, and in many cases, cancer will be recurrent. Therefore, need for treatments that are more effective is well felt. Researchers have recently shifted their attention towards antipsychotic dopamine antagonists to treat cancer. The anticancer activities of aripiprazole remain unknown. OBJECTIVE: This study aimed to evaluate the efficacy and safety of aripiprazole on gastric cancer and normal cell lines. METHODS: In this regard, the cytotoxicity and genotoxicity of aripiprazole were investigated in MKN45 and NIH3T3 cell lines by methyl tetrazolium assay and on peripheral blood lymphocytes by micronucleus assay. For this purpose, cells were cultured in 96 wells plate. Stock solutions of aripiprazole and cisplatin were prepared. After cell incubation with different concentrations of aripiprazole (1, 10, 25, 50, 100 and 200 μL), methyl tetrazolium solution was added. For micronucleus assay fresh blood was added to RPMI culture medium 1640 supplemented, and different concentrations of aripiprazole (50, 100 and 200 μL) were added. RESULTS: The finding of present study showed that the IC50 of aripiprazole in the cancer cell line (21.36 μg/mL) was lower than that in the normal cell line (54.17 μg/mL). Moreover, the micronucleus assay showed that the frequency of micronuclei of aripiprazole at concentrations below 200 μM was much less than cisplatin. CONCLUSION: Aripiprazole can be a good cytotoxic compound and good candidate for further studies of cancer therapy.


RESUMO CONTEXTO: O câncer gástrico é conhecido como o quarto câncer mais comum. Os tratamentos atuais para o câncer danificaram os tecidos sensíveis do corpo saudável e, em muitos casos, o cancro será recorrente. Portanto, a necessidade de tratamentos que são mais eficazes é desejada. Recentemente, os pesquisadores mudaram sua atenção para os antagonistas antipsicóticos da dopamina para tratar o câncer. As atividades anticâncer de aripiprazol permanecem desconhecidas. OBJETIVO: Este estudo objetivou avaliar a eficácia e a segurança do aripiprazol no câncer gástrico e nas linhagens celulares normais. MÉTODOS: A este respeito, a citotoxicidade e a genotoxicidade do aripiprazol foram investigadas em linhas celulares MKN45 e NIH3T3 por ensaio de metil tetrazólio e em linfócitos periféricos de sangue por ensaio de micronúcleos. Para este efeito, as células foram cultivadas em 96 placas. As soluções de estoque de aripiprazol e cisplatina foram preparadas. Após incubação celular com diferentes concentrações de aripiprazol (1, 10, 25, 50, 100 e 200 μL), a solução de metil tetrazólio foi adicionada. Para o ensaio do micronúcleo o sangue fresco foi adicionado ao meio de cultura RPMI 1640 suplementado, e as concentrações diferentes de aripiprazole (50, 100 e 200 μL) foram adicionadas. RESULTADOS: O presente estudo mostrou que o IC50 de aripiprazol na linhagem celular cancerosa (21,36 μg/mL) foi menor do que na linha celular normal (54,17 μg/ mL). Além disso, o ensaio de micronúcleos demonstrou que a frequência de micronúcleos de aripiprazol em concentrações inferiores a 200 μM foi muito inferior à cisplatina. CONCLUSÃO: O aripiprazol pode ser um bom composto citotóxico e bom candidato para estudos adicionais da terapia do câncer.

Abstract The effects of aging, gender and lifestyle factors on inducing chromosomal damage (micronuclei) and nuclear degenerative changes were assessed using the micronucleus test on exfoliated cells of the oral mucosa. The sample included 80 healthy subjects divided into four groups according to age and gender: men and women aged 19-29 years (M19, W19) and men and women aged over sixty years (M60, W60). An interview questionnaire was used to characterize the sample and to determine an index reflecting lifestyle (HLI). The frequency of micronuclei and nuclear degenerative changes was significantly higher among the elderly (p<0.001) and did not differ by gender among young people (p>0.05). The occurrence of micronuclei was similar among elderly men and women (p>0.10), but karyorrhexis and karyolysis were more frequent among men (p<0.005 and p<0.025, respectively), who also had a lower HLI than the other groups (p<0.0004). The results of the study indicate that age is the main factor associated with the induction of genetic material damage. AU


