Résumé

Immature hematopoietic progenitors are a constant source for renewal of hemocyte populations and the basic component of the tissue and cell repair apparatus. A unique property of these cells of internalizing extracellular double-stranded DNA has been previously shown. The leukostimulatory effect demonstrated in our pioneering studies was considered to be due to the feature of this cell. In the present research, we have analyzed the effects of DNA genome reconstructor preparation (DNAgr), DNAmix, and human recombinant angiogenin on both hematopoietic stem cells and multipotent progenitors. Treatment with bone marrow cells of experimental mice with these preparations stimulates colony formation by hematopoietic stem cells and proliferation of multipotent descendants. The main lineage responsible for this is the granulocyte-macrophage hematopoietic lineage. Using fluorescent microscopy as well as FACS assay, co-localization of primitive c-Kit- and Sca-1-positive progenitors and the TAMRA-labeled double-stranded DNA has been shown. Human recombinant angiogenin was used as a reference agent. Cells with specific markers were quantified in intact bone marrow and colonies grown in the presence of inducers. Quantitative analysis revealed that a total of 14,000 fragment copies of 500 bp, which is 0.2% of the haploid genome, can be delivered into early progenitors. Extracellular double-stranded DNA fragments stimulated the colony formation in early hematopoietic progenitors from the bone marrow, which assumed their effect on cells in G0. The observed number of Sca1+/c-Kit+ cells in colonies testifies to the possibility of both symmetrical and asymmetrical division of the initial hematopoietic stem cell and its progeny.

Abstract In advanced biotechnology, the utilization of enzymes to achieve new or modified compounds with antibacterial, fungicidal, and anti-cancer specifications is crucial. Mushroom lactases are a hopeful biocatalyst for the synthesis and modification of different compounds. They are an accessible and inexpensive enzyme for the preparation of reaction objects and have recently received attention. Laccase purification was performed from basidiomycete Lentinus strigosus (LS) in several stages: Stage 1. On ion-exchange chromatography on TEAE Servacell 23 (400 ml), two distinctly separated laccase activity peaks were observed, eluted from the carrier at 0.21 and 0.27 M NaCl. In order to reduce the loss of enzymes, all fractions with laccase activity were collected, concentrated, and desalted using an ultrafiltration cell (Amicon, United States) with a UM-10 membrane. Stage 2. The resulting preparation with laccase activity was applied to a Q-Sepharose column (60 ml). Two well-separated peaks with laccase activity were obtained during the elution: laccase I (0.12 M NaCl) and laccase II (0.2 M NaCl). Stage 3. In the course of further purification of both enzymes, carried out on anion-exchange carrier Resource Q (6 ml), a broken gradient was used: 0 - 10%, 10 - 20%, and 20 - 100% with 1M NaCl. Stage 4. Both laccase I and laccase II, obtained after Resource Q, were desalted, concentrated to 1 ml each, and applied to a Superdex 75 gel filtration column. As a result, two laccases were obtained in a homogeneous form.


Resumo Na biotecnologia moderna, o uso de enzimas para obter compostos novos ou modificados com propriedades antibacterianas, antifúngicas e anticancerígenas é crucial. Lactases de cogumelos são biocatalisadores promissores para síntese e modificação de diferentes compostos, por serem enzimas baratas e disponíveis para a preparação de componentes de reação, e vem recebendo a devida atenção recentemente. A purificação da lacase foi realizada a partir do basidiomiceto Lentinus strigosus em vários estágios: Etapa 1 - na cromatografia de troca iônica em TEAE Servacell 23 (400 ml), foram observados dois picos de atividade da lacase distintamente separados, com eluição do transportador a 0,21 e 0,27 M de NaCl. Para reduzir a perda de enzimas, todas as frações com atividade de lacase foram coletadas, concentradas e dessalinizadas em uma célula de ultrafiltração (Amicon, Estados Unidos) com membrana UM-10; Etapa 2 - a preparação resultante com atividade de lacase foi aplicada a uma coluna Q-Sepharose (60 ml). Durante a eluição, foram obtidos dois picos bem separados com atividade de lacase: lacase I (NaCl 0,12 M) e lacase II (NaCl 0,2 M); Etapa 3 - no decurso da purificação adicional de ambas as enzimas, realizada no Recurso Q de transportador de troca aniônica (6 ml), um gradiente quebrado foi usado: 0-10%, 10-20% e 20-100% com NaCl 1M; Etapa 4 - tanto a lacase I como a lacase II, obtidas após o Recurso Q, foram dessalinizadas e concentradas para 1 ml cada e aplicadas a uma coluna de filtração em gel Superdex 75. Como resultado, duas lacases foram obtidas de forma homogênea.

