Résumé

Aim To predict the key targets and signaling pathways of Semiliquidambar cathayen. sis Chang (JLBFH) by network pharmacology and molecular docking, etc, then to explore the mechanism of JLBFH' s effect on inflammatory response to depression through reserpine-induced depression rat model. Methods The target of drug and disease was predicted by network pharmacological database, protein interaction network diagram was constructed, biofunctional enrichment and pathways were analyzed, and molecular docking prediction was performed by AGFR software. Based on reserpine-induced depression, the role of JLBFH in depression inflammation was verified by behavior, molecular biology and pathological examination, and so on. Results A total of 13 active ingredients were screened, 11 key targets of JLBFH modulation of depression were selected, and the bioenrichment results were mainly related to cognition, prominent plasticity regulation, etc. The pathways were mainly related to Rapl signaling pathway, Toll-like receptor signaling pathways. The results of validation experiments showed that high and low doses of JLBFH extract significantly shortened the forced swimming immobility time in mice, markedly reduced the retention time in the circle of rats, increased serum levels of 5-HT and DA, decreased serum levels of IL-6, improved inflammatory infiltration in the prefrontal cortex, decreased brain tissue levels of IL-6,IL-1β ,TNF-α mRNA expression, and increased AKT1 mRNA expression in brain tissue. Conclusions The present study reveals that JLBFH can exert antidepressant effects through multi-component, multi-target and multi-pathway, and the experimental validation results show that JLBFH can improve the d¬pression-like symptoms by improving the inflammatory response of depression through TOLL-like signaling pathway.

Résumé

ObjectiveTo observe the effect of Flemiphilippinin D on collagen-induced arthritis (CIA) in rats and explore its mechanism. MethodForty rats were randomly divided into normal group, CIA group, methotrexate (MTX) group (1.35 mg·kg-1), low-dose Flemiphilippinin D group (1.5 mg·kg-1), and high-dose Flemiphilippinin D group (3.0 mg·kg-1), with eight rats in each group. Except for the normal group, the CIA model was induced by type Ⅱ collagen. Each group was given corresponding liquid medicine or normal saline, once a week in the MTX group, and once a day in the Flemiphilippinin D groups for a total of 28 days. The arthritis score and joint swelling degree of rats were experimentally recorded. Pathological changes in the ankle joint of rats were observed by hematoxylin-eosin (HE) staining. Serum levels of inflammatory cytokines interleukin (IL)-1β, IL-6, IL-8, and tumor necrosis factor (TNF)-α were detected by enzyme-linked immunoabsorbent assay (ELISA), and the mRNA expression of Toll-like receptor 2 (TLR2), myeloid differentiation factor 88 (MyD88), and nuclear transcription factor-κB (NF-κB) p65 were detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR), and the protein expressions of TLR2, MyD88, and NF-κB p65 were detected by Western blot. ResultCompared with the normal group, the ankle joint of the CIA group was significantly swollen, and the clinical score of arthritis and the degree of joint swelling were significantly increased (P<0.01). The ankle joint tissue structure was significantly damaged, and the levels of inflammatory factors IL-1β, IL-6, IL-8, and TNF-α in serum were significantly increased (P<0.01). The mRNA levels and protein levels of TLR2, MyD88, and NF-κB p65 were significantly increased(P<0.01). Compared with the CIA group, arthritis clinical score and joint swelling of rats in each administration group were significantly reduced (P<0.05, P<0.01), and the pathological changes in the ankle joint were significantly improved. The contents of serum IL-1β, IL-6, IL-8, and TNF-α were significantly decreased (P<0.05, P<0.01). The mRNA levels and protein levels of TLR2, MyD88, and NF-κB p65 in the ankle joint were significantly decreased (P<0.05, P<0.01). ConclusionTo a certain extent, Flemiphilippinin D can reduce the expression of inflammatory factors in rheumatoid arthritis rats and play a good therapeutic effect. It works perhaps by inhibiting the activation of the TLR2/MyD88/NF-κB signaling pathway and thus shows an anti-inflammatory effect.

