Résumé

Abstract Visceral leishmaniasis (VL) is an infectious disease predominant in countries located in the tropics. The prediction of occurrence of infectious diseases through epidemiologic modeling has revealed to be an important tool in the understanding of its occurrence dynamic. The objective of this study was to develop a forecasting model for the incidence of VL in Maranhão using the Seasonal Autoregressive Integrated Moving Average model (SARIMA). We collected monthly data regarding VL cases from the National Disease Notification System (SINAN) corresponding to the period between 2001 and 2018. The Box-Jenkins method was applied in order to adjust a SARIMA prediction model for VL general incidence and by sex (male or female) for the period between January 2019 and December 2013. For 216 months of this time series, 10,431 cases of VL were notified in Maranhão, with an average of 579 cases per year. With regard to age range, there was a higher incidence among the pediatric public (0 to 14 years of age). There was a predominance in male cases, 6437 (61.71%). The Box-Pierce test figures for overall, male and female genders supported by the results of the Ljung-Box test suggest that the autocorrelations of residual values act as white noise. Regarding monthly occurrences in general and by gender, the SARIMA models (2,0,0) (2,0,0), (0,1,1) (0,1,1) and (0,1,1) (2, 0, 0) were the ones that mostly adjusted to the data respectively. The model SARIMA has proven to be an adequate tool for predicting and analyzing the trends in VL incidence in Maranhão. The time variation determination and its prediction are decisive in providing guidance in health measure intervention.

Effects of aqueous seed extracts of Sphenostylis stenocarpa on the reproductive indices of male rats were investigated. A total of 104 adult rats were used for the experiment, and were divided into 4 groups (group A – D) and replicated in triplicate. Group A served as the normal control, while groups B, C and D received three graded doses (800mg/kg, 1200mg/kg and 1600mg/kg) of the extracts, respectively, by oral intubation. The gonad characteristics, sperm parameters and hormonal analyses of the male rats were determined using standard procedures. These were ascertained prior to the commencement of treatment, and on weekly basis. Data were analyzed statistically using SPSS and R software at 95% confidence interval. An overall dose and time dependent showed significant differences in the mean weekly gonad characteristics of the male rats in the treatment groups when compared with the control. There was a significant reduction (p < 0.05) in the body weights of the male rats, but a significant increase (P < 0.05) in the testes weights, gonad somatic index, sperm count and sperm motility in the rats. The gonadal hormone testosterone, responded to the plant extracts, while follicle-stimulating and luteinizing hormones were largely undetected. There were significant increases in the testosterone levels of all the treated rats. Conclusively, aqueous seed extracts of Sphenostylis stenocarpa seems to possess ability to enhance reproductive health in male rats.

Résumé

The impact of linseed oil as a lipid source on liver disease induced by a high-carbohydrate diet (HCD) was evaluated. Adult male Swiss mice received an HCD containing carbohydrates (72.1%), proteins (14.2%), and lipids (4.0%). The Control HCD group (HCD-C) received an HCD containing lard (3.6%) and soybean oil (0.4%) as lipid sources. The L10 and L100 groups received an HCD with 10 and 100% linseed oil as lipid sources, respectively. A group of mice were euthanized before receiving the diets (day 0) and the remaining groups after 56 days of receiving the diets (HCD-C, L10, and L-100 groups). Morphological and histopathological analyses, as well as collagen deposition were evaluated. Perivenous hepatocytes (PVH) of the HCD-C group were larger (P<0.05) than periportal hepatocytes (PPH) in the median lobe (ML) and left lobe (LL). There was a greater (P<0.05) deposition of type I collagen in PPH (vs PVH) and in the ML (vs LL). The ML exhibited a higher proportion of apoptotic bodies, inflammatory infiltrate, and hepatocellular ballooning. All these alterations (hepatocyte size, deposition of type I collagen, apoptotic bodies, inflammatory infiltrate, and hepatocellular ballooning) induced by HCD were prevented or attenuated in L10 and L100 groups. Another indicator of the beneficial effects of linseed oil was the lower (P<0.05) number of binucleated hepatocytes (HCD-C vs L10 or L100 group). In general, the L100 group had greater effects than the L10 group. In conclusion, linseed oil impedes or reduces the liver injury progression induced by an HCD.