Resumo O objetivo deste estudo foi avaliar os efeitos do envelhecimento, do gênero e de fatores relacionados ao estilo de vida na indução de danos cromossômicos (micronúcleo) e de alterações nucleares degenerativas com o uso do teste de micronúcleo em células esfoliadas da mucosa oral. A amostra incluiu oitenta indivíduos saudáveis divididos em quatro grupos de acordo com a faixa etária e o gênero: homens e mulheres de 19-29 anos (H19, M19), homens e mulheres com mais de sessenta anos (H60, M60). Questionário de entrevista foi aplicado para caracterizar a amostra e para determinar um índice refletindo o estilo de vida (IVS). A frequência de micronúcleos e alterações nucleares degenerativas foi significativamente maior entre os idosos (p<0,001) e não diferiu em função do gênero entre os jovens (p>0,05). Ocorrência de micronúcleos foi similar entre homens e mulheres idosos (p>0,10), mas cariorréxis e cariólise foram mais frequentes entre os homens (p<0,005 e p<0,025, respectivamente), que também apresentaram o menor IVS em relação aos demais grupos (p<0,0004). El uso creciente de nanomateriales en productos industriales y de consumo ha incrementado la preocupación mundial respecto a sus posibles efectos adversos en los sistemas biológicos. Como consecuencia de la falta de un marco legislativo y la ausencia de un consenso sobre los protocolos experimentales, las investigaciones ecotoxicológicas se llevan a cabo a un ritmo mucho más lento que la producción de nuevas nanopartículas. Por esta razón, existe una necesidad creciente de realizar estudios que aporten conocimiento sobre el riesgo de estos contaminantes emergentes de propiedades únicas. El objetivo del presente trabajo fue evaluar la frecuencia de micronúcleos (FMN) en eritrocitos de ejemplares juveniles de pacú (Piaractus mesopotamicus) expuestos a nanopartículas de plata (Nano-Ag) a las concentraciones de 0 μg·L-1 (control); 2,5 μg·L-1; 10 μg·L-1; y 25 μg·L-1, durante 24 horas. Se observó que la FMN se incrementó significativamente (p<0,01) en la concentración de 25 μg·L-1, mientras que no hubo diferencias significativas entre los grupos expuestos a 2,5 y 10 μg·L-1 y el control. Estos resultados sugieren que los eventos aneugénicos o clastogénicos podrían representar un posible mecanismo de toxicidad de Nano-Ag en esta especie.


The growing use of nanomaterials in consumer and industrial products has aroused global concern about possible adverse effects on biological systems. Due to the lack of a regulation framework and the absence of a consensus on the experimental protocols, ecotoxicological investigations are carried out much slower than the production of new nanoparticles. For this reason, there is a growing need for studies that provide knowledge about the risk of these emerging contaminants of unique properties. The objective of the present study was to evaluate the frequency of micronuclei (FMN) in erythrocytes of juvenile Piaractus mesopotamicus exposed to silver nanoparticles (nano-Ag; Nanotek SA) at concentrations of 0 μg·L-1 (control); 2.5 μg·L-1; 10 μg·L-1; and 25 μg·L-1, for 24 hours (n = 10 per treatment). The FMN show a significant increase (p <0.01) in fish exposed to 25 μg·L-1 of Nano-Ag, while there were no significant differences among the groups exposed to 2.5 and 10 μg·L-1 with the control. These results suggest that the aneugenics or clastogenics events may represent a possible mechanism of toxicity of Nano-Ag in this specie.

OBJECTIVES: This study intends to provide a quick, easy, and inexpensive way to assess nuclear abnormalities such as micronuclei and bud frequencies; binucleated, karyorrhectic, karyolytic, pycnotic, and condensed chromatin cells in nasal scrapings of infants, which are particularly important for conducting genotoxic studies related to the inhaled atmosphere in pediatric populations. METHODS: Nasal swab samples were collected from 40 infants under 12 months of age using a small cytobrush. 2,000 cells from each infant sample were analyzed and classified according to the frequency of nuclear abnormalities. RESULTS: Rates of nuclear abnormalities found agree with values reported in other studies of neonates and children. This study found 0.13% of cells with micronuclei; 1.20% karyorrhexis; 0.03% pyknosis; 10.85% karyolysis; 1.11% condensed chromatin; 0.54 binucleated cells; and 0.02% nuclear bud. Differences were not observed between genders or environmental passive smoking, nor was any age correlation found. CONCLUSION: The assay proposed here is suitable for assessing the frequency of nuclear abnormalities from nasal cells in infants. .


OBJETIVOS: Este estudo pretendeu fornecer uma forma rápida, fácil e barata de avaliar anormalidades nucleares, como frequências de micronúcleos e gêmea, células binucleadas, cariorréticas, cariolíticas, picnóticas e com cromatina condensada, em esfregados nasais de neonatos, o que é particularmente importante para a realização de estudos genotóxicos relacionados ao ar inalado nas populações pediátricas. MÉTODOS: Foram coletadas amostras de esfregaço nasal de 40 neonatos com menos de 12 meses de idade, utilizando uma pequena escova citológica. Foram analisadas 2.000 células da amostra de cada neonato e classificadas de acordo com a frequência de anormalidades nucleares. RESULTADOS: As taxas de anormalidades nucleares encontradas neste estudo são compatíveis com os valores relatados em outros estudos de neonatos e crianças. Encontramos 0,13% de células com micronúcleos, 1,20% com cariorrexe, 0,03% com picnose, 10,85% com cariólise, 1,11% com cromatina condensada, 0,54 com células binucleadas e 0,02% com células nucleares gêmeas. Não observamos diferenças entre os gêneros, tabagismo passivo e nenhuma correlação entre idades. CONCLUSÃO: O ensaio proposto neste estudo é adequado para avaliar a frequência de anormalidades nucleares de células nasais em neonatos. .