Résumé

This study aimed to investigate the mechanism of Jiu Wei Bu Xue Oral Liquid on insomnia rats combining the methods of network pharmacology, molecular docking and experimental verification. UPLC-Q-TOF-MS/MS method and TCMIP, TCMSP databases were used to collect the ingredients and targets of Jiu Wei Bu Xue Oral Liquid. Protein-protein interactions and network analysis were performed to screen the key network targets and putative active ingredients of Jiu Wei Bu Xue Oral Liquid in treatment of insomnia, and then following by biological function and KEGG pathway analysis. Then binding ability for key network targets and putative active ingredients were predicted with molecular docking. The prediction targets were validated in para-chlorophenylalanine (PCPA) induced insomnia rats with administration of Jiu Wei Bu Xue Oral Liquid (2, 4, 8 mL·kg-1) for 7 days. Pentobarbital sodium induced sleeping test were performed to evaluate the synergistic sleep-aiding effect of Jiu Wei Bu Xue Oral Liquid. Then glutamic acid (Glu), γ-aminobutyrate (GABA) content and glutamate decarboxylase 1 (GAD67) activity in hypothalamus or hippocampus were evaluated, and the expressions of GAD67, γ-aminobutyric acid receptor subunit α1 (GABRA1) and γ-aminobutyric acid receptor subunit β2 (GABRB2) in hippocampus were detected by qRT-PCR and Western blot methods. Animal experiments were approved by the Institutional Committee on Animal Care of Guangxi Institute of Chinese Medicine & Pharmaceutical Science (the number of permission: 2022060802). Results showed that 16 key network targets and 16 putative active ingredients were obtained by analyzing the herbs-ingredients-targets network of Jiu Wei Bu Xue Oral Liquid in treatment of insomnia. Network pharmacology and molecular docking all indicated these active ingredients, for example atractylenolide Ⅲ, showed better binding ability with GABRA1 and GABRB2. Animal study indicated that, compared to PCPA-induced insomnia model, Jiu Wei Bu Xue Oral Liquid remarkably shortened the sleeping latency and increased the sleeping duration, increased GAD67 activity and the production of GABA in hippocampus of insomnia rats, as well as the expressions of GAD67, GABRA1 and GABRB2, while decreased Glu content in hypothalamus, leading to decreasing of Glu/GABA ratio and recovery of Glu-GABA balance. These results indicated that Jiu Wei Bu Xue Oral Liquid improved insomnia symptoms and helped maintain the Glu-GABA balance within hypothalamus and hippocampus, and reduced the excitatory neurotoxicity within brain. The mechanism may due to the elevation of GAD67 expression and enzyme activity, and the enhancement of type-A GABA receptor (GABAAR)-mediated neurons inhibition.

Résumé

Objective: To grasp the epidemiological characteristics of influenza outbreaks in Guangdong Province by analyzing the outbreaks of influenza-like cases reported in Guangdong Province from January 2015 to the end of August 2022. Methods: In response to the outbreak of epidemics in Guangdong Province from 2015 to 2022, information on on-site epidemic control was collected, and epidemiological analysis was conducted to describe the characteristics of the epidemics. The factors that influence the intensity and duration of the outbreak were determined through a logistic regression model. Results: A total of 1 901 influenza outbreaks were reported in Guangdong Province, with an overall incidence of 2.05%. Most outbreak reports occurred from November to January of the following year (50.24%, 955/1 901) and from April to June (29.88%, 568/1 901). A total of 59.23% (1 126/1 901) of the outbreaks were reported in the Pearl River Delta region, and primary and secondary schools were the main places where outbreaks occurred (88.01%, 1 673/1 901). Outbreaks with 10-29 cases were the most common (66.18%, 1 258/1 901), and most outbreaks lasted less than seven days (50.93%,906/1 779). The size of the outbreak was related to the nursery school (aOR=0.38, 95%CI:0.15-0.93), the Pearl River Delta region (aOR=0.60, 95%CI:0.44-0.83), the time interval between the onset of the first case and the time of report (>7 days compared with ≤3 days: aOR=3.01, 95%CI:1.84-4.90), the influenza A(H1N1) (aOR=2.02, 95%CI:1.15-3.55) and the influenza B (Yamagata) (aOR=2.94, 95%CI: 1.50-5.76). The duration of outbreaks was related to school closures (aOR=0.65, 95%CI: 0.47-0.89), the Pearl River Delta region (aOR=0.65, 95%CI: 0.50-0.83) and the time interval between the onset of the first case and the time of report (>7 days compared with ≤3 days: aOR=13.33, 95%CI: 8.80-20.19; 4-7 days compared with ≤3 days: aOR=2.56, 95%CI: 1.81-3.61). Conclusions: An influenza outbreak in Guangdong Province exhibits two peaks, one in the winter and spring seasons and the other in the summer. Primary and secondary schools are high-risk areas, and early reporting of outbreaks is critical for controlling influenza outbreaks in schools. Furthermore, comprehensive measures should be taken to prevent the spread of the epidemic.

