Résumé

Abstract Fish protein is serving as a source of nutrition for protein starving world. However, sustainable aquaculture products require inexpensive plant by-products due to finite sources of fish meal. Therefore, this study was conducted to examine nutrient utilization, growth performance and hematological indices of Cirrhinus mrigala fingerlings fed on Moringa oleifera leaf meal (MOLM) based diets. Fish were fed with six isonitrogenous and isoenergetic diets having MOLM as a substitute of fish meal (FM) at the levels of 0%, 10%, 20%, 30%, 40% and 50% for the period of 90 days. Fingerlings having initial weight 6.35±0.04g were reared in triplicate tanks at the stocking density of 15 and hand fed at the rate of 5% of total biomass twice regularly. Chromic oxide inclusion level was 1% in diets. After analysis, maximum growth performance and improved digestibility of nutrients were found in fish fed with diet at 10% replacement level as compared to fish fed on control diet and other test diets. Additionally, it was found that the red blood cells, white blood cells, hemoglobin and mean corpuscular hemoglobin concentration of fish showed a significantly (p 0.05) inverse correlation with the increase in MOLM. In present research, it was concluded that MOLM has good potential to be used as a FM substitute in C. mrigala diet with maximum effect at 10% showing positive hematological indices.


Resumo A proteína do peixe está servindo como fonte de nutrição para o mundo faminto por proteína. No entanto, produtos de aquicultura sustentáveis requerem subprodutos vegetais baratos devido às fontes finitas de farinha de peixe. Portanto, este estudo foi conduzido para examinar a utilização de nutrientes, o desempenho do crescimento e os índices hematológicos de alevinos de Cirrhinus mrigala alimentados com dietas à base de farelo de folhas de Moringa oleifera (MOLM). Os peixes foram alimentados com seis dietas isonitrogênicas e isoenergéticas contendo MOLM como substituto da farinha de peixe (FM) nos níveis de 0%, 10%, 20%, 30%, 40% e 50% pelo período de 90 dias. Os alevinos com peso inicial de 6,35 ± 0,04g foram criados em tanques triplicados com densidade de 15 animais e alimentados à mão a uma taxa de 5% da biomassa total duas vezes regularmente. O nível de inclusão de óxido crômico foi de 1% nas dietas. Após análise, desempenho máximo de crescimento e melhor digestibilidade de nutrientes foram encontrados em peixes alimentados com dieta em nível de reposição de 10% em comparação com peixes alimentados com dieta controle e outras dietas teste. Além disso, verificou-se que as concentrações de hemácias, leucócitos, hemoglobina e hemoglobina corpuscular média dos peixes apresentaram correlação inversa significativa (p 0,05) com o aumento do MOLM. Na presente pesquisa, concluiu-se que o MOLM tem bom potencial para ser utilizado como substituto do FM na dieta de C. mrigala com efeito máximo a 10% apresentando índices hematológicos positivos.

Abstract A ninety days nutritional trial was directed to explore the effects of dietary chromium on body composition, gut enzyme activity and physiological status of Cirrhinus mrigala by using G & NG corn. Six experimental diets were prepared by using different levels of chromium chloride hexahydrate (0, 0.2, 0.4 mg/kg, each with G & NG corn). For this experimental trial, 480 fingerlings, irrespective of sex were distributed in six aquariums each with replicate. Results revealed that gelatinized corn along with increasing level of Cr2Cl3.6H2O have a positive impact upon body composition of fish. Hematology was positively correlated with chromium chloride hexahydrate supplementation in gelatinized corn. Amylase gut enzyme also showed significant (P 0.05) increase in group fed with chromium chloride hexahydrate supplemented diet (G corn). However, corn with chromium chloride hexahydrate supplementation did not revealed any significant impact on gut protease enzyme activity. From these results it can be concluded that both chromium chloride hexahydrate and gelatinized corn in fish feed are very beneficial to improve body composition, enzymes activity and physiological health status of fish.


Resumo Um ensaio nutricional de 90 dias foi dirigido para explorar os efeitos do cromo dietético na composição corporal, atividade enzimática intestinal e estado fisiológico de Cirrhinus mrigala usando milho G & NG. Seis dietas experimentais foram preparadas usando diferentes níveis de cloreto de cromo hexa-hidratado (0, 0,2, 0,4 mg/kg, cada um com milho G e NG). Para este ensaio experimental, 480 alevinos, independentemente do sexo, foram distribuídos em seis aquários, cada um com réplicas. Os resultados revelaram que o milho gelatinizado, juntamente com o aumento do nível de Cr2Cl3.6H2O, tem um impacto positivo na composição corporal dos peixes. A hematologia foi positivamente correlacionada com a suplementação de cloreto de cromo hexa-hidratado em milho gelatinizado. A enzima amilase intestinal também apresentou aumento significativo (P 0,05) no grupo alimentado com dieta suplementada com cloreto de cromo hexa-hidratado (milho G). No entanto, o milho com suplementação de hexahidrato de cloreto de cromo não revelou nenhum impacto significativo na atividade da enzima protease intestinal. Desses resultados, pode-se concluir que tanto o cloreto de cromo hexa-hidratado quanto o milho gelatinizado na alimentação de peixes são muito benéficos para melhorar a composição corporal, a atividade de enzimas e o estado fisiológico de saúde dos peixes.