RESUMEN

El cáncer gástrico (CG) es la cuarta causa de muerte por cáncer a nivel global. La dieta y el consumo de alcohol y tabaco, además de la infección por Helicobacter pylori determinan un gran número de casos de esta neoplasia. Algunos alimentos contienen sustancias que podrían influir en el proceso de carcinogénesis gástrica, aunque los mecanismos subyacentes no están completamente dilucidados. En México y el mundo, la disminución en el consumo de frutas, vegetales no feculentos y allium, leguminosas y alimentos fuente de selenio, así como el aumento en el consumo de sal, alimentos salados, salmuera y ahumados, chile, carnes procesadas y asadas o a la parrilla se han asociado respectivamente con un aumento de riesgo de CG. Con la evidencia disponible, se podrían desarrollar y evaluar programas para la prevención y control del CG.


Gastric cancer (GC) is the fourt leading cause of cancer death at global level. Diet, alcohol and tobacco, in addition to Helicobacter pylori infection, account for a large number of cases. Some substances contained in foods may influence GC carcinogenesis process; however, the underlying mechanisms have not been fully elucidated. In Mexico and worldwide, a low intake of fruits, non-starchy and allium vegetables, pulses, and foods containing selenium, as well as high intake of salt, salty, salted and smoked foods, chili pepper, processed and grilled/barbecued meats, have been respectively associated with an increased risk of GC. Based on the available evidence, programs for GC prevention and control could be developed and evaluated.

Los anfibios son constituyentes integrales de ecosistemas naturales y agrícolas, y debido a su sensibilidad pueden ser utilizados como bioindicadores. La frecuencia de micronúcleos se emplea como un biomarcador que evalúa la respuesta a agentes genotóxicos. El objetivo del trabajo fue determinar y comparar la frecuencia de micronúcleos en eritrocitos de Bufo arenarum que habitan ambientes antrópicos de la ciudad de Río Cuarto. Se escogieron tres sitios modificados: un lago urbano Lago Villa Dálcar (33º06´S-64º22´W) y dos sitios de cultivo denominados Cultivo1 (33º05'S-64º26'W) y Cultivo 2 (33º05'S-64º25'W). Como control se escogió un área ubicada en las sierras de Comechingones, en el Campo Experimental Las Guindas (32º35'S- 64º42'W), pertenecientes a la UNRC. En cada sitio se recolectaron ocho individuos utilizando trampas de caída y relevamientos por encuentros visuales. A cada individuo se le extrajo sangre de la vena angularis inmediatamente después de la captura. Se realizaron dos extendidos por cada individuo, se dejaron secar al aire por 24 hs, luego se fijaron con metanol por 20 minutos y se colorearon con Giemsa al 5% durante 10 minutos. Posteriormente los frotis fueron analizados con microscopio (1000X) y se tomaron fotografías con cámara digital. La frecuencia de micronúcleos fue contabilizada para 4000 células por individuo. Se realizaron comparaciones utilizando el test de Kruskal-Wallis. Las frecuencias de micronúcleos encontradas son: Las Guindas: 0,17 ± 0,23; Cultivo1: 0,78 ± 0,28; Cultivo 2: 0,61 ± 0,45 y Villa Dálcar: 0,76 ± 0,47. Existen diferencias significativas entre el control y los demás sitios (p< 0,01). Los resultados obtenidos demuestran la relevancia en la aplicación del test de micronúcleos in vivo en anfibios anuros para comparar distintos ambientes. Estos resultados indicarían la necesidad de nuevas investigaciones y de un monitoreo más prolongado en el tiempo para estos ambientes, incluyendo por ejemplo otros tipos de biomarcadores, así como otras posibles especies representativas de la región.


Amphibians are integral constituents of natural and agricultural ecosystems, and because of their sensitivity can be used as bioindicators. Micronuclei test is used as a biomarker for determining the response to genotoxic agents. The purpose of this study was to determine and compare the frequency of micronuclei in peripheral blood erythrocytes of Bufo arenarum toads that inhabiting anthropic environments for the city of Rio Cuarto. Organisms used in the MN test were collected from three sites modified: an urban lakeVilla Dalcar (33º06´S-64º22´W) and two cultivation sites called Crop 1 (33º05'S-64º26'W) and Crop 2 (33º05'S-64º25'W). As a control site, an area in the mountains of Comechingones, Las Guindas (32º35'S-64º42'W), belonging to the UNRC was sampled. At each site, eight individuals were collected using pitfall traps and by visual encounter surveys. To each individual is bled from the vein angularis immediately after capture. We performed two extended for each individual, air-dried for 24 hours, then fixed with methanol for 20 minutes and stained with 5% Giemsa for 10 minutes. Then the smears were analyzed with microscope (1000X) and took pictures with digital camera. For each individual the frequency of micronuclei for 4000 cells was recorded. Comparisons were made using the Kruskal-Wallis test. Micronuclei frequencies found are: Las Guindas: 0.17± 0.23; Crop 1: 0.78 ± 0.28, Crop 2: 0.61 ± 0.45 and Villa Dalcar: 0.76 ± 0.47. There are significant differences between the control and the other sites (p< 0.01). These findings show the relevance in the application of micronucleus test in vivo for anuran amphibians to compare different environments. These results suggest the need for further research and monitoring longer time for these environments, including such other biomarkers, as well as other possible species representative of the region.

