Résumé

OBJECTIVE: To develop an HPLC method for simultaneous determination of seven active constituents including neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, quercetin-3-O-β-D-glucuronide, isofraxidin, kaempferol-3-O-β-D-glucuronide and rosmarinic acid in different parts and various collecting time of Sarcandra glabra. METHODS: The DIKMA Diamonsil C18 (4.6 mm × 250 mm, 5 μm) column was used. acetonitrile(A) -0.2% phosphoric acid(B) was used as the mobile phase with gradient elution(0 - 10 min, 5% A→15% A; 11 - 18 min, 15% A→25% A; 18 - 25 min, 25% A; 25 - 40 min, 25% A→40% A), at a flow rate of 1.0 mL•min-1. The detection wavelength was set at 344 nm and the column temperature was maintained at 35 ℃. RESULTS: The calibration curves of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, quercetin-3-O-β-D-glucuronide, isofraxidin, kaempferol-3-O-β-D-glucuronide and rosmarinic acid were in good linearity over 9.32 - 466.00, 11.25 - 562.50, 10.94 - 547.00, 8.68 - 434.00, 10.48 - 524.00, 9.66 - 483.00 and 10.86 - 543. 00 μg•mL-1 (r = 0.999 6 - 0.999 9), respectively. The average recoveries (n = 6) were in the range of 99.0% - 101.5% and RSD were in the range of 0.85% - 1.8%. The optimal parts and collecting time of Sarcandra glabra are aerial parts and from September to November according to the comprehensive evaluation. CONCLUSION: The method is precise and reliable, can be used to determin the content of seven chemical constituents in Sarcandra glabra and provide a reference for collecting in cultivate area.

Résumé

OBJECTIVE: To optimize the formulation of self-microemulsion containing total saponins from Lysimachia capillipes by central composite design-response surface methodology. METHODS: Based on the study of solubility, compatibility test and ternary phase diagram, central composite design-response surface methodology was adopted to optimize the best prescription with the emulsifying time, particle size, and Zeta pontential as indexes, physicochemical properties of this self-microemulsion were also determined. RESULTS: Optimum formulation was 12.93% of ethyl oleate, 57.25% of Kolliphor RH40, 29.82% of Transcutol. The a verage particle size, polydispersity index, Zeta pontential and drug loading of self-microemulsion containing total saponins from Lysimachia capillipes were 23.0 nm, 0.160, -20.28 mV and 10.52 mg·g-1.CONCLUSION: With good predictability, this methods can be used for the formulation optimization of self-microemulsion loaded total saponins from Lysimachia capillipes.

Résumé

Objective To understand the effects of social demographic characteristics and social support on the quality of life among patients with diabetes. Methods By using multi -stage random sampling,a total of 300 people from Tonglu County were enrolled.Social demographic characteristics were investigated with questionnaire.WHOQOL -BREF was used to evaluate the quality of life of diabetic patients and Social Support Rating Scale to test the situation of social support. Results A total of 114 men and 186 women participated in this study.People with primary school or below were accounted for 64.33%.Famers and workers were accounted for 76.00% and 12.33% respectively. The average monthly income of family was below 3 000 yuan in 82.00% of the participants.People who participated in the rural cooperative medical insurance were accounted for 77.33%.Subjective support had significant influence on the score of physical, mental health and social relationship (P <0.05).People with high education had higher scores on the field of mental health and the surrounding environment (P <0.05 ).People who participated the new rural cooperative medical insurance had lower scores on the field of physiological and social relationship (P <0.05).People whose monthly income of family above 5 000 yuan had a higher score on the field of surrounding environment (P <0.05).Conclusion The quality of life of diabetes patients in rural areas may be mainly affected by average monthly income of family, education, health care and other social demographic characteristics factors.Social support was also an important factor for the quality of life.

Résumé

BACKGROUNDRadiotherapy plays a critical role in the management of non-small cell lung cancer (NSCLC). This study was conducted to identify gene expression profiles of acquired radioresistant NSCLC cell line established by fractionated ionizing radiation (FIR) by cDNA microarray.

METHODSThe human lung adenocarcinoma cell line Anip973 was treated with high energy X-ray to receive 60 Gy in 4 Gy fractions. The radiosensitivity of Anip973R and its parental line were measured by clonogenic assay. Gene expression profiles of Anip973R and its parental line were analyzed using cDNA microarray consisting of 21 522 human genes. Identified partly different expressive genes were validated by quantitative reverse transcription-polymerase chain reaction (Q-RT-PCR).

RESULTSFifty-nine upregulated and 43 downregulated genes were identified to radio-resistant Anip973R. Up-regulated genes were associated with DNA damage repair (DDB2), extracellular matrix (LOX), cell adhesion (CDH2), and apoptosis (CRYAB). Down-regulated genes were associated with angiogenesis (GBP-1), immune response (CD83), and calcium signaling pathway (TNNC1). Subsequent validation of selected eleven genes (CD24, DDB2, IGFBP3, LOX, CDH2, CRYAB, PROCR, ANXA1 DCN, GBP-1 and CD83) by Q-RT-PCR was consistent with microarray analysis.

CONCLUSIONSFractionated ionizing radiation can lead to the development of radiation resistance. Altered gene profiles of radioresistant cell line may provide new insights into mechanisms underlying clinical radioresistance for NSCLC.

Résumé

OBJECTIVEThe purpose of the present study was to explore the relationship between the expression of S100A4 and E-cad protein and some clinicopathological factors such as histological types, TNM stages, lymph node metastasis and prognosis in non-small cell lung cancer (NSCLC), and analyze whether there is a correlation between them.

METHODSThe expression of S100A4 protein and E-cad protein was detected with immunohistochemical SP technique in 116 non-small-cell lung cancers.

RESULTSThe positive immunohistochemical staining for S100A4 protein was observed in 64 out of the 116 patients, with a positive rate of 55.2%. The expression rate of S100A4 protein was associated with age, tumor size, lymph node metastasis and prognosis of NSCLC. The expression of S100A4 protein was not significantly related with histological types, sex and TNM stages. The positive rate of E-cad protein expression was 65.5%. The expression of E-cad protein was associated with TNM staging, lymph node metastasis and prognosis of NSCLC. The expression of E-cad protein had no significant correlation with histological types, patient age and sex. An inverse correlation between the levels of S100A4 and E-cad protein expression was found (P < 0.005).

CONCLUSIONExpression of S100A4 protein and loss of E-cad protein expression are significantly associated with tumor progression, and may become valuable markers in prediction of biological behavior and tendency of metastasis of non-small-cell lung cancers.
