Résumé

Gut microbiome is closely related to human health. Changes in intestinal microbial composition promote the development of tumors. Specific intestinal microorganisms and their metabolites regulate host physiological functions and tumor microenvironment, significantly affecting the anti-cancer treatment response and its adverse events. Strategies targeting gut microbiome have shown promising prospects in diagnosis and treatment of cancer.

Objective@#To investigate the distribution of the gene subtypes of human papillomavirus (HPV) in male patients with condyloma acuminatum (CA) and analyze the characteristics of the gene subtypes.@*METHODS@#We extracted genomic DNA of the HPV virus from the genital tissue of 70 male CA patients, detected the DNA subtypes of HPV using the PCR-reverse dot hybridization technique, and analyzed the rates of different subtypes identified and their characteristics of distribution in different age groups.@*RESULTS@#The male HPV-positive patients were mainly infected at the age of 20-39 years, primarily with high- and low-risk mixed infection of various subtypes, which accounted for 61.54% in the 20- to 29-year-olds and 42.86% in the 30- to 39-year-olds. Among the 70 CA patients, 22 HPV subtypes were identified, the top five subtypes including HPV 11 (21.08%), HPV 6 (19.46%), HPV 42 (6.49%), HPV 59 (6.49%) and HPV 53 (5.95%); 20 infected with a single subtype (28.57%), 19 with two subtypes (27.14%) and 31 with three or more (44.29%); and 30 infected with a low-risk single subtype (42.86%) and 40 with both high- and low-risk multiple subtypes (57.14%).@*CONCLUSIONS@#Male patients with CA are mainly infected with HPV 11 and HPV 6, with a significantly higher rate of multi-subtype than single-subtype infection, and the multi-subtype patients chiefly with high- and low-risk mixed infection. Men aged 20-39 years old are most commonly affected by CA.

Objective To investigate the relationship between methylation of CpG island in glutathione S-transferase P1(GSTP1) promoter region and injury induced by isoniazid in HL-7702 cells.Methods HL-7702 cells were divided into the control group and three isoniazid groups(200,400,800 mg/L).Colorimetric method was used to detect the activity level of lactate dehydrogenase in the medium of HL-7702 cells;the mRNA expression of GSTP1,DNA methyltransferases 1(DNMT1),DNMT3a and DNMT3b were de?tected by real-time fluorescence quantitative PCR;the protein expression levels of DNMT1,DNMT3a and DNMT3b were detected by enzyme-linked immunosorbent assay;the methylation of the CpG island in the GSTP1 promoter region was determined by the bisulfite sequencing PCR.Results The activity level of lactate dehydrogenase in supernatants of the HL-7702 cells in isoniazid group(400, 800 mg/L)was higher than that in the control group(P<0.01).Compared with the control group,the mRNA expression of DNMT 1、3a、3b and GSTP1 were elevated in 400 and 800 mg/L isoniazid groups(P<0.05,P<0.01).The proteins expression of DNMT1 and 3a in the 400 and 800 mg/L isoniazid groups were higher than that in the control group(P<0.05,P<0.01),and the protein expression of DNMT 3b in the 800 mg/L isoniazid groups were higher than that in the control group(P<0.01).The methylation level of CpG is?land in GSTP1 promoter region of three isoniazid groups were decreased.Conclusion The CpG island in the promoter of GSTP1 has hypomethylation in hepatocyte cells damaged by isoniazid.

Objective:The process of myocardial infarction is generally characterized by the activation of host immune cells and the occurrence of inflammation. However, it is unknown which immune cells are preferentially activated and participated into the progression of myocardial infarction. Methods:A total of 55 patients with myocardial ischemia including 13 of stable angina ( SA) ,25 of unstable angina (UA) and 17 of acute myocardial infarction (AMI) as well as 12 of healthy controls (HCs) were enrolled in the study. The frequency and the immune activation marker CD38 expression by peripheral CD3 T cells,CD4 T cells,CD8 T cells,CD4+NKT cells, CD4- NKT cells, CD3-CD56+ NK cells and B cells were comprehensively analyzed. Results:There was no significant difference in the frequencies of these immune cell subsets in peripheral blood among these four groups. Importantly,it was found that CD38 expression was significantly increased on CD8 T cells,NKT cells and NK cells in patients with acute coronary syndromes ( ACS) including UA and AMI patients as compared with those in SA and HC subjects. These data indicated that multiple immune cells were activated in ACS patients,which were possibly participated into the pathogenesis of ACS. Conclusion:The activation of multiple immune cells was closely associated with the progression and outcome in ACS patients. This study provides immune hyper-activation mechanism underlying the development of ACS and may favor for finding a novel immune marker to predict the progression of ACS.