RESUMEN

Los micronúcleos son fragmentos o cromosomas completos que quedan fuera del núcleo durante la mitosis; mediante su estudio se pueden evaluar los efectos de genotóxicos ambientales y ocupacionales. Esta prueba es ampliamente utilizada y es una alternativa eficaz, sencilla y económica para detectar la perdida de material genético. Por otra parte, la cavidad oral puede reflejar el estado de salud de los individuos, debido a que la mucosa que la recubre, puede presentar evidencias a nivel microscópico como macroscópico de cambios indicativos de enfermedad local sistémica o por exposición a sustancias tóxicas así como efectos secundarios por tratamientos. Dichas ventajas, favorecen su utilización en pruebas para evaluar genotóxicos o citotóxicos. La mucosa es una barrera protectora del resto del organismo, es un punto de contacto de agentes potencialmente peligrosos; por tanto, se torna susceptible de sufrir daños. El epitelio de revestimiento oral (60 por ciento) es estratificado no queratinizado formado por células con abundante citoplasma, permite la penetración de colorantes y facilita la observación e identificación de características morfológicas del núcleo y membrana celular. Además la mucosa tiene elevada capacidad proliferativa y aunque esta particularidad mantiene la población celular constante, por otro lado, se vuelve más vulnerable a daño al ADN. Esto cobra relevancia ya que el 90 por ciento del cáncer tienen origen epitelial, así que la mucosa oral es usada para monitorear eventos genotóxicos tempranos causados por cancerígenos inhalados o ingeridos. Este epitelio es de fácil acceso, poco invasivo, por lo que al tomar la muestra a los individuos, se les genera mínimo estrés. Por todo lo anterior, el epitelio oral es un tejido ideal para aplicar la técnica de micronúcleos y detección de anormalidades nucleares sin necesidad de cultivos celulares, lo que representa una oportunidad para realizar estudios epidemiológicos en poblaciones de alto riesgo.


Micronucleus are fragments or whole chromosomes that are outside the nucleus during mitosis. Through this study we can evaluate the environmental and occupational the genotoxic effects. This test is widely used because it is a very effective alternative, it is a simple, fast and inexpensive way to detect the loss of genetic material. Meanwhile a healthy oral cavity is evidenced because in the overlying mucosa changes indicative of local or systemic disease, toxic exposure and side effects of treatments can be observed. This favors their use in tests to assess the presence of genotoxins or cytotoxins. Although protective barrier from the rest of the body is the point of contact of potentially dangerous agents thus becoming susceptible to damage. Coating and oral epithelium (60 percent) are formed by stratified non-keratinized cells with abundant cytoplasm, allowing the absorption of dyes and facilitating microscopic observation and identification of nucleus and membrane morphological characteristics. It has a particularly proliferative capacity, and even though this particularity maintains constant cell population, on the other hand, becomes more vulnerable to DNA damage. This information is relevant as 90 percent of all cancers are of epithelial origin. Therefore, the oral mucosa is used to monitor early events caused by inhaled or ingested genotoxic carcinogens. Epithelium is easily accessible and minimally invasive, thereby generating less stress when samples are obtained from study participants. In view of the above, oral epithelium tissue is ideal for implementing micronucleus assay and for the detection of nuclear abnormalities without the need for cell cultures, which presents a unique opportunity for epidemiological studies in high-risk populations.

Different concentrations of H. rhodopensis total extract (HRE; 0.03, 0.06 and 0.12 g/kg body weight) were injected im, into rabbits 2 h before collecting the blood samples. The whole blood samples were exposed in vitro to 2.0 Gy 60Co -radiation. The radiation-induced changes were estimated by using the chromosome aberration test (CA) and cytokinesis blocked micronucleus assay (CBMN) in peripheral lymphocytes, and by determining the malondialdehyde levels (MDA) in blood plasma and the superoxide dismutase (SOD) and catalase (CAT) activity in erythrocytes. Radiation significantly increased the chromosome aberration and micronuclei frequencies as well as MDA levels and decreased the antioxidant enzyme activity. On the other hand, the HRE pretreatment significantly decreased the CA, MN frequencies and MDA levels and increased the SOD and CAT activity in a concentration dependent manner. The most effective was the highest concentration of HRE (0.12 g/kg body weight). The results suggest that HRE as a natural product with an antioxidant capacity could play a modulatory role against the cellular damage induced by -irradiation. The possible mechanism involved in the radioprotective potential of HRE is discussed.

Oral submucosus fibrosis [OSMF] is a collagen-related disorder seen in habitual betel quids and smokers. This is a high risk precancerous condition in which the connective tissue fibers of the lamina propria and deeper parts of the mucosa becomes stiff with restricted mouth opening. Patients with severe cases have symptoms like difficulties in chewing, swallowing and speaking. In the present study 25 individuals were gutkha chewers and 25 were OSMF patients [chewing gutkha along with smoking] and 25 individuals were taken as controls. A significant increase in the frequency of micronuclei was observed in OSMF patients [34.4 +/- 1.79] as compared to gutkha chewers [14.4 +/- 0.73] and controls [4.36 +/- 0.27]. The number of micronucleated cells in OSMF, gutkha chewers and control groups were 19.84 +/- 0.69, 12.6 +/- 0.51 and 4.20 +/- 0.27, respectively and are significantly different at p < 0.05. Acridine orange is used due its fluorescence nature and easier visibility of the micronucleus present in the buccal epithelial cells. It is concluded that chewing gutkha along with smoking is more dangerous for human health as it hastens the incidence of OSMF

