RÉSUMÉ

<p><b>OBJECTIVE</b>To explore the safety and efficacy of pegylated recombinant human granulocyte colony-stimulating factor (PEG-rhG-CSF) in preventing chemotherapy-induced neutropenia in patients with breast cancer and non-small cell lung cancer (NSCLC), and to provide the basis for clinical application.</p><p><b>METHODS</b>According to the principle of open-label, randomized, parallel-group controlled clinical trial, all patients were randomized by 1∶1∶1 into three groups to receive PEG-rhG-CSF 100 μg/kg, PEG-rhG-CSF 6 mg, or rhG-CSF 5 μg/kg, respectively. The patients with breast cancer received two chemotherapy cycles, and the NSCLC patients received 1-2 cycles of chemotherapy according to their condition. All patients were treated with the combination chemotherapy of TAC (docetaxel+ epirubicin+ cyclophosphamide) or TA (docetaxel+ epirubicin), or the chemotherapy of docetaxel combined with carboplatin, with a 21 day cycle.</p><p><b>RESULTS</b>The duration of grade 3-4 neutropenia in the PEG-rhG-CSF 100 μg/kg and PEG-rhG-CSF 6 mg groups were similar with that in the rhG-CSF 5 μg/kg group (P>0.05 for all). The incidence rate of grade 3-4 neutropenia in the PEG-rhG-CSF 100 μg/kg group, PEG-rhG-CSF 6 mg group, and G-CSF 5 μg/kg group were 69.7%, 68.4%, and 69.5%, respectively, with a non-significant difference among the three groups (P=0.963). The incidence rate of febrile neutropenia in the PEG-rhG-CSF 100 μg/kg group, PEG-rhG-CSF 6 mg group and G-CSF 5 μg/kg group were 6.1%, 6.4%, and 5.5%, respectively, showing no significant difference among them (P=0.935). The incidence rate of adverse events in the PEG-rhG-CSF 100 μg/kg group, PEG-rhG-CSF 6 mg group and G-CSF 5 μg / kg group were 6.7%, 4.1%, and 5.5%, respectively, showing a non-significant difference among them (P=0.581).</p><p><b>CONCLUSIONS</b>In patients with breast cancer and non-small cell lung cancer (NSCLC) undergoing TAC/TA chemotherapy, a single 100 μg/kg injection or a single fixed 6 mg dose of PEG-rhG-CSF at 48 hours after chemotherapy show definite therapeutic effect with a low incidence of adverse events and mild adverse reactions. Compared with the continuous daily injection of rhG-CSF 5 μg/kg/d, a single 100 μg/kg injection or a single fixed 6 mg dose of PEG-rhG-CSF has similar effect and is more advantageous in preventing chemotherapy-induced neutropenia.</p>

<p><b>OBJECTIVE</b>To evaluate the effect of calcification of autogenous bone marrow stem cell transplantation in periodontal tissue regeneration.</p><p><b>METHODS</b>Bone marrow stem cells derived from the same dog were cultured with alpha-MEM. 1 x 10(7) cells of first passage were allowed to attach to the collagen membrane for 24 hours. The membrane-cells were transplanted into periodontal defect in the same dog. Then the defects were covered with e-pTFE membranes. The defects covered only with e-pTFE without membrane-cells were served as control. Eighteen teeth of 6 dogs for every group were studied. The dogs were sacrificed after 6 weeks.</p><p><b>RESULTS</b>The results showed that new bone formation in test group was significantly higher than that of control group. The calcification of new bone in test group was better than control group.</p><p><b>CONCLUSIONS</b>The results suggested that autogenous bone marrow stem cell transplantation with guided tissue regeneration technique could enhance periodontal tissue regeneration and could form new bone tissue fast and could shorten times of periodontal tissue regeneration in dogs.</p>

Objective:To study the biological characteristics and mineralization activity of strom cells (MSC) isolated from rat bone marrow.Methods: MSCs were isolated from rat bone marrow using gradient centrifuge (460 g,20 min) with 70% percoll and cultured with DMEM. The cells were then incubated with mineralization culture medium (MCM) in the presence of Dex (10~7 mol/L), β-GP (10 mmol/L) and AcA (50 g/L).Cell growth was studied with cell counting,alkaline phosphatase (ALP) expression with a kit and mineralization with a saturated solution of alizarin red S (pH 4.3) stainning.Results: The isolated cells from bone marrow of rat were epithelial-like. The doubling time (DT) of the cells in MCM or DMEM were 50.3 h and 66.1 h respectively. Mineralization was observed after the cells had been cultured in MCM for 28 days. bFGF and rhBMP-2 increased ALP expression of MSC in a dosage dependent manner. Conclusion:In vitro cultured MSCs may be induced to differentiate towards osteoblasts.

To observe repair of dental defect and its curative effect of the vertical root split of molar in the seniors, every affected section of 36 cases (40 teeth) of the senior's root split was extracted by hemisection, the other sections of them were repaired by either complete crown restoration (15 cases) or fixed bridge restoration (21 cases). The results showed that after 6~12 months, excellent curative effect was achieved in 32 cases, good curative effect in 3 cases, poor curative effect in 1 case. Objective To study the effect of hemisection in the treatment of the molars. Methods 136 molars were selected and hemisection technique were used. If the pathological change was concentrated to the root, such as periodontics, apical inflammation, root fractures, the pathological root was extracted. If the pathologicalchange was concentrated to the divergence of the roots or the bottom of pulp chamber, hemisection was performed. then all the tooth was restoredwith fixed restoration according to the solid conditions. Results These were followed up and evaluated in three years 94. 85% of the all molars were treated satisfactory. Conclusions The hemisection techniqueoperation is simple, but the indications are extensive,and its effect in the treatment of the molars is regarded as a positive method

The effects of polytetrafluoroethylene (e PTFE) membrane on periodontal ligament cell (PDLC) growth and periodontal tissues and new cement regeneration were studied, and the prospects of clinical application of e PTFE membrane were discussed. The results showed that PDLC grew well on the e PTFE membrane and no toxicity or inhibition was found. Close adhesion of the cells to the membrane was observed by scanning electron microscope. Histological studies suggested that the peculiar porous structure of e PTFE membrane protects and promotes healing of alveolar bone defect when the membrane edge was sealed perfectly on the crown side,exhibiting inhibition of growth of epithelium towards tooth root. The membrane was beneficial to regeneration of new alveolar bone and cement and establishment of new adhesion,and was regarded as a good substitute of old materials.