Résumé

Leishmaniasis is a neglected disease that affects millions of people worldwide, and special attention should be given to treatment because the available drugs have limitations, which can lead to low therapeutic adherence and parasitic resistance. This study evaluated the activity of the bioactive naphthoquinones, lapachol and β-lapachone, against Leishmania amazonensis. The cell alterations were evaluated in vitro on promastigote and amastigote forms. The lethal dose (LD50) at 24, 48, and 72 h on the promastigote's forms using lapachol was 75.60, 72.82, and 58.85 μg/mL and for β-lapachone was 0.65, 1.24, and 0.71 μg/mL, respectively. The naphthoquinones significantly inhibited the survival rate of L. amazonensis amastigotes at 83.11, 57.59, and 34.95% for lapachol (82.28, 41.14, and 20.57 µg/mL), and 78.49, 83.25, and 80.22% for β-lapachone (3.26, 1.63, and 0.815 µg/mL). The compounds on the promastigote's forms led to the loss of mitochondrial membrane potential, induced changes in the integrity of the membrane, caused damage to cells suggestive of the apoptotic process, and showed inhibition of tumor necrosis factor (TNF)-α and interleukin (IL)-6 production. The results showed that these naphthoquinones are promising candidates for research on new drugs with anti-Leishmania activity derived from natural products.

Purpose: To describe the etiology, clinical profile, duration of lagophthalmos cases and thereby, framing a decision for the management based on the severity of Exposure keratitis (EK), Facial palsy (FP) with each etiology and to describe the outcome of the management options. Methods: The method was a prospective review of 120 lagophthalmos cases treated at a single tertiary center from January 2018 to January 2019. The main outcome measures were analysing the association between age, etiology, duration and management of lagophthalmos. Results: Of the 120 patients studied, paralytic etiology was noted in 86 and eyelid etiology in 34 patients. The percentage of various lagophthalmos etiology documented were Bell’s palsy (35.83%), lagophthalmos in ICU patients (15%), traumatic facial palsy(FP) (10.80%), stroke associated FP (6.67%), infection associated FP (6.67%), iatrogenic FP, cicatricial lagophthalmos (5%), lagophthalmos post eyelid surgeries (5%), neoplastic FP(3.33%), congenital FP (1.67%), proptosis induced lagophthalmos (1.67%), floppy eyelid syndrome induced lagophthalmos (0.83%) and lid coloboma associated lagophthalmos (0.83%). A statistically significant correlation was noted between exposure keratitis and age, with an increased prevalence age advances. The management showed significant variation with individual etiology, with some etiologies unquestionably requiring surgical management. Surgical management is crucial as the duration of lagophthalmos increases more than 6 weeks, EK involving pupillary axis and poor FP recovery. Conclusion: This study concludes that the conservative management was sufficient in all cases when the duration is less than 1 week, Exposure keratitis not involving the pupillary axis (EK< Grade II) and FP with good functional recovery ( FP < Grade III). The predominant causes being Bell’s palsy, lagophthalmos in ICU patients and vascular FP. Whereas, cases with poor functional recovery of facial palsy(FP) and permanent eyelid deformation require definitive surgical management like Traumatic FP & cicatricial lagophthalmos

Résumé

2,4-Dichlorophenoxyacetic acid (2,4-D) is a herbicide of the chlorophenoxy class and the second most widely used herbicide applied to several different crops worldwide. Environmental factors, especially those related to diet, strongly affect the risk of developing cancer of the gastrointestinal tract. There is currently no evidence to determine whether there is an association between 2,4-D exposure and gastrointestinal disorders. We evaluated the histological effect of chronic oral and inhalation exposure to 2,4-D on the digestive tract of rats. Eighty male adult albino Wistar rats were divided into 8 groups (n=10): two control groups, one for inhalation and one for oral exposure, and 6 groups exposed orally or by inhalation at three different concentrations of 2,4-D [3.71×10-3 grams of active ingredient per hectare (gai/ha), 6.19×10-3 gai/ha, and 9.28×10-3 gai/ha]. The animals were exposed for 6 months. The esophagus, stomach, and intestine were collected for histopathological analysis. Animals exposed to 2,4-D had hyperkeratosis of the esophagus, regardless of the exposure route. All animals exposed to a higher concentration of 2,4-D orally presented mild dysplasia of the large intestine. In the small intestine, most animals exposed to moderate and high concentrations of 2,4-D had mild dysplasia. No gastric changes were observed in any of the groups studied. Chronic exposure to 2,4-D, especially at moderate and high concentrations, regardless of the exposure route, caused reactive damage to the esophagus (hyperkeratosis) and dysplastic changes to the intestine.

