Résumé

The TP63 gene, as the oldest homologue of TP53, encodes two main N-terminal variants by different promoters: the trans-activating variant, TAp63 with tumor suppressor activity and the amino terminal truncated variant, delta Np63 with oncogenic activity. As the result of the deletion of exon 4 in each of the variants, two further N-terminal variants have been reported for p63 in the last decade: d4TAp63 and delta Np73L, but the exact function of these variants have not been determined in normal and tumoral cells. In this study we evaluated the expression pattern of p63 variants and usefulness of d4TAp63 and delta Np73L as potential diagnostic molecular markers in breast cancer. In this study 30 tumoral and 20 non tumoral samples of marginal tissues were studied by use of Semi-quantitative Reverse Transcriptase-nested PCR [RT-nPCR] method. SPSS16 software was used for data analysis. In all cases with expression of TAp63 and delta Np63 variants, d4TAp63 and delta Np73L variants were always expressed with their long variants simultaneously. In tumoral and marginal samples delta Np73L mean expression level was significantly higher than the mean expression level of d4TAP63 [P = 0.009 and P=0.008 respectively]. delta Np63 expression levels in tumoral and marginal samples were also higher than those of TAp63, which had a statistically significant difference in tumoral samples [P=0.03], but no significant difference was detected in marginal samples [P = 0.11]. The results indicated dominant negative inhibitory activity of delta Np63 and its potential role as an oncogene in breast cancer. delta Np73L variant compared to d4TAp63, in tumoral and non tumoral breast tissues can act as dominant negative variant. Both variants [delta Np73L and d4TAp63] with similar expression patterns to their respective longer variants [delta Np63 and TAp63] and simultaneous expression with their variants are likely to be involved in strengthening the tumor suppressor activity in normal and tumoral breast cells. On the other hand, according to the expression patterns of delta Np73L and d4TAp63 variants, they cannot be considered as molecular markers in the diagnosis of breast tumors

Résumé

Impairment in the function of the brain caused by deficiency in the brain circulation is called stroke. Stroke is the most important factor of disability in adults. One of the most prevalent consequences of stroke is disability of walking or so-called [gait disorder] so that more than 50 percent of the patients who are in the acute phase of the disease are not capable of walking, and need to a rehabilitation period to reach for walking. In order to compare the effect of exercise therapy on the function of walking and balance and Improvement of depression in left and right hemiplegics patients, a Non-randomized clinical study was done. This study consisted of 13 right- hemiplegics patients with an average age of [60.46 +/- 1.47] and 13 left hemiplegics patients with an average age of [61.92 +/- 2.01] who were matched in age, height and weight. To measure the function of walking the Gait Dynamic Index, function of balance the Berg Balance Scale and Depression Beck Depression Questionnaire were used. The data were analyzed by SPSS software. To compare the effect of exercise therapy on the improvement of walking and balance before and after the treatment in each group t-paired -test and for more comparison of the two groups an independent T-test was used. With analyzing the received data the following results were derived: From the viewpoint of walking, a significant difference in the right hemiplegics group [p<0.001] and the left hemiplegics one [P=0.0] before and after the treatment is derived. From the viewpoint of balance, a significant difference in the right hemiplegics group [p<0.001] and the left hemiplegics group [P=0.0] before and after the treatment was noticed. About the improvement of depression, a significant difference in the right hemiplegics group [p<0.001] and the left one [p<0.00l] before and after the treatment was noticed. There was no significant difference between two groups before [p=0.85] and after [0.36] the treatment from the point of view of walking. There was no significant difference between two groups before [p=0.056] and after [p=0.067] the treatment in balance. There was no significant difference between two groups before [p=0.067] and after [p=0.054] treatment from the viewpoint of depression. This study showed that performing the exercise therapy protocol improves the function of walking, balance and depression in herniplegic patients; never the less, no differences were observed between the two right and left hemiplegics groups

The study of genetically polymorphism of population have been done on mitochondrial genome in many developed countries. Azeri is one of the largest Iranian ethnic groups settled in north west of Iran. The aim of this study is the evaluation of mtDNA polymorphism by PCR- RELP method. In this study 120 blood samples were collected from Azerbaijan people settled in different regions of this state, and the buffy coat of blood samples were isolated. The DNA of white blood cells were extracted, and the mitochondrial D-loop fragment was amplified by using special primers with PCR. Then, PCR product was digested by restriction enzymes. D-Loop Fragment with the size of 1024bp amplified by PCR and related bands showed on gel electrophoresis. In cutting with Hae III enzyme, six diversities were observed, one of which had heteroplasmy. Also, we detected two diversities including one case of heteroplasmy by AluI digestion. Seven genetically diversities were determined in 120 samples. This diversity in our samples can be the result of the extent of a smaller ancestral population or the shorter time genetic history in Azeri ethnic group
