RÉSUMÉ

Objective@#To investigate the characteristics of mortality of injury among residents in Gusu District, Suzhou City, Jiangsu Province from 2013 to 2023, so as to provide the evidence for developing targeted measures of injury prevention and control.@*Methods@#Gender, age and underlying cause of deaths due to injury in Gusu District were collected through Death Reporting Information System of Chinese Disease Prevention and Control Information System and Jiangsu Death Reporting Information System from January 1, 2013 to December 31, 2023. The crude mortality, Chinese-standardized mortality and world-standardized mortality of injury were analyzed, and the trend in mortality was analyzed using annual percent change (APC).@*Results@#Totally 4 217 deaths due to injury were reported in Gusu District from 2013 to 2023. The crude, Chinese-standardized and world-standardized mortality rates were 51.58/105, 23.24/105 and 21.98/105, respectively, all showing a tendency towards a rise (APC=6.802%, 2.688% and 2.823%, all P<0.05). The crude mortality rate of injury was higher in women than in men (54.61/105 vs. 48.41/105, P<0.05). The five most common causes of injury included fall (32.99/105), traffic accidents (6.03/105), suicide (4.23/105), drowning (3.00/105) and asphyxia (2.16/105), accounting for 93.86% of the total number of deaths. The crude mortality rates of fall, suicide and asphyxia appeared a tendency towards a rise (APC=9.724%, 6.333% and 5.638%, all P<0.05). The crude mortality rates of injury among men, women and overall residents appeared a tendency towards a rise with age (all P<0.05). Fall was the primary cause of injury death among residents aged 65 years and above, and suicide was the primary cause of injury death among residents aged 15 to 44 years.@*Conclusions@#The crude mortality of injury appeared a tendency towards a rise in Gusu District from 2013 to 2023. The main causes of death were fall, traffic accidents, suicide, drowning and asphyxia, with the crude mortality of fall, suicide and asphyxia showing an upward trend.

@#Objective To investigate the infection effects of different strains(Prototype,Delta,Beta,and Omicron BA. 1)of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)on transgenic mice K18-hACE2 expressing angiotensinconverting enzyme 2(ACE2)by nasal drip.MethodsMice were challenged with each strain of different gradients of doses by nasal drip,which were monitored for body mass and survival daily for 6 d in Beta group and 14 d in the other three groups. The median lethal dose(LD_(50))was calculated by IBM SPSS Statistics 26 analysis software,and the virulence of different SARS-CoV-2 strains were compared by detection of the viral nucleic acid loads of lung,brain and turbinate tissues and the lesion of lung tissues.ResultsBeta strain showed the strongest virulence and the highest lethality,and the lowest dose(1 CCID_(50))was 100% lethal with a LD_(50)less than 1 CCID_(50);The LD_(50) of Delta,Prototype and Omicron BA.1 strains were 0. 52,0. 60 and 52. 92 CCID50respectively. The copy number of viral nucleic acid in lung tissue of mice infected with different strains elevated with the increase of challenge dose,and the copy number of viral nucleic acid in brain tissue of dead mice reached 10~8~ 10~(11) copies/mL. The lung tissues of mice infected with different strains of different doses showed different degrees of pathological damage.ConclusionThe virulence of different SARS-CoV-2 strains in K18-hACE2 mice from high to low were Beta,Delta,Prototype and Omicron BA.1.

@#AIM: To observe the clinical effect of trabeculectomy combined with dexamethasone iontophoresis in the treatment of acute angle-closure glaucoma(PACG)with diabetes mellitus.<p>METHODS: Totally 42 eyes of 42 patients with acute PACG combined with diabetes in our hospital were selected and divided into trabeculectomy combined with iontophoresis group(group A)and trabeculectomy combined with peribulbar injection of methylprednisolone group(group B)according to different ways of controlling inflammation after surgery. Intraocular pressure, anterior chamber inflammation and complications before and after surgery were compared between the two groups.<p>RESULTS: The intraocular pressure of the two groups after surgery was significantly improved compared with that before surgery, and the difference was statistically significant(<i>P</i><0.05). Postoperative anterior chamber inflammation was well controlled in both groups without statistical significance(<i>P</i>>0.05). In terms of complications, group A was significantly superior to group B.<p>CONCLUSION: Trabeculectomy combined with dexamethasone iontophoresis or peribulbar injection of methylprednisolone can effectively control postoperative intraocular pressure and anterior chamber inflammation in patients with acute angle-closure glaucoma complicated with diabetes mellitus. Among them, the effect of combined dexamethasone iontophoresis in reducing complications is better and patients suffer less pain.

Objective To observe the therapeutic efficacy of balance cupping plusYang He decoction in treating granulomatous mastitis (GM).Method Eighty GM patients were randomized into a treatment group and a control group according to the last digit of registered number, odd or even, 40 cases each. The control group was treated withYang He decoction; the treatment group was intervened by balance cupping in addition to the treatment given to the control group. They were treated for 6 months in total. The size of lump, swollen size and infection size by the ultrasound B-scanning were observed before and after the treatment, and the clinical efficacies were compared between the two groups.Result The total effective rate was 82.5% in the treatment group versus 60.0% in the control group, and the between-group difference was statistically significant (P<0.05). There was a significant difference in comparing the clinical efficacy between the two groups (P<0.05). The size of lump, swollen area and infection size by the ultrasound B-scanning were significantly changed in the two groups after the treatment (P<0.05); the between-group comparisons showed significant differences after the treatment (P<0.05).Conclusion Balance cupping plusYang He decoction can produce a certain efficacy in treating GM.

