RÉSUMÉ

The objective of this study was to observe the infection of human cytomegalovirus (HCMV) to human umbilical vein endothelial cells, and its effect on the expression of single-stranded DNA-binding protein (SSBP1) and on lipid metabolism in endothelial cells. We screened the differential expression of mRNAs after HCMV infection by suppression subtractive hybridization and the expression levels of SSBP1 mRNA and protein after HCMV infection by real-time PCR and western blot. After verification of successful infection by indirect immunofluorescent staining and RT-PCR, we found a differential expression of lipid metabolism-related genes including LDLR, SCARB, CETP, HMGCR, ApoB and LPL induced by HCMV infection. The expression levels of SSBP1 mRNA and protein after HCMV infection were significantly down-regulated. Furthermore, we found that upregulation of SSBP1 inhibited the expression of atherosclerosis-associated LDLR, SCARB, HMGCR, CETP as well as the accumulation of lipids in the cells. The results showed that the inhibition of SSBP1 by HCMV infection promotes lipid accumulation in the cells.

Diarrhoea is a major public health problem in Thailand. During November 1998-January 1999, a cross-sectional survey and a nested qualitative study were conducted to understand the relationship between feeding practices and weanling diarrhoea, and to describe the related local beliefs and practices in a subdistrict of northeast Thailand. A cluster-sampling method was used for selecting 156 weanlings aged 3-24 months. A structured interview was conducted with the main caregivers of these weanlings. The questionnaire used for the interview included items about feeding practices and diarrhoea-history of the weanlings in 2 months prior to the interview. Seven focus-group discussions with an opportunistic sample of the caregivers were held in the villages. A series of vignettes and unstructured questions were used for eliciting the local beliefs about weanling diarrhoea and its causes. Thirty-six (23%) of the 156 weanlings had diarrhoea in 2 months prior to the interview. The factors that were significantly related to reported weanling diarrhoea included consumption of unboiled water by weanlings (OR = 10, p = 0.03), not covering perishable foods (OR = 3, p = 0.02), and washing feeding utensils of weanlings without dishwashing detergent (OR = 3.1, p = 0.02), 'Su' and 'tongsia'--two common local terms--were used for describing different types of weanling diarrhoea. Many caregivers considered 'su' a natural occurrence in a child's development. The results suggest that some poor feeding practices may contribute to the higher risk of weanling diarrhoea in northeast Thailand. Some local beliefs about weanling diarrhoea may mask the true causes, and mislead messages about its prevention.

A community-based seroepidemiologic survey on Taenia solium cysticercosis in humans was carried out in Shandong Province, China. Blood specimens from 2,898 residents were collected and examined for anti-cysticercus antibody. Information on demographic and potential risk factors was obtained using a standardized questionnaire. The overall seroprevalence of cysticercosis was 3.2%. Seropositivity tended to increase with age ranging from 1.8% in children under 6 years of age to 5.7% in those over 60 years old. Distance between village residence and the town of the community was negatively associated with seropositivity (Chi-square for trend test p = 0.02). Multivariate logistic regression analysis identified four risk factors for cysticercosis infection in the community: defecating indiscriminately (OR = 1.35, 95% CI 1.01-1.81), being unable to identify diseased pork (OR = 4.09, 95% CI 1.53-10.97), raising pigs (OR = 1.36, 95% CI 1.09-1.69), and more than 60 years old (OR = 1.49, 95% CI 1.10-2.02. These findings have implications for developing appropriate strategies for the control of Taenia solium cysticercosis in the community.