Los ecosistemas acuáticos cercanos a las zonas urbanas sufren un deterioro constante ocasionado por diversos factores físico-químicos relacionados con la actividad antrópica, representando un riesgo para los organismos que los habitan, en especial los peces. Estos efectos pueden ser biomonitoreados mediante técnicas de amplia utilización, como el Test de Micronúcleos (Mn) y el de Anormalidades nucleares (AN), entre otros. El objetivo de este trabajo fue verifcar y comparar la frecuencia de micronúcleos y anormalidades nucleares en tres especies ícticas colectadas en un lago urbano, Cyprinus carpio, Astyanax eigenmanniorum y Cheirodon interruptus. Luego de la captura y anestesia de los peces, se extrajo de cada individuo una muestra de sangre y se registró el peso y la longitud total. Las muestras fueron fjadas y luego teñidas con Giemsa al 10%, posteriormente se las analizó con microscopio (1000X) y se les tomaron fotografías con cámara digital. Se calculó, además, el índice de condición corporal (K) para cada uno de los ejemplares capturados. Se aplicó un análisis de correlación de rangos de Spearman (rs) entre frecuencia de Mn y AN vs K para cada especie. La prueba de Kruskal-Wallis se aplicó para evaluar diferencias de Mn y AN entre las especies. Los resultados no mostraron una correlación estadísticamente signifcativa (P> 0,05) entre las frecuencias de Mn y AN vs K. Las frecuencias de Mn entre las diferentes especies no arrojaron diferencias signifcativas (P> 0,05), mientras que las AN sí (P< 0,05). Astyanax eigenmanniorum fue la especie que mayor sensibilidad presentó. Las diferencias preliminares de AN entre las especies utilizadas, convierten a A. eigenmanniorum en una especie "centinela" y posible biomonitor de agentes xenobióticos.


Aquatic ecosystems near urban areas suffer a steady deterioration caused by physical and chemical factors related to human activity, representing a danger to organisms that inhabit them, especially the fsh. These effects may be biomonitored by widely used techniques, such as the Micronucleus Test (Mn) and Nuclear Abnormalities (NA), among others. The aim of this study was to verify and compare the frequency of micronuclei and nuclear abnormalities in three fsh species collected in an urban lake, Cyprinus carpio, Astyanax eigenmanniorum and Cheirodon interruptus. After the capture and anesthesia of the fsh, weight and length were registered, and a blood sample was obtained from each individual. The samples were fxed and stained with 10% Giemsa, subsequently they were analyzed with the microscope (1000X) and photographed with a digital camera. The body con-dition index (K) was calculated for each fsh caught. A correlation analysis Spearman rank (rs) between frequency of Mn and NA vs. K was performed for each species. The Kruskal-Wallis test was applied to assess differences in Mn and NA between species. The results showed no statistically signifcant correlation (P> 0.05) between the frequencies of Mn and NA vs K. The frequencies of Mn between different species yielded no signifcant differences (P> 0.05), whereas NA (P <0.05) did. A. eigenmanniorum was the most sensitive species. NA preliminary differences between the species used, convert A. eigenmanniorum in a sort of "sentinel specie" and a possible biomonitor of xenobiotic agents.

The Trad-MCN bioassay was used to investigate the genotoxicity on Tradescantia pallida var. purpurea plants exposed to variations in the environmental conditions in urban and rural sites in the metropolitan area of Porto Alegre, southern Brazil, over a one-year period. In spring 2009 and in summer, autumn and winter 2010, potted plants of T. pallida var. purpurea were exposed at two sites with different characteristics: the urban area of the municipality of Estância Velha, with leather and footwear industrial activity, and a Site of Special Environmental Interest in the rural area of the municipality of Novo Hamburgo. Other plants comprised the control group and were kept indoors. Frequencies of micronuclei (MCN) were determined in early tetrads of pollen mother cells and expressed as MCN/100 tetrads. Climate data were also registered during the experiment. MCN frequencies in the urban area were significantly higher (up to 8.13) than those found in the rural area (up to 1.26) and in the control group (up to 1.10), which did not differ statistically from each other over the year. The higher MCN frequencies observed in the urban site can be attributed to air pollution, but also may have been influenced by microclimatic and daily thermal variation differences between sites. Higher temperatures recorded in spring and summer may have influenced MCN frequencies observed in the urban site. No clear relation was observed between rainfall and MCN frequencies. Similar and high relative humidity percentages were registered over the period of the study. Considering that the bioindicator plant presents an integrated response to abiotic factors such as pollutants and weather conditions, it can be used as an additional tool that can point to synergistic effects of environmental variables on organisms.


