Résumé

Objective:To evaluate the role of Dorea in glucose intolerance of people with obesity. Methods:This study recruited 113 young individuals with obesity and varying degree of glucose tolerance [body mass index(BMI)≥30 kg/m 2] and 105 controls, comparing the metabolic phenotypes and Dorea abundance. Correlation analysis and ROC analysis were performed to assess the association between Dorea and clinical parameters and its predictive role in predicting glucose intolerance. Results:(1) Metabolic parameters were higher in obesity group than the control group. There was no difference in body weight, BMI and WHR among subgroups classified by glucose tolerance in people with obesity. (2) The abundance of Dorea, Dorea formicigenerans were higher in obese individuals, however showing a downward trend in accordance with glucose intolerance. The abundance was inversely associated with OGTT-2 h plasma glucose and HbA 1C, while positively associated with HOMA-β. Logistic regression demonstrated that Dorea formicigenerans was an independent protective factor after adjusting confounders such as age and gender in the prevention of glucose intolerance. (3) ROC analysis exhibited that the AUC values of Dorea formicigenerans was 0.73 in the total population. Conclusion:Dorea and Dorea formicigenerans exert protective effect on glucose metabolism in obese subjects. The abundance of Dorea and Dorea formicigenerans can be used as predictors of glucose intolerance risk in obese subjects, which facilitate the early screening and monitoring.

Objective: To study the contents of hyperoside in Semen Cuscutae,which grew in different host and different environment.Methods: Hyperoside was determined by HPLC.Results: The content of hyperoside is 0.28%-0.65% in the semen Cuscutae growing in different conditions in Shandong.The content is higher in semen of C.chinensis in Linyi,whose host is Scutellaria baicalensis Georgi.The content of hyperoside is lower in semen of C.australia which grew in Humulus scandens(lour.) Merr.The content has no obvious difference between C.chinensis and C.australis.Conclusions: The content of hyperoside in semen Cuscutae has relation with the hosts.The content increased with the seed mature degree.

Objective: To explore the differences among contents of quercetin and kaempferol in four kinds of Semen Cuscutae in Shandong province. Methods : Quercetin and kaempferol are determined by HPLC. Results : The contents of both quercetin and kaempferol are lower in four kinds of Semen Cuscutae. The former is within 0.0002% to 0.0502% and the latter varies from 0.0002% to 0.0205%, respectively. In seeds of Cuscuta chinensis Lam., C. japonica Choisy and C.lupuliformis Krocker, quercetin contents are much higher than kaempferol contents. while in seed of C. australis R.Br., by contrast, its quercetin content is lower than kaempferol. The contents of quercetin and kaempferol have close relation with both the variety and the hostplant. Conclusion : HPLC can be used for the quality evaluation of Semen Cuscutae.

Objective: To compare the effects of invigorating the kidney and strengthening yang on four kinds of Semen Custutae. Methods: Sexual organs of juvenile rat and shen yang deficiency rat were weighed to analyze its contents of SOD and MDA. Results: Alcohol extracts of four kinds of Semen Custutae have conspicuous effects on increasing weight of sexual organs. Water extracts of Semen Custutae have better effects on raising the contents of SOD and MDA and increasing weight of immune organs of shen yang deficiency rat. Conclusion: The effects of Cuscuta chinensis Lam. and C.australis R. Br. have better than that of C. japonica Choisy and C.lupuliformis Krockr in invigorating the kidney and strengthening yang.