The aim of this study was to determine how sacubitril/valsartan compared with valsartan in an outpatient setting affects left ventricular remodeling in heart failure with reduced ejection fraction and functional (or secondary) mitral regurgitation (SMR) due to the effect of dual inhibition of the renin-angiotensin system and neprilysin. The outpatient study included 90 patients with chronic SMR who were followed up for 12 months. They received sacubitril/valsartan or valsartan instead of the more commonly used angiotensin-converting enzyme inhibitor enalapril for heart failure, in addition to standard medical therapy for heart failure. The difference in NT-proBNP change between groups was the primary endpoint. Changes in effective regurgitation orifice area, left ventricular ejection fraction, left ventricular end-systolic and end-diastolic volume indices, left atrial volume index, E/e' index, and exercise tolerance on the 6-minute walk test were secondary endpoints. In the treatment efficacy analysis, NT-proBNP levels decreased significantly by 37% in the sacubitril/valsartan group and by 11% in the valsartan group (P<0.001). Ejection fraction and exercise tolerance (increase in walking distance in the 6-min test) increased in the sacubitril/valsartan group (P<0.05). Also, in this group, the effective area of the regurgitation orifice, the left atrial volume index, the E/e' index, and the indices of the end-systolic and end-diastolic volume of the left ventricle (P<0.05) decreased more pronouncedly. Compared with valsartan, treatment with sacubitril/valsartan led to a significant improvement in cardiac remodeling in patients with SMR and heart failure with reduced ejection fraction.

Aims: The objective of this work was to analyze the anti-inflammatory, cytotoxic and antioxidant activity of the fractions of ethyl acetate from the leaves of D. pubipetala. Methodology: Liquid-liquid partitioning was performed, the ethyl acetate partition was eluted in classical liquid chromatography, followed by thin layer chromatography. Antioxidant activity was performed by the DPPH photochlorimetric method and the levels of interleukin (IL?6), tumor necrosis factor ? (TNF??), interleukin-10 (IL-10) and nitric oxide (NO) were identified in supernatants of cell cultures. Results: Fractions F4, F5 and F10 showed antioxidant activity. The F10 fraction indicated efficient anti-inflammatory activity by the production of IL-10 and nitric oxides. Fractions F4, F5 and F9 stimulated the production of IL-6 and TNF-?. Conclusion: These results indicate a possible synergistic action of the extract and the partition of ethyl acetate, since after fractionation the potential presented in previous studies was lower.

Flavoring additives are of great technological importance for the food industry. However, there is little information regarding the toxicological properties of these micro-ingredients, especially at the cellular level. The present study used meristematic root cells of Allium cepa L. to evaluate the toxicity of a liquid, aroma and flavor synthetic chocolate additive, manufactured and widely marketed throughout Brazil and exported to other countries in South America. The flavoring concentrations evaluated were 100.00; 50.00; 25.00; 1.00; 0.50 and 0.25 µL/L, where the highest concentration established was one-hundred times lower than that commercially suggested for use. The concentration 100 µL/L substantially reduced cell division of meristems within 24- and 48-hours exposure. Concentrations from 100.00 to 0.50 µL/L resulted in a significant number of prophases to the detriment of the other phases of cell division, indicating an aneugenic activity, and induced a significant number of cellular changes, with emphasis on micronuclei, nuclear buds and chromosomal breaks. Under the established analysis conditions, with the exception of concentration 0.25 µL/L, the flavoring of chocolate caused cytotoxicity, genotoxicity and mutagenicity to root meristems.

