Résumé

Staphylococcal enterotoxin B [SEB] is a potent inducer of cytotoxic T-cell activity, cytokine production and necrosis induction in vivo. Monophosphoryl lipid A [MPL] is an adjuvant derived from the lipopolysaccharide of E.coli, Salmonella Minnesota Re595 and other gram negative bacteria. In this research, The antitumor and antimetastatic effect of intra-venus injection of Monophosphoryl Lipid A [MPL], Staphylococcal enterotoxin B [SEB] and SEB+ MPL was evaluated using Balb/C male mice bearing inoculable mice Fibrosarcoma. The anti tumor effect of SEB+ MPL, SEB and MPL in mice with inoculated fibrosarchoma tumor [Wehi-164] was examined by IV injection and the sizes of the inoculated tumors were determined. The inoculated tumors were also examined histologically. Moreover, histophatologic study in lung tissue didn't showed any metastasis. In the mice IV injected group with SEB4- MPL, reduction of tumor size show a significant difference compared with mice in the SEB and MPL injected and negative control group. A significantly higher frequency of necrosis in tumor tissues was also observed in mice in the IV [SEB+ MPL]-injected group in comparison with other group. Moreover, histophatologic study in lung tissue didn't show any metastasis Our findings suggest that tumor cell death and the prevention of metastasis be caused by increased Cytotoxic T-cell activity in response to IV injection of SEB+ MPL that need to more investigation

Résumé

The regeneration of axon and myelin sheet after crush injury of peripheral nerves involves interaction of several types of cells. including Schwann cells, monocyte, niacrophage and fibrohiast. Among them, haeniatogenous macrophages invading into the peripheral nervous system play a major role in myelin uptake during Wallerian degeneration. In this study 35 C57IBL6male mice 10 weeks old were used and classified in 7 groups of 5 mice. The localization of epidermal-type fatty acid binding protein [E-FABF] in the mature mouse sciatic nerve after crush injury was examined by immuno-light microscopy. Numerous macrophages immunopositive for both anti-E-FABP and F4/80 a macrophage marker, were found in degeneration process of sciatic nerve. Macrophages contained phagosomes of various sizes and they were regarded as those actively involved in the phagocytosis of sciatic nerve debrise. Conclusion: The present dietection of E-FABP immunopositivity selectively in invading macrophages suggests possible involvement of E-FABP and/or its fatty acids ligand in the process of peripheral nerve regeneration

Résumé

Effect of lipocalin 2 on the expression of heme oxygenase I, II and NF-kB transcription factor was the purpose of this survey. Lcn2 was cloned to pcDNA3.1 plasmid by using genetic engineering method. The recombinant vector was transfected to CHO and HEK293T to establish stable cell expressing lipocalin 2. The presence of lipocalin 2 gene in these cells was confirmed by using through RT-PCR and western blot analysis. Expression of Lcn2 was also down-regulated by siRNA in A549 cell line. Expression of heme oxygenase I, II and NF-kB transcription factor were determined in both ectopic expression Lcn2 cells and Lcn2 down regulated cells by using of RT-PCR and western blot analysis. The results showed that the expression of heme oxygenase I and those of NF-kB were higher in cells expressing recombinant lipocalin2 compared with the control cells. On the other hand, expression of heme oxygenase I and NF-kB in siRNA transfected cells was down-regulated. These findings indicate that lipocalin2 induces the expression of HO-1 and suggest Lcn2 through NF-kB induces HO-1 expression
