Résumé

Objective To investigate the protective effect of excretory-secretory protein (AES) from adult Trichinella spiralis on ovalbumin (OVA)-induced allergic rhinitis in mice. Methods Eighteen female BALB/c mice were randomly divided into three groups, including the blank control group (Group A), OVA-induced rhinitis group (Group B) and AES treatment group (Group C). Mice in Group A were given PBS. Mice in Group B were intraperitoneally injected with antigen adjuvant suspension for systemic sensitization, once every other day for seven times; then, local excitation was intranasally induced with 5% OVA solution once a day for seven times to establish a mouse model of allergic rhinitis. In addition to induction of allergic rhinitis, mice in Group C were given 25 μg AES at baseline sensitization and local excitation. Following the final challenge, mice were observed for 30 min in each group, and the behavioral score was evaluated. The serum levels of IFN-γ, IL-4, IL-5, IL-10 and TGF-β were determined using an enzyme-linked immunosorbent assay in mice, and the pathological changes of mouse nasal mucosa were observed under a microscope. Results There was a significant difference in the mouse behavioral scores among the three groups (F = 110.12, P < 0.01). The mouse behavioral score was significantly higher in Group B than in Group A (7.17 ± 0.75 vs. 1.33 ± 0.52, P < 0.01), and more remarkable pathological damages of mouse nasal mucosa were seen in Group B than in Group A, while the mouse behavioral score was significantly decreased in Group C than in Group B (P < 0.01), and the pathological damages of mouse nasal mucosa remarkably alleviated in Group C relative to Group B. There was a significant difference in serum IFN-γ level among the three groups (F = 7.50, P < 0.01) and the serum IFN-γ level in Group B was significantly lower than in group A and C (both P < 0.05). There were significant differences in serum IL-4 (F = 470.81, P < 0.01) and IL-5 levels (F =68.20, P < 0.01) among the three groups, and significantly greater serum IL-4 and IL-5 levels were detected in Group B than in Group A (P < 0.01), while significantly lower serum IL-4 and IL-5 levels were detected in Group C than in Group B (P < 0.01). There were significant differences in serum IL-10 (F = 174.91, P < 0.01) and TGF-β levels (F = 9.39, P < 0.01) among the three groups, and significantly greater serum IL-10 and TGF-β levels were seen in Group C than in Group B (both P < 0.05). Conclusion T. spiralis AES has a remarkable protective activity against OVA-induced allergic rhinitis in mice.

5-HT as an important neurotransmitter which takes part in the development of many diseases, including bronchial asthma,schizophrenia and bone metabolism as reported in literatures in recent years. Central 5-HT can inhibit sympathetic excitations and promote bone formation through Tph2. Lrp5 can reduce the level of peripheral 5-HT and act on Tph1 to promote bone formation. Peripheral 5-HT blocks the association between transcription factor FOXO1 and CREB and inhibits bone formation. Peripheral 5-HT can also promote IL-6 expression and induce bone reabsorption. LP533401 as a Tph1 inhibitor could effectively reduce the level of peripheral 5-HT and induce bone formation as reported. In this paper, we try to summarize the effect of 5-HT on bone metabolism and its mechanism, and further using 5-HT to regulate bone quantity effectively.

