RÉSUMÉ

ObjectiveTo explore the pharmacodynamic effect of gramine on 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis (AD) in mice and its potential mechanism. MethodThe mice were divided into the normal control group, model group, dexamethasone (0.05 g·kg-1) group, and high- and low-dose (0.12,0.06 g·kg-1) gramine groups. Mice in all groups except for the normal control group were stimulated with DNCB, followed by medication 13 d later. The changes in skin lesions were then observed, and the skin thickness, moisture content, and transepidermal water loss (TWEL) in each group were measured. The pathological changes in skin lesions were observed by hematoxylin-eosin (HE) staining, and the effects of drugs on CD4+/CD8+T-cell ratio in the spleen were detected by flow cytometry. The levels of immunoglobulin E (IgE), interleukin (IL)-4, and IL-6 in serum were detected by enzyme-linked immunosorbent assay (ELISA), and the changes in serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), and creatinine (CRE) by microplate method. The mRNA expression levels of inflammatory cytokines γ-interferon(IFN-γ), IL-13, IL-17, IL-1β, IL-6, and tumor necrosis factor-α (TNF-α) in skin lesions were assayed by real-time polymerase chain reaction (Real-time PCR), and the protein expression levels of nuclear transcription factor -κB (NF-κB) and NF-κB inhibitory protein α (IκBα) in skin lesions by Western blot. ResultCompared with the normal control group, the model group showed skin edema, erythema, scab, scratch, and lymphocyte and neutrophil infiltration, decreased skin moisture content, as well as increased skin thickness, TWEL (P<0.01), spleen index, CD4+/CD8+ T-cell ratio in the spleen (P<0.05), mRNA expression of IFN-γ, IL-13, IL-17, IL-1β, IL-6, and TNF-α in the skin lesions (P<0.05), serum contents of IgE, IL-4, and IL-6 (P<0.05), and protein expression of IκBα and NF-κB in skin lesions (P<0.05). Compared with the model group, dexamethasone and gramine at different doses alleviated skin erythema, scale, scab, and inflammatory cell infiltration, elevated skin moisture content, inhibited skin thickening and TWEL, and decreased spleen index, CD4+/CD8+T-cell ratio in the spleen, mRNA expression of inflammatory factors in the skin lesions, serum contents of IgE and inflammatory factors, and protein expression of IκBα and NF-κB in skin lesions, especially in the dexamethasone group and the high- dose gramine group(P<0.05,P<0.01). ConclusionGramine can inhibit the expression of related inflammatory factors and regulate the immune function of AD mice via the IκBα/NF-κB pathway, enabling it become a potential drug for treating AD.

ObjectiveIn order to establish a systematic quality evaluation system for Fritillariae Cirrhosae Bulbus adulteration, portable near-infrared (NIR) spectroscopy was used to identify Fritillariae Cirrhosae Bulbus and its adulterants and detect their adulteration quantity. MethodA total of 72 batches of Fritillariae Cirrhosae Bulbus samples were collected and 570 batches of adulterated products (dry bulbs of Fritillaria thunbergii, F. ussuriensis, F. pallidiflora and F. hupehensis, Bulbus Tulipae, flour) were prepared, NIR spectral data of samples were collected by the portable NIR spectrometer. Linear discriminant analysis (LDA) was used to establish the qualitative correction models of Fritillariae Cirrhosae Bulbus-adulterants and adulterants of different categories, partial least squares (PLS) was used to establish the quantitative correction models of adulteration quantity of different kinds of adulterants. ResultThe recognition rates of qualitative analysis model of Fritillariae Cirrhosae Bulbus and its adulterants were 99.49% (calibration set) and 100.00% (validation set), respectively. In different adulterant models, the recognition rates of calibration set and validation set were 70.47% and 73.68%, respectively. Moreover, the correlation coefficients of validation set (R2P) of the six quantitative models of adulteration ratio were 0.840 2 (Fritillariae Cirrhosae Bulbus adulterated with F. thunbergii dry bulbs), 0.960 2 (Fritillariae Cirrhosae Bulbus adulterated with F. ussuriensis dry bulbs), 0.765 7 (Fritillariae Cirrhosae Bulbus adulterated with F. pallidiflora dry bulbs), 0.902 5 (Fritillariae Cirrhosae Bulbus adulterated with F. hupehensis dry bulbs), 0.957 4 (Fritillariae Cirrhosae Bulbus adulterated with Bulbus Tulipae), 0.976 1 (Fritillariae Cirrhosae Bulbus adulterated with flour), the root mean square error of prediction (RMSEP) were 10.948 5, 5.463 9, 13.256 4, 8.549 2, 5.655 3, 4.235 6, respectively. The two qualitative models and six quantitative models showed good prediction performance. ConclusionThe portable NIR spectroscopy can be used to identify Fritillariae Cirrhosae Bulbus and its adulterants in real time, the method is rapid and accurate, which can meet the requirements of nondestructive identification of Fritillariae Cirrhosae Bulbus on site.

