Detecção da citotoxicidade de materiais biocompatíveis nas linhagens celulares MRC-5, HeLa e RC-IAL / Detection of cytotoxicity of biocompatible materials in MRC-5, HeLa, and RC-IAL cell lines

The sensitivity of diploid and heteroploid cell lines for detection of cytotoxicity using the agar diffusion method on cell culture, was tested with ascorbic acid solution of different concentrations. A total of 562 samples of 21 various materials were tested. The heteroploid cell line, RC-IAL, showed in relation to the MRC-5 and HeLa cell lines, greater sensitivity because it showed the presence of cytotoxic effect with the lowest concentration used (10 and 25 micrograms/ml) of ascorbic acid and showed greater diameter of cytotoxic halo in 15 samples and equal diameter in 16 of the 43 positive samples (7.6). Out of 43 positive samples, the MRC-5 line did not show cytotoxicity in 3 sponge samples and 1 of acrylic resin. The PVC (polyvinylchloride) and polyethylene rarely showed positivity, while, the plastic, cotton and acrylic resin demonstrated cytotoxicity in about 5 of samples. We thus suggest the use of the RC-IAL and HeLa cell lines for continuation of this type of analysis at Adolfo Lutz Institute.

Culturas celulares na detecção da toxicidade de materiais médico-hospitalares e outros que entram em contato com o ser humano / Cell culture for detecting toxicity of biocompatible materials

Em testes de citotoxicidade in vitro, pelo método de difusão em ágar, para materiais de uso médico hospitalar e outros, foram usadas 6 linhagens celulares estabelecidas: AV3, Vero, RC-IAL, NCTC clone 929, RV-IAL, HeLa e cultura primária de embrião de galinha (EG). Nos 20 materiais testados, as células EG demonstraram maior sensibilidade, seguidas das células RC-IAL e HeLa. O efeito citotóxico foi avaliado pela presença e pelas medidas dos diâmetros dos halos difundidos. Devido às dificuldades operacionais com a cultura primária EG recomenda-se, para os testes de citotoxicidade in vitro dos materiais citados, o uso de placas, em duplicata, das células RC-IAL e HeLa (AU).

Proteção "in vitro" conferida pela gama-globulina contra o vírus do sarampo / Protection "in vitro" by gamma globulin against the measles virus

A gamaglobulina fornecida por um laboratório nacional experimentada em culturas celulares de linhagem contínua de rim humano conferiu proteção apreciável contra a ação do vírus do sarampo, em doses de 0,1 mg/ml, e proteção total em doses de 0,2 ou 0,3 mg/ml. (AU)

The gamma globulin obtained from the ISA Laboratory was tested on contínuous line of human kidney cells, In experimental condítíons the gamma globulin showed a sígnifleant protectíon against measles vírus, when used in doses of 0,1 mg per ml, and total protection with 0,2 or 0,3 mg per ml. (AU)