ObjectiveTo investigate the effect and mechanism of total flavones of Spatholobi Caulis (TFSC) against depression in rats. MethodThe fifty KM mice were randomly divided into the normal group and high-, medium-, and low-dose (1, 0.5, 0.25 g·kg-1) TFSC groups and gavaged with the corresponding drugs for 12 successive days. One hour after the last administration, the immobility time in forced swimming test and tail suspension test was recorded. The SD rats were randomly divided into the normal group, model group, fluoxetine (5 mg·kg-1) group, and high- and low-dose (1, 0.25 g·kg-1) TFSC groups. Following the exposure of rats to two different kinds of stimuli daily for inducing chronic unpredictable stress, they were administered with the corresponding drugs for 21 d. After the experiment, the levels of serum neurotransmitters and inflammatory factors in rats were detected by enzyme-linked immunosorbent assay (ELISA). The changes in hippocampal neurons of rats were observed by hematoxylin-eosin (HE) and Nissl staining. The mRNA expression levels of nuclear factor-κB (NF-κB) and tumor necrosis factor-α (TNF-α) in the hippocampus of rats were detected by real-time fluorescence quantitative polymerase chain reaction (Real-time PCR), and the protein expression levels of cAMP-response element binding protein (CREB), phosphorylated CREB (p-CREB), and brain-derived neurotrophic factor (BDNF) in hippocampal tissues by Western blot. ResultCompared with the normal group, TFSC significantly shortened the immobility time of mice in tail suspension and swimming tests (P<0.05). Compared with the normal group, the model group exhibited reduced sucrose intake and wilderness activity (P<0.01), decreased 5-HT, DA, NE (P<0.05, P<0.01), MAO, IL-6, TNF-α (P<0.05, P<0.01), damaged neurons, increased mRNA levels of TNF-α and NF-κB (P<0.01), and down-regulated BDNF and CREB protein expression (P<0.05). Compared with the model group, TFSC significantly enhanced sucrose intake and wilderness activity of rats (P<0.05), increased the serum 5-HT, DA and NE (P<0.05, P<0.01), and decreased the serum MAO, IL-6, and TNF-α (P<0.05, P<0.01) as well as NF-κB and TNF-α mRNA expression (P<0.01), up-regulated the protein expression levels of BDNF and CREB (P<0.01), and improved the pathological symptoms of hippocampus. ConclusionTFSC improved the hippocampal neurons of rats via CREB/BDNF signaling pathway and reduced depressive pathological damage, thus relieving depression.

