Résumé

Purpose: IL-1 is a multifunctional proinflammatory cytokine. As the proliferative effects of IL-6 and IL-6 receptor expressions on prostatic cancer cells in response to IL-1 have not been determined, the effects of IL-1 on prostatic cancer cell lines were investigated. Materials and Methods: PC-3 and DU-145 cells were cultured in RPMI-1640 medium containing 10% fetal bovine serum. Cell cultures were supplemented with various concentrations of IL-1 (0, 1, 10, 20 and 40ng/ml), and the MMT growth assay performed. PC-3 and DU-145 cells were treated for 2, 4, 8, 12 and 24 h both with and without IL-1. IL-6 and IL-6 receptor (IL-6R) mRNA expressions were investigated using RT-PCR, and the IL-6 levels in cultured supernatant measured by ELISA. Results: The viability of both PC-3 and DU-145 cells decreased after IL-1 treatment (10, 20 and 40ng/mul). With 40ng/ml the IL-1, IL-6 and IL-6RmRNA expressions were lower in PC-3 cells, but unchanged in DU-145 cells, whereas the IL-6 protein production was higher in both PC-3 and DU-145 cells. Conclusions: IL-1 inhibited the proliferation of both PC-3 and DU145 cells. In the PC-3 cells, IL-1 decreased the expressions of IL-6 and IL-6R mRNA, but paradoxically increased the IL-6 production. In the DU-145 cells, IL-1 treatment did not affect the IL-6 or IL-6R mRNA expressions, but the IL-6 production was increased. This discrepancy between IL-1-induced IL-6 mRNA and protein production may be mediated by modification to the protein synthesis or an increased cellular excretion.

Résumé

A primary squamous cell carcinoma of the thyroid is extremely rare disease. We evaluated this disease by fine needle aspiration cytology in a 43 year-old female. On physical examination, the thyroid was moderately enlarged and palpated with hard consistency. The cytologic aspirate revealed nests of squamous cells, malignant squame and thyroid follicular cells in the necrotic background. We diagnosed this tumor as squamous cell carcinoma with cytologic evidence. Surgically resected thyroid revealed well differentiated squamous cell carcinoma. Immunohistochemical staining for antithyroglobulin proved that this tumor was primary in origin and clinical study confirmed this lesion to be of primary thyroid origin rather than metastasis or direct invasion from contiguous structures.

Résumé

Forty cases of infiltrating duct carcinoma of the breast were examined immunohistochemically for expression of TGF-beta and angiogenesis in order to analyze significant correlation with prognostic parameters including tumor size, axillary lymph node metastasis, clinical stage, histologic grade, estrogen receptor and progesterone receptor status. The TGF-beta expression was observed in tumors center and advancing edges of tumors. To determine microvessel density for angiogenesis, we stained endothelial cells for Factor VIII related antigen and counted microvessel within tumor. The results were as follows: 1) The strong immunohistochemical expression of TGF-beta and higher counts of microvessels were observed in advancing edges of tumors (p0.05). Therefore, the results suggested that the TGF-beta expression and angiogenesis in infiltrating duct carcinoma of the breast may play an important part in prognostic factors, closely related to the lymph node metastasis and clinical stage.

All solid tumors require the induction of new blood vessel to grow. The neovascularization of tumor tissue(angiogenesis) is considered essential for tumor growth, proliferation and eventually metastasis. Recent reports have demonstrated that the intensity of tumor angiogenesis in prostate and other tumors may be of prognostic value. We analyzed the prognostic significance of microvessel quantitation in bladder carcinoma. Microvessels were identified by immunohistochemistry using antibodies to endothelial marker, factor VIII-related antigen. The three most vascular area within a tumor were selected, and the microvessels within a x200 microscopic field of each area were counted by an investigator. The significant relationship was observed between microvessel counts and recurrence rate in 17 patients with superficial bladder cancer. Microvessel counts correlated with stage, grade, lymph node and distant metastasis, and 2 year disease free survival rate in 28 patients with invasive bladder cancer. In 15 patients with invasive cancer who were not showed distant metastasis, tumors from patients who experienced distant metastasis had higher microvessel counts than did tumors from patients who were disease- free(75.7118.59 and 61.7517.78), these values were not significantly different(p=0.081). These findings suggest that assessment of angiogenesis by microvessel quantitation may be a valuable method to predict metastatic potential of tumors, survival and the candidates for adjuvant therapy in patients with invasive bladder cancer.

Nucleolar organizer regions(NORs) are intranucleolar segments of DNA coding for ribosomal RNA and contribute to the regulation of cellular protein synthesis. Since NOR-associated proteins are argyrophilic, silver staining method has been used for demonstration of NORs. The numbers of argyrophilic NORs(AgNORs) have been shown to be correlated with DNA ploidy and have prognostic value in diverse human neoplasms. However, the prognostic value of AgNORs in renal cell carcinoma(RCC) remain ill defined. We herein investigated the prognostic value of AgNORs in 39 patients with RCC. There was no significant relationship between mean number of AgNORs (m AgNORs) per nucleus and Robson stage and Fuhrman nunlear grade. However, there was significant relationship between the percentage of tumor cells with more than five AgNORs per nucleus (p AgNORs) and Robson stage and nuclear grade (p<0.05). There was significant difference in survival rate between patients with RCC in whom AgNORs were two or less and in whom AgNORs were greater than two(p<0.05). The patients with a low p AgNORs(less than 8%) have a better prognosis than those with a high p AgNORs. These results suggest that number of AgNORs may be a clinically useful prognostic marker for patients with RCC.

Résumé

Phenytoin is an anticonvulsant compound which modulates the voltage-dependent sodium channels. It has a neuroprotective effect in vitro against hypoxic damage in hippocampal slices of adult rats. The authors studied the efficacy of phenytion on cerebral ischemia in an vivo model of hypoxic-ischemic brain injury in developing rat brain. To elicit injury, 7 days old(P7) Sprague-dawley rats subjected to right common carotid ligation followed by 8% O2 exposure(humidified, balanced with nitrogen) for 3 hours under the halothane anesthesia(control group, N=58). Body temperature of the rats was accurately controlled before and during hypoxia. Before hypoxia, pups received intraperitoneal phenytoin(30mg/kg)(phenytoin-treated group, N=17). The animals were sacrificed one week later and histopathological evaluation of ischemic neuronal damage were conducted employing hematoxylin-eosin staining and measurement of the hemispheric weight differences were performed. Phenytoin was found to be effective in reducing neuronal damage in terms of weight comparison(24+/-2.4% atrophy of control vs. 5+/-2.9% atrophy of phenytoin group, p<0.001) and ischemic changes in hippocampal region(p<0.05 in CA1, CA2, and CA3 area). These data suggest that compounds like phenytoin, which modulates voltage-dependent sodium channels, can reduce the degree of injury from hypoxic-ischemic insults to the developing rat brain.