O bioensaio Trad-MCN foi utilizado para investigar a genotoxicidade em plantas de Tradescantia pallida var. purpurea expostas a variações nas condições ambientais, em uma área urbana e uma área rural, na região metropolitana de Porto Alegre, Região Sul do Brasil, durante o período de um ano. Na primavera de 2009 e no verão, no outono e no inverno de 2010, plantas de T. pallida var. purpurea em vasos foram expostas em dois pontos amostrais, com diferentes características: a área urbana do município de Estância Velha, com atividade industrial do setor coureiro calçadista, e uma Área de Especial Interesse Ambiental (AEIA), na zona rural do município de Novo Hamburgo. Outras plantas compuseram o grupo controle e foram mantidas em ambiente interno. Frequências de micronúcleos (MCN) foram determinadas em tétrades jovens de células-mãe de grãos de pólen e expressas como MCN/100 tétrades. Dados climáticos também foram registrados durante o experimento. As frequências de MCN na área urbana foram significantemente superiores (até 8,13) àquelas encontradas na área rural (até 1,26) e no grupo controle (até 1,10), as quais não diferiram estatisticamente entre si ao longo do ano. As maiores frequências de MCN observadas na área urbana podem ser atribuídas à poluição atmosférica, mas também podem ter sido influenciadas por diferenças microclimáticas e de variação térmica diária entre áreas. Temperaturas mais altas registradas na primavera e no verão podem ter influenciado as frequências de MCN observadas na área urbana. Não foi observada uma clara relação entre a pluviosidade e as frequências de MCN. Similares e altas porcentagens de umidade relativa foram registradas ao longo do período de estudo. Considerando‑se que a planta bioindicadora apresenta uma resposta integrada a fatores abióticos, tais como poluentes e condições climáticas, esta pode vir a ser utilizada como uma ferramenta adicional, que pode apontar para efeitos sinérgicos de variáveis ambientais sobre os organismos.

Há poucas informações sobre o efeito tóxico in vivo do extrato etanólico das folhas de Carica papaya (ECP). Portanto, esse estudo caracteriza quimicamente ECP por meio de HPLC-RP e avalia a atividade antimutagênica e citotóxica da fração aquosa de ECP422 Antimutagenic activity of Carica papaya L.Rev Ciênc Farm Básica Apl., 2011;32(3):419-423(CA), empregando ensaio do micronúcleo (MN). O extrato tem predominância de substâncias polares, entre elas a rutina. O ensaio do MN foi realizado em ratos Wistar. Grupos: Controle Negativo (CN), veículo; Controle Positivo (CP40), ciclofosfamida (40 mg/Kg i.p.), 24 horas; Grupo tratado (CA), ECP (500 mg/Kg, p.p.) 24h; Grupo Tratado (CACP40), CA (500 mg/Kg, p.o.) 48, 36 e 24 horas , a última dose de ECP foi administrada junto com ciclofosfamida (40 mg/Kg, i.p.). O índice de MN para CN e CA foi de 3,2 ± 1,79 e 1,6 ± 0,5, e razão PCE/NCE foi de 1,38 ± 0,52 e 1,13 ± 0,28, respectivamente, indicando baixa toxicidade. Por outro lado o índice de MN foi de 20 ± 4,9 para CP40 e 3,0 ± 1,6 para o CACP40, indicando efeito antimutagênico de ECP. Este estudo sugere que ECP possui efeito antimutagênico e baixa toxicidade e que a rutina pode estar envolvida nesse efeito.


There is little information about in vivo toxic effects of the ethanolic extract of leaves of Carica papaya L. (ECP). Therefore, in this study ECP was characterized chemically by HPLC-RP and the antimutagenic and cytotoxic activities of an aqueous solution of ECP (CA) were assessed by the micronucleus (MN) bioassay. The extract consisted mainly of polar substances, one of which was rutin. The MN test was performed on groups of Wistar rats, as follows: negative control (NC) - vehicle; positive control (CP40) - cyclophosphamide (40 mg/kg, ip), 24h; extract-treated (CA) - ECP (500 mg/kg, po), 24h; extract + cyclophosphamide-treated (CACP40) - ECP (500 mg/kg, po), 48, 36 and 24 h, plus cyclophosphamide (40 mg/kg, ip), 24h. The MN index was 3.2 ± 1.79 and 1.6 ± 0.5, for NC and CA, and the PCE-to-NCE ratio was 1.38 ± 0.52 and 1.13 ± 0.28, respectively, indicating low cytotoxicity of CA. CP40 showed a high MN index of 20 ± 4.9, but CACP40 only 3.0 ± 1.6, the same as NC, indicating an antimutagenic effect. The study suggests that ECP has low toxicity and possesses an antimutagenic protective effect in which rutin may be involved.

Background & objectives: Radioiodine (131I) or radioactive iodine in low doses is used worldwide as the first line of management in the treatment of hyperthyroidism. Information is available on the extent and severity of cell damage after a high dose radioiodine (131I) therapy for thyroid cancer, but information is scanty on its cellular effects, its extent and severity of cell damage after a low dose 131I therapy. The present investigation was aimed to study the cytotoxic effects of a low dose 131I therapy in varying doses as is normally being used in routine clinical practice in the treatment of various forms of hyperthyroidism. Methods: Peripheral blood lymphocytes were analyzed in 32 hyperthyroid patients. All of them received 131I in the form of sodium iodide solution orally. Blood lymphocytes were studied for the presence of chromosomal aberrations (CA) and micro nucleus (MN) using micronucleus assay. Blood samples of these patients were drawn prior to the treatment, on 7 thand 30 thdays after the treatment. Results: The results indicated a positive relationship between 131I dose, CA and MN frequency. A statistically significant increase in CA and MN frequency in day 7 post- therapy and a decrease in mean levels of CA and MN on day 30 post-therapy were observed when compared to pre-therapy. Interpretation & conclusions: This study showed that the cytogenetic damage induced by 131I in low doses i.e., less than 555MBq was minimal and reversible. Patients can be motivated to undertake this safe and easy procedure as a first line of therapy in the treatment of hyperthyroidism.