Contudo, poucas são as informações quanto as propriedades toxicológicas desses microingredientes, especialmente, em nível celular. No presente estudo avaliou-se, sobre as células meristemáticas de raízes de Allium cepa L., a toxicidade de um aditivo sintético líquido de aroma e sabor de chocolate, fabricado e amplamente comercializado em todo Brasil, e exportado para outros países da América do Sul. As concentrações de aromatizante avaliadas foram 100,00; 50,00; 25,00; 1,00; 0,50 e 0,25 µL/L, onde a maior concentração estabelecida foi cem vezes menor que a sugerida comercialmente para uso. Com base na interpretação dos resultados, a concentração 100 µL/L reduziu substancialmente a divisão celular dos meristemas nas 24 e 48 horas de exposição. Códigos da Classificação Internacional de Doenças como Rastreadores de Eventos Adversos a MedicamentosARTIGO DE REVISÃOJosé Romério Rabelo Melo1,2 , Elisabeth Carmen Duarte,3 Silvia Maria de Freitas4 , Eduardo Gabriel Pinheiro5 , Eudiana Vale Francelino6,7 , Paulo Sergio Dourado Arrais7 Introdução: Os Eventos Adversos a Medicamentos-EAM representam riscos à saúde e sua subnotificação representa um desafio para a saúde pública. A busca ativa de casos suspeitos de EAM nos bancos de dados de saúde utilizando a Classificação Internacional de Doenças-CID é uma das estratégias que pode reduzir as subnotificações desses eventos. Objetivo: O objetivo desse estudo é identificar os códigos da CID mais usados como rastreadores de EAM e avaliar a sua concordância entre os pesquisadores. Métodos: Foi realizada uma revisão sistemática da literatura utilizando as bases de dados PubMed, Scopus, Web of Science, MEDLINE e LILACS com os descritores "Classificação Internacional de Doenças", "CID-10", "Efeitos Colaterais e Reações Adversas Relacionados a Medicamentos", "Envenenamento", "Erros de Medicação". Os artigos incluídos tiveram seus códigos CID identificados, comparados e sua qualidade avaliada. A análise de concordância dos códigos foi feita usando o modelo de ensaios de Bernoulli, testes de proporções binomial exata e a técnica de false discovery rate para analisar as hipóteses postas. A análise estatística foi feita com o software R. O estudo está registrado no PROSPERO sob n.º CRD42019120694. Resultados: Foram identificados 5.167 artigos e após os critérios de seleção, 33 foram incluídos nessa revisão. Foram identificados 1.105 códigos da CID. O coeficiente de prevalência dos EAM variou entre 0,18% e 18,4% em internações hospitalares e a taxa de mortalidade variou entre 0,12 a 45,9 óbitos por 100 mil óbitos. Somente 195 (17,7%) códigos tiveram alta concordância entre os pesquisadores. Muitos códigos CID utilizados para detectar EAM possuem baixa concordância entre os pesquisadores e produziram diferentes taxas do evento. Conclusão: Os códigos rastreadores de EAM identificados representam um método simples e eficiente para captação de eventos adversos em grandes bancos de dados em saúde, contribuindo na redução da subnotificação nos tradicionais sistemas de notificações de EAM.Palavras-chave: Informática médica, Farmacoepidemiologia, Segurança do paciente, Classificação internacional de doenças, Efeitos colaterais e reações adversas relacionadas a medicamentos. The active search for suspected cases of ADE in health databases using the International Classification of Diseases-CID is one of the strategies that can reduce underreporting of these events. Objective: The aim of this study is to identify the ICD codes most commonly used as tracers of ADE and to assess their concordance among researchers. Methods: A systematic literature review was conducted using the PubMed, Scopus, Web of Science, MEDLINE and LILACS databases with the descriptors "International Classification of Diseases", "ICD-10", "Drug-Related Side Effects and Adverse Reactions", "Poisoning", "Medication Errors". The included articles had their ICD codes identified, compared and their quality assessed. The analysis of concordance of the codes was done using Bernoulli's test model, exact binomial proportions tests and the false discovery rate technique to analyze the hypotheses posed. Statistical analysis was done using R software. The study is registered in PROSPERO under CRD42019120694. Results: A total of 5,167 articles were identified and after the selection criteria, 33 were included in this review. A total of 1,105 ICD codes were identified. The prevalence coefficient of ADEs ranged from 0.18% to 18.4% in hospital admissions and the mortality rate ranged from 0.12 to 45.9 deaths per 100,000 deaths. Only 195 (17.7%) codes had high concordance among researchers. Many ICD codes used to detect ADEs have low inter-rater concordance and produced different event rates. Conclusion: The identified ADE tracking codes represent a simple and efficient method for capturing adverse events in large healthcare databases, contributing to the reduction of underreporting in traditional ADE reporting systems. The objective of this study was to estimate genetic parameters and genetic trends of different conformation and management traits regularly measured within the context of the National Dairy Gir Breeding Program (PNMGL). The estimation of genetic and residual variances for each trait was performed using average information restricted maximum likelihood (AI-REML) procedure in AIREMLF90 program software. The population was divided into three subpopulations constituted by measured females (with phenotype records), all females, and males. Linear regressions were applied for each trait, considering two periods of birth (1st period: 1938-1996; 2nd period: 1997-2012). The estimated heritability of conformation and management traits varied from 0.01 to 0.53, denoting a perspective of genetic improvement through selection and corrective matings for purebred Dairy Gir populations. The average genetic changes in conformation and management traits were, in general, variable and inexpressive, showing that the selection of Dairy Gir may have had been directed essentially to increase milk yield. The analysis of the two periods of birth indicated that some linear traits present progress (although inexpressive) in the 2nd period (more recent period).(AU)


