Résumé

OBJECTIVE@#To explore the treatment conditions of acid decalcified specimens and improve the poor quality of sections and unclear structure of hematoxylin-eosin (HE) staining caused by the change in pH in tooth and hard tissue after acid decalcification.@*METHODS@#A total of 20 cases of oral pathological specimens that contain hard tissues were decalcified and treated with routine treatment, concentrated ammonia water immersion treatment, and saturated lithium carbonate solution immersion treatment. The quality and HE staining effects of hard tissue sections treated with different methods were compared.@*RESULTS@#Compared with routine treatment, lithium carbonate saturated solution treatment showed complete sections. Hematoxylin is strongly stained, the nucleus is clear, and the cytoplasm is bright.@*CONCLUSIONS@#Soaking acid decalcified specimens in lithium carbonate saturated solution before embedding in dehydration can neutralize the acidic environment of the tissue. The quality of sections and HE staining effect are improved and are suitable for the pretreatment of acid decalcified tissue samples of oral pathology.

Résumé

Ghost cell odontogenic carcinoma (GCOC) is an exceptionally rare and malignant odontogenic tumor with aggressive growth characteristics. We describe a case of GCOC which was considerably derived from a previously resected calcifying cystic odontogenic tumor (CCOT). Cellular atypia, mitotic activity, Ki-67 labeling index and matrix metalloprotease-9 positive expression rate were all increased in the currently resected specimen compared to the initial one. This is a rare case of malignant transformation of CCOT to GCOC with respect to its histopathological and immunohistochemical findings.

Soft tissue clear cell sarcoma usually occurred in the end of the limbs, especially foot and ankle. Soft tissue clear cell sarcoma occurring in the torso and head and neck were even fewer. In this article, parapharyngeal soft tissue clear cell sarcoma was reported.

<p><b>OBJECTIVE</b>To report eight cases of central mucoepidermoid carcinoma of the jaws (CMCJ) and to analyze its clinical and pathological features.</p><p><b>METHODS</b>Eight cases of central mucoepidermoid carcinoma were diagnosed between 1989 and 2008. The clinical manifestation, radiological and histopathological changes were analyzed.</p><p><b>RESULTS</b>The mean age of this group of patients was 43.3 years, with 5 male and 3 female. Seven cases occurred in mandible, mainly in the molar, angle and ramus areas, and one in maxilla. The first complain usually was local swelling, pain or paraesthesia of the jaw and loosening and pain of the tooth. X-ray displayed unilocular or multilocular radiolucent lesion with distinct or ill-distinct border, and the light microscopic findings were similar to the mucoepidermoid carcinoma originated in the salivary gland.</p><p><b>CONCLUSIONS</b>To diagnose a CMCJ, it's necessary to synthetically analyze the case history, clinical examination, radiological and histopathological features. The treatment is wide local resection. Selective neck dissection and radiochemotherapy may improve curative effect and prognosis.</p>

<p><b>OBJECTIVE</b>To investigate the roles of matrix metalloproteinases (MMP) and tissue inhibitors of metalloproteinases (TIMP) in dentinogenic ghost cell tumor (DGCT) and ghost cell odontogenic carcinoma (GCOC).</p><p><b>METHODS</b>The expressions of MMP-2, MMP-9, MMP-14, TIMP-1 and TIMP-2 were examined in 15 DGCT cases and 9 GCOC cases by immunohistochemistry. Their mRNA expression in one DGCT case and one GCOC case were investigated by RT-PCT.MMP-2 and MMP-9 protein activities in the two cases were analyzed by gelatin zymography.</p><p><b>RESULTS</b>MMP-9 and TIMP-1 expressions elevated greatly in GCOC, and there was a significant difference (P < 0.05) in TIMP-1 expression between GCOC and DGCT.Pro-MMP-9, MMP-9 activated form, pro-MMP-2, and MMP-2 activated forms were detected in the GCOC case, while pro-MMP-9 and MMP-9 activated form were very faint in the DGCT case. The mRNA level of MMP-9 elevated obviously in the GCOC case, which was similar to that of TIMP-1.</p><p><b>CONCLUSIONS</b>The elevated expression of MMP-9 and TIMP-1 may influence the behaviour of GCOC.</p>