Funções da via auditiva eferente incluem a modulação das células ciliadas externas, proteção contra ruído e melhora na detecção da fonte sonora em ambientes ruidosos. Genotoxicidade são danos ao DNA. OBJETIVOS: Analisar associação entre funções da via auditiva eferente com marcadores genotóxicos. Adicionalmente, considerou-se tabagismo e gênero como principais variáveis intervenientes. MATERIAL E MÉTODO: Estudo prospectivo-clínico, quantitativo, transversal, contemporâneo. Foi realizada uma análise da função da via auditiva eferente e indicadores de genotoxicidade em 60 voluntários adultos jovens. RESULTADOS: Idade média dos voluntários foi de 24,86±3,68 anos; 30 do gênero masculino e 30 do gênero feminino, 15 de cada gênero tabagistas e 15 não tabagistas; indivíduos do gênero masculino tabagistas apresentaram maior ocorrência de efeito de supressão das EOAEPDs nas frequências de 2000 e 6000Hz na orelha do lado esquerdo; mulheres tabagistas apresentaram maior prevalência de queixa de dificuldade de ouvir em ambiente ruidoso; indivíduos tabagistas e mulheres apresentaram maiores danos ao DNA; indivíduos com queixas de dificuldade auditiva e zumbido apresentaram maiores índices de genotoxicidade. CONCLUSÕES: Em adultos jovens normo-ouvintes que referem queixas relacionadas às funções da via auditiva eferente, como zumbido e dificuldade auditiva, já é possível observar associação com genotoxicidade considerando interações entre gênero e tabagismo.


Efferent auditory pathways modulate outer hair cells of the cochlea, protect against noise, and improve the detection of sound sources in noisy environments. Genotoxicity is DNA damage. AIM: To study the association between auditory pathway efferent functions with genotoxic markers. The study also considered smoking and gender as two main variables. METHODS: A prospective-clinical, quantitative, cross-sectional, contemporary study. The function of efferent auditory pathways and genotoxicity tests in 60 healthy young subjects were assessed. RESULTS: The mean age of subjects was 24.86 years +/- 3.68 years; there were 30 males and 30 females, 15 of each gender smokers and 15 non-smokers. Male smokers had a higher incidence of DPOEA suppression effect at 2000 and 6000 Hz in the left ear; female smokers had a higher prevalence of complaints of difficulty to hear in noisy environments; smokers and women had a higher mean DNA damage; subjects with complaints of hearing loss and tinnitus had higher genotoxicity. CONCLUSIONS: In young normal-hearing adults that complain about efferent auditory pathways functions, such as tinnitus and hearing impairment, there are possible associations with genotoxicity; interactions between gender and smoking are considered.

BACKGROUND: For the past few decades, it has been widely known in developed countries that tobacco is dangerous, but it is still insufficiently realized how big these dangers really are. AIMS: To determine and evaluate micronuclei (MN) frequencies of young smokers and nonsmokers in three different tissues (peripheric blood lymphoctes, buccal mucosa, and exfoliative urothelial cells) at the same time. MATERIALS AND METHODS: MN assay was performed on buccal mucosa, urothelial cells, and peripheric blood lymphocyte samples obtained from 15 healthy male smokers (>5 pack-years) and 15 healthy male nonsmoker controls who had not been exposed to any known genotoxic agent. STATISTICAL ANALYSIS USED: The statistical differences between smoker and nonsmoker groups were calculated by using student t test. The differences between smoker-group tissues were compared by ANOVA. RESULTS: It was found that MN frequency (mean value ± standard deviation) in oral mucosa cells from smokers and controls were 1.20 ± 0.22% and 0.26 ± 0.10%; in urothelial exfoliative cells, 1.29 ± 0.28% and 0.12 ± 0.08%; in peripheric blood lymphocytes, 1.53 ± 0.23% and 0.38 ± 0.12%, respectively. The mean MN frequencies in buccal mucosa, urothelial exfoliative cells, and peripheric blood lymphocytes were significantly higher in smokers than in those of controls (P<0.05). All tissues were affected from smoking, but the most destructive effect was seen in urothelial cells of smokers (P<0.05). CONCLUSIONS: Our data showed that cigarette smoke is a DNA damage causitive agent on exfoliative buccal mucosa and urothelial cells and peripheric blood lymphocytes of young smokers, but it has most destructive effect on urothelial cells.

