Résumé

Objective To investigate the effects of survivin antisense oligonucleotide (ASODN) on human uterine endometrial cancer HEC-1B cell line. Methods The experiment included four groups: blank control group (blank), lipofectin control group (Lip), oligonucleotide (SODN) control group and antisense oligonucleotide (ASODN) test group. Artificially synthesized survivin antisense oligonucleotide was transfected for 48 hours into endometrial cancer cells using liposome after cultivation. The expression of survivin protein was determined by Western blot, and the cellular apoptotic rate was tested by flow cytometric analysis. MTT method was used to assay cellular growth rate. Results In comparing with the blank, Lip and SODN control groups, survivin protein expression was obviously decreased in HEC-1B cancer cells after transfected with survivin ASODN. The apoptotic rate and proliferation inhibition rate in ASODN test group were significantly higher than that in the control groups (P<0.05). There was no significant difference between other control groups (P>0.05). Conclusions The expression of survivin is decreased in endometrial cancer cells after ASODN transfection. Survivin ASODN causes cellular apoptosis and inhibits the proliferation of HEC-1B cells. It may be a useful gene therapy for endometrial cancer.

Objective To investigate the possible role of nerve growth factors (NGF) and its receptors in ovarian follicle development,and detect the difference between polycystic ovarian symdrome (PCOS) and normal control.Methods Immunohistochemisty was applied to detect the expression of NGF,p75NTR,TrkA in ovarian follicle granulosa cells.Results In 54 cases (including 9 cases of IVM-PCOS,16 cases of IVF-PCOS and 29 cases of IVF-Normal),49 cases showed NGF protein positive in granulose cells with significance difference between groups (P<0.05),and especially NGF was the strongest expressed factor in IVF-PCOS group but there was no remarkable difference between the other two groups in the expression of NGF.41 were TrkA protein positive,almost all IVF cases were TrkA protein positive while only 2 in IVM and there was no remarkable difference in expression of TrkA between IVF PCOS and IVF Normal(P>0.05).The cases with stronger expression of NGF or TrkA had higher estradiol level than that of lower expression group (P<0.05).51 cases had positive expression of p75 NTR protein but there was no significant difference between groups (P>0.05).Conclusions TrkA is expressed in mature ovarian follicle,and the interaction of NGF-TrkA may be involved in ovarian follicle development,and overexpression of NGF may be associated with PCOS development.

<p><b>OBJECTIVE</b>To study the significance of Fas and Fas-L expression in adenocarcinoma of uterine cervix.</p><p><b>METHODS</b>Both carcinoma tissue and their surrounding tissues from 36 patients with adenocarcinoma of uterine cervix, previously untreated either by radiation or chemotherapy, were studied for the expression of Fas and Fas-L by immunohistochemical stain with DNA apoptosis fragment detected by TUNEL.</p><p><b>RESULTS</b>The TUNEL labeling index was negatively correlated with differentiation of adenocarcinoma of cervix. Compared to highly differentiated and moderately differentiated tumor, the TUNEL labeling index was reduced obviously in poorly differentiated adenocarcinoma (P < 0.01). Fas expression was detected in 31 cases (86%) while there were only 3 weakly stained in the normal endocervical glands around the carcinoma. The 5 unstained carcinomas were 3 highly differentiated and 2 moderately differentiated. The positively stained Fas was associated with differentiation; the stronger the stain, the less differentiation there was. The Fas-L expression was detected in all adenocarcinomas while there was only 1 weakly stained in the normal ones. No significant difference was found in the expression of Fas-L in carcinomas with different degrees of differentiation. No correlation was observed between Fas and Fas-L expression.</p><p><b>CONCLUSIONS</b>The Fas expression is positively correlated with the different degrees of differentiation and Fas-L expression may be associated with the escape from of immunal surveillance.</p>
