Résumé

OBJECTIVETo study the effect of the two arms of miR-590, miR-590-5p and miR-590-3p, on hepatoma cell proliferation and their roles in tumor development.

METHODSWe analyzed and verified the expression pattern of miR-590 in liver cancer specimens and cell lines by miRNA microarrays and QPCR. MiR-590 mimic or inhibitor was transfected into normal liver cells or liver cancer cells via liposome, and the changes in cell proliferation and survival were determined by MTT assay and soft agar colony formation assay. The target genes of miR-590-5p and miR-590-3p were predicated with Targetscan and validated by luciferase reporter system and Western blotting.

RESULTSThe expressions of miR-590-5p and miR-590-3ps were up-regulated in 3 hepatocellular carcinoma (HCC) tissues and their synchronization was significantly up-regulated in 8 out of 10 HCC tissues as compared with the adjacent tissues. QPCR further showed that miR-590-5p/3p was up-regulated in 3 HCC cell lines (HepG2, Hep3B, and Huh7) in comparison with the normal liver cell line L-O2. L-O2 cells over-expressing miR-590-5p and miR-590-3p exhibited significantly increased proliferation (P<0.05), while down-regulation of miR-590-5p and miR-590-3p caused significantly suppressed proliferation in HepG2, Hep3B, and Huh7 cells. Targetscan predicted PDCD4 and PTEN as the potential target genes of miR-590-5p and miR-590-3p, which was verified by luciferase reporter system and Western blotting. miR-590-3p was found to activate PI3K-AKT signaling pathway by down-regulating PTEN to promote AKT1-S473 phosphorylation.

CONCLUSIONMiR-590 is an important tumorigenic factor for HCC, and its two arms can both promote tumorigenesis by regulating the expression of their target tumor suppressor gene, PDCD4 and PTEN, to promote HCC cell proliferation and survival and activate the core tumor signal pathway PI3K-AKT.

Résumé

Objective:To develop an appearance-management behavior scale for college female students. Methods:Based on literature analysis and open questionnaire, the preliminary scale was formed.A total of 822 college female students completed the scale.Exploratory and confirmatory factor analytic approaches were used to develop and evaluate the intra-structure of the scale. Results:Exploratory principle factor analysis of the items indicated that the scale had three factors.Internal consistency analysis showed that the Cronbach' ? coefficient,split-half and test-retest correlation coefficients were 0.88,0.86,0.77 respectively.Confirmatory factor analysis showed that the fit indexes for ?2/df,GFI,AGFI,NFI,NNFI,RMSEA,CFI,IFI were 3.20,0.87,0.83,0.82,0.85,0.085,0.87,0.87.Total scale had no significant correlation with SAAS,but with APS,PSS,IMS,SES (r=0.23,0.21,0.38,0.28,P

Résumé

Objective: To explore the reliability and validity of Ruminative Responses Scale in Chinese version. Methods: A total of 912 Chinese college students completed the RRS. Results: All the indexes of confirmatory analysis including ?2、RMSEA、GFI、AGFI、NNFI、CFI、IFI met the requirements of psychological assessment. The ? coefficients of the scale and subscales and test -retest reliabilities were over 0.63. There was gender difference in rumination level. Conclusion: RRS is appropriate for the assessment of rumination in Chinese colleges.

Résumé

Objective: To explore the mediation of stress between perfectionism and life satisfaction. Methods: 317 college students were surveyed with questionnaires. The data were analyzed by hierarchical regression. Results: ①Regarding general perfectionism, both male and female students' stress fully mediated negative perfectionism and life satisfaction but did not mediate positive perfectionism and life satisfaction;②Regarding domain-specific perfectionism, male students' appearance stress fully mediated related perfectionism and satisfaction, but for female students its mediation was partial. Both male and female students' personality stress fully mediated personality perfectionism and satisfaction.Mediation models involved academic, love, interpersonal relationship perfectionism and their related satisfaction were not found. Conclusion: The mediations of stress between perfectionism and life satisfaction are different in different life domains.

Résumé

Objective:To develop a domain-specific perfectionism scale for college students. Methods:Exploratory and confirmatory factor analytic approaches were used to develop and evaluate the intra-structure of the scale. Results:Exploratory principle factor analysis of the items indicated that the scale had five factors, explaining 57.538% of the total variance.Internal consistency analysis showed that the Cronbach's ? coefficient,split-half and test-retest correlation coefficients were 0.648~0.781,0.664~0.790,0.720~0.779 respectively for the factors.Confirmatory fator analysis showed that the fit indexes for ?2/df,GFI,AGFI,NFI,NNFI,RMSEA,CFI,IFI were 2.32,0.91,0.88,0.84,0.88,0.06,0.90,0.90.Conclusion:Psychometric properties analysis shows that the scale achieves from good to exellent levels of internal consistency,test-retest rliability and content, construct and criteria-related validity.

Résumé

Objective:To explore the mediating effect of attitudes going into the of undergraduates world or leaving the mundane world between internet addiction disorder (IAD) and undergraduates' well-being. Methods:603 undergraduate students were surveyed with the Scale of Going into the World and Leaving the World, the Scale for Internet Addiction and the Multiple Happiness Questionnaire. Correlation, regressions and structural equation were adopted. Results:The rate of IAD in our sample was 9.1%, which was higher in male (47persons, 13.2%) than in female (8persons, 3.2%)(?2=16.47,P