O objetivo deste estudo foi estimar os parâmetros genéticos e as tendências genéticas para diferentes características de conformação e manejo de animais puros da raça Gir Leiteiro, pertencentes ao Programa Nacional de Melhoramento do Gir Leiteiro (PNMGL). A estimativa das variâncias genéticas e residuais para cada característica foi realizada usando-se o procedimento de máxima verossimilhança restrita (AI-REML), por meio do programa AIREMLF90. A população foi dividida em três subpopulações, constituídas por fêmeas mensuradas (com registros de fenótipo), todas fêmeas e machos. As regressões lineares para cada característica foram ainda divididas em dois períodos de anos de nascimento (1º período: 1938 a 1996; 2º período: 1997 a 2012). As herdabilidades estimadas variaram de 0,01 a 0,53 para as características de conformação e manejo, possibilitando a perspectiva de melhoramento mediante seleção e acasalamentos corretivos na população pura da raça Gir Leiteiro. As mudanças genéticas nas características conformação e manejo foram, em geral, variáveis e inexpressivas, sugerindo que a seleção no Gir Leiteiro possa ter sido direcionada essencialmente para maior produção de leite. Ao serem observados os dois períodos distintos de anos de nascimento, infere-se que algumas características lineares apresentaram progresso (embora inexpressivo) no 2º período analisado.(AU)

Introducción: Las infecciones por SARS-CoV-2 representan un problema de salud pública a nivel mundial. En los niños se reporta menor incidencia y cuadros clínicos más leves. Se realizó el presente estudio con el objetivo de describir las características clínicas y evolutivas de los niños con diagnóstico de infección por SARS CoV-2 en el Hospital Juan P. Garrahan. Material y métodos: estudio de cohorte prospectivo. Se incluyeron todos los pacientes con diagnóstico confirmado por PCR de COVID-19 desde 20.4.20 hasta el 3.07.21 y con seguimiento en el hospital de Pediatría Juan P. Garrahan. Resultados: n: 1644. Eran varones 836 (51%). La mediana de edad fue 75 meses (RIC 22- 143). Tenían alguna enfermedad de base previa al diagnóstico de COVID-19: 884 pacientes (53,7%), la más frecuente fue la enfermedad oncohematológica. Estaban asintomáticos 423 pacientes (25,7%). De los pacientes sintomáticos, 1071 (65,1%) presentaron cuadro leve, 5 (0,3%) moderado, 69 (4,2%) grave y 76 (4,6%) crítico. La fiebre fue el hallazgo más frecuente n: 782; (47,5%). Se internaron 900 pacientes (54,7%), 33 en UCI (2%). Fallecieron 7 pacientes (0,4%), todos ellos con comorbilidades graves. Conclusiones: En este estudio de cohorte de niños con infección por SARS-CoV-2 confirmada, predominaron los pacientes con enfermedad de base y las formas leves de COVID-19. El ingreso a UCI fue menor al 2%. Fallecieron 7 pacientes (0.4%) todos ellos con comorbilidades y coinfecciones (AU)


