RÉSUMÉ

ObjectiveTo evaluate the curative effect of Jiedu Huayu granules (JDHY) in the treatment of chronic liver failure (CLF) with the syndrome of toxic heat and stasis and investigate the influence on the inflammatory state. MethodA total of 136 patients were randomly divided into a control group and an observation group with 68 cases in each group. In addition to the comprehensive western medicine treatment, patients in the control group received Yinchen Haotang granules orally at 1 dose/day and those in the observation group received JDHY at 10 g/time,3 times/day. The treatment lasted for eight weeks. The endotoxin (ET),diamine oxidase (DAO),aromatic amino acids (AAA),branched chain amino acids (BCAA),blood ammonia,calcitonin (PCT),tumor necrosis factor-α (TNF-α),interleukin (IL)-1,IL-6,IL-17,regulatory T cells (Treg cells),helper T cells 17 (Th17),Th17/Treg ratio,total bilirubin (TBil),albumin (Alb),alanine aminotransferase (ALT),aspartate aminotransferase (AST),prothrombin activity (PTA), and D-dimer (D-D) levels before and after treatment were detected. The Child-Pugh grading scores of liver function, toxic heat and stasis syndrome scores, and the model scores of end-stage liver disease(MELD) before and after treatment were recorded. The fatality rate and survival were recorded at the follow-up for 48 weeks. ResultCompared with the control group after treatment, the observation group showed decreased ET,DAO, and blood ammonia, increased BCAA/AAA ratio (P<0.01), reduced PCT,TNF-α,IL-1,IL-6, and IL-17 (P<0.01), elevated Treg cells, dwindled Th17 and Th17/Treg ratio (P<0.01), diminished TBil,ALT,AST, and D-D levels, and up-regulated Alb and PTA(P<0.01). The Child-Pugh grading score,MELD score, and toxic-heat and stasis syndrome score of the observation group were lower than those of the control group (P<0.01). The total response rate in the observation group was 93.65% (59/63),which was higher than 79.03% (49/62) in the control group (χ2=5.683,P<0.05). The fatality rate of the observation group eight weeks after treatment was 6.35% (4/63),which was lower than 19.35% (12/62) of the control group (χ2=4.757,P<0.05). There was no significant difference in mortality between the two groups 16,24, and 48 weeks after treatment. As revealed by the Log-rank test,the difference in survival curves between the two groups was not statistically significant. ConclusionJDHY can remove toxins from the body,regulate immune function,relieve inflammation,improve liver function, and reduce the severity of the disease in CLF patients with the syndrome of toxic heat and stasis. It is significant in clinical efficacy and worthy of clinical application.

OBJECTIVE To explore the mechanism of oridon in(Ori)reversing the drug resistance of breast cancer cell MCF-7 to fulvestrant (Ful). METHODS Ful-resistant breast cancer cell strains MCF- 7/Ful were induced and constructed in vitro . The relative cell viability of MCF- 7 cells and MCF- 7/Ful cells was detected by MTT assay ,inhibitory rate of Ori to MCF- 7/Ful cells was also detected. CompuSyn software was used to analyze the synergistic effect of Ori and Ful. MCF- 7/Ful cells were randomly divided into blank control group ,Ful group (5 μmol/L),Ori group (8 μmol/L),Ful(5 μmol/L)+Ori(8 μmol/L)group. The phosphorylation of phosphatidylinositol 3 kinase(PI3K)/protein kinase B (Akt)signaling pathway related protein in each group was detected by Western blot. MCF-7/Ful cells were used to prepare drug resistance model of transplanted tumor in nude mice,and they were randomly divided into blank control group ,Ful group (80 μmol/g),Ori group (50 μmol/g),Ful(80 μmol/g)+ Ori(50 μmol/g)group. The tumor weight and tumor inhibition rate were calculated ,to verify the reversal effect of Ori and Ful in vivo. RESULTS MTT assay showed that when Ful ≥10 μmol/L,the relative cell viability of MCF- 7/Ful cells was significantly higher than that of MCF- 7 cells(P<0.05),and the drug resistance was significantly enhanced ;Ori had a significant inhibitory effect on MCF- 7/Ful cells ,the inhibition rate of Ori combined with Ful to MCF- 7/Ful cells was significantly increased (P<0.05 or P<0.01),and the effect of reversing drug resistance was significantly increased . The results of Western blot showed that compared with Ful group ,the phosphorylation levels of PI 3K and Akt protein in Ful+Ori group were significantly decreased (P<0.05 or P< 0.01). In vivo results showed that the tumor volume and mass of Ful+Ori group were significantly decreased ,and the tumor inhibition rate was (63.90±4.11)%,which was significantly higher than that of Ful group (P<0.01). CONCLUSIONS Ori can reverse drug resistance of breast cancer cell MCF- 7 to Ful , and this effect may be through regulating PI 3K/Akt signaling pathway.

