RÉSUMÉ

Objective:To investigate the effects of matrine on proliferation, apoptosis and radiotherapy sensitivity of uveal melanoma cells.Methods:An animal experiment study. In vitro experiment: MuM2B cells of human choroidal melanoma were randomly divided into control group and matrine 0.25, 0.50, 1.00, 2.00 g/L groups. The cell morphology was observed by transmission electron microscope. Cell proliferation was detected by thiazole blue colorimetry. The mRNA and relative expression levels of CyclinD D (CyclinD), B lymphoblastoma-2 (Bcl-2) and Bcl2-associated X protein (Bax) were detected by real-time polymerase chain reaction and Western blot. In vivo experiment: BALB/C mice were injected with MuM2B cell suspension subcutaneously on the back of forelimb to prepare transplanted tumor model. After successful modeling, they were randomly divided into blank group and matrine treatment group with different concentrations. Mice in blank group were injected with phosphate buffer subcutaneously. Mice in different matrine treatment groups were injected with 15, 25, 50, 100 mg/kg matrine subcutaneously, respectively, for 7 consecutive days. The tumor was weighed and its volume was measured after the last administration. Single factor analysis of variance was used to compare different groups. The t test was used for pairwise comparison between groups. Results:In the control group, the cell structure was normal, the distribution was uniform, and no or rare nuclear pyknosis was seen. With the increase of matrine dosage, the nuclear pyretosis increased gradually and cell morphology changed obviously. Compared with the control group, the cell survival rate in 0.50, 1.00 and 2.00 g/L groups gradually decreased with matrine concentration increasing and treatment time prolongating, the relative expression levels of CyclinD and Bcl-2 mRNA and protein gradually decreased, and the relative expression levels of Bax mRNA and protein gradually increased. Under the same radiation dose X-ray irradiation, the cell survival rate of 0.50, 1.00 and 2.00 g/L groups gradually decreased, and the differences were statistically significant ( P<0.05). Compared with blank group, the tumor weight and volume of mice in different doses of matrine group were significantly decreased, and the differences were statistically significant ( P<0.05). Conclusion:Matrine can down-regulate the expression of CyclinD and Bcl-2, up-regulate the expression of Bax, promote the apoptosis of MuM2B human melanoma cells, inhibit cell proliferation, and enhance cell radiosensitivity.

Objective To analyze the effects of position shift and dose adjustment on the pass rate of IMRT dose verification to facilitate to obtain rapidly high-pass-rate IMRT dose verification report.Methods At first,the dose unit of measuring dose map image and planning Dose Profile was unified.Secondly,the planning Dose Profile was moved at lateral and cephal-ocaudal directions respectively in contrast mode so as to determine the position error with the maximum pass rate.Thirdly,the highest pass rate point of dose verification was found in the range of dose adjustment.Fourthly,the plan report with the highest pass rate of dose verification was found out by adjusting position error and dose coefficient.Results The highest pass rate was obtained in case the displacement was-3 mm at lateral direction or +3 mm at cephal-ocaudal direction;high pass rate was got when dose adjustment coefficient was 1.02;high pass rate was achieved in case the displacement was-3 mm at lateral direction,+2 mm at cephal-ocaudal direction and the dose adjustment coefficient was 1.02,which was significantly different from those with other combined values (P<0.05).Conclusion The dose verification technique is convenient and quick when used to obtain a high-pass-rate dose verification report.

<p><b>OBJECTIVE</b>To identify potential mutation of the colony stimulating factor 1 receptor gene (CSF1R) in a large Chinese family affected with hereditary diffuse leukoencephalopathy with spheroids (HDLS) and analyze the genotype-phenotype correlation.</p><p><b>METHODS</b>The proband was evaluated physically and radiologically to ascertain the HDLS phenotype. Genomic DNA was extracted from peripheral blood samples from family members. The coding region of the CSF1R gene was amplified with PCR and subjected to direct DNA sequencing.</p><p><b>RESULTS</b>There were 9 affected members (5 alive) in this five-generation family (1 member had died during the follow-up). A missense mutation c.2563C>A (p.P855T) of the CSF1R gene has been identified in the proband. The same mutation was identified in 3 affected and 1 unaffected members of the family.</p><p><b>CONCLUSION</b>The family was consistent with autosomal dominant inheritance. CSF1R gene mutation is also a disease-causing mutation in Chinese patients.</p>

Objective: The reliability of multiple exposure technique in QA imaging was evaluated by combining IP plate of X-ray radiography and homemade QA verification plate. Methods: The QA verification films with different properties derived from placing the QA verification plate and the IP plate on corresponding location were compared with established traditional QA methods. Results: The data from verification films of light and radiation field overlapping derived from combining QA verification plate and IP plate were analyzed by eFilm software in 10cm×10cm radiation field. The results were similar with that of 3D water tank scanning, and were basically same with the analytical results collected from the PTW 2D-ARRAY729 matrix. Conclusion:Using described method in this paper, the result was accurate, the operation was easy, and the cost was low. This method was of good practicability.

