Résumé

Objective@#To evaluate the inhibitory effect of tumor vaccines in colon carcinoma model mice.@*Methods@# Mouse bone marrow⁃derived dendritic cells(BMDCs)were stimulated by using CpG β⁃glucan nanoparticles(CNP)in vitro. The BMDCs were divided into PBS group,NP group(without CpG nanoparticles),Lysate group(MC38 cell lysate)and CpG group(CpG1826),which were determined for the expression of marker molecules on the surface by flow cytometry and for the contents of interleukin⁃6(IL⁃6)and IL⁃12p40 in the culture supernatant by ELISA. The tumor lysate nano⁃vaccine was pre⁃ pared by mixing 50 mg/mL tumor lysate(MC38 cell lysate)with 200 mg/mL CNP in a volume ratio of 1∶1,with which mice were subcutaneously immunized as Vaccine group. Vaccine group,PBS group,CNP group and Lysate group were im⁃ munized once a week,for three times in total. Mice were subcutaneously inoculated with MC38 cells,2 × 105 cells for each, in the right lower limb 1 h after the last immunization,and measured for tumor volume once every three days to plot the tumor growth curve. The ratios of CD3+ CD4+ T and CD3+ CD8+ T cells in the blood were analyzed by flow cytometry and the levels of tumor necrosis factor⁃α(TNF⁃α)and interferon γ(IFNγ)in the blood and spleen of mice were determined by ELISA.@*Results@# CNP effectively increased the expression of CD11c+ CD80+,CD11c+ CD86+,CD11c+ MHC⁃Ⅱ+ and the secretion of IL⁃6 and IL⁃12p40 in BMDCs in vitro,which were significantly higher than those in other 4 groups(t = 4. 3 ~ 46. 2,each P < 0. 05). Compared with that of the other three groups,the tumor volume of mice in Vaccine group decreased significantly(t =2.6~3.4,eachP <0. 05);TherewasnosignificantdifferenceinCD3+ CD8+ TandCD3+ CD8+ Tcellratios(t = 0.5~ 1. 9,each P > 0. 05);The content of IFNγ in blood increased significantly(t = 3. 8 ~ 4. 6,P < 0. 05),while thatof TNF⁃α showed no significant difference(t = 0. 4 ~ 2. 0,each P > 0. 05);However,the contents of IFN γ and TNF⁃α in spleen increased significantly(t = 6. 3 ~ 13. 0,each P < 0. 001).@*Conclusion@#The prepared nano⁃vaccine of tumor lysate improvedtheimmune level in mice and effectively inhibited the growth of colon carcinoma.

Résumé

[Objective] To observe the cure effect of Qianlishu Pil combined with hydrochloric acid doxycycline and Hytrine on chronic nonbacterial prostatitis(CNP). [Method] Select 264 cases of CNP, randomly divide them into control and treatment groups, n=132 for each. The control group was treated with hydrochloric acid doxycycline and Hytrine for 2m; the treatment group, with combined Qianlieshu Pil with hydrochloric acid doxycycline and Hytrine for 2m;make analysis and comparison of 2 groups after treatment. [Result] The effect and clinical signs recovery time were better in treatment group than control one; the total effective rate was 88.9% for treatment group, and 53.8% for control one, the comparison of them had difference of statistical meaning. [Conclusion] The combination of Qianlieshu Pil , hydrochloric acid doxycycline and Hytrine has marked cure effect on CNP.

Résumé

Objective To observe the application of clinical nursing pathways in obstetrics and gyne-cology department, and explore the service methods during perinatal period by clinical record. Methods 150 eases parturient woman in our hospital from July, 2005 to June, 2008 were divided into the CNP group and the control group with 75 cases in each group. Results The treatment expense was lower, the hospitaliza-tion time was shorter and the satisfaction degree was higher in the CNP group than those in the control group. Conclusions The normalization and routinization of CNP application can turn nursing from pas-siveness to initiative, so that the patients can get best nursing service.

BACKGROUND: Head-down suspension (HDS) of rats has been used as a model for the simulation of a microgravity environment. Natriuretic peptide family including atrial natriuretic peptide (ANP), brain natriuretic peptide (BNP) and C-type natriuretic peptides (CNP), and their receptors are responsible for body fluid homeostasis and blood pressure control. However, little is known about the long-term responses and the simultaneous observations of both natriuretic peptide family and their receptors following HDS. This study was analyzed the regulation of cardiac, hypothalamic and renal natriuretic peptides and their receptors syntheses to 4 weeks of HDS in rats. METHODS: Unanesthetized, unrestrained, male Sprague-Dawley rats were subjected to either a horizontal position (control rats) or a -45degrees head-down tilt using the tail-traction technique (HDS rats). This study observed the cardiac, hypothalamic and renal syntheses of natriuretic peptides as a expression of ANP, BNP and CNP mRNA. The mRNA expressions of A-type natriuretic peptide receptor (NPR-A), B-type NPR (NPR-B) and clearance receptor (NPR-C) were also determined. The expressions of natriuretic peptide and their receptor mRNA were measured by reverse transcription-polymerase chain reaction with [32P]-dCTP following 4 weeks of HDS in both control and HDS rats. RESULTS: After 4 weeks of HDS, the expressions of ANP mRNA were significantly decreased in the right atrium and the kidney, and showed the non-significant decreasing trend in the left atrium and the hypothalamus of HDS rats. BNP mRNA expressions were decreased in the heart especially in the left atrium, while its mRNA in the left ventricle was showed the non-significant increasing trend following 4 weeks of HDS. The expressions of CNP mRNA were increased in the hypothalamus and showed the non-significant increasing trend in the kidney of the HDS rats. After 4 weeks of HDS, NPR-A mRNA expressions were decreased in the kidney and hypothalamus, but NPR-C mRNA expression was showed the non-significant increasing trend in the kidney of HDS rats compared with controls. CONCLUSION: These results suggest that the regulation of natriuretic peptide mRNA following 4 weeks of HDS exerts to maintain the blood volume through an attenuation of syntheses in natriuretic peptide family of the heart and the kidney.