OBJECTIVE:To study the effects of Plantago asiatica polysaccharide on the proliferation ,migration and invasion of breast cancer cells ,and to investigate its mechanism preliminarily. METHODS :Using human breast cancer cell MDA-MB- 231 as subjects ,MTT method was adopted to detect the effects of different concentrations of P. asiatica polysaccharide(8,16,32,64 mg/L)on the cell proliferation ability ,and survival rate of the cells was calculated. Scratch test and Transwell invasion test were used to detect the effects of different concentrations of P. asiatica polysaccharide(8,16 mg/L)on cell migration ability and invasion ability. Western blot assay was used to detect the expression of epithelial-mesenchymal transition (EMT)-related proteins [matrix metalloproteinase- 2(MMP-2),MMP-9,E-cadherin,N-cadherin,vimentin]. RESULTS :Results of MTT assay showed that survival rate of the cells in 32,64 mg/L P. asiatica polysaccharide groups were significantly lower than control group (P<0.05 or P<0.01),so that 8,16 mg/L,which did not affect the cell survival rate ,were used as the follow-up drug concentrations. Compared with control group ,relative mobility (12,24 h),relative invasion rate and relative expression of MMP- 2,MMP-9, N-cadherin and vimentin protein were decreased significantly in 8,16 mg/L P. asiatica polysaccharide groups (P<0.05 or P< 0.01),while relative expression of E-cadherin protein was increased significantly (P<0.05 or P<0.01). CONCLUSIONS :P. asiatica polysaccharide can inhibit the proliferation of breast cancer cells MDA-MB- 231,and inhibit the migration and invasion of the cells by regulating the expression of metastasis and EMT-related proteins.

Aim Toinvestigatetheeffectofhigh,me-dium and low doses of Mahuang decoction on the re-lease amount of rat hippocampal neural transmitter (Glu,Gly,Asp,GABA),then compare Mahuang de-contion with ephedra alkaloids and ephedra.Methods Ratswererandomlydividedinto6groupsandgiven orally with Mahuang decoction of high dose (calculated by ephedra 4 g·kg-1 ),medium dose (calculated by ephedra 2 g·kg-1 ),low dose (calculated by ephedra 1 g · kg-1 ),ephedra (calculated by ephedra 2 g · kg-1 ),ephedra alkaloids (ephedrine 7 mg · kg-1 , pseudoephedrine 2. 4 mg · kg-1 , methylephedrine 1. 12 mg · kg-1 )and blank control group.Samples were obtained from the hippocampus of conscious rat by microdialysis sampling technique.The content of ami-no acid neurotransmitters in dialysates was detected u-sing the established HPLC-ECD with OPA pre-column derivationmethod.Results Fouraminoacidneuro-transmitters could be well separated in 28 min.High, medium and low doses of Mahuang decoction,ephedra and ephedra alkaloids significantly increased the con-tent of these four amino acid neurotransmitters,com-pared with blank control group (P<0. 05 ).Ephedra alkaloids significantly reduced the levels of inhibitoryamino acid neurotransmitters GABA and Gly in 90 min,compared with the medium dose of Mahuang de-coction.Excitatory neurotransmitters of ASP and Glu in hippocampus showed the trend of increase first and then decrease after oral administration of Mahuang de-coction and ephedra. The levels of Glu and Asp reached peaks from 90 min to 120 min after treatment with Mahuang decoction,and also increased along with dose increase of Mahuang decoction.In comparison with the medium dose of Mahuang decoction group,the level of Glu reached peak at 90 min and 150 min in ephedra alkaloids group and ephedra group respective-ly,and the content of Glu significantly increased at peaktime.Conclusions Increasedcontentofexcita-tory amino acid neurotransmitters (Asp and Glu ) shows positive correlation with the dose of Mahuang de-coction.Other components in Mahuang decoction in-hibits the up-regulation effect of ephedra and ephedra alkaloids on Glu,and promotes the up-regulation effect of ephedra alkaloids on GABA and Gly.

Objective To construct a human tumor necrosis factor (hTNF-a) plasmid and identify it to optimize the fermentation conditions of hTNF-α protein so as to achieve high expression in Escherichia coli.Methods The gene of hTNF-a was cloned into pET24a vector to obtain the pET24a-hTNF-a expression plasmid that was transformed into Escherichia coli BL21(DE3),and the expression conditions of BL21 (DE3) were optimized.Results The plasmid of pET24a-hTNF-α was successfully constructed and identified by PCR and digestion,which was consistent with the target fragment hTNF-α.The plasmid was transformed into Escherichia coli BL21(DE3),the best induced expression conditions of Escherichia coli BL21 (DE3) were as follows:M9+LB medium,37℃,0.5 mmol/L IPTG,pH =7.5,and induction time was 5 h.The results showed that dry weight of the cells and the rate of TNF were increased by 2.56 times and 3.68 times,respectively,and the expression rate of hTNF-α was increased by 3.49 times from 9.38% to 32.74%.Conclusion The optimal conditions for the expression of plasmid pET24a-hTNF-α in Escherichia coli were determined.