Résumé

Adipose tissue-derived mesenchymal stromal/stem cells (ASCs) are considered important tools in regenerative medicine and are being tested in several clinical studies. Porcine models are frequently used to obtain adipose tissue, due to the abundance of material and because they have immunological and physiological similarities with humans. However, it is essential to understand the effects and safe application of ASCs from pigs (pASCs) as an alternative therapy for diseases. Although minipigs are easy-to-handle animals that require less food and space, acquiring and maintaining them in a bioterium can be costly. Thus, we present a protocol for the isolation and proliferation of ASCs isolated from adipose tissue of farm pigs. Adipose tissue samples were extracted from the abdominal region of the animals. Because the pigs were not raised in a controlled environment, such as a bioterium, it was necessary to carry out rigorous procedures for disinfection. After this procedure, cells were isolated by mechanical dissociation and enzymatic digestion. A proliferation curve was performed and used to calculate the doubling time of the population. The characterization of pASCs was performed by immunophenotyping and cell differentiation in osteogenic and adipogenic lineages. The described method was efficient for the isolation and cultivation of pASCs, maintaining cellular attributes, such as surface antigens and multipotential differentiation during in vitro proliferation. This protocol presents the isolation and cultivation of ASCs from farm pig as an alternative for the isolation and cultivation of ASCs from minipigs, which require strictly controlled maintenance conditions and a more expensive process.

Résumé

Glial cells have been implicated in temporal lobe epilepsy in humans and in its models. Astrocytes are lost in several brain regions after acute seizures induced by pilocarpine and may suffer hyperplasia at subsequent time points. This study investigated the effect of N-methyl-(2S,4R)-trans-4-hydroxy-L-proline (NMP) on astrocytes exposed to cytotoxic concentrations of pilocarpine. Astrocytes were incubated with pilocarpine (half maximal inhibitory concentration (IC50)=31.86 mM) for 24 h. Afterwards, they were treated with NMP at concentrations ranging from 3.12 to 100 μg/mL for 24 h. Cell viability was assessed by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Cytoplasmic reactive oxygen species (ROS) and mitochondrial transmembrane potential (ΔΨm) were analyzed by flow cytometry using 2',7'-dichlorofluorescein diacetate (DCFH-DA) and rhodamine-123 (Rho123), respectively. Expression of glial fibrillary acidic protein (GFAP) and voltage-dependent anion channel-1 (VDAC-1) were measured by western blot. Pilocarpine significantly decreased cell viability and mitochondrial potential and increased ROS concentration significantly by 6.7 times compared to the control. NMP concentrations ≥25 µg/mL protected astrocytes against pilocarpine-induced injury in a concentration-dependent manner. Concomitantly, NMP reduced cytoplasmic ROS accumulation to 27.3, 24.8, and 12.3% in the groups treated with 25, 50, and 100 µg/mL NMP, respectively. NMP also protected mitochondria from pilocarpine-induced depolarization. These effects were associated with improvement of pilocarpine-induced GFAP and VDAC-1 overexpression, which are important biomarkers of astrocyte dysfunction. In conclusion, the improvement of ROS accumulation, VDAC-1 overexpression, and mitochondrial depolarization are possible mechanisms of the NMP protective action on reactive astrocytes.

Abstract Aim: Prejudice against women's futsal players is reported in the literature. The perception of prejudice in university futsal may be lower due to the higher educational level of the players and a more open-minded context. This study aimed to describe the experience of young women university futsal players and to explore their perspective on gender prejudice in that sports practice. Methods: Ten women university futsal players (aged 18 to 30 years old) were interviewed. Data were produced and analyzed following a grounded theory approach. Results: The results showed that the athletes perceived they suffered gender prejudice for playing futsal, however, in the university context this was attenuated. The participants experienced diversified practice before specializing in futsal and felt that the support of male family members and friends was important to their engagement in futsal and soccer in childhood and adolescence. However, they also suffered from prejudice against their futsal practice coming from family and friends, struggling with the constant association between futsal practice and hegemonic masculinity. Conclusion: The participants of this study perceived less prejudice in university women's futsal in comparison to other sports contexts. Thus, the university context may have the potential to boost the practice of futsal among women.

