Objective To investigate the expressions of CD133 and CD44 and their prognostic significance in gastrointestinal stromal tumors (GIST).Methods Streptavidin perosidase (SP) method of immunohistochemistry was used to detect expressions of CD133 and CD44 proteins in 42 cases of GIST, and the relationship between their expressions and tumor size, mitotic count were analyzed by univariate and multivariate factor analyses.Results The expressions of CD133 and CD44 proteins in GIST were 21.4% (9/42) and 78.6% (33/42), respectively.The expressions of CD133 and CD44 proteins were significantly correlated with tumor size and mitotic count (P < 0.05).Univariate factor analysis showed that the overall survival of GIST patients with positive CD133 protein (23.2 months) was shorter than that of patients with negative CD133 protein(63.1 months) (P < 0.05).The overall survival of GIST patients with negative CD44 protein (23.2 months) was shorter than that of patients with positive CD44 protein (63.3 months) (P < 0.05).Multivariate factor analysis showed that tumor size, mitotic count and CD44 protein were independent prognostic indicators for survival time after operation.Conclusions The positive expressions of CD133 and CD44 proteins might be the prognostic factors of GIST patients.

Objective To investigate the expressions and clinical significance of survivin, and lymphatic vessel endothelial hyaluronan receptor 1 ( LYVE-1) in gastric cancers.Methods The expres-sions of survivin and LYVE-1 were detected with streptavidin avidin-peroxidase ( SP) immunohistochemical method in 70 cases of gastric cancers and 50 cases of normal gastric tissues.The relationship between the expressions of survivin and LYVE-1 with the clinicopathological parameters was analyzed.Results The ex-pression rate of survivin was 65.71%(46/70) in gastric cancers, and 6.00% (3/50) in normal gastric tissues, with statistically significant difference between two groups (χ2 =43.048, P 0.05).The expression rate of LYVE-1 was 32.86%(23/70) in gastric cancers, and 4.00%(2/50) in gastric ulcers, with significant difference between two groups (χ2 =14.726, P 0.05 ) .There was a positive correlation between expressions of survivin and LYVE-1 in gastric cancers ( r =0.271, P <0.05).Conclusions Survivin and LYVE-1 were abnormally expressed in gastric cancers with a colose relationships between two parameters.They might play important roles in tumorigenesis and progression of a gastric cancer.

Résumé

Objective To validate the possibility of CD133 CD34 CD44 be served as biomarkers in cancer stem cell of human lung adenocarcinoma. Methods Two kinds of culturing methods were performed to generate adhesive tumor cells and floating aggregates, and the differences of expression of CD133 CD34 CD44 between 2 kinds of cultured cells were observed by immunofluorescence. Results Floating aggregates grew more slowly, kept activity for longer period than adhesive cells (72.5% vs 47.5%,P<0.05). Floating aggregates expressed higher level of CD133, CD34 and CD44 than adhesive cells (68.97%,82.76%,93.10% vs 5.26%,15.79%,5.26%,P<0.01). Conclusions The combination of CD133, CD34 and CD44 probably can be used as surface markers of cancer stem cells for human lung adenocarcinoma.