Introduction: SARS-CoV-2 infections represent a worldwide public health problem. A lower incidence and milder clinical pictures are reported in children. The aim of this study was to describe clinical and outcome characteristics of children diagnosed with SARS-CoV-2 infection at Hospital de Pediatría Juan P. Garrahan. Methods: A prospective cohort study was conducted. All patients with a PCR-confirmed diagnosis of COVID-19 seen between 20.4.20 and 3.07.21 and followed-up at Hospital de Pediatría Juan P. Garrahan were included. Results: n: 1644; 836 males (51%) were male. Median age was 75 months (IQR, 22-143). Overall, 884 patients (53.7%) had an underlying disease prior to COVID-19 diagnosis, most frequently hematologic/ oncologic disease. 423 patients (25.7%) were asymptomatic. Of the symptomatic patients, 1071 (65.1%) had mild, 5 (0.3%) moderate, 69 (4.2%) severe, and 76 (4.6%) critical disease. Fever was the most frequent finding (n: 782; 47.5%). A total of 900 patients (54.7%) were admitted, 33 of whom to the ICU (2%). Seven patients (0.4%) died, all with severe comorbidities. Conclusions: In this cohort study of children with confirmed SARSCoV-2 infection, patients with underlying disease and mild forms of COVID-19 predominated. ICU admission occurred in less than 2%. Seven patients (0.4%) died, all of them with comorbidities and coinfections. (AU)

Introducción: según estudios previos en México señalan que la esperanza de vida era de 75.5 años y de acuerdo con nuevos datos, se estima que esta aumentara a 77.8 años para 2030¹, por lo cual es necesario fortalecer y fomentar el autocuidado en el adulto mayor, mediante el apoyo educativo, debido a la pandemia a través del uso de las tecnologías de la información y la comunicación (TICs) con la finalidad de aumentar su nivel de calidad de vida. Objetivo: evaluar el nivel de calidad de vida de los adultos mayores del Valle del Mezquital antes y después del apoyo educativo para fomentar el autocuidado mediante el uso de las TICs. Material y métodos: investigación cuantitativa, nivel de alcance exploratorio, con un diseño metodológico experimental, prolectivo longitudinal prospectivo. Se ocupó la prueba estadística paramétrica t de student. Resultados: de acuerdo al instrumento WHOQOL-BREF preintervención se obtuvo que el 80% de la población reflejo una calidad de vida media, posterior a un mes de intervención 67 % de la población reflejo calidad de vida alta, de acuerdo con la prueba estadística t se obtuvo una significan la bilateral de .041 posterior a la intervención. Conclusiones: la calidad de vida de los adultos mayores mejoró gracias a las intervenciones educativas ofrecidas médiate las TICs logrando así una nueva estrategia de acercamiento en tiempos de pandemia demostrando que estas herramientas ayudan a mejorar el autocuidado.


