Objective To analyze the correlations of the expressions of miR-4500 and nerve growth factor(NGF)in breast cancer patients with preoperative magnetic resonance(MR)signs.Methods A total of 105 patients with breast cancer admitted to our hospital were selected,the mRNA expression levels of miR-4500 and NGF in breast cancer tissues and adjacent tissues of patients were detected by qRT-PCR,and the positive expression rates of NGF in breast cancer tissues and adjacent tissues were determined by immunohistochemistry method.The correlations of the expressions of miR-4500 and NGF in breast cancer tissues with clinicopathological features and MR signs of patients were analyzed.The targeting relationship between miR-4500 and NGF was predicted by the bioinformatics website,and the correla-tion between the expressions of the two was analyzed.Results Compared with the adjacent tissues,the expression level of miR-4500 in breast cancer tissue decreased(P<0.05),while the level of NGF mRNA and the positive expression rate of NGF increased(P<0.05).There was no significant difference in age,pathological type,tumor classification,parenchymal background,apparent diffusion coefficient or enhancement curve among different expressions of miR-4500 and NGF(P>0.05).The histological grade,lymph node metastasis,tumor diameter,tumor morphology,ring-like enhancement,and peritu-moral brain edema were related to the expressions of miR-4500 and NGF(P<0.05).The bioinformatics website prediction showed that miR-4500 and NGF had binding sites,and the expressions of miR-4500 and NGF mRNA in breast cancer tissues were negatively correlated(r=-0.576,P<0.05).Conclusion The expression of miR-4500 in breast cancer tissue is low,and the expression of NGF is high,which are correlated with the preoperative MR signs in patients,providing a molecu-lar basis for MR signs.

Objective To evaluate the clinical efficacy and safety of vitamin D combined with atomoxetine hydrochloride in the treatment of attention deficit hyperactivity disorder(ADHD)in children.Methods A retrospective analysis was performed for the data of children with ADHD admitted to Huzhou First People's Hospital from September 2022 to September 2023,and the children were divided into combination group and control group according to the diagnosis and treatment plan.The efficacy of treatment and cognitive function[Mini-Mental State Examination Scale(MMSE)and Wechsler Intelligence Scale for Children,Fourth Edition(WISC-IV.)],behavioral dysfunction[Conners]and neurofilament markers[neurential-specific enolase(NSE)and neurofilament light chain protein(NFL))before and after 3 months of treatment were observed and compared after treatment.The occurrence of adverse reactions during the treatment period was observed and recorded,and the safety was evaluated.Results A total of 101 children with ADHD were enrolled,including 51 in the combined group and 50 in the control group.After treatment,the total effective rate of the combination group was significantly higher than that of the control group(P<0.05).Before treatment,there were no significant differences in MMSE score,WISC-IV.score,Conners score,serum NSE level and serum NFL level between the two groups(P>0.05).After treatment,the MMSE score,WISC-IV.score,Conners score,serum NSE level and serum NFL level of the two groups were significantly improved compared with those before treatment(P<0.05),and the improvement degree in the combination group was better than that in the control group(P<0.05).There was no significant difference in the incidence of adverse reactions between the two groups during treatment(P>0.05).Conclusion Compared with atomoxetine hydrochloride monotherapy,vitamin D combined with atomoxetine hydrochloride has better efficacy in children with ADHD,and can improve cognitive function,behavioral dysfunction and neurological impairment,with high safety.

Abstract Objective The current work aimed to investigate the expression and potential clinical significance of C-type Lectin domain family 14 (CLEC14A) in hepatocellular carcinoma. Methods The relative expressions of CLEC14A in the Hepatocellular Carcinoma (HCC) tissue and adjacent normal tissue of 105 HCC patients were examined using RT-qPCR methods. Furthermore, Receiver Operating Characteristic (ROC) curve was drawn for exploring the diagnostic value of CLEC14A. Next, the expressions of CLEC14A in HCC cell lines and normal liver epithelial cells were compared, and the effects of knockdown of CLEC14A on the growth and apoptosis of HCC cells were examined. Results The authors found that the expression of CLEC14A was markedly increased in hepatocellular carcinoma tumors in comparison with the adjacent tissue, and the expression level of CLEC14A was positively correlated with the size and differentiation of the tumor. Moreover, results of ROC analysis showed CLEC14A might function as a sensitive diagnostic biomarker for HCC. Furthermore, CLEC14A was up-regulated in HCC cell lines, and transient over-expression of CLEC14A decreased the proliferation and increased the apoptosis of HCC cells in vitro. Conclusions Our results suggested that CLEC14A was up-regulated in HCC and might function as a potential diagnostic marker.