The processing of Chinese herbal medicine is a form of pharmaceutical technology developed over thousands of years, in order to increase efficiency and decrease toxicity of herbs in traditional Chinese medicine (TCM). Herbal processing is essential for safe and effective application of TCM in clinical practice, as it alters the active chemical components and therefore the functions of herbal medicines. Alkaloid-rich herbal medicines in TCM are commonly processed by cleansing, cutting, processing by dry stir-frying, stir-frying with liquid adjuvants, and processing by water decoction. In addition, commonly used adjuvants for processing alkaloid-rich herbal medicines are river sand, wine, vinegar, brine, honey and herbal juice. For alkaloid-rich herbal medicines, the main chemical reactions that occur during processing include hydrolysis, oxidation, replacement, decomposition and condensation. This paper aimed to summarize the processing methods and mechanisms for alkaloid-rich Chinese herbal medicines, and provide much-needed theoretical support and scientific evidence for understanding those mechanisms and effects. Information on processing methods for alkaloid-rich herbal medicines was collected from classic books of herbal medicine, PhD and MSc dissertations, online scientific databases including PubMed, SciFinder, Scopus, Web of Science, Baidu Scholar and Google Scholar. This paper should help to advance our knowledge of the processing mechanisms and aid in the development of processing methods for alkaloid-rich Chinese herbal medicines.

Objective:To propose a new method for detecting and evaluating traditional Chinese medicine (TCM) by artificial intelligence and machine vision technology. Method:Taking Fritillariae Cirrhosae Bulbus, Crataegi Fructus and Pinelliae Rhizoma as the research objects, big data of pictures was collected by machine vision and the image database was established. Through the intelligent analysis of the external characteristics of TCM, the deep convolutional neural network model was established to realize the functions of location detection and variety identification by means of deep learning, so as to significantly improve the accuracy of rapid identification of TCM. Result:The classification accuracy of 11 kinds of Chinese herbal pieces (raw, fried, parched and charred products of Crataegi Fructus, Pinelliae Rhizoma, Pinelliae Rhizoma Praeparatum Cum Zingibere et Alumine, Pinelliae Rhizoma Praeparatum, Pinelliae Rhizoma Praeparatum Cum Alumine and three products of Fritillariae Cirrhosae Bulbus) could be more than 99%, and the average recognition accuracy of specific categories could reach more than 97%. Conclusion:The intelligent identification technology of TCM decoction pieces realized by deep learning algorithms has the advantages of simplicity, rapidity, high precision and quantifiable detection, which can provide technical support for the quality detection and evaluation of TCM, and enrich the research ideas of quality evaluation of TCM.

Objective:To discuss the effect of Arisaema Rhizoma and Pinelliae Pedatisectae Rhizoma on water metabolism and hemorheolog in rats. Method:A total of 50 healthy SD rats were randomly divided into 5 groups, namely normal group, high-dose Arisaema Rhizoma group (1.2 g·kg·d-1), low-dose Arisaema Rhizoma group (1.2 g·kg·d-1), high-dose Pinelliae Pedatisectae Rhizoma group (1.2 g·kg·d-1) and low dose Pinelliae Pedatisectae Rhizoma group (0.3 g·kg·d-1), with 10 rats in each group. The effects of Arisaema Rhizoma and Pinelliae Pedatisectae Rhizoma on water metabolism and hemorheology were compared, with the micturition index, fecal volume, fecal water content, pathological morphology of submandibular gland, aquaporin (AQP2) content in kidney, adenosine cyclic 3′,5′-(hydrogen phosphate)/current good manufacture practices (cAMP/cGMP) and Na+-K+-ATP enzyme content in serum as the evaluation indexes of water metabolism, and the whole blood viscosity (WBV) and erythrocyte aggregation index (RE) as the evaluation indexes of hemorheolog. Result:Compared with normal group, the fecal volume, fecal water content and AQP2 expression decreased significantly, acinus of submandibular gland were atrophied, micturition index, cAMP/cGMP and Na+-K+-ATP enzyme content in serum were increased significantly, WBV and RE were increased significantly in high-dose Arisaema Rhizoma and Pinelliae Pedatisectae Rhizoma groups(P<0.05), but with no significant difference between them. Conclusion:High-dose Arisaema Rhizoma and Pinelliae Pedatisectae Rhizoma groups had significant effects on water metabolism and hemorheology in rats, suggesting that attention shall be given to the safety of Arisaema Rhizoma and Pinelliae Pedatisectae Rhizoma. And this study provides a theoretical basis for the rational use of the two traditional Chinese medicines in clinic.

In recent years, near-infrared spectroscopy has developed into an analytical technique widely used in various fields. Because of its advantages of fast, green and non-destructive, it plays an increasingly prominent role in the field of traditional Chinese medicine (TCM) analysis. However, due to the complexity and overlap of spectra, near-infrared spectroscopy needs to be combined with chemometrics for analysis and calculation. The principle, application scope, advantages and limitations of near infrared spectroscopy and chemometrics are summarized in detail, in addition, their combined applications in the identification of the origin, authenticity, processed products, composition prediction and water content detection of TCM are reviewed. The authors discussed and analyzed the joint application of near-infrared spectroscopy and chemometrics in the field of TCM analysis, and summarized the unique advantages of the combined technology in the field of TCM, which had certain guiding significance for medical workers to better use this technology.