Atrial natriuretic peptide (ANP), C-type natriuretic peptides (CNP) and their receptors are found in the kidney, suggesting that these peptides could play a significant physiological role in the kidney. Therefore, this study was investigated the changes in the adaptations of renal natriuretic peptides and their receptors syntheses after 4 weeks of HDS in rats. METHODS: Unanesthetized, unrestrained, male Sprague-Dawley rats were subjected to either a horizontal position (control rats) or a -45degreeshead-down tilt using the tail-traction technique (HDS rats). This study observed the renal syntheses of natriuretic peptides as a expression of ANP and CNP mRNA, and also determined the expression of A-type natriuretic peptide receptor (NPR-A) mRNA and B-type NPR (NPR-B) mRNA. The expressions of natriuretic peptide and NPR mRNA were measured by reverse transcription-polymerase chain reaction with [(32)P]-dCTP following 4 weeks of HDS in the kidney of both control and HDS rats. RESULTS: After 4 weeks of HDS, the expression of ANP mRNA significantly (P<0.01) decreased, while CNP mRNA expression was showed the non-significant increasing trend in the kidney of HDS rats. NPR-A, which binds with ANP, was significantly (P<0.001) decreased in renal mRNA expression of HDS rats compared with controls. Expression in mRNA of NPR-B, which binds with CNP, showed a slightly decreasing trend in the kidney of rats following HDS. CONCLUSION: These results suggest that the renal adaptation following 4 weeks of HDS exerts to maintain the blood volume and electrolyte balance through attenuation of syntheses in the natriuretic peptide and its binding receptor, especially in ANP rather than in CNP systems.

Therefore, this study investigated the changes in the central adaptations of hypothalamic ANP and CNP syntheses to 4 weeks of HDS in rats. METHODS: Unanesthetized, unrestrained, male Sprague-Dawley rats were subjected to either a horizontal position (control rats) or a -45 degrees head-down tilt using the tail-traction technique (HDS rats). We determined the hypothalamic syntheses of natriuretic peptides as an expression of ANP and CNP mRNA. The expression of natriuretic peptide mRNA was measured by reverse transcription-polymerase chain reaction with [32P]-dCTP following 4 weeks of HDS in the hypothalamus of control and HDS rats. RESULTS: After 4 weeks of HDS, the expression of ANP mRNA showed a decreasing trend in the hypothalamus of HDS rats. In contrast with ANP, CNP mRNA expression was significantly (p<0.01) increased in the hypothalamus of HDS rats. There were different changes in the hypothalamic CNP and ANP mRNA expressions of HDS rats compared with that of the control rats.CONCLUSION: These results represent that the hypothalamic syntheses of natriuretic peptides are differently responded and the role of CNP is augmented to compensate for the decrement of ANP action in the central nervous system following 4 weeks of HDS.

Résumé

The 2',3'-cyclic nucleotide 3'-phosphodiesterase (CNP), a protein of unknown function in vivo, is abundantly expressed in myelinating glia in two isoforms, CNP1 and CNP2. In this study, immunoblot analysis showed that CNP1 is the major isoform in adult forebrain, and that both isoforms are included in the postsynaptic density (PSD) fraction and tyrosine-phosphorylated at the basal level. However, subcellular distribution and detergent extraction data showed that CNP is nonspecifically associated with the PSD fraction. Immunocytochemistry revealed that CNP is detected, in a weak but punctate pattern, in dissociated rat hippocampal neurons of 3 days to 2 weeks in vitro. The CNP-positive punctae were distributed throughout soma and dendrites, and distinct from PSD95-positive ones. Immunoblot analysis indicated that CNP is also expressed in neuronal stem cell lines, HiB5 and F11. Interestingly, in addition to the known two isoforms, a new CNP isoform of MW 45 kDa was expressed in these cell lines and was the major type of isoform in F11 cells. Taken together, our data suggest that CNP is expressed in the early stage of in vitro development and nonspecifically included in the adult rat PSD fraction.

BACKGROUND: C-type natriuretic peptide (CNP), secreted from endothelial cells, dilates the vessel and increases the renal excretion of sodium and water. Plasma CNP level is not elevated in patients with congestive heart failure (CHF), but the myocardial content of CNP is increased, suggesting paracrine or autocrine role of CNP. We studied to compare the effect of ANP and CNP on vasorelaxation in CHF rat model. METHODS: Coronary artery ligation was performed in Sprague Dawley rats, resulting in CHF (n=5) and they were fed with ordinary chow for 10 weeks. After measuring hemodynamics, aorta and pulmonary artery were incised and studied in organ chamber to measure the isotonic force to KCl and to observe the relaxation response to atrial natriuretic peptide (ANP), and CNP in vessels precontracted with phenylephrine. Normal Sprague Dawley rats (n=5) were used as control. RESULTS: There were no difference in vasorelaxation responses to ANP between control and CHF rats. The response to CNP was significantly impaired in both aorta and pulmonary artery of CHF rats compared to control rats.
