Comparative bioavailability and pharmacokinetics of diazepam in various formulations by HPLC technique

Four different brands of diazepam were tested and compared for the absolute bioavailability of N-desmethyldiaze-parm as its major metabolite by HPLC technique in the same six nomral mixed breed dogs. The drug was given P.O. by LV. routes. The results revealed significant differences for Cp-max between formulation 2 and 3 and 2 and 4. Ever results of fontutlation 3 and 4 were quite similar. Formulation 1 being an LV. solution was taken as standard. The T-max for formulation 2 was significantly different from formulation 3 and 4. The A UC for formulations 2, 3 4 did not show much difference. The absolute bioavailability for formulation No. 2,3 and 4 was 81%, 79% and 81% respectively