The disease caused by the new coronavirus (SARS-CoV-2), denominated COVID-19, has spread rapidly across five continents, and has been causing serious health and socioeconomic implications for the affected countries. Although most affected individuals have mild illness, vulnerable patients can have interstitial pneumonia with serious or even fatal complications. The objective of the present qualitative and quantitative study was to collect and analyze, under a homeopathic perspective, the symptoms of COVID-19, as well as to identify the homeopathic remedies most similar to the symptomatic picture of the disease (epidemic genius method). Forty-six individuals from four Brazilian regions (Fortaleza, Campo Grande, Florianópolis and São Paulo) were interviewed by homeopathic physicians. There was a consistent pattern of symptoms between the different centers, with little difference in frequency and intensity of symptoms. The symptomatic analysis of the entire sample, with the aid of different repertoire techniques, indicated the homeopathic remedies Arsenicum album, Phosphorus and Bryonia alba as the epidemic genius for this COVID-19 epidemic. Our results may provide support information for the intervention experiments assessing the effectiveness of Homeopathy in the prevention and treatment of COVID-19.

Résumé

Seizures are a disorder caused by structural brain lesions, life-threatening metabolic derangements, or drug toxicity. The present study describes the behavior related to proconvulsant activity induced by thiocolchicoside (TCC) in rats and investigates the electrocorticographic patterns of this behavior and the effectiveness of classic antiepileptic drugs used to control these seizures. Forty-nine adult male Wistar rats were used and divided into two phases of our experimental design: 1) evaluation of seizure-related behavior and electrocorticographic patterns induced by TCC and 2) evaluation of the efficacy of classical antiepileptic drugs to control the proconvulsive activity caused by TCC. Our results showed that TCC induced tonic-clonic seizures that caused changes in electrocorticographic readings, characteristic of convulsive activity, with average amplitude greater than that induced by pentylenetetrazole. Treatment with anticonvulsants, especially diazepam, reduced the electrocorticographic outbreaks induced by TCC. The results suggested that TCC caused seizures with increased power in brain oscillations up to 40 Hz and that diazepam may partially reverse the effects.

Résumé

More information is needed on asthma control and health-related quality of life (HRQoL) in smokers with severe asthma. The main study objective was to characterize the association of HRQoL and disease control with cigarette smoking in individuals with severe asthma. A secondary objective was to analyze subject characteristics according to asthma onset: asthma that developed before smoking initiation versus asthma that developed after smoking initiation. This cross-sectional study included subjects with severe asthma aged 18-65 years. HRQoL was assessed using the Asthma Quality of Life Questionnaire (AQLQ) and asthma control was assessed using the Asthma Control Test (ACT) and Global Initiative for Asthma (GINA) criteria. Of the 87 patients studied, 58 (66.7%) were classified as asthmatics who had never smoked and 29 (33.3%) as asthmatics with smoking exposure. The proportion of subjects with uncontrolled asthma was higher in the asthma and smoking group (GINA criteria: P=0.032 and ACT criteria: P=0.003. There were no between-group differences in overall AQLQ score (P=0.475) or AQLQ domain scores (P>0.05). Fifty-eight subjects (66.7%) were nonsmokers, 20 (23%) had asthma onset before smoking, and 9 (10.3%) had asthma onset after smoking. Asthma onset before smoking was associated with uncontrolled asthma (P=0.013). Résumé

A four-week feeding trial on the simultaneous replacement of 0 to 50% of soybean meal and 0 to 100% of soybean oil on the growth performance, carcass composition and profitability was conducted using 45 randomly chosen one-week-old growing Japanese quail. The five experimental diets were; D0 = Diet containing 20% soybean meal and 4% soybean oil (control diet), D1 = Diet, where 12.5% of soybean meal and 25% of soybean oil has been replaced by RSC; T2 = Diet, where 25% of soybean meal and 50% of soybean oil has been replaced by RSC; T3 = Diet, where 37.5% of soybean meal and 75% of soybean oil has been replaced by RSC; T4 = Diet, where 50% of soybean meal and 100% of soybean oil has been replaced by RSC. Results demonstrated that in raw rubber seed the proportion of kernel to hull is 64: 36. Proximate components of the boiled and sundried full fat rubber seed kernel (RS) were, moisture = 96.6%, ME = 5305 kcal / kg DM, CP = 17.6%, EE = 51.05%, CF = 8.5%, NFE = 18.25% and Ash = 4.6%. The body weights of the birds at 35 days of age in all dietary treatments were statistically similar. The feed intake of the quail among different dietary groups was varied significantly (p < 0.05) and there was a tendency of decreasing the feed intake at 25% and above inclusion level of RSC. The body weight gain and FCR were also statistically similar in all dietary groups. The feed cost and total production cost /kg BW of quail was also found to become steadily higher (p> 0.05) at higher RSC inclusion level. The muscle development was found to better at lower (%) RSC level, whereas the organs were enlarged at higher RSC inclusion. Results on the majority of the production parameters, and economics and carcass yield parameters suggested that the simultaneous replacement of maximum 50% soybean oil 25% soybean meal by processed RSC might be suggested, particularly, there will be a crisis in the availability of these two ingredients.