Résumé

OBJECTIVE@#To explore the intervention effect of recombinant human interleukin-11 (rhIL-11) and recombinant human granulocyte-colony stimulating factor (rhG-CSF) on the duration and severity of agranulocytosis in patients with hematological malignancies after chemotherapy, and to analyze the influencing factors.@*METHODS@#The data of hematological malignancy patients treated with rhIL-11 and rhG-CSF after chemotherapy in the hematology department of The First Hospital of Lanzhou University from July 2017 to July 2020 were collected retrospectively. The duration and differences of agranulocytosis in differeent groups were compared by univariate analysis, and the influencing factors of agranulocytosis duration were further analyzed by multiple regression analysis.@*RESULTS@#The duration of agranulocytosis in 97 patients was 6.47±2.93 days. The results of univariate analysis showed that there were no statistical differences in the duration of agranulocytosis among patients with different sex, age, height, weight, body surface area, body mass index (BMI), dose of rhG-CSF, dose of rhIL-11, spontaneous bleeding after administration of rhG-CSF and rhIL-11, and the duration of agranulocytosis in patients with different red blood cell count (RBC), hemoglobin(HGB) level, platelet count (PLT) and absolute neutrophil count (ANC), before administration of rhG-CSF and rhIL-11. There were significant differences in agranulocytosis time among patients with different disease types, chemotherapy cycle, fever after rhG-CSF and rhIL-11 administration, and different white blood cell count (WBC) baseline level before rhG-CSF and rhIL-11 administration (P<0.05). Compared with patients with acute lymphoblastic leukemia (ALL) and non-Hodgkin lymphoma (NHL), patients with acute myeloid leukemia (AML) had the longest duration of agranulocytosis, which was 7.07±3.05 d. Compared with patients with chemotherapy cycles of 4-6 and ≥7, patients with total chemotherapy cycle of 1-3 had the shortest duration of agranulocytosis, which was 5.25±2.48 d. Compared with patients without fever, patients with fever within 1 day after administration of cytokines and patients with fever within 2-5 days after administration of cytokines, the duration of agranulocytosis was the longest in patients with fever 6 days after administration of cytokines, which was 8.85±2.85 d. Compared with patients with WBC baseline <1.0×109/L, (1.0-1.9)×109/L and (2.0-3.9)×109/L, patients with WBC baseline ≥4.0×109/L had the shortest duration of agranulocytosis, which was 4.50±2.56 d. Multiple linear regression analysis showed that chemotherapy cycle, different fever after administration of rhG-CSF and rhIL-11, diagnosis of ALL and NHL, and WBC baseline level before administration of rhG-CSF and rhIL-11 were the influencing factors of the duration of agranulocytosis (P<0.001).@*CONCLUSION@#The risk of prolonged agranulocytosis is higher in patients diagnosed with AML, with more chemotherapy cycles, lower WBC baseline before cytokines administration and fever later after cytokines administration, which should be paid more attention to.

Résumé

Objective:To investigate the effects and mechanism of Gecko extract for treatment of depression in rats. Method:The depression rats were induced by intraperitoneal injection of reserpine (0.5 mg·kg-1). The successfully modeled rats were randomly divided into model group, fluoxetine group (1.8 mg·kg-1), high dose and low dose groups of Gecko extract (12, 6 g·kg-1). The rats were given corresponding dose of drugs once a day for 10 days. After administration, the levels of neurotransmitters and inflammatory factors in serum and prefrontal cortex of rats were detected by enzyme-linked immunosorbent assay (ELISA). The cell changes in hippocampal tissues were observed by hematoxylin-eosin (HE) staining. The mRNA levels of interleukin-6 (IL-6), nuclear factor-κB (NF-κB), and tumor necrosis factor (TNF-α) in the hippocampus of rats were detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). The protein levels of Toll-like receptor 4 (TLR4) and NF-κB in hippocampal tissues of rats were detected by Western blot. Result:Compared with the normal group, Gecko extract significantly shortened the immobility time of tail suspension and swimming in mice. Compared with model group, Gecko extract significantly reduced blepharoptosis and retention time in circles for the rats (P<0.05), increased the levels of 5-hydroxytryptamine (5-HT) and dopamine (DA) in serum (P<0.05), decreased the levels of Monoamine oxidase (MAO), IL-6, and TNF-α in serum (P<0.05) and prefrontal cortex (P<0.05), decreased the mRNA levels of inflammatory cytokines IL-6, NF-κB and TNF-α and the protein expressions of TLR4 and NF-κB in the hippocampus of rats (P<0.05, P<0.01), and improved the pathological symptoms of the hippocampus. Conclusion:Gecko extract can significantly alleviate the pathological damage of depression and improve the symptoms of depression, and its mechanism may be due to inhibiting TLR4/NF-κB signaling pathway and reducing the expression of NF-κB, IL-6 and other inflammatory factors in the hippocampus of rats.

Résumé

OBJECTIVE:To compare the chemical components in Sinapis alba before and after stir-frying. METHODS : UPLC-Q-Exactive Obitrap MS was adopted to analyze chemical constituents of S. alba before and after stir-frying. The determination was performed on Waters CORTECS T 3 column with mobile phase consisted of methanol- 0.1% formic acid solution (gradient elution )at the flow rate of 0.25 mL/min. The column temperature was 30 ℃ and the sample size was 2 μL. High resolution MS adopted heating electrospray electron source ,positive ion scanning mode ,scanning range m/z 120-1 000. The chemical constituents of S. alba before and after stir-frying were identified by Compound Discover 3.2 software combined with relevant database ,and the content changes of chemical constituents were analyzed by using peak area. Chemometrics analysis was performed for the content changes of chemical constituents using peak area as variable. RESULTS :A total of 54 chemical components were identified in S. alba ,mainly fatty acids (represented by erucic acid ),alkaloids(represented by sinapine ), flavonoids. After stir-frying ,the contents of 19 chemical components changed significantly ,of which the contents of 10 components decreased significantly and those of 9 components increased significantly (P<0.05). Principal component analysis and orthogonal partial least squares discriminant analysis could clearly distinguish S. alba from stir-fried S. alba . CONCLUSIONS :The contents of some chemical components of S. alba change significantly after stir-frying ,which may be one of the important reasons for the change of efficacy after stir-frying.

