Objective:Preparation of monoclonal antibodies against Carprofen,which lays a foundation for the rapid detection of Carprofen.Methods:Via an active ester method,Carprofen was conjugated to bovine serum albumin (BSA) as immune antigen and ovalbumin (OVA) as detection antigen,respectively.Hybridomas were obtained by fusing mouse myeloma cells SP2/0 with splenocytes from the mice immunized with Carprofen-BSA.Hybridomas 51C3 secreting antibodies against Carprofen were obtained and subcloned.Results: Ascites of monoclonal antibodies (McAb) were prepared by injecting cells of hybridoma 51C3 into mice abdomen.The titer of purified McAb was 3.2×105and the McAb was IgG1 subtype.McAb was highly specific for Carprofen and the cross-reactivity (CR50%) was less than 0.04% with Ibuprofen, Ketoprofen, Naproxen, Feinuoluofen, Indoprofen, Fenbufen respectively.The sensitivity of the McAb to carprofen was 0.32 ng/ml and the IC50value was 0.882 ng/ml.The recoveries of Carprofen in the pork samples were from 81.4% to 104.4 % and coefficients of variation were from 16.3% to 25.8%.Conclusion:All results indicate that the McAb 51C3 is suitable to develop an immunoassay colloidal gold rapid dipstick test.

Efficacy of carprofen, administered by different routes, was studied in experimental uveitis in dogs. Anterior chamber paracenteses was accomplished at two different moments (M0 and M1), with a five hour interval between them. At M0 and M1, 0.2mL of aqueous humor was collected and quantitation of total protein and prostaglandin E2 (PGE2) were determined. Four groups were formed (n=8), which received carprofen at the end of M0, by the following routes: subcutaneous (GIm), subconjunctival (GII), and topical (GIII). A fourth group that received no treatment was instituted (Control). Conjunctival histopathology of the GII animals was performed. In all groups, values of protein and PGE2 significantly enhanced at M1; however, they did not significantly change among groups at M1. Inflammatory exudate of acute character and mild hemorrhage were seen at histopathology after carprofen administration. Carprofen was unable to inhibit PGE2 synthesis and the protein influx to the anterior chamber by any of the tested routes. However, the reduction of 44 percent in protein levels (topical) suggests that the agent can be used by this route as an adjuvant to control uveitis in dogs.
Estudaram-se os efeitos do carprofeno, aplicado por diferentes vias, em uveítes experimentais em cães. Realizou-se paracentese de câmara anterior em dois momentos (M0 e M1), com intervalo de cinco horas entre si. Em M0 e M1, colheram-se 0,2mL de humor aquoso e determinaram-se as concentrações de proteína total e de prostaglandina E2 (PGE2). Constituíram-se quatro grupos (n = 8), que receberam carprofeno ao final de M0 pelas vias subcutânea (GI), subconjuntival (GII) e tópica (GIII). Um quarto grupo não recebeu tratamento (controle). Procedeu-se à avaliação histopatológica nos indivíduos do GII. Em todos os grupos, encontrou-se aumento significativo dos níveis proteicos e de PGE2 em M1. Não se observou diferença significativa, em M1, entre os grupos para nenhum dos parâmetros estudados. Exsudado inflamatório de caráter agudo e hemorragia discreta foram vistos à histopatologia após a aplicação do fármaco. O carprofeno foi ineficaz em inibir a síntese de PGE2 e o influxo de proteínas para a câmara anterior, por qualquer uma das vias testadas. Contudo, a redução de 44 por cento nos níveis de proteínas (via tópica), sugere que por esta via ele pode ser utilizado como adjuvante no controle da uveíte em cães.

In this study the authors aimed to compare the efficiency of carprofen, ketoprofen and vedaprofen for alleviating postoperative pain in bitches submitted to ovariohysterectomy (OH). Pre- and postoperative assessment of pain was made using serum levels of cortisol and glucose, the visual analogue scale (VAS) and the University of Melbourne pain scale (UMPS) in twenty-one bitches undergoing OH. Dogs were randomly assigned to one of three groups: vedaprofen at 0.5mg kg-1, carprofen at 2.2mg kg-1 or ketoprofen at 2.2mg kg-1. All analgesics were given orally 2 hour before surgery. Assessments were made before surgery and at 1, 2, 3, 4, 5, 6, 7, 8, 12 and 24 hours post-extubation. No dog of this study required additional doses of analgesics. There were no significant differences on serum cortisol and glucose concentrations among groups or from basal values, excepted one hour after extubation. No significant differences on pain scores were observed. It was concluded that vedaprofen provided as good a level of postoperative analgesia as carprofen and ketoprofen.
O objetivo deste estudo foi comparar a eficiência do carprofeno, do cetoprofeno e do vedaprofeno no alívio da dor pós-operatória em cadelas submetidas à ovariohisterectomia (OH) eletiva. Para tal, foram realizadas avaliações pré e pós-operatórias de cortisol e glicose séricos e foram usadas a escala análoga visual (VAS) e a escala da Universidade de Melbourne (University of Melbourne Pain Scale - UMPS). Vinte e uma cadelas foram distribuídas aleatoriamente em três grupos de sete animais cada: vedaprofeno 0,5mg kg-1, carprofeno 2,2mg kg-1 ou cetoprofeno 2,2mg kg-1. Todos os analgésicos foram administrados pela via oral, duas horas antes da cirurgia. As avaliações foram realizadas antes da cirurgia e uma, duas, três, quatro, cinco, seis, sete, oito, 12 e 24 horas após a extubação. Nenhum dos cães precisou de analgesia de resgate. Com exceção dos valores observados uma hora após a extubação, não houve diferenças significativas nas concentrações séricas de cortisol e glicose entre grupos ou quando foram comparadas aos valores basais. Tampouco houve diferenças significativas nos escores de dor entre as escalas usadas, nos tempos de avaliação para cada grupo, nem entre os grupos. Concluiu-se que o vedaprofeno fornece analgesia pós-operatória de qualidade similar à obtida com carprofeno e com cetoprofeno.