Objective To investigate the efficacy of the Ivor-Lewis cervical stapled esophagogastrostomy via thorax in the treatment of middle esophageal carcinoma.Methods The clinical data of 303 patients with middle esophageal carcinoma who were admitted to the Rugao Boai Hospital (107 patients) and the Rugao People's Hospital (196 patients) from March 2005 to March 2013 were prospectively analyzed.All the patients received Ivor-Lewis stapled cervical esophagogastrectomy (Ivor-Lewis group,151 patients) or Sweet stapled cervical esophagogastrostomy (Sweet group,152 patients) according to the admission order.The intraoperative condition,perioperative complications,lymph node dissection and postoperative follow-up of the 2 groups were analyzed.All the patients were followed up via out-patient examination till December 2012.The measurement data,enumeration data and the ranked data were analyzed using the independent samples t-test,chi-square test or Fisher exact probability and Wilcoxon rank sum test,respectively.The survival curve was drawn by the Kaplan-Meier method,and the postoperative mortality rate was analyzed using the Cox proportional hazard model.Results The operation time of the Ivor-Lewis group was (239 ± 21)minutes,which was significantly longer than (188 ± 30)minutes of the Sweet group (t =11.32,P < 0.05).The surgical resection rate of the Ivor-Lewis group was 98.68% (149/151),which was significantly higher than 92.76% (141/152) of the Sweet group (x2 =6.45,P < 0.05).The positive rate of the upper resection margin of the esophagus,postoperative morbidity rate and operative were 0.67%(1/149),10.07% (15/149) and 0 in the Ivor-Lewis group,and 0.71% (1/141),11.35% (16/141) and 0.71%(1/141) in the Sweet group,with no significant difference between the 2 groups (P > 0.05).The number of lymph nodes dissected from the cervical-thoracic junction and the upper abdomen were 3.6 ± 1.1 and 3.5 ± 1.1 in the Ivor-Lewis group,which were significantly greater than 2.3 ± 0.8 and 2.4 ± 0.8 in the Sweet group (Z =9.96,9.02,P < 0.05).The number of positive lymph nodes dissected from the cervical-thoracic junction was 0.7 ± 1.1 in the Ivor-Lewis group,which was greater than 0.3 ± 0.6 of the Sweet group,with significant difference between the 2 groups (Z =3.26,P < 0.05).Of the 290 patients who received surgical treatment,273 were followed up with the follow-up rate of 94.14% (273/290),and the median time for follow-up was 28.0 months.The 1-,2-,3-year tumor recurrence rates were 8.21% (11/134),19.64% (22/112) and 29.35% (27/92) of the Ivor-Lewis group,which was significantly lower than 19.05% (24/126),35.24% (37/105) and 44.19%(38/86) of the Sweet group (x2=6.55,7.33,5.03,P < 0.05).There were significant differences in the 1-,2-,3-year locoregional recurrence rate of the lymph nodes between the 2 groups (x2 =7.03,9.68,6.87,P <0.05).The 1-,2-,3-year accumulative survival rates of the Ivor-Lewis group were 90.30% (121/134),80.36%(90/112) and 71.74% (66/92),which were significantly higher than 80.95% (102/126),59.05% (62/105)and 51.16% (44/86) of the Sweet group (x2=4.65,1 1.73,7.97,P < 0.05).Conclusion Ivor-Lewis stapled cervical esophagogastrostomy via thorax has advantages of high resection rate,better safety and better quality of life of patients,and it could be an optimized design of the treatment for patients with middle esophageal cancer without intumescent lymph node of neck.

Agitation plays an important role in the hyaluronic acid (HA) fermentation process. However, views about the effect of agitation on HA production remain controversial. We investigated the effect of agitation on cell growth and HA synthesis during HA fermentation process by using Computational Fluid Dynamics (CFD) technology. The results showed that the biomass and HA yield changed a little with the increase of impeller speed, but the HA molecular weight firstly increased and then decreased. The results of phase agitation control strategy demonstrated that the influence of agitation on the HA molecular weight mainly exhibited at the stage of HA synthesis. Moreover, the CFD simulation results indicated that when impeller speed increased, the mixing time reduced while the shear rate increased significantly. The removal of anchor could moderate the contradiction between the mixing time and shear rate, and finally the HA molecular weight increased by 23.9%. The results of this work could provide guidelines for optimizing the HA fermentation, as well as the bioreactor design and scaling up.

RÉSUMÉ

[Objective]To explore the treatment effect of acute Lisfranc fracture-dislocations by cannulated screw fixation.[Method]From February 2001 to August 2005,21cases of acute Lisfranc fracture-dislocations were treated with open reduction and cannulated screw fixation.[Result] According to the AOFAS for mid foot,1 case scored 70 to 80,3 cases scored 80 to 90 and 17 cases scored 90 to 100.[Conclusion]Treatment effect of acute Lisfranc fracture-dislocations by cannulated screw fixation in early stage can lead to satisfactory reduction and clinical results,it is a satisfying method.

AIM: To observe the effect of phagotrophic ability of macrophage on pseudomonas aeruginosa which was cocultured with low concentration of erythromycin. METHODS: The pseudomonas aeruginosa ATCC 27853 was cultured in LB both with low concentration of erythromycin (Group A) and without erythromycin (Group B) for 18 h separately, then incubated with macrophage from murine abdominal cavity for 2 h. The ratio of phagocytosis of macrophage was determined by microscope detection. RESULTS: The ratio of phagocytosis of group A in which erythromycin was added was higher than the group B, and the pseudopodia of macrophage were increased in the group A under electron microscope. CONCLUSION: Low concentration of erythromycin may enhance the phagocytosis ability of macrophage in vitro by influencing the virulent factors of PA.