Objective To investigate the clinical efficacy of cupping treatment for recurrent mycotic vaginitis. Method Three hundred and eighty-three patients with recurrent mycotic vaginitis were randomly allocated, including 188 cases to a control group and 195 cases to an observation group. The control group received vaginal administration of clotrimazole and oral administration of sporanox and the observation group, cupping treatment in addition. Re-examination was made after two weeks of treatment. The clinical therapeutic effects and the clinical scores were compared between the two groups. Result In the observation group, clinical cure occurred in 96 patients, accounting for 49.2%; marked effectiveness, in 45 patients, accounting for 23.1%; effectiveness, in 34 patients, accounting for 17.4%; ineffectiveness, in 20 patients, accounting for 10.3%, with a total efficacy rate of 89.7%. In the control group, clinical cure occurred in 78 patients, accounting for 41.5%; marked effectiveness, in 34 patients, accounting for 18.1%; effectiveness, in 33 patients, accounting for 17.6%; ineffectiveness, in 43 patients, accounting for 22.9%, with a total efficacy rate of 77.1%.The clinical therapeutic effect was significantly better in the observation group than in the control group (P<0.05). There was a statistically significant pre-/post-treatment difference in the clinical score in the two groups (P<0.05) and a statistically significant post-treatment difference in the clinical score between the two groups (P<0.05).Conclusion Cupping can improve the clinical therapeutic effect on recurrent mycotic vaginitis and relieve its clinical symptoms.

This study was aimed to investigate the effect of Honokiol (HNK) on proliferation and apoptosis of acute myeloid leukemia HL-60 cells and its potential mechanism. Inhibitory effect of HNK on the HL-60 cell proliferation was detected by MTT assay. Flow cytometry was used to detect the change of cell cycle and AnnexinV/PI staining was used to detect apoptosis. Western blot was applied to analyze the cell cycle protein (cyclins), cyclin-dependent kinase (CDK), P53, P21, P27, BCL-2, BCL-XL, Bax, caspase-3/9 and proteins for MAPK signal pathway. The results showed that HNK could inhibit the proliferation of HL-60 cells in time- and dose dependent ways. HNK arrested HL-60 cells in G0/G1 phase, and S phase cells decreased significantly (P < 0.05). The expression of cyclin D1, cyclin A, cyclin E and CDK2/4/6 were significantly down-regulated (P < 0.05), the expression of P53 and P21 was significantly upregulated after treating for 24 h with HNK (P < 0.05). After 24 h treatment with HNK, HL-60 cell apoptosis increased significantly with the upregulation of activated caspase-3, -9, BAX expression and the downregulation of BCL-2, BCL-XL expression. The MAPK subfamily, P38 and JNK were not significantly changed, but the expression of MEK1/2-ERK1/2 was significantly downregulated (P < 0.05). It is concluded that HNK arrestes the cells at G0/G1 phase and induces HL-60 cell apoptosis through the intervention of MEK1/2-ERK1/2 signaling pathway.

This study was aimed to explore the effect of arsenic trioxide combined with curcumin on proliferation and apoptosis of KG1a cells and its potential mechanism. The cell survival rate was mesured by MTT; colony formation capacity was examined by methylcellulose colony formation test; flow cytometry was used to analyse the cell surface molecules, cell apoptosis rate and cell cycle; the cell morphology was observed with Wright-Giemsa staining and the protein expression of BCL-2, BAX, PARP was detected by Western blot. The results showed that the phenotype of KG1a cells was CD34(+)CD38(-), while the phenotype of HL-60 cell was CD34(+)CD38(+). The former possessed a stronger colony ability than the latter. Effect of curcumin and arsenic trioxide alone on cell proliferation and inhibition was in dose-dependent manner. Compared with single drug-treatment group, the cell survival rate and colony number were lower, and the apoptosis rate was higher in combined drug-treatment group. Protein expression of BCL-2 and PARP was upregulated, while the protein expression of PARP was downregulated in the combined treatment group. It is concluded that compared with HL-60 cells, KG1a cells are the earlier leukemia stem/progenitor cells. Arsenic trioxide combined with curcumin can effectively inhibit the KG1a cell proliferation and induce apoptosis, which may be associated with the downregulation of BCL-2 and PARP protein expression and the upregulation of BAX protein expression.

To study the gene variation and the distribution of HPV16 variant in Hubei, China, DNA was extracted from cervical cancer tissue samples. The E6 and E7 genes of HPV16 were amplified and the PCR products were sequenced using E6- and E7-specific primers. Fortyseven cases were found mutations at nucleotide position 178 of HPV16 E6 gene in 80 cervical cancer samples. This mutation resulted in amino acid change from Asp to Glu. The rate of mutation at nucleotide position 178 of E6 gene was 58. 75%. Twenty two cases were found mutations at nucleotide position 647 of HPV16 E7 gene in 31 cervical cancer samples. This mutation resulted in amino acid change from Asn to Ser. The rate of mutation was 70.97%. These results showed that mutations at nucleotide position 178 of E6 gene, nucleotide position 647 of E7 gene of HPV16 in cerveical cancer samples were prevalent in Hubei, China. Phylogenetic analysis showed that Asian (As) variants of HPV16 are predominated in Hubei, China. European (Ep) varinats were also found in Samples in Hubei areas. None of Asian American (AA), African-1 (Af-1), African-2 (Af-2) variants of HPV16 was found in this region. Whether Asian (As) variants of HPV16 are more oncogenic and play a much more important role in the progress of cervical cancer than European (Ep) variants is not clear. More sequences of E6 and E7 gene in CIN and normal cervical tissue samples and study of the function of E6 and E7 protein of these HPV16 variants are needed to adress above question.