Objective To investigate the effect of cysteine protease inhibitor derived from S chistosoma japonicum(SjCys-tatin)on dextran sodium sulfate(DSS)-induced acute ulcerative colitis in mice.Methods Eighteen C57BL/6 mice were ran-domly divided into three groups:a control group treated with PBS(Group A),a DSS-induced-colitis group treated with PBS(Group B),and a DSS-induced-colitis group treated with SjCystatin(Group C).Colitis was induced in mice by giving 3%DSS orally for 7 days.During this period,the mice were daily injected with 10μg of SjCystatin or PBS only as a control intraperitone-ally.The mice were monitored daily for their clinical manifestations and given scores based on disease activity index(DAI).The severity of colonic inflammation was monitored by the macroscopic score and pathological change.The cytokine profile including TNF-α,IL-4,IL-6 and IL-10 in the supernatants of colon homogenate was detected by ELISA.Results Compared with Group A(0.50 ± 0.28),the DAI score increased significantly in Group B(9.30 ± 1.30)(F=86.86,P<0.01),with remarkable path-ological damages seen in colon tissues.and the levels of TNF-α and IL-6 were(321.33±67.01)and(403.58 ±180.51)pg/mL.The DAI score significantly reduced in Group C(6.67±1.57)as compared to Group B(F=86.86,P<0.01),with improve-ments in the macroscopic and microscopic pathology in mouse colon specimens.As compared to Group B,the levels of TNF-α [(188.14 ± 40.14)pg/mL] and IL-6 [(209.71 ± 48.47)pg/mL] significantly decreased(F=17.46 and 9.89,both P<0.01).Con-clusion SjCystatin has a significantly inhibitory effect for alleviating DSS-induced acute ulcerative colitis in C57BL/6 mice.

Objective The relationship between osteoblast autophagy induced by wear particles and secretion of inflammatory cytokines has not been illuminated.The purpose of this study was to investigate the effect of wear particles on the secretion of TNF -α and IL-6 by autophagy of osteoblasts in aseptic loosening. Methods The mouse MC3T3-E1 osteoblasts in logarithmic growth phase were divided into 5 groups:control group(in α-MEM medium),CoCrMo particle group(100 μg/mL CoCrMo particles was added in α-MEM medium),3-MA group(10 mmol/L autophagy inhibitor 3-MA prestimulated osteoblasts in α-MEM medium,3h later 100 μg/mL CoCrMo particles were added),the expression of autophagy marker protein LC-3 in osteoblasts was detected by Western blotting, and the levels of TNF-αand IL-6 were detected by ELISA. Results Compared with the control group,the expression level of auto-phagy marker protein LC-3 was significantly increased in CoCrMo particles group(P<0.05).Compared with CoCrMo particles group,the expression level of LC-3 in 3-MA group was significantly decreased(P<0.05).Compared with control group TNF-α[(0.95± 0.00)pg/mL]and IL-6[(4.67±0.23)pg/mL], CoCrMo particles group TNF-α[(10.35±3.63)pg/mL]and IL-6[(14.27±1.07)pg/mL]secretion level was significantly increased(P<0.05),compared with CoCrMo particles group,the effects of 10 mmol/L 3-MA group to TNF-α[(13.13±5.08)(14.71±5.46)pg/mL]and IL-6[(15.31±0.88),(16.75±3.87)pg/mL]secretion were not significant, while 10 mmol/L 3-MA group TNF-α[The secretion levels of(302.61±45.63)pg/mL]and IL-6[(216.67±27.71)pg/mL]in-creased significantly(P<0.01). Conclusion The osteoblast autophagy induced by CoCrMo particles can inhibit the secretion of in -flammatory cytokines TNF-αand IL-6 in osteoblasts.

Objective@#To investigate the effect of modified Bianchi (single incision in the midline of the scrotum) orchiopexy (MBO) versus that of traditional surgery in the treatment of median or low cryptorchidism.@*METHODS@#Eighty-two children with median or low cryptorchidism were treated from February 2013 to February 2014, 46 (53 testes) by MBO and the other 36 by the traditional method of inguinal incision (control, 40 testes). Comparisons were made in the operation time and postoperative complications between the two surgical strategies.@*RESULTS@#The mean operation time was significantly shorter in the MBO group than in the control ([25±6] vs [35±4] min, P0.05). The incision scar was obvious in all the controls, with 1 case of postoperative inguinal hematoma, but almost invisible in all the MBO cases.@*CONCLUSIONS@#Modified Bianchi orchiopexy is superior to traditional surgery in the treatment of median or low cryptorchidism for its advantages of short operation time, few complications, and satisfactory appearance of the healed incision.