Pueraria mirifica is a Thai phytoestrogen-rich herb traditionally used for the treatment of menopausal symptoms. Pueraria lobata is also a phytoestrogen-rich herb traditionally used in Japan, Korea and China for the treatment of hypertension and alcoholism. We evaluated the mutagenic and antimutagenic activity of the two plant extracts using the Ames test preincubation method plus or minus the rat liver mixture S9 for metabolic activation using Salmonella typhimurium strains TA98 and TA100 as indicator strains. The cytotoxicity of the two extracts to the two S. typhimurium indicators was evaluated before the mutagenic and antimutagenic tests. Both extracts at a final concentration of 2.5, 5, 10, or 20 mg/plate exhibited only mild cytotoxic effects. The plant extracts at the concentrations of 2.5, 5 and 10 mg/plate in the presence and absence of the S9 mixture were negative in the mutagenic Ames test. In contrast, both extracts were positive in the antimutagenic Ames test towards either one or both of the tested mutagens 2-(2-furyl)-3-(5-nitro-2-furyl)-acrylamide and benzo(a)pyrene. The absence of mutagenic and the presence of anti-mutagenic activities of the two plant extracts were confirmed in rec-assays and further supported by a micronucleus test where both plant extracts at doses up to 300 mg/kg body weight (equivalent to 16 g/kg body weight plant tuberous powder) failed to exhibit significant micronucleus formation in rats. The tests confirmed the non-mutagenic but reasonably antimutagenic activities of the two plant extracts, supporting their current use as safe dietary supplements and cosmetics.

This work investigated the cytotoxic and genotoxic potential of water from the River Paraíba do Sul (Brazil) using Allium cepa roots. An anatomo-morphological parameter (root length), mitotic indices, and frequency of micronuclei were analysed. Eight bulbs were chosen at random for treatment for 24 to 120 hours with the River water collected in the years of 2005 and 2006 from sites in the cities of Tremembé and Aparecida (São Paulo state, Brazil). Daily measurements of the length of the roots grown from each bulb were carried out throughout the experiment. Mitotic index (MI) and frequency of micronuclei (MN) were determined for 2000 cells per root, using 3-5 root tips from other bulbs (7-10). Only in the roots treated with samples of the River water collected in 2005 in Tremembé city was there a decrease in the root length growth compared to the respective control. However, a reduction in MI values was verified for both sites analysed for that year. Considering the data involving root length growth and especially MI values, a cytotoxic potential is suggested for the water of the River Paraíba do Sul at Tremembé and Aparecida, in the year of 2005. On the other hand, since in this year the MN frequency was not affected with the river water treatments, genotoxicity is not assumed for the river water sampled at the aforementioned places.


Este trabalho é parte de uma investigação sobre o potencial citotóxico e genotóxico da água do Rio Paraíba do Sul (Brasil) utilizando raízes de Allium cepa. Foi analisado um parâmetro anátomo-morfológico (crescimento das raízes), bem como o índice mitótico e a frequência de micronúcleos. Aleatoriamente oito bulbos foram submetidos aos tratamentos de 24 a 120 horas com água do Rio proveniente dos pontos de coleta das cidades de Tremembé e Aparecida nos anos de 2005 e 2006. O comprimento das raízes de cada bulbo foi acompanhado diariamente ao longo do experimento. O índice mitótico (IM) e a frequência de micronúcleos (MN) foram determinados pela análise de 2.000 células por raiz, sendo utilizadas 3-5 raízes de outros bulbos (7-10). Somente as raízes tratadas com amostras de água coletadas em 2005 na cidade de Tremembé, apresentaram decréscimo no comprimento das raízes quando comparadas com o controle. Entretanto, foi observada redução do IM nas raízes tratadas com água de ambos os pontos de coleta no mesmo ano. Considerando os dados de crescimento de raiz e especialmente IM, um potencial citotóxico é sugerido para a água do Rio Paraíba do Sul em Tremembé e Aparecida, no ano de 2005. Por outro lado, para este mesmo ano, a frequência de micronúcleos não foi alterada; assim, a genotoxicidade não foi assumida para a água do Rio nos pontos mencionados.

El D-004 es un extracto lipídico del fruto de la palma real (Roystonea regia), que ha demostrado ser eficaz en prevenir la hiperplasia prostßtica inducida por testosterona y por fenilefrina en modelos experimentales en ratas. El objetivo fue determinar si el D-004 induce cambios en la frecuencia de aparición de micronúcleos al realizar la administración oral a dosis repetida durante 8 semanas. Se formaron 5 grupos experimentales (7 animales/grupo): un grupo control solvente, tres tratados con D-004 (500, 1 000 y 1 500 mg/kg) y un control positivo tratado con ciclofosfamida. No ocurrieron muertes ni se detectaron signos clínicos de toxicidad. No hubo diferencias significativas entre controles y tratados en cuanto a la frecuencia de eritrocitos policromatófilos micronucleados y el índice de citotoxicidad. En conclusión, el D-004 administrado por vía oral a las dosis empleadas no presenta actividad clastogénica ni citotóxica en médula ósea de ratones OF-1 machos in vivo.


D-004 is a lipid extract from the real palm fruit (Roystonea regia), which has demonstrated that is effective to prevent prostatic hyperplasia from testosterone and from phenylephrine in rats' experimental models. Aim of this paper was to determine if D-004 provoke changes in appearance frequency of micronuclei during oral administration of repeated doses for 8 weeks. Five experimental groups were created (7animals/group): a solvent control-group, three treated with D-004(500, 1 000, and 1 500 mg/kg), and a positive control-group treated with cyclophosphamide. There were no deaths or clinical signs of toxicity neither significant differences among controls and treated ones as regards the frequency of micronucleated polychromatophils erythrocytes, and the cytotoxic index. In conclusion, D-004 administered per os at doses used has neither clastogenic nor cytotoxic activity in bone marrow male OF-1mice in vivo.