Resumo Um experimento referente à alimentação foi realizado para avaliar o efeito da suplementação dietética de cromo, no coeficiente de digestibilidade aparente do nutriente (%) do milho gelatinizado e não gelatinizado em alevinos de Cirrhinus mrigala por 90 dias. Résumé

Indian carp (Cirrhinus mrigala) exposed to two sub-lethal concentrations (0.815 mg/L and 1.63 mg/L) of diazinon for 30 days showed a significant decrease in erythrocyte count, hemoglobin concentration, hematocrit, mean corpuscular volume, mean corpuscular hemoglobin, plasma levels of total protein, albumin, globulin, activity of enzyme acetylcholinesterase and a significant increase in plasma glucose levels when compared with controlled fish (P<0.05). Compared to the controlled fish, leukocyte count decreased significantly in fish exposed to both sub-lethal concentrations and the plasma activities of enzyme asparate aminotransferase and alanine aminotransferase increased significantly after 20 and 30 days in fish exposed to 0.815 mg/L of diazinon, while the activities of these enzymes in fish exposed to 1.63 mg/L of diazinon increased significantly at all sampling periods (P<0.05). On the other hand, the plasma activity of the enzyme lactate dehydrogenase decreased significantly in both diazinon treated fish groups after 10 days of exposure (P<0.05) and returned to the normal value after 20 and 30 days of exposure. In conclusion, long-term exposure to diazinon at sub-lethal concentrations induces severe physiological alterations in C. mrigala that may potentially disrupt their survival in their natural habitat.

Résumé

Nucleotide binding and oligomerization domain (NOD)1 and NOD2 are important cytoplasmic pattern recognition receptors (PRRs) and key members of the NOD-like receptor (NLR) family. They sense a wide range of bacteria or their products and play a key role in inducing innate immunity. This report describes the role of NOD1 and NOD2 receptors signalling in innate immunity in the Indian major carp, mrigal (Cirrhinus mrigala). Tissue-specific expression analysis of NOD1 and NOD2 genes by quantitative real-time PCR (qRT-PCR) revealed their wide distribution in various organs/tissues. In the untreated fish, the highest expression of NOD1 and NOD2 was detected in liver and blood, respectively. Stimulation with NOD1- and NOD2-specific ligands, i.e. iE-DAP and MDP, activated NOD1 and NOD2 receptor signalling in vivo and in vitro resulting in significant (p<0.05) induction of downstream signalling molecule RICK, and the effector molecules IL-1β, IL-8 and IFN-γ in the treated group as compared to their controls. In response to both Gram-positive and Gram-negative bacterial infections, NOD1 and NOD2 receptors signalling were activated and IL-1β, IL-8 and IFN-γ were induced. These findings highlight the important role of NOD receptors in eliciting innate immune response during the pathogenic invasion to the fish.

Résumé

The present study was carried out to evaluate the immunostimulant potential of Nelumbo nucifera (Lotus). Formulated diets with different concentrations of ethanolic extract of N.nucifera (D1=0%, D2=1% and D3=2%) were fed to Cirrhinus mrigala for 40 days, the Specific Growth Rate (SGR) and Feed Conversion Ratio (FCR) were calculated significant increase was observed in SGR &FCR. Then the experimental fishes were infected with Pseudomonas aeruginosa and after 5 days the haematological parameters like Total Erythrocyte count (TEC), Haemoglobin (Hb), Total leucocyte count (TLC), Differential leucocyte count (DLC), serum total protein, serum albumin and globulin levels were analyzed. The TEC, Hb, TLC, Lymphocytes and monocyte counts increased significantly in D3 diet fed fishes. Highly significant increase was observed in serum total protein and albumin levels only. Globulin levels remain unchanged. Basophil and Eosinophil counts decreased significantly. Thus N.nucifera in fish feed preparations may be included as growth promoter and immunostimulator.

Résumé

Disease outbreak in fish culture system interferes with productivity. Oral Immunostimulant containing vitamin C can result in activating the immune system in a nonspecific way thus providing resistance against pathogens. In the present study the fish Cirrhinus mrigala was fed with a feed supplemented with vitamin C (100mg/100g) for 40 days and post challenged with two different dilutions (102 and 105) of Aphanomyces invadans. The haematological parameters like TEC, TLC and differential leukocyte counts were analyzed 24hours, 72 hours and 7th day after infection. TEC counts and lymphocyte counts of infected fishes previously fed with control diet decreased significantly (43%, 36%) whereas it was minimal in fishes (21%, 8%) fed with Vitamin C supplemented diet. Post challenged fishes exhibited an increase in TLC in Vitamin supplemented diet (20%, 32%) over control diet fed fishes. In differential leucocyte counts the lymphocytes decreased in both the experimental groups (9%, 11%) whereas neutrophils increased in both, however the increase was higher in control diet fed group (34%). The basophils and eosinophils increased significantly in Vitamin C supplemented diet only (10%, 60%). Thus supplementation of feed with vitamin C enhanced fish’s tolerance to infection.

Résumé

The histopathological effects of dichlorvos, an organophosphate pesticide, on the gill and liver tissues in Cirrhinus mrigala were determined by light microscopy. The fishes were exposed to sublethal concentrations (0.91 and 1.82 ppm) of dichlorvos for 10 days with parallel untreated control. No histopathological effects were observed in control group. Hyperplasia, desquamation, and necrosis of epithelial, epithelial lifting, oedema, lamellar fusion, collapsed secondary lamellae, curling of secondary lamellae and aneurism in the secondary lamellae were observed in gill tissues exposed to dichlorvos. Hepatic lesions in the liver tissues of fishes exposed to dichlorvos were characterized by cloudy swelling of hepatocytes, congestion, vacuolar degeneration, karyolysis, karyohexis, dilation of sinusoids and nuclear hypertrophy.