<p><b>OBJECTIVE</b>To compare the effect of transumbilical single-site single-port with that of transumbilical single-site double-port laparoscopic varicocelectomy in the treatment of varicocele in adolescents.</p><p><b>METHODS</b>We randomly assigned 80 varicocele patients aged 10 - 16 years to two groups of equal number to receive transumbilical single-site single-port and single-site double-port laparoscopic varicocelectomy, respectively. We compared the operation time, postoperative hospital stay, incisional pain, complications and satisfaction with the abdominal cosmetic outcomes between the two groups.</p><p><b>RESULTS</b>All the operations were successfully performed. The double-port group showed a significantly higher score on the Visual Analogue Scale than the single-port group (4.8 +/- 1.4 vs 3.6 +/- 1.1, t = -4.986, P < 0.01), but there were no significant differences between the two groups in the operation time ([29.8 +/- 4.2] vs [31.2 +/- 4.6] min, t = 1.383, P = 0.171), postoperative hospital stay ([1.95 +/- 0.7] vs [1.82 +/- 0.8] d, t = -0.784, P = 0.436), complications (0 vs 0) and scores on the satisfaction with abdominal cosmetic outcomes (4.6 +/- 0.6 vs 4.8 +/- 0.5, t = 1.253, P = 0.214). No recurrence, umbilical hernia, hydrocele and orchiatrophy were found in the two groups of patients at 6 months after operation, and no visible scar was observed on the abdominal surface.</p><p><b>CONCLUSION</b>With strict surgical indications, single-site single-port and single-site double-port laparoscopic varicocelectomies have similar clinical effects in the treatment of varicocele, which leave no scar on the abdominal surface. Single-site double-port laparoscopy needs no special instruments and therefore is worthier of wide clinical application.</p>

<p><b>OBJECTIVE</b>To establish the method of detecting the concentrations of chlorpyrifos in air of workplace with high performance liquid chromatographic (HPLC).</p><p><b>METHODS</b>According to standards of methods for determining the chemical substances in workplace air, chlorpyrifos in the air was collected by silicone tube, then dissolved by acetonitrile and determined by high performance liquid chromatography with UV-detector.</p><p><b>RESULTS</b>There was a linear relationship within the range of 0 ∼ 10.0 µg/ml, and regression equation was y = 5206.1x - 104.7, correlation coefficient was 0.9999, the detection limit was 0.006 µg/ml. The lowest detected concentration was 0.001 mg/m(3) (sampling volume 4.5 L). The average recoveries was 98.3% ∼ 102.5%. The within-run precision was 1.96% ∼ 4.39%, the between-run precision was 2.76% ∼ 5.87%. The desorption efficiencies were 99.0% ∼ 103.3% and the sampling efficiencies were 94%. The samples in silicone tube could be stored for 15 days at room temperature.</p><p><b>CONCLUSION</b>The present method could meet with the requirements of Guide for establishing occupational health standards-Part 4 Determination methods of air chemicals in workplace and be feasible for determination of chlorpyrifos in workplace air.</p>

As the progress of organ and tissue transplantation technology, pancreas and islet transplantation has been introduced to treat diabetes and became hot. However, many shortcomings and re-lated issues need to be improved concerning the islet cell sources. Bone marrow mesenchymal stem cells (BMSCs) has become the hot topic on induction of islet-like cell because it was simple to access, easy to culture and possess the function of immunosuppressian. In recent years, artificial construction of pancreatic islet cells, which uses gene recombinant and gene transfection to reforge islet-like cells, has improved the induction rate of BMSCs and increased the amount of insulin serection, and it also provide new ideas and direction for the treatment of diabetes.