Objective To explore the clinical significance of the heart rate viability decrease in the elderly patients with multiple organ disorders.Methods Two hundred and fifty-nine elderly patients with multiple organ disorders, with underlying diseases as coronary heart disease, hypertension, diabetes, cerebrovascular disease,chronic obstructive pulmonary disease,chronic bronchitis and malignant tumor were enrolled in this study,a retrospective analysis was made on the dynamic electrocardiogram data of the research objects.According to SDNN,123 cases were assigned into the normal HRV group(SDNN≥100ms); 50 cases were divided into the moderately decreased HRV group(50ms≤SDNN<100ms),and 36 cases were divided into the severely decreased HRV group(SDNN<50ms).The average heart rate within 24 hours,clinical diagnosis,and discharge record of the three groups were analyzed.Results As HRV decreased,SDNN decreased gradually (normal HRV group 116(104~131)ms,moderately decreased HRV group 85(67.5~91)ms,severely decreased HRV group 40(35~48)ms),the average heart rate within 24 hours gradually increased(normal HRV group 67(60~76)times/min,moderately decreased HRV group 78(66~84)times/min,severely decreased HRV group 91(80~112)times/min),the differences among the three groups were statistically different(P<0.05).The differences among the groups in the rates of coronary heart disease,hypertension, diabetes,cerebrovascular diseases,chronic bronchitis and chronic heart failure were not statistically significant(P>0.05).The rate of malignant tumor in severely decreased HRV group was 8.3%,significantly higher than that in normal HRV group(0),the difference was statistically significant(P<0.01).In severely decreased HRV group,the rates of chronic obstructive pulmonary disease(25%),chronic renal failure(30.56%),respiratory failure(25%),uremic cardiomyopathy(13.89%),hypoproteinaemia(41.67%)and anemia(41.67%)were significantly higher than those in normal HRV group(1.63%,0.9%,0.81%,0,6.5%,8.94%)and moderately decreased HRV group(7%,11%,2%,3%,11%,11%),the differences were statistically significant(P<0.05) .In moderately decreased HRV group,the rate of chronic renal failure(11%)was higher than that in normal HRV group(0.9%),the difference was statistically significant(P<0.05).Mortality in severely decreased HRV group(41.66%)was significantly higher than that in normal HRV group(2.44%)and moderately decreased HRV group(8%),the difference among the three groups was statistically significant(P<0.05).Conclusion For elderly patients with multiple organ disorders,along with severely decreased HRV,they face the risks of severe imbalances of automatic nervous system,fast heart rate,serious illness,poor nutrition,high mortality.

Objective To compare the efficacy of ultrasound ablation and laparoscopic lesion resection with uterine artery liga-tion in treating adenomyosis .Methods By using the prospective ,non-randomized controlled clinical study method ,the adenomyosis patients with clinical symptoms and requiring reserved uterus in our hospital from March 2011 to June 2012 were treated by the ul-trasound ablation technique(ultrasound ablation group ,40 cases) and the laparoscopic lesion resection with uterine artery ligation (operation group ,38 cases) .The menstrual blood volume ,dysmenorrhea and treatment satisfaction in postoperative1 ,3 ,6 months were compared between the two groups .Results The menstrual blood volumes in postoperative 1 ,3 ,6 months in the two groups were decreased significantly(P0 .05) .Conclu-sion The two methods of the ultrasound ablation technique and the laparoscopic lesion resection with uterine artery occlusion for treating adenomyosis could significantly reduce the menstrual blood volume ,improve dysmenorrheal and obtain higher satisfaction . Both can achieve the same therapeutic effect .

<p><b>OBJECTIVE</b>To investigate the inhibitory effects of an antisense PC cell derived growth factor (PCDGF) vector on proliferation and invasion of highly malignant ovarian cancer cell lines Sw626 and A2780 cells, and preliminarily explore the related mechanisms.</p><p><b>METHODS</b>MTT assay and Boyden chamber in vitro invasion assay were employed to detect the changes of proliferation and invasion ability in the Sw626 and A2780 cells transfected with anti-sense PCDGF. The expression levels of cyclin D1 and CDK4 proteins before and after transfection were detected by Western blotting. The effects on the expression and activity of MMP-2 were evaluated by quantitative RT-PCR and zymography, respectively.</p><p><b>RESULTS</b>Comparing with the blank group, the proliferation inhibition rate of the Sw626 and A2780 cells transfected with anti-sense PCDGF was 72.9% and 70.9%, respectively, and the invasion ability was inhibited by 62.9% and 59.0%, respectively. The levels of cyclin D1 and CDK4 protein expression in antisense PCDGF transfected cells were 0.38 +/- 0.08 and 0.37 +/- 0.13, respectively, all significantly lower than 0.84 +/- 0.11 and 0.64 +/- 0.11, respectively, in the blank group (P < 0.01). The MMP-2 mRNA expression level in antisense PCDGF transfected cell group was 0.66 +/- 0.11, not significantly decreased in comparison with 0.89 +/- 0.09 in the blank group (P > 0.05), but the activity of MMP-2 was inhibited significantly.</p><p><b>CONCLUSION</b>The antisense PCDGF vector may inhibit markedly the proliferation and invasion of highly malignant ovarian cancer cells, and partially reverses their malignant phenotype. It seems to be related with down-regulating the expression of cyclin D1 and CDK4 and inhibiting the activity of MMP-2. Our findings indicate that PCDGF may become a new target for antisense gene therapy of ovarian cancer.</p>