<p><b>OBJECTIVE</b>To study the clinicopathologic features and prognostic factors of Chinese patients with mantle cell lymphoma.</p><p><b>METHODS</b>One hundred and two cases of mantle cell lymphoma occurring in Chinese patients were studied by light microscopy and immunohistochemistry. The follow-up information was also analyzed. The cases were classified as mantle zone, nodular or diffuse patterns and as typical or blastoid variants. Age, Ann-Arbor staging, B symptoms, hematologic parameters, histologic variants, mitotic index and immunophenotype were assessed for possible prognostic implication.</p><p><b>RESULTS</b>The median age of the patients was 59 years (range: 30 to 79 years) and the male-to-female ratio was 2.92:1. Seventy-one patients (87.65%) presented with advanced stage disease (Ann Arbor stage III to IV). B symptoms were present in 45.45% of patients. The commonest site of involvement was lymph node (100%). The other involved sites included bone marrow (64.44%), spleen (63.16%), Waldeyer's ring (31.25%), peripheral blood (29.41%), liver (22.64%) and gastrointestinal tract (14.71%). All cases expressed B-cell markers but were negative for T-cell marker. Majority of cases were positive for cyclin D1 (94.12%) and CD5 (71.43%). Blastoid variant accounted for 24.51% of cases. Amongst the 68 cases with follow-up data available, the median survival was 10 months. Parameters associated with shorter survival included diffuse pattern, blastoid variant, high mitotic index, high proliferative activity and presence of bone marrow involvement.</p><p><b>CONCLUSIONS</b>The clinicopathologic features and prognostic factors of mantle cell lymphoma occurring in Chinese are similar to those in Caucasians. Diffuse pattern, blastoid variant, high mitotic index, high proliferative activity and involvement of bone marrow indicate poor prognosis.</p>

<p><b>OBJECTIVE</b>To investigate the role of MMP-2, MT1-MMP and TIMP-2 in the carcinogenesis of oral lichen planus (OLP).</p><p><b>METHODS</b>Immunohistochemistry was performed to examine the expression of OLP and compare with that of NOM.</p><p><b>RESULTS</b>The expression of these proteinases significantly increased from NOM, non-atrophic OLP, to atrophic OLP and OSCC. The expression of MMP-2 and MT1-MMP in atrophic OLP was significantly higher than in non-atrophic OLP. Furthermore, the expression of TIMP-2 consequently increased with the increasing of the MMP, but the increase of TIMP-2 was less than that of MMP.</p><p><b>CONCLUSIONS</b>MMP may be useful marker to judge the possibility of malignant change of OLP.</p>

<p><b>OBJECTIVE</b>To examine the expression of MMPs and TIMPs in pleomorphic adenoma of salivary gland and to investigate the relationship between the expression and the biological behaviour of the tumor.</p><p><b>METHODS</b>Twenty-three cases of pleomorphic adenoma were divided into active type and common type according to their biological behavior. Immunohistochemistry for MMP-2, MMP-9, TIMP-1, TIMP-2 and gelatin zymography analysis were performed in these 23 cases and in 6 malignant and 6 benign salivary gland tumors.</p><p><b>RESULTS</b>The immunoreactivity of MMP-2 protein and MMP-2/TIMP-1, 2 ratio were significantly higher in active pleomorphic adenoma than in common pleomorphic adenoma (P = 0.028, P = 0.009, P = 0.045). The expression of active MMP-2, proMMP-9 and active MMP-9 were significantly higher in active pleomorphic adenoma than in common pleomorphic adenoma (P = 0.034, P = 0.021, P = 0.001). There was no significant difference in expression of MMP-2, MMP-9, TIMP-1 and TIMP-2 between the salivary malignant tumor and active pleomorphic adenoma, also between the salivary benign tumor and common pleomorphic adenoma.</p><p><b>CONCLUSIONS</b>The expression of MMPs and TIMPs in active pleomorphic adenoma is similar to that in salivary carcinomas, and the expression in common pleomorphic adenoma also resembled to that in salivary adenoma. The expression of MMP-2, 9 and TIMP-1, 2 is related to the biological behavior of pleomorphic adenoma of salivary gland.</p>

<p><b>OBJECTIVE</b>To detect protein expression of MMPs and TIMPs in various salivary gland neoplasms and to investigate their roles in invasion and metastasis of the malignant salivary gland tumors.</p><p><b>METHODS</b>Immunohistochemistry and Gelatin zymography analyses for MMP-2, MMP-9, MT1-MMP, TIMP-1 and TIMP-2 were performed in 26 malignant and 28 benign salivary gland tumors.</p><p><b>RESULTS</b>The expression of MMP-2 and MMP-9 was significantly higher in carcinomas than in adenomas (P < 0.05). The MMP-2/TIMP-1 and MMP-2/TIMP-2 was also significantly higher in carcinomas than in adenomas (P < 0.05). There was a cooperated effect among MMP-2, MT1-MMP and TIMP-2. The expression of active MMP-2, proMMP-9 and active MMP-9 was significantly higher in malignant tumors than in benign tumors (P < 0.05).</p><p><b>CONCLUSION</b>MMP-2 and MMP-9 may play important roles in invasion of malignant salivary gland tumors. A disturbed balance between MMP-2, MMP-9, TIMP-1 and TIMP-2 in malignant salivary gland tumors was detected. It was the absolute increase of MMP-2 and MMP-9 to induce the unbalance.</p>