Introduction: according to previous studies in Mexico indicate that life expectancy was 75.5 years and according to new data, it is estimated that this will increase to 77.8 years by 2030, so it is necessary to strengthen and encourage self-care in the elderly, through educational support, due to the pandemic through the use of Information and communication technologies (ICTs) to increase their quality of life. Objective: to assess the quality of life of older adults in the Mezquital Valley before and after educational support to promote self-care through the use of ICTs. Material and methods: basic research with a quantitative approach, exploratory scope level, with an experimental methodological design, prospective longitudinal prolective. The parametric statistical test of the student was taken. Results: according to the instrument WHOQOL-BREF pre intervention was obtained that 80 % of the population reflected an average quality of life, and after a month of intervention was obtained 67 % of the population reflected high quality of life, according to the statistical test was obtained a bilateral mean of .041 after the intervention. Conclusions: the quality of life of older adults improved thanks to the educational interventions offered by ICTs, thus achieving a new approach strategy in times of pandemic, demonstrating that these tools help to improve self-care

Tracheal fungal infections in horses are rare. This case report describes surgical and clinical management of a filly with a Curvularia sp. infection within the trachea and skin that caused severe intraluminal granulomas and cutaneous nodules, respectively. The patient was successfully treated with itraconazole and surgical excision.(AU)

OBJECTIVE: To assess the possible factors that influence sleep quality in adolescents with and without chronic immunosuppressive conditions quarantined during the coronavirus disease 2019 (COVID-19) pandemic. METHODS: This cross-sectional study included 305 adolescents with chronic immunocompromised conditions and 82 healthy adolescents. Online surveys were completed, which included questions on socio-demographic data and self-rated healthcare routine during COVID-19 quarantine and the following validated questionnaires: the Pittsburgh Sleep Quality Index (PSQI), Pediatric Quality of Life Inventory 4.0 (PedsQL4.0), and Pediatric Outcome Data Collection Instrument (PODCI). RESULTS: The median current age [14 (10-18) vs. 15 (10-18) years, p=0.847] and frequency of female sex (62% vs. 58%, p=0.571) were similar in adolescents with chronic conditions compared with healthy adolescents. The frequency of poor sleep quality was similar in both groups (38% vs. 48%, p=0.118). Logistic regression analysis, including both healthy adolescents and adolescents with chronic conditions (n=387), demonstrated that self-reported increase in screen time (odds ratio [OR] 3.0; 95% confidence interval [CI] 1.3-6.8; p=0.008) and intrafamilial violence report (OR 2.1; 95% CI 1.2-3.5; p=0.008) were independently associated with poor sleep quality in these adolescents. However, the PODCI global function score was associated with a lower OR for poor sleep quality (OR 0.97; 95% CI 0.94-0.99; p=0.001). Further logistic regression, including only adolescents with chronic conditions (n=305), demonstrated that self-reported increase in screen time (OR 3.1; 95% CI 1.4-6.8; p=0.006) and intrafamilial violence report (OR 2.0; 95% CI 1.2-3.4; p=0.011) remained independently associated with poor quality of sleep, whereas a lower PODCI global function score was associated with a lower OR for sleep quality (OR 0.96; 95% CI 0.94-0.98; p<0.001). CONCLUSION: Self-reported increases in screen time and intrafamilial violence report impacted sleep quality in both healthy adolescents and those with chronic conditions. Decreased health-related quality of life was observed in adolescents with poor sleep quality.

Risk factors that determine the severity of Covid-19 have not been fully elucidated. The aim of this study was to evaluate the role of coronary artery calcification (CAC) as a risk factor for death or mechanical ventilation (MV) of patients without known heart disease infected with Covid-19. We analyzed 283 consecutive in-patients with acute respiratory symptoms with chest computed tomography (chest-CT), without previous heart disease, and criteria for Covid-19 (RT-PCR positive and/or typical clinical and chest-CT findings). CAC was classified by the number of coronary segments affected as absent (0), mild (1-3), and severe calcification (more than 3). The association between CAC, CAC severity, and death or MV due to severe respiratory failure was assessed by logistic regression. The mean age was 58.7±15.7 years and 54.1% were men. Patients with CAC were older, more likely to have hypertension, and less likely to be obese. CAC was present in 75 patients (26.5%), of which 42 had a mild calcification and 33 had severe calcification, and was associated with death (OR=2.35, 95%CI: 1.01-5.48) or MV (OR=2.72, 95%CI: 1.20-6.20) adjusted for multiple confounders, with significant and increased odds ratio for the severe form of CAC (death: OR=3.70, 95%CI: 1.20-11.42; MV: OR=3.30, 95%CI: 1.09-9.95). We concluded that CAC was an independent risk factor for death or MV in Covid-19 patients without previous heart disease, particularly for those with severe calcification. CAC can be easily visualized on common chest-CT, widely used in evaluation of moderate to severe Covid-19.