Objective To reveal and forecast the incidence trend of Brucellosis, in order to provide acientific basis for future intervention and policy-making. Methods Descriptive epidemiological method was used to analyze and statistically describe the distribution of the disease in different times, different locations and different (7.0783/10 million to 13.1257/10 million) and Qingxu ( 1.4811/10 million to 8.5241/10 million) were higher,followed by Yangqu county(0 to 5.8232/10 million), Xiaodian(0.8108/l0 million to 2.4229/10 million) and Jinyuan district ( 0.5329/ 10 million to 1.5896/10 million), and the remaining counties(districts) in the annual There were 223 cases of Brucellosis patients from 2006 to 2009 in Taiyuan. Vocational high risk population was farmers, with a total of 140 cases, accounting for 62.78% of the total number of incidence, followed by students and workers, respectively, 13, 14 cases, accounting for 5.83% and 6.28%, other occupational groups, 56 cases,77.58%;28 cases aged above 60 years, accounting for 12.56%;22 cases aged younger than 19 years, accounting identical in the four years, most cases occurred in spring and summer and showing a clear seasonal high.Conclusions The incidence trend of Brucellosis is on the rise from 2006 to 2009. High risk population is farmer,and the number of younger patients is on the rise, we propose strengthen protection for high risk groups.

Résumé

Objective To study the effect of Coxsackie virus B3 (CVB3) infection on islet cells in vitro, and to explore the mechanism of islet cells caused by CVB3. Methods Bone marrow mesenchymal stem cells( BMSCs) were separated from the bone marrow and cultured. Then they were induced to differentiate into islet-like cells using nicotinamide and mercaptoethanol. Differentiated cells were detected by morphology , special staining and RT-PCR. Observe CVB3 infection on islet cells under inverse microscope and detect the specific gene fragment by RT-PCR. Results BMSCs showed half suspended shape and gathered to form a cluster after induction. Cells became red brown by dithizone specific staining. RT-PCR also proved the existence of mRNA expressing insulin. Infected islet cells appeared typical pathological changes like shrinks, refraction decreases. RT-PCR detected the desired specific gene fragment of 299 bp in infected islet cells. Conclusion CVB3 can directly injury islet cells, and damage the function of islet cells of secreting insulin.

Four hundreds and ninety sheep sera from seven breeds raised at eight counties and one city of Aksu region in Xinjiang were tested by ELISA for the presence of antibodies against hepatitis E virus (HEV), and the positive rate differences were compared based on administrative areas, breeds and age by Chi-square test. The result showed that the general positive rate was 28.98% (142/490), the positive rate were 35.44% (28/79), 29.67% (27/91), 20% (4/20), 40% (12/30), 32.5% (26/80), 38% (19/50), 22.5% (9/40), 8% (4/50) and 26% (13/50) respectively in eight counties and one city, there was a significant difference between Xayar and other administrative areas (P<0.01); there was also a significant difference among age ranges (P<0.01), being 38.75% (31/80) over 2 years old, 15.45% (17/110) below 1 year old; The seroprevalence was still related to breeds, i. e. there was a significant difference between Mongolia sheep and other breeds (P<0.01). From these data, it is confirmed that there is a possibility of previous and potential infection of sheep HEV in Aksu region of Xinjiang Autonomous.

