RESUMEN

No ovário das abelhas as células germinativas e as células foliculares são interconectadas por pontes intercelulares mantidas abertas por reforços do citoesqueleto na membrana plasmática. As pontes entre as células germinativas têm comportamento dinâmico e provavelmente atuam na determinação do ovócito entre as células do clone formado pelas mitoses pré meióticas formando posteriormente uma via de transporte para que os produtos sintetizados pelas células nutridoras atinjam o ovócito durante sua maturação. Os elementos do citoesqueleto presentes nas pontes intercelulares das gônadas das abelhas são basicamente microfilamentos e microtúbulos, mas nas pontes entre os cistócitos pré-meióticos outro tipo de filamento (espesso de natureza não definida, associado a elementos do retículo endoplasmático) está presente, atravessando a ponte e prendendo-se através dos microfilamentos à membrana plasmática. Estes filamentos aparentemente controlam o vão da ponte. Terminada a fase de proliferação os cistócitos tomam a forma de uma roseta e um fusoma, formado pela convergência das pontes, aparece no centro desta. Nesta conformação os filamentos grossos não estão presentes. Nova mudança ocorre com a diferenciação do ovócito e das células nutridoras, com a reorientação de todas as pontes de maneira a canalizar o conteúdo das futuras células nutridoras para o ovócito.

RESUMEN

PURPOSE: To investigate the morphological characteristics of keratocytes and the interconnection of keratocytes with adjacent keratocytes using the flat preparation method and scanning electron microscopy with a frontal section of the human corneal stroma. METHODS: The thin, corneal collagen lamellae were carefully dissected from the cornea (n=7), which had been stained by the flat preparation method. The remaining tissue was fixed in 3% glutaraldehyde and observed by transmission electron microscopy following the frontal section. RESULTS: The flat preparation revealed the corneal fibroblasts between the lamellae of the collagen fibers and showed that the ramifying cellular processes of the keratocytes were in contact with the cytoplasmic processes or cell bodies of neighboring fibroblasts. Two types of discrete subpopulations of keratocytes were identified: a smaller, cellular type of keratocyte with spindle-shaped nucleus with heterochromatin, and a larger, cellular type with a large indented nucleus with relatively scanty cytoplasm. Collagen fibers ran parallel to each other toward the fenestration of the cytoplasmic wall of the keratocyte. CONCLUSIONS: These flat preparation method results showed that the keratocytes within the corneal stroma are interconnected with the adjacent keratocytes, which indicates the presence of a functional communicating network through the keratocyte circuits within the stroma. A smaller, cellular type of keratocyte with spindle-shaped nucleus was morphologically differentiated from a larger, cellular type with a large, indented nucleus by flat preparation and transmission electron microscopy.

RESUMEN

Seventeen chronic tobacco chewers and three control subjects underwent clinical evaluation, upper gastrointestinal endoscopy and esophageal mucosal biopsies. The esophageal biopsies were processed and examined under the electron microscope. A large number of ultrastructural abnormalities such as discontinuous, fragmented basement membrane, with reduction in hemidesmosomes, widened intercellular spaces were found in the esophageal mucosa of chronic tobacco chewers which resembled the ultrastructural features of experimental carcinogenesis and leukoplakia. It is concluded that chronic chewing of tobacco produces ultrastructural abnormalities in the esophageal mucosa which could be important precursors for esophageal malignancy.

RESUMEN

El número de uniones estrechas intercelulares fue cuantificado en islotes aislados de rata, incubados durante diferentes lapsos en un medio conteniendo distintas concentraciones de glucosa. El número de uniones estrechas aumentó en función de la duración de la incubación y de la concentración de glucosa en el medio. Estos resultados sugerirían la participación de las uniones estrechas en la regulación de la secreción de insulina en respuesta a la glucosa
