BACKGROUND: Salmonella pathogenicity island (SPI)-13 is conserved in many serovars of S. enterica, including S. Enteritidis, S. Typhimurium and S. Gallinarum. However, it is absent in typhoid serovars such as S. Typhi and Paratyphi A, which carry SPI-8 at the same genomic location. Because the interaction with macrophages is a critical step in Salmonella pathogenicity, in this study we investigated the role played by SPI-13 and SPI-8 in the interaction of S. Enteritidis and S. Typhi with cultured murine (RAW264.7) and human (THP-1) macrophages. RESULTS: Our results showed that SPI-13 was required for internalization of S. Enteritidis in murine but not human macrophages. On the other hand, SPI-8 was not required for the interaction of S. Typhi with human or murine macrophages. Of note, the presence of an intact copy of SPI-13 in a S. Typhi mutant carrying a deletion of SPI-8 did not improve its ability to be internalized by, or survive in human or murine macrophages. CONCLUSIONS: Altogether, our results point out to different roles for SPI-13 and SPI-8 during Salmonella infection. While SPI-13 contributes to the interaction of S. Enteritidis with murine macrophages, SPI-8 is not required in the interaction of S. Typhi with murine or human macrophages. We hypothesized that typhoid serovars have lost SPI-13 and maintained SPI-8 to improve their fitness during another phase of human infection.

El NaOH al 1% es efectivo para obtener pleurodesis como tratamiento paliativo del derrame pleural maligno (DPM). El porcentaje de fracasos de obtener pleurodesis con los diferentes agentes y métodos es variable, fluctúa entre 30% al 100% y las causas no han sido bien establecidas. Analizamos 49 pacientes con DPM cuyos promedios de edad fueron de 56 años y que se estudiaron con radiología, broncoscopia y toracoscopia. De ellos, el cáncer primario fue: pulmonar 26 casos(44.06%), mama en 17 casos (28,81%) y los restantes en otras localizaciones, con 9 (15,25%), sin determinar el primario. Instilamos a través de pleurotomía 20 cc de NaOH al 1% (0,3 ml por kg) y previa toracocentesis total (controlado por toracoscopia) hasta obtener adhesión de las pleuras y cese del líquido pleural. En 47 casos (79,7%) obtuvimos pleurodesis completa. En 9 pacientes (15,3%) pleurodesis parcial, y en 3 (5%) fracaso. No hubo complicaciones ni mortalidad con el método. En conclusión el NaOH es un agente útil, barato y bien tolerado para el tratamiento paliativo del DPM. Las causas del fracaso de la pleurodesis se deben a : 1) Presencia de linfangitis carcinomatosa, que evita la expansión del pulmón y se evalúa con radiología; 2) Estenosis bronquial tumoral, que también lleva al colapso pulmonar y se evalúa con broncoscopia 3) Siembra tumoral pleural visceral y parietal y/o engrosamiento pleural, que puede evitar expansión pulmonar o movilidad diafragmática y que se evalúa con la toracoscopia