<p><b>OBJECTIVE</b>To study the features of blood supply and results of transarterial infusion and embolization in spinal metastases.</p><p><b>METHODS</b>Forty-one patients with spinal metastasis received transarterial infusion and embolization between March 2001 and June 2008. The inclusion criteria were: The metastatic lesion caused back pain; The metastatic lesion involved vertebra at or below T3 level. There were 29 males and 12 females with a mean age of 56.0 (33 - 71) years. Epirubicin was used as the chemotherapeutic agent. Lipoid Ultra-Fluid, Contour SE or gelfoam particles were used as embolitic material.</p><p><b>RESULTS</b>The technical success of therapy was achieved in 52 vertebrae (100%) including 14 thoracic, 35 lumbar and 3 sacral vertebrae. 105 arteries were used for infusion and embolization (16 intercostal arteries, 78 lumbar arteries, 4 iliolumbar arteries, 4 branches of iliac arteries, and 3 median sacral arteries). Lipoid Ultra-Fluid (2 - 8 ml) was used in 15, Contour SE (300 approximately 500 microm, 20 - 100 mg) in 20, and gelfoam particles in 33 arteries. Three days after treatment, complete pain relief (CR) was achieved in 17 patients, partial pain relief (PR) in 20, and moderate pain relief (MR) in 4, with an effective rate of 90.2%. Two weeks after treatment, CR was achieved in 17 patients, PR in 21, and MR in 3, with an effective rate of 92.7%. No adverse nervous system effect occurred. 16 patients developed swelling and pain of normal tissues which were alleviated after symptomatic treatment.</p><p><b>CONCLUSION</b>Transarterial infusion and embolization is an effective therapy in relieving pain resulting from spinal metastases.</p>

Objective: To determine whether there exist the cells expressing both Clara cell antigen (CCA) and surfactant protein C (SP-C) in different lung tissues from mouse, rat, and human by dual immunofluorescent labeling technique. The CCA+ and SP-C+ cells were named as the double positive cells (DPCs) which may provide the basis for further investigation of lung stem cells. Methods: The normal lung tissues were isolated from adult and newborn mice and rats. The human lung squamous carcinoma and adenocarcinoma tissues and para-cancerous lung tissues were isolated and subjected to cryosection and dual immunofluorescent staining. The DPCs in different lung tissues were identified by using laser confocal microscopy. For each specimen, four to six visual fields were selected and totally 100-200 cells were counted to calculate the percentage of DPCs. The data were analyzed by the SPSS 11.0 software. Results: The DPCs were found in normal lung tissues from adult and newborn mice and rats. DPCs were first discovered in human lung adenocarcinoma tissues but not found in lung squamous carcinoma tissues. Furthermore, the number of DPCs in the paracancerous tissues was significantly increased than the corresponding lung cancer tissues (P < 0.05). Conclusion: We not only validate the wide distribution of DPCs in the normal lung tissues, but also discover the existence of DPCs in the lung adenocarcinoma tissues and the number of DPCs in para-cancerous tissues is significantly higher than that in lung cancer tissues. Our study provide the experimental basis for further isolation and identification of DPCs and investigation of the biological features of normal lung stem cells and lung cancer stem cells.

Objective To cultivate,identify and sort bronchoalveolar stem cells(BASC)derived from normal adult mouse lung.Methods After enzymatic digestion of lung tissue with dispase and collagenase in combination,the Sca-1+ cells were isolated from the obtained pulmonary cells by magnetic cell sorting.These Sca-1+ cells were cultured in dishes coated with collagen and mouse fibroblast cell line Swiss-3T3 under a serum-free culture system for BASC,which were identified by the dual-color immunofluorescent staining clara cell specific antigen(CCA)and surfactant protein C(SP-C).Finally,these pure BASC were isolated by the flow cytometry.Results One lung of normal adult mouse could yield(1.6-1.8)?107 nucleated cells in this enzyme digestion procedure.The percentage of Sca-1+ cells we sorted from lung tissue was much higher than the unsorted [(87.3?5.9)% and(9.6?1.8)%,P

<p><b>OBJECTIVE</b>To investigate the manufacture method and the effects of Cosmopost ceramic posts, to summarize the indications of Cosmopost ceramic post in clinic.</p><p><b>METHODS</b>228 pieces Cosmopost ceramic posts and cores were made for 96 patients. 2l7 pieces full ceramic crowns and 8 pieces full ceramic bridgs for Cosmopost ceramic posts and cores were made. The fitness of Cosmopost ceramic post were checked, and the color of all full ceramic crowns and full ceramic bridgs were checked in clinic. The amount of loosening and broken in all restorations were checked every year in clinic. follow up 2 - 3 years.</p><p><b>RESULTS</b>1 Cosmopost ceramic post was fracture about 1.5 mm before the full ceramic crown was worn. 227 pieces Cosmopost ceramic posts were perfect. All of the full ceramic crowns and bridgs for full ceramic posts were excellent in color. No loosening and broken cases were checked in 0.5 - 3 years. But the indications of Cosmopost ceramic posts were strict in clinic.</p><p><b>CONCLUSION</b>Cosmopost ceramic post has natural aesthetic effect and good strength. it can be used in clinic.</p>

Objective To evaluate the treatment of hypervascular hepatic metastasis with TACE. Methods One hundred and twenty nine cases of hepatic metastasis treated by TACE were selected retrospectively and then analyzed the survival rate,clinical effectiveness and lipidol deposition quantity in tumor.Results Malformation of tumor vessels and rich blood supply were found in all cases of this study.The survival rates of 6 months,1 year and 3 years were 100%,73.6% and 26.4% respectively.The clinical effective rate was 68.2%(88/129)and no-progress rate was 23.3%(30/129).The satisfactory lipidol deposition quantity was obtained in 80.9%(97/129).Conclusions TACE is a favorable method for hepatic metastasis,and discerning the hypervascular subgroup could improve the treating effectiveness and be useful to make an appropriate planning.
