Résumé

Histochemical studies and SEM observations on the morphological changes in the head of the spermatozoa in general, and the acrosome in particular, in A. indica treated rats are reported. In the treated rats change in the shape and size of the sperm head, with a dorso-ventral constriction of the middle region of the sperm head i.e., between the anterior and posterior regions was observed. It was rather difficult to differentiate the outer acrosomal and outer plasma membranes. A decrease in the perforatorium or sub-acrosomal material, post nuclear cap and the nuclear material near the basal plate at the base of the sperm head were also observed. The results suggest that the effects are probably due to androgen deficiency and a general disturbance in carbohydrates or polysaccharides located in the sperm head, caused by the antiandrogenic property of the leaves of A. indica.

Résumé

Indomethacin (2 mg/100 g body weight), induces haemorrhagic gastric ulcers in albino rats. The incidence and severity of ulceration increased with starvation period. Indomethacin caused little or no effect on the cellular and the nuclear diameter of parietal and chief cells while reduction was observed in mucus and endocrine cells. The effect was enhanced with increased duration of starvation. Both mucous and endocrine cells decreased in their number after 72 hr of starvation. Thus prolonged starvation enhanced the gastric mucosal damage induced by indomethacin.

Résumé

Histological and biochemical changes in the testis of rats treated with the leaf powder of A. indica are reported. The pattern of recovery is also studied at 8, 16 and 24 day after withdrawal of the treatment. In the treated rats, a general reduction in the diameters of seminiferous tubule, nuclei of the germinal elements and a mass atrophy of the spermatogenic elements has been observed. The Leydig cells are found to be atrophic. Biochemically, a decrease in the protein content and the activity of acid phosphatase and an increase in the total free sugar, glycogen, cholesterol contents and the activities of alkaline phosphatase and lactate dehydrogenase have been observed. A gradual recovery is observed in both the histological and biochemical parameters after 8.16 and 24 day of cessation of the treatment. The result suggest a possible reversible antiandrogenic property of the leaves of A. indica in male albino rats.

Résumé

The histological and biochemical changes in the caput and cauda epididymis of albino rat treated with 20, 40 and 60 mg dry powder of the leaves of A. indica per day for 24 days are reported. In the treated rats, the height of the epithelium and the diameter of the nucleus in both the regions were reduced. The lumen of the caput was packed with lymphocytes. Biochemically, a decrease in the protein content and acid phosphatase activity and an increase in the alkaline phosphatase and lactate dehydrogenase activities were observed in both the regions. The effect was dose dependent. Further, serum testosterone concentration in the higher dose treated animals decreased significantly. The results suggest a possible antiandrogenic property of the leaves of A. indica.

Résumé

The fraction containing high hemagglutinating activity was prepared from raw winged bean tubers and orally administered to growing rats. The food intake and body weights of these rats decreased as the level of lectin increased and significant lectin activity was detected in the faeces extracted from these rats which is anti-genically similar to the native lectin preparation. Microscopic examination has revealed morphological changes in the intestinal epithelial cells. The binding action of the lectin to the mucosal epithelia of the gastrointestinal tract is indicative of the deleterious effects caused by the winged bean tuber lectin.

Résumé

The gastric ulcerogenic effect of the glucocorticoid, prednisolone, was studied in albino rats. Shay operation in forestomachectomized rats with intact adrenals induced acute gastric ulcers in the corpus, whereas adrenalectomy prevented the development of gastric ulcers in another group of forestomachectomized Shay rats. Only superficial erosion of gastric epithelium was noticed in 66.67% of these rats. Administration of prednisolone to another group of similarly operated rats increased the number and severity of gastric ulceration, whether the adrenals were ablated or otherwise. From the observation of acid and mucous content of the stomach, it is postulated that the reduced mucous secretion may be an important predisposing factor in gastric ulcerogenic effect of steroids.

Résumé

Oral administration of 2.5 mg of prednisolone/100 gm B.W. to starved forestomachectomized albino rats for three days induces 5-7 acute gastric ulcers with a severity of 3+ in 86% of the rats in the corpus comparable to human gastric ulcers with an ulcer index of 17.1. When 15 mg of Phenobarbital/100 gm B.W. was concomitantly administered with prednisolone either orally or subcutaneously prevents the formation of steroid induced ulcers probably due to their insensitization of gastric epithelium to corticoid ulcerogenic induction. This preliminary observation may have a therapeutic value as this sedative has a common usage. It is is suggested that forestomachectomized rats are best suited for ulcerogenic experiments.
