Résumé

Objective:To investigate the effects of emodin on proliferation, adhesion, migration, c-kit mRNA and protein expression of human epidermal melanocytes. Methods:Human epidermal melanocytes cultured in vitro were treated with different concentrations of emodin, a blank control group (containing medium + human epidermal melanocytes) and an experimental group (containing medium + human epidermal melanocytes + different concentrations of emodin) were set up. Cell proliferation was measured by CCK-8 method, cell adhesion was measured by microplate assay, cell migration was measured by Transwell membrane assay, and c-kit mRNA expression was measured by reverse transcription-polymerase polymerase chain reaction; Western blot was used to detect the expression of c-kit protein. Results:Compared with the blank control group, emodin decreased the proliferation, adhesion and migration of human epidermal melanocytes, the difference was statistically significant ( F=391.48, P<0.0001; F=10.93, P=0.003; F=7.75, P=0.009). Compared with the blank control group, emodin in the experimental group had a bidirectional effect on the expression of c-kit mRNA and protein. High concentration of emodin inhibited the expression of c-kit mRNA and protein, and the expression of c-kit mRNA and protein was significantly increased by Emodin ( F=11.491, P=0.003; F=2155.11, P<0.001). Conclusions:Emodin inhibits the proliferation, adhesion and migration of human epidermal melanocytes, high concentration of emodin inhibits the expression of c-kit mRNA and protein, and low concentration of emodin promotes the expression of c-kit mRNA and protein, and the results provide a basis for the clinical application of emodin in pigmented dermatosis.

Résumé

Objective To investigate human papillomavirus (HPV) genotypes in adult patients with plantar warts, and to explore their relationship with clinical features of adult plantar warts. Methods PCR,gene sequencing and TA-cloning technique were performed to detect HPV types in 221 patients with plantar warts.Meanwhile, clinical data were recorded, including patients' age, disease duration, symptoms and number of warts. Results Of 221 specimens,HPV DNA was detected in 215,with the HPV-positive rate being 97%.Single HPV infections were detected in 206 specimens (96%,206/215),among which,HPV types included HPV-27 (44.7%,92/206),-2 (12.1%,25/206),-57 (7.3%,15/206),-7 (Alpha genus;0.5%,1/206),-65 (Gamma genus;0.5%,1/206),-1 (33.5%,69/206) and-63 (Mu genus;1.5%,3/206).There were no significant differences in age, disease duration, number of warts, incidence of pain and gender among patients with HPV-27,-2 or-57 infections.Compared with patients with HPV-27 infection, patients with HPV-1 infection were more related to age ≤ 30 years, disease duration ≤ 1 year, number of warts ≤ 2 and high incidence of pain. Conclusion HPV-27,-2,-57 and-1 are the main types in adult patients with plantar warts, and HPV types are correlated with clinical features of adult plantar warts.

Résumé

The effect of OQP, a pormula of traditional Chinese medieine belonging to the category "Precription of the Reinforeing Qi and Enriching Blood for keep the Original Qi", on cell strain of human hepatic carcinoma SMMC-7721 was observed. The results showed that OQP depressed the incorporation of both ~3H-TdR and ~(14)C-UR in cells of hepatic carcinoma. This indicated that OQP inhibited the synthesis of DNA and RNA, and the growth of the cancer cell. Such effect may result from the low ratio Cu/Zn and the high content of Se in OQP. In conclusion, in this study it has been demonstrated that OQP can not only improve the general condition of patients in advanced liver cancer and lcukopenia of patients in chemotherapy and radiotherapy, but also can inhibite the growth of the cell of hepatic carcinoma directly.

Résumé

In the paper, the effect of Xin Ji Rong liquer for oral on the plasm membrane fluidity of cultured cardiac myocytes in neonatal rats was observed with 1,6-dipheny-1,3,5-hexatriene (DPH) as a fluorescence probe. The result showed that adding isoprenaline to the cultured medium could make the membmne fluidity reduce signalicantly as compared with the control (P

Résumé

The glutathione peroxidase cDNA has been acquired by reverse transcriplate-polymerase chain reaction (RT-PCR). mRNA was isolated from the culture liver cancer cells firstly, then trancriplate if into cDNA with AMV reverse transcriptase. By using this cDNA as PCR template and two synthetic special primers, we get about 600 bp DNA fragment after 35 circles. The result is identical with our expectation.