Résumé

Aim To explore changes of gut microbiota in experimental model rats with hyperuricemia-induced mild cognitive impairment. Methods The experimental rats were provided with a diet containing 2% W/W uric acid (UA) and 2% W/W oxonic acid for period ranging from 1 day to 12 weeks. Morris water maze, blood biochemical indexes and other auxiliary models were used to evaluate the experimental animal model. Changes of gut microbiota were studied by 16S rDNA high-throughput sequencing. Results Compared with control group, the amount of UA of rats was significantly elevated. In addition, the rats showed significantly reduced spatial learning and memory in an escape latency and probe trial. The diversity of gut microbiota in model rats changed, and the relative abundances of 14 species of gut microbiota changed markedly. The abundance of Butyrivibrio, Erysipelotrichia, Erysipelotrichales, Erysipelotrichaceae, Erysipelatoclostridium, Lachnoclostridium 5, Anaeroplasmatales, Anaeroplasmataceae and Anaeroplasma in model mice increased, while that of Dorea, RuminococcaceaeUCG_005, Butyricicoccus, Tyzzerella 3 and Parasutterella decreased. Conclusions The continuous increase of UA can regulate gut microbiota, and the imbalance of gut microbiota is closely related to the development of mild cognitive impairment, which may be one of the mechanisms of hyperuricemia-induced mild cognitive impairment.

Résumé

Parkinson's disease is a common neurodegenerative disease in middle-aged and elderly people, in which the pathogenic factors are not yet clear. Genetics, dietary habits, environmental toxins, immunological abnormalities, inflammation and oxidative stress response, apoptosis, and mitochondrial dysfunctions which caused by a variety of physiological and biogenic changes are likely to exacerbate the occurrence of Parkinson's disease. In recent years, studies have shown that the activity of microglia is closely related to Parkinson's disease, and that the active microglia can promote the release of inflammatory factors, while the differentiation of dopamine neurons in the substantial nigra of midbrain area is also closely related to Parkinson's disease. As a histone H3K27me3 demethylase, JMJD3 is involved and affects the activity of microglia, which can regulate the polarization of microglia as well and affect the survival of dopaminergic neurons in the mesencephalon. This provides new methods and strategies for treating Parkinson' s disease. This paper summarizes the structure and function of JMJD3, as well as its role in neuro-inflammation mediated by microglia and its effect on neurons, and explores the functions and related research progress of JMJD3 in Parkinson's disease.

Résumé

Objective: By summarizing the technical points and therapeutic outcomes of combing infratemporal fossa approach (IFA) and internal carotid artery (ICA) reconstruction for the colossal skull base tumor invading ICA in petrous bone, the clinical application value was discussed. Methods: Five patients (2 males, 3 females, aging from 27 to 55 years old) who received surgeries between July 2015 and May 2017 for lateral skull base pathology involved petrous ICA using technique combined IFA and pre-reconstruction, were reviewed. Results: Among the five patients, three were paraganglioma of head and neck, one was carotid aneurysms, and one was recurrent adenoid cystic carcinoma (ACC). The median tumor size in the largest cross-section was 60 mm × 51 mm (range, 28 mm × 22 mm-72 mm × 58 mm). Complete excision was achieved with IFA and ICA reconstruction. The median blood loss volume was 1 000 ml (range, 600-2 500 ml). Four cases showed no new long-term neurologic sequelae, while one showed hemiplegia due to graft vessel occlusion. Except for the one with ACC having facial nerve cut, others achieved good facial nerve function of HB grade Ⅰ to Ⅱ during 3 to 12 months, follow-up. No tumor recurrence was observed over the median duration of follow-up for above 36 months (range, 36-58 months). Conclusion: For lesions involved superior part of ICA, which is unable to separate from ICA, IFA and ICA reconstruction can achieve complete excision.
