RESUMEN

<p><b>OBJECTIVE</b>To study tissue culture of Vinca minor and determine the content of vincamine.</p><p><b>METHOD</b>Leaf blades, stalks, root segment of V. minor were used as explants to study the effect of 2, 4-D,6-BA,NAA on its callus induction and vincamine contents in the orthogonal design experiment. In the peak period of callus formation, vincamine content in callus of V. minor and sterile plants was determined by HPLC. The experimental data was statistically analyzed.</p><p><b>RESULT</b>The content of 6-BA and NAA had no significant effect on its callus induction. But the content of 2, 4-D had significant effect on its callus induction. Within 20,40,60 d, the content of vincamine in sterile plant was (0.015 +/- 0.003)%, (0.097 +/- 0.001)% , (0.113 +/- 0.06)%, respectively. In the peak period of callus formation, vincamine content in callus of leaf blades, stalks, root segment was (0.024 +/- 0.0025)%, (0.016 +/- 0.0015)%, (0.010 +/- 0.0015)%, respectively. To 30 days of subculture, vincamine content in callus of leaf blades, stalks, root segment was (0.041 +/- 0.002)%, (0.019 +/- 0001)%, (0.016 +/- 0.002)%, respectively.</p><p><b>CONCLUSION</b>The optimal hormone combination for callus initiation was MS +2, 4-D 1.0 mg x L(-1) +6-BA 0.5 mg x L(-1) + NAA 0.5 mg x L(-1). In different growth periods, vincamine content in sterile plants is significantly different. From different explants in callus vincamine content is different, in which leaves callus is significantly higher than that of stems, roots produced callus organization.</p>

RESUMEN

Kneaded mixtures as well as physical mixtures, of an acidic drug, indomethacin, a basic drug vincamine, and a neutral drug, prednisolone, with gelatin and casein hydrolysates were prepared. Their in vitro dissolution profiles were examined. The dissolution of drugs from the kneaded mixtures was significantly increased compared to the drugs alone as well as their physical mixtures. This increase was most prominent for the acidic drug indomethacin. The casein and gelatin hydrolysates enhanced the dissolution of the three drugs by improving the wettability of the drug particles. Solubility also contributes to the enhancement of dissolution of indomethacin and prednisolone. Buffering was an additional reason in case of indomethacin

The pineal tissue was studied in four groups of male albino rats: a group of adult rats, a second group of senile rats, the third and fourth groups consisted of senile rats which were injected with the vehicle of vincamine drug and the drug itself respectively in their old adult stage of life. The investigations showed that the adult pineal gland was characterized by high cellularity, and two types of cells were observed; the pinealocytes and the glial cells. The pinealocytes constituted the majority of cells and were of two types; light and dark, while the glial cells were fewer, smaller with ramifying thin processes. Using electron microscopy, the cytoplasm of light pinealocytes was noted to be rich in secretory organelles, containing euchromatic nuclei and with prominent nucleoli. On the other hand, the dark pinealocytes appeared more electron dense with less amount of organelles and heterochromatic nuclei in their cytoplasm. Glial cells appeared as small cells of high electron density with thin ramifying processes full of filaments and large dark heterochromatic nuclei. Senile pineal glands of both the second and third groups showed marked decrease in the cellularity, irregularity and in folding of the nuclei were a prominent feature. By electron microscopy, the light pinealocytes showed marked decrease in the amount of secretory organelles together with the appearance of lipid droplets, lysosmes, and dense bodies. In addition, the nuclei of this type showed deep indentations. The cytoplasm of dark pinealocytes showed abundant multivesicular bodies and deeply indented heterochromatic nuclei. The glial cells had to same structure as in the adult glands. The study of the pineal tissue in senile animals which were previously treated with vincamine showed marked increase in the cellularity, and the majority of cells were of the light variety. By electron microscopy, the cells had the picture of highly active cells. The light pinealocytes were rich in mitochondria and secretory organelles such as dilated cisterns of endoplasmic reticulum, and Golgi apparatus. On the other hand, lysosomes, lipid droplets and dense bodies were hardly observed. The nuclei of these cells were either oval or irregular and of the euchromatic appearance. The dark pinealocytes showed secretory organelles in their cytoplasm but less abundant than those observed in the cytoplasm of the light cells. The nuclei, although of high electron density, yet they were of the euchromatic appearance. According to the present investigation, it could be concluded that the currently discussed drop in melatonin synthesis in old age is due to regressive changes in the morphology of the gland. Moreover, the use of vincamine may prevent or delay the appearance of such degenerative changes. This may preserve the melatonin synthesis in the body and consequently delays the aging process

RESUMEN

Se realizó un estudio simple ciego paralelo con duración de seis meses, para evaluar la eficacia de buflomedil y vincamina en pacientes con síndrome demencial. De sesenta y dos pacientes que reunieron los criterios de inclusión (7 ó más puntos de la escala isquémica de Haschinski), hubo 14 deserciones; los 48 pacientes restantes se dividieron en dos grupos: 25 recibieron buflomedil (450 mg/día) y 23 vincamina (60mg/día) durante 180 días sin interrupción. Se les practicó: a) examen neurológico, b) psicométrico, c) registro de signos vitales, d) datos de tolerancia al medicamento y electroencefalograma los días 0,90 y 180. Los estudios a, c y d, también se hicieron los días 30, 60, 120 y 150. Los dos medicamentos modificaron estadísticamente los parámetros psicométricos y no hubo cambios neurológicos, modificación electroencefalográfica, ni en los signos vitales, ni hubo datos de intolerancia al medicamento. El buflomedil modificó favorablemente los parámetros de inteligencia en la prueba de Wais tanto para el cociente verbal como de ejecución. Así mismo, los pacientes menores de 75 años mejoraron el cociente mnésio. Cambios similiares también se observaron en otros parámetros (cociente mnésico global) en la prueba de Weschler y Bender en los tratados con vincamina. Ambos medicamentos pueden ser útiles como un recurso más en la terapéutica del paciente con síndrome demencial

RESUMEN

Trescientos cincuenta y tres pacientes, divididos en tres grupos, con manifestaciones de insuficiencia circulatoria cerebral, pérdida de la autonomía y sintomatología neurosensorial, fueron tratados con 60 mg de vinvamina* al día, durante 30,45 y 60 días. Se observó mejoría de la sintomatología objetiva y subjetiva que acompaña a la insuficiencia circulatoria cerebral y se comprobó mediante análisis estadístico con la prueba t de Student. Todos los parámetros mostraron resultados altamente significativos con p <0.001. La tolerancia fue excelente, sólo 5% de los 353 pacientes tratados presentaron efectos secundarios leves y pasajeros que no impidieron la contaminación del estudio

RESUMEN

Estudio multicéntrico en diferentes zonas de Venezuela en 78 pacientes con síntomas de envejecimiento cerebral, tratados con vincamina 20 mg t.i.d. durante 6 semanas. Los pacientes se evaluaron por método de Student y Willcoxon. Se demonstró la mejoría estadísticamente significativa de todos los parámetros con una buena tolerancia, sólo 5 epigastralgias que no ameritaron interrupción de la terapia