Objective: To observe the effect of Pinelliae Rhizoma Praeparatum Cum Alumine polysaccharides(PRPCAP)on airway mucus secretion in rats with allergic asthma, in order to study the material basis of the "macromolecule" component of the polysaccharides as the original medicinal materials. Method: The 60 SPF-grade Wistar rats were induced by intraperitoneal injection of chicken ovalbumin (OVA) and aluminum-magnesium adjuvant, except for the normal control group. The OVA solution was aerosolized to establish a rat model of allergic asthma. After successful modeling, the rats were randomly divided into 5 groups, namely allergic asthma model group, positive drug group (montalurast sodium,5 mg·kg-1), high-dose PRPCAP group (400 mg·kg-1), middle-dose PRPCAP group (200 mg·kg-1) and low-dose PRPCAP group (100 mg·kg-1). The contents of interleukin-4 (IL-4) and interferon-γ (IFN-γ) in serum and bronchoalveolar lavage fluid (BALF) supernatant were determined by enzyme-linked immunosorbent assay (ELISA), and the count of eosinophils (EOS) was detected by BALF sediment. The histopathological changes were observed by hematoxylin-eosin (HE) staining in lung tissue. The mRNA expression of mucin 5AC (MUC5AC) was detected by Real-time fluorescent quantitative polymerase chain reaction (Real-time PCR). Result: Compared with the normal control group, serum IL-4 level in the allergic asthma model group was significantly increased (Pγ level was significantly decreased (PPPPPγ in serum (PPPPConclusion: The "macromolecule" component of polysaccharides in the Pinelliae Rhizoma Praeparatum Cum Alumine may be the material basis for the efficacy of eliminating dampness and eliminating phlegm.

AIM To analyze the optimal "Huohou" (fermentation time and temperature) of fermentation process of Jianqu Leaven [fermentation product composed of medicinal materials (Artemisiae annuae Herba,Menthae haplocalycis Herba,Angelicae dahuricae Radix,etc.),wheat bran and flour).METHODS The colors and odors of twelve batches of samples during fermentation process were detected by machine vision and electronic nose technologies,respectively.The contents of total sugar and total protein,together with activities of amylase,protease and lipase,were determined to analyze constituent changes.RESULTS The content of total sugar was significantly decreased,but that of total protein was slightly increased,and the activities of three enzymes were increased to different degrees.At temperature of 28-30 ℃ and relative humidity of 70%-80%,the optimal fermentation terminal point of Jianqu Leaven was 30-36 h.CONCLUSION The Jianqu Leaven obtained by this fermentation method is totally covered with white mould,and has wine flavor,which meets the requirement of traditional processing.

To study the variation of six ester-type alkaloids and characteristic fingerprints in the process from Radix Aconite Lateralis to Heishunpian and lay a foundation for the study of the processing principle of Heishunpian, HPLC. analysis was performed on a Phenomenex Gemini C18 (4.6 mm x 250 mm, 5 microm) with acetonitrile and 40 mmol x L(-1) ammonium acetate (adjusted to pH 10 with concentrated ammonia water) as mobile phase. The detection wavelength was set at 235 nm. The flow rate was set at 0.8 mL x min(-1) and the injection volume was 10-20 microL. Six ester-type alkaloids were determined and characteristic fingerprints of the process were established. As the process continues, the contents of diester diterpene alkaloids were decreased step by step, while the contents varia tion of monoester diterpene alkaloids were not obvious. Each sample showed significant difference in characteristic fingerprints. With the exception of 6 known monoester diterpene alkaloids and diester diterpene alkaloids, 13 peaks were marked in the characteristic fingerprints, of which the total change rule of the other 7 unknown peaks were similar with 3 diester diterpene alkaloids. The established method is accurate, reliable and repeatable, and can provide reference for revealing change rule of index components and illuminating processing principle in the process of Heishunpian.

RÉSUMÉ

Though parched Chinese herbal medicines contain less effective or index components, their pharmacological actions do not reduce or even become improved to some extent. However, the current studies related to material basis could not explain the changes in property, flavour and efficacy of parched Chinese herbal medicines. Meanwhile, due to the lack of objective and specific evaluation indexes, the quality evaluation could not reflect features of parched Chinese herbal pieces. Therefore, how to break the bottleneck for the studies on parched Chinese herbal pieces, make further innovation and conduct in-depth studies on the material basis of parched Chinese herbal medicines are common problems that medical scholars are facing. According to the findings in the previous studies, the author proposed to explain the material basis of parched Chinese herbal medicines by studying Maillard reaction and establish specific quality evaluation indexes according to the features of parched Chinese herbal pieces, and conducted relevant studies.