Abstract The objective of this study was to evaluate the internal morphology of seeds of Campomanesia pubescens (Mart. ex DC.) O. Berg with varying water content through X-ray image analysis, and correlate the images with the germination of these seeds. Fruits were collected in Jataí, Goiás, Brazil, and taken to the Seed Laboratory of the Federal Institute Goiano, Rio Verde Campus, where they were processed and the seeds were extracted. To establish the predetermined water content (43%, 37%, 34%, 31%, and 28% wet basis), seeds with an initial water content of 43% were kept in an oven with forced air circulation at a temperature of 40 °C until they reached the predetermined water content levels. Next, the seeds were placed in acrylic holders with double-sided tape and transported to the Federal University of Lavras (UFLA), where they were exposed to different intensities of radiation. The X-ray test was conducted with 200 seeds per treatment, and images of the internal structures of the seeds were evaluated. The seeds were returned to the Seed Laboratory of the Federal Institute Goiano, Rio Verde Campus, where the germination test was initiated. The experimental design was completely randomized, in a factorial design (5 × 4), in which there were five water content levels and four seed classes, with four replications. The X-ray test was efficient in the identification of filled, malformed, damaged and empty seeds, which germination rate above 50% in the filled seeds. This visualization of the internal morphology of the seed can be useful to select seeds of better quality, improving the percentage of germination of C. pubescens seeds.


Resumo O objetivo deste trabalho foi avaliar a morfologia interna de sementes de Campomanesia pubescens (Mart. ex DC.) O.Berg com diferentes teores de água por meio de análise de imagens de raios-X, e correlacionar as imagens com a germinação dessas sementes. Os frutos foram coletados no município de Jataí, Goiás, Brasil, e levados ao Laboratório de Sementes do Instituto Federal Goiano, Campus Rio Verde, onde foram processados ​​e as sementes foram extraídas. Para estabelecer o teor de água predeterminado (43%, 37%, 34%, 31% e 28% base úmida), as sementes com um teor inicial de água de 43% foram mantidas em estufa com circulação de ar induzida, a uma temperatura de 40 °C até atingirem os níveis de conteúdo de água predeterminados. Em seguida, as sementes foram colocadas em suportes acrílicos com fita dupla face e transportadas para a Universidade Federal de Lavras (UFLA), onde foram expostas a diferentes intensidades de radiação. O teste de raios-X foi realizado com 200 sementes por tratamento, e imagens das estruturas internas das sementes foram avaliadas. As sementes foram devolvidas ao Laboratório de Sementes do Instituto Federal Goiano, Campus Rio Verde, onde foi iniciado o teste de germinação. O delineamento experimental foi inteiramente casualizado, em esquema fatorial (5 × 4), no qual foram encontrados cinco níveis de umidade e quatro classes de sementes, com quatro repetições. O teste de raios-X foi eficiente na identificação de sementes preenchidas, malformadas, danificadas e vazias, com taxas de germinação acima dos 50% nas sementes cheias. Essa visualização das morfologia interna da semente pode ser útil para selecionar sementes de melhor qualidade, melhorando a porcentagem de germinação das sementes de C. pubescens.

Résumé

A male 15-year-old captive Siberian tiger (Panthera tigris altaica) developed pelvic limb hypermetry over the past 10 years. Recently, an ulcerated black nodule located caudally to the right ear was observed. The animal was submitted to surgery for removing the tumor, but died during anesthetic recovery. At necropsy, another infiltrative nodule was observed caudally to the right ear. Histologically, both nodules corresponded to melanocytic neoplasia, varying from heavily pigmented to amelanotic, with metastasis to mediastinal lymph nodes, spleen and lung. Lipofuscinosis and corpora amylacea were histologically observed in the central nervous system. Macroscopic and histologic findings confirmed the diagnosis of skin metastatic melanoma in a captive adult Siberian tiger.(AU)