Several studies emphasize the use of owl pellets in small mammal inventories in natural areas harboring high richness of rare species, but few Brazilian Atlantic forest localities have been surveyed by this method. The present study documents the species composition and abundance of small mammals in the diet of Tyto furcata in an urban area of the municipality of Campos dos Goytacazes, Rio de Janeiro, Brazil, remarking on a new record of the dwarf mouse opossum genus Cryptonanus in the Atlantic forest. We analyzed 265 pellets regurgitated by a pair of T. furcata from November 2016 to September 2017 found in a nesting box. Analysis of the samples enabled finding a total of 596 individuals of four small mammal species. Mus musculus was predominant among the prey items (98.3%), while the native rodents Necromys lasiurus (1.3%) and Holochilus brasiliensis (0,17%) were much rarer. A single specimen of Cryptonanus sp. was identified among the diet items based on distinctive dental characters. The identification of this genus in the present study represents the second record in the state of Rio de Janeiro, and the sixth in the Atlantic Forest biome, suggesting that this marsupial occupies a wider ecological and biogeographic range. The present study underscores the relevance of owl pellets for small mammal surveys, even in urban and highly disturbed areas.

Several records of associated fauna, including parasitoids, inquilines, predators, and successors, have been reported by insect gall inventories in Brazilian restingas. Although most guilds are well established, inquilines have frequently been misinterpreted. In this paper, the inquilinous fauna of insect galls is revised based on five criteria: food habit; coexistence with the inducer; modification of gall tissues or production of new tissues; phylogenetic relationship with the inducer; and mobility. Gall inventories dated from 1988 to 2019 were examined, totaling 16 publications, eight of them with inquiline records. This guild was reported in 53 gall morphotypes in 44 plant species and four morphospecies distributed among 36 genera of 24 host families for a total of 65 records. Most inquilines were repositioned into the cecidophage guild and others into the kleptoparasite guild, resulting in a large reduction in the frequency of inquilines (from 65 to five records), and in first reports of cecidophages and kleptoparasites, with 46 and 13 records, respectively. Cecidophage was the most diverse guild with insects of five orders (Diptera, Coleoptera, Lepidoptera, Hemiptera, and Thysanoptera) while kleptoparasites were represented only by two orders (Diptera and Hymenoptera) and inquiline solely by Hymenoptera. Other results indicate that Leptothorax sp. (Formicidae) could be a successor and not an inquiline.

Helminths are endoparasites that infect a variety of bird species. Endoparasite infections can cause severe diseases, including kill captive avian hosts and represents a problem to maintenance and conservation. The eggs and larval forms of these parasites are usually eliminated in the host feces. The main interest of this study is to report the occurrence of eggs and oocysts in feces from captive wild birds in Goiânia Zoo and free-living birds in its surroundings. The fecal samples were subjected to parasitological examination to identify the presence of helminths and to classify their eggs based on morphological characteristics. Eggs of nematode parasites (Positive/N) were identified as Ascaridia spp. in Brotogeris chiriri (2/2), Dromaius novaehollandiae (1/1) and Rhea americana (2/2); Ascarididae in Pavo cristatus nigripensis (1/1); Capillaria spp. in Ara chloropterus (1/1) and Penelope jacucaca (1/1); Capillaria plagiaticia in Anodorhynchus hyacinthinus (1/2) and Ara spp. (2/2); Capillaria venusta in Rhamphastus tucanus (1/2); and unidentified nematode eggs from Amazona amazonica (2/2). Eimeria spp. oocyst was founded in feces from A. hyacinthinus (1/2). Considering free-living birds, Diphyllobothrium spp. eggs were founded in Ardea alba (2/2) and Nycticorax nycticorax (2/3). Co-infection of Eustrongylides spp. was founded in A. alba (1/2). This is the first occurrence of: Ascaridia spp. parasitizing B. chiriri; and C. venusta parasitizing R. tucanus. In conclusion, the helminth eggs found in the abovementioned host bird species are consistent with those reported in the literature, and treatment and control protocols were based on their identification.