<p><b>OBJECTIVE</b>To study the clinical efficacy of Shufeng Liangxue Decoction (SLD) in treating hormone dependence dermatitis (HDD).</p><p><b>METHODS</b>One hundred and sixteen patients with HDD were randomly assigned to two groups. Both were treated with symptomatic Western medical therapy, including oral taken of loratadine 10 mg, and intravenous injection of 10% calglucon 10 mL and vitamin C 3.0 g adding in 20 mL 50% glucose, once per day, and to the test group, one dose of SLD consisting of imperata rhizome 30 g, rehmannia root 30 g, moutain bark 15 g, schizonepeta spike 15 g, divaricate saposhnikovia root 10 g, scutellaria root 15 g, forsythia fruit 15 g, cape-jasmine fruit 10 g, red peony root 10 g, white-stiff silkworm 10 g, broom cypress fruit 15 g, Indian bread 15 g and licorice root 10 g, was given every day by decocting with water. The course of treatment was 4 weeks. Symptoms of patients, including redden-swollen skin, capillary dilatation, inflammatory papula, itching and other discomfort sensation (pain, burning, dry, buckled) were scored before and after treatment according to their severity. The efficacy was evaluated by the change in scores of symptoms, the adverse reactions occurring in the therapeutic course and the relapse rate after treatment were observed as well.</p><p><b>RESULTS</b>The cure-obvious effective rate was 76.32% in the test group and 42.50% in the control group; the total effective rate in them was 94.74% and 77.50% respectively, all showing significant difference (P < 0.05) between groups. In the test group, slight diarrhea appeared in 5 patients and slight nausea with upper abdominal discomfort in 3; while in the control group, slight somnolence appeared in 2, but all these adverse reactions did not affect the treatment. A 3-month follow-up study showed that the relapse rate in the test group was significantly lower than that in the control group (16.00% vs. 42.50% , P < 0.01).</p><p><b>CONCLUSION</b>SLD is effective and safe in treating HDD, with the efficacy better and relapse rate lower than those of treatment with Western medicine alone.</p>

<p><b>OBJECTIVE</b>To assess the feasibility, safety and outcome of anatomical laparoscopic left lateral hepatic lobectomy for benign and malignant liver tumors.</p><p><b>METHODS</b>From April 2005 to May 2008, 11 patients (7 male, 4 female; mean age 51.7 years) underwent anatomical laparoscopic left lateral hepatic lobectomy. Four patients presented with hepatocellular carcinoma and cirrhosis, while 1 patient had metastatic liver tumors from postoperatively colon cancer, five patients had hemangioma (2 cases with gallstones underwent cholecystectomy), 1 patient had a huge symptomatic angiolipoleiomyoma. Mean tumor size was 5.8 cm (range 2.1 to 12.0 cm). All the lesions were localized in the anatomical left lateral lobe (segments II to III).</p><p><b>RESULTS</b>The mean operative time was 147 min (range 120 to 180 min). There were no intraoperative or postoperative complications, and blood transfusions were not required. The mean postoperative hospital stay was 5.9 days.</p><p><b>CONCLUSIONS</b>Anatomical laparoscopic left lateral hepatic lobectomy are feasible and safety.</p>

<p><b>OBJECTIVE</b>To investigate the correlation between the expression of the kinase insert domain-containing receptor (KDR) and the histological grade of prostate adenocarcinoma.</p><p><b>METHODS</b>Forty-eight samples of prostate adenocarcinoma tissues and 20 samples of benign prostatic hypertrophy (BPH) tissues were studied by LsAB immunohistochemical staining.</p><p><b>RESULTS</b>The positive expression rate of KDR was 73% in prostate adenocarcinoma and 30% in BPH. The expression of KDR was stronger in prostate adenocarcinoma, and there was no relationship between staining intensity and the histological grade of carcinoma.</p><p><b>CONCLUSION</b>KDR, expressed more highly in prostate adenocarcinoma, promises to be a new target in the treatment of prostate adenocarcinoma.</p>

Résumé

The objective of the study is to find out the effect of shark chondroitin on T lymphocyte subsets in cancer patients. Patients were divided into two groups. One group was treated with chemotherapy alone, and the other group was treated with chemotherapy plus shark chondroitin. Using immunofluorescence technique, T lymphocyte subsets in the peripheral blood were determined in two groups before and after chemotherapy. The results showed that CD4(+)/CD8(+) ratio increased in the patients received shark chondroitin. In the chemotherapy group, CD3(+) had no change, but CD4(+) decreased while CD8(+) increased significantly. The results suggest that shark chondroitin could enhance immune function in cancer patients, especially during chemotherapy.