Um tigre-siberiano (Panthera tigris altaica) de cativeiro, macho, de 15 anos de idade, apresentou hipermetria dos membros pélvicos nos últimos 10 anos, e recentemente, foi observado um nódulo preto e ulcerado caudalmente à orelha direita. O animal foi submetido à cirurgia para remoção do nódulo e morreu durante a recuperação anestésica. À necropsia, outro nódulo infiltrativo, foi detectado caudalmente a orelha direita. Histologicamente, ambos os nódulos correspondiam à neoplasia melanocítica, com células variando de fortemente pigmentadas a amelanóticas, com metástase para linfonodos mediastinais, baço e pulmão. Havia lipofucsinose e corpora amilácea no sistema nervoso central. Os achados macroscópicos e histológicos confirmam o diagnóstico de melanoma cutâneo metastático em um tigre-siberiano adulto de cativeiro.(AU)

The aim of the study was to investigate the effect of a new anti-stress drug on protein and lipid metabolism in thecultivation and fattening of pigs. The experiment was carried out on 5 groups of pigs of Irish Landrace breed (4experimental and 1 control) with 10 heads in each in the period from 60 to 210 days of age. Animals of 1, 2, 3and 4 experimental groups daily during the entire period of rearing and fattening received lithium ascorbate inthe form of powder at a dose of 10, 5, 2 and 0.5 mg/kg of live weight, respectively. Weighing was carried outbefore the introduction of the drug, for the 4th month and before slaughter. Before setting the animals in theexperiment and on the 180th day of the experiment, blood samples were taken. Triacylglycerols, total protein andglobulins, and fractions of lipoproteins were determined in blood plasma. Acting on protein and lipid metabolism,lithium ascorbate activates the functions associated with the participation of α-, β - globulins in the transport oflipids, as well as in the performance of γ-globulins protective functions. Lithium ascorbate has a positive effecton lipid-cholesterol metabolism and, as a result, contributes to the increase in live weight and quality of meat.With the introduction of feed lithium ascorbate from the 60th day prior to slaughter at a dosage of 10, 5 and 2mg/kg of body weight, lithium ascorbate exhibits a pronounced adaptogenic and stress-protection properties,prevents the accumulation of lipoproteins of low and very low density and activates the production of high-densitylipoproteins. And in the control animals, it is is observed in the reverse reaction. The use of lithium ascorbateaffects protein synthesis. Given that γ-and β-globulins serve as raw materials for the production ofimmunoglobulins, it can be noted that the animals treated with lithium ascorbate had the best opportunities forhumoral specific protection for 180 days of experience, which contributes to the increase of nonspecific immunityand resistance. The revealed effects of lithium ascorbate indicate the prospects for the development of neweffective ways to increase stress resistance, nonspecific resistance and productivity of animals using drugs basedon organic lithium salts.

Leptospirosis is a zoonotic disease caused by bacteria of the genus Leptospira, which can cause lipid changes in the erythrocyte membrane. Optical tweezers were used to characterize rheological changes in erythrocytes from patients with leptospirosis in the late stage. Biochemical methods were also used for quantification of plasma lipid, erythrocyte membrane lipid, and evaluation of liver function. Our data showed that the mean elastic constant of erythrocytes from patients with leptospirosis was around 67% higher than the control (healthy individuals), indicating that patient's erythrocytes were less elastic. In individuals with leptospirosis, several alterations in relation to control were observed in the plasma lipids, however, in the erythrocyte membrane, only phosphatidylcholine showed a significant difference compared to control, increasing around 41%. With respect to the evaluation of liver function of individuals with leptospirosis, there was a significant increase in levels of alanine transaminase (154%) and aspartate transaminase (150%), whereas albumin was 43.8% lower than control (P<0.01). The lecithin-cholesterol acyltransferase fractional activity was 3.6 times lower in individuals with leptospirosis than in the healthy individuals (P<0.01). The decrease of the erythrocyte elasticity may be related to the changes of erythrocyte membrane phospholipids composition caused by disturbances that occur during human leptospirosis, with phosphatidylcholine being a strong candidate in the erythrocyte rheological changes.
