The effect of the antineoplastic agents methotrexate and vinblastine on odontogenesis of the continuously growing rat incisor has been investigated. Methotrexate resulted in marked degenerative changes of the odontoblasts and their replacement with large rounded cells having giant or double nuclei, these cells occupy the subodontoblastic zone. Ameloblasts expressed distal movement of their nuclei and signs of degeneration in more severly affected areas. Vinblastine resulted in hydropic degeneration, karyolysis and atrophy of the odontoblasts, fatty infiltration of the subodontoblastic cells, disorganization and degeneration of the ameloblasts. Both drugs resulted in marked decrease of staining reactivity of the odontoblasts and subodontoblastic cells to alkaline phosphatase. Tetracycline labelling revealed that treatment with methotrexate or vinblastine resulted in very highly significant decrease in dentin formed during the period of the experiment
